1Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16

6Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
7Alendronate loaded graphene oxide functionalized collagen sponge for the dual effects of osteogenesis and anti-osteoclastogenesis in osteoporotic rats显示文摘Graphene Oxide(GO)-related hydrogels have been extensively studied in hard tissue repair,because GO can not only enhance the mechanical properties of polymers but also promote osteogenic differentiation of mesenchymal stem cells.However,simple GO-related hydrogels are not ideal for the repair of osteoporotic bone defects as the overactive osteoclasts in osteoporosis.Alendronate(Aln)is known to inhibit osteoclasts and may bind to GO through covalent connection.Therefore,delivering Aln in GO-related hydrogels may be effective to repair osteoporotic bone defects.Here,we developed a control-released system which is constructed by collagen(Col)-GO sponges loaded with Aln(Col-GO-Aln)for osteoporotic bone defect repair.In vitro,Col-GO-Aln sponges prolonged the release period of Aln,and the sponge containing 0.05%(w/v)GO released Aln faster than sponge with 0.2%GO.Furthermore,tartrate-resistant acid phosphatase(TRAP)and F-actin staining demonstrated that Col-GO-Aln sponges effectively inhibited osteoclastogenesis of monocyte-macrophages.In vivo,micro-CT scan showed that the volume of newborn bone in defect site by 0.05%GO sponge was nearly three times larger than that of other groups.Moreover,the CT and histological examinations of rat femur proved that Col-GO-Aln sponges decreased the number of osteoclasts and suppressed the systemic bone loss in osteoporotic rats.These findings reveal that the application of GO as carriers of anti-osteoporosis drugs is a viable treatment for osteoporosis.The results also underscore the potential of GO-related hydrogels with Aln-releasing capacity for bone regeneration in osteoporosis.Yuyang Zeng Muran Zhou Lifeng Chen Huimin Fang Shaokai Liu Chuchao Zhou Jiaming Sun Zhenxing Wang 2020Bioactive Materials2020,5,4:10

9Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients.Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong 2020Cellular & Molecular Immunology2020,17,3:8
10Top-emitting thermally activated delayed fluorescence organic light-emitting devices with weak light-matter coupling显示文摘Resonance interaction between a molecular transition and a confined electromagnetic field can lead to weak or strong light-matter coupling.Considering the substantial exciton–phonon coupling in thermally activated delayed fluorescence(TADF)materials,it is thus interesting to explore whether weak light-matter coupling can be used to redistribute optical density of states and to change the rate of radiative decay.Here,we demonstrate that the emission distribution of TADF emitters can be reshaped and narrowed in a top-emitting organic light-emitting device(OLED)with a weakly coupled microcavity.The Purcell effect of weak microcavity is found to be different for TADF emitters with different molecular orientations.We demonstrate that radiative rates of the TADF emitters with vertical orientation can be substantial increased in weakly coupled organic microcavity.These observations can enhance external quantum efficiencies,reduce efficiency roll-off,and improve color-purities of TADF OLEDs,especially for emitters without highly horizontal orientation.Chunxiu Zang Shihao Liu Mengxin Xu Ruifang Wang Chen Cao Zelin Zhu Jiaming Zhang Hui Wang Letian Zhang Wenfa Xie Chun-Sing Lee 2021Light(Science & Applications)2021,10,7:7
11Correlation of Gut Microbiome Between ASD Children and Mothers and Potential Biomarkers for Risk Assessment显示文摘Variation of maternal gut microbiota may increase the risk of autism spectrum disorders(ASDs) in offspring. Animal studies have indicated that maternal gut microbiota is related to neurodevelopmental abnormalities in mouse offspring, while it is unclear whether there is a correlation between gut microbiota of ASD children and their mothers. We examined the relationships between gut microbiome profiles of ASD children and those of their mothers, and evaluated the clinical discriminatory power of discovered bacterial biomarkers. Gut microbiome was profiled and evaluated by 16S ribosomal RNA gene sequencing in stool samples of 59 mother–child pairs of ASD children and 30 matched mother–child pairs of healthy children. Significant differences were observed in the gut microbiome composition between ASD and healthy children in our Chinese cohort. Several unique bacterial biomarkers, such as Alcaligenaceae and Acinetobacter, were identified. Mothers of ASD children had more Proteobacteria, Alphaproteobacteria, Moraxellaceae, and Acinetobacter than mothers of healthy children. There was a clear correlation between gut microbiome profiles of children and their mothers; however, children with ASD still had unique bacterial biomarkers, such as Alcaligenaceae, Enterobacteriaceae, and Clostridium. Candidate biomarkers discovered in this study had remarkable discriminatory power. 13An eFP browser for visualizing strawberry fruit and flower transcriptomes显示文摘Wild strawberry Fragaria vesca is emerging as an important model system for the cultivated strawberry due to its diploid genome and availability of extensive transcriptome data and a range of molecular genetic tools.Being able to better utilize these tools,especially the transcriptome data,will greatly facilitate research progress in strawberry and other Rosaceae fruit crops.The electronic fluorescent pictograph(eFP)software is a useful and popular tool to display transcriptome data visually,and is widely used in other model organisms including Arabidopsis and mouse.Here we applied eFP to display wild strawberry RNA sequencing(RNA-seq)data from 42 different tissues and stages,including various flower and fruit developmental stages.In addition,we generated eight additional RNA-seq data sets to represent tissues from ripening-stage receptacle fruit from yellow-colored and red-colored wild strawberry varieties.Differential gene expression analysis between these eight data sets provides additional information for understanding fruit-quality traits.Together,this work greatly facilitates the utility of the extensive transcriptome data for investigating strawberry flower and fruit development as well as fruit-quality traits.Charles Hawkins Julie Caruana Jiaming Li Chris Zawora Omar Darwish Jun Wu Nadim Alkharouf Zhongchi Liu 2017Horticulture Research2017,4,1:6

15Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 17Single-nucleus transcriptomic landscape of primate hippocampal aging显示文摘The hippocampus plays a crucial role in learning and memory,and its progressive deterioration with age is functionally linked to a variety of human neurodegenerative diseases.Yet a systematic profiling of the aging effects on various hippocampal cell types in primates is still missing.Here,we reported a variety of new aging-associated phenotypic changes of the primate hippocampus.These include,in particular,increased DNA damage and heterochromatin erosion with time,alongside loss of proteostasis and elevated inflammation.To understand their cellular and molecular causes,we established the first single-nucleus transcriptomic atlas of primate hippocampal aging.Among the 12 identified cell types,neural transiently amplifying progenitor cell(TAPC)and microglia were most affected by aging.In-depth dissection of gene-expression dynamics revealed impaired TAPC division and compromised neuronal function along the neurogenesis trajectory;additionally elevated pro-inflammatory responses in the aged microglia and oligodendrocyte,as well as dysregulated coagulation pathways in the aged endothelial cells may contribute to a hostile microenvironment for neurogenesis.This rich resource for understanding primate hippocampal aging may provide potential diagnostic biomarkers and therapeutic interventions against age-related neurodegenerative diseases.Hui Zhang Jiaming Li Jie Ren Shuhui Sun Shuai Ma Weiqi Zhang Yang Yu Yusheng Cai Kaowen Yan Wei Li Baoyang Hu Piu Chan Guo-Guang Zhao Juan Carlos Izpisua Belmonte Qi Zhou Jing Qu Si Wang Guang-Hui Liu 2021Protein & Cell2021,12,9:5

19Weapon configuration, allocation and route planning with time windows for multiple unmanned combat air vehicles显示文摘Unmanned combat air vehicles(UCAVs) mission planning is a fairly complicated global optimum problem. Military attack missions often employ a fleet of UCAVs equipped with weapons to attack a set of known targets. A UCAV can carry different weapons to accomplish different combat missions. Choice of different weapons will have different effects on the final combat effectiveness. This work presents a mixed integer programming model for simultaneous weapon configuration and route planning of UCAVs, which solves the problem optimally using the IBM ILOG CPLEX optimizer for simple missions. This paper develops a heuristic algorithm to handle the medium-scale and large-scale problems. The experiments demonstrate the performance of the heuristic algorithm in solving the medium scale and large scale problems. 20A CCAAT-binding factor,SlNFYA10,negatively regulates ascorbate accumulation by modulating the D-mannose/L-galactose pathway in tomato显示文摘Ascorbic acid(AsA),an important antioxidant and growth regulator,and it is essential for plant development and human health.Specifically,humans have to acquire AsA from dietary sources due to their inability to synthesize it.The AsA biosynthesis pathway in plants has been elucidated,but its regulatory mechanism remains largely unknown.In this report,we biochemically identified a CCAAT-box transcription factor(SlNFYA10)that can bind to the promoter of SlGME1,which encodes GDP-Man-3’,5’-epimerase,a pivotal enzyme in the D-mannose/L-galactose pathway.Importantly,SlNFYA10 simultaneously binds to the promoter of SlGGP1,a downstream gene of SlGME1 in the Dmannose/L-galactose pathway.Binding assays in yeast and functional analyses in plants have confirmed that SlNFYA10 exerts a negative effect on the expression of both SlGME1 and SlGGP1.Transgenic tomato lines overexpressing SlNFYA10 show decreased levels of SlGME1 and SlGGP1 abundance and AsA concentration in their leaves and fruits,accompanied by enhanced sensitivity to oxidative stress.Overall,SlNFYA10 is the first CCAAT-binding factor identified to date to negatively regulate the AsA biosynthetic pathway at multiple sites and modulate plant responses to oxidative stress.Weifang Chen Tixu Hu Jie Ye Bing Wang Genzhong Liu Ying Wang Lei Yuan Jiaming Li Fangman Li Zhibiao Ye Yuyang Zhang 2020Horticulture Research2020,7,1:3
