1Endocrine impact of Helicobacter pylori:Focus on ghrelin and ghrelin o-acyltransferase显示文摘Ghrelin is predominantly produced by the gastric enteroendocrine cell compartment and is octanoylated by the recently discovered ghrelin o-acyltransferase (GOAT) before secretion into the bloodstream. This octanoylation is essential for many of the biological properties of ghrelin including appetite stimulation and anti-inflammatory properties as only the acylated form of ghrelin binds to the ghrelin receptor, the growth hormone secretagogue receptor (GHS-R). Given the gastric location of ghrelin production, it is perhaps not surprising that insult to the gastric mucosa affects circulating ghrelin levels in humans. Helicobacter pylori (H. pylori) infects more than f ifty percent of the world's population and once established within the gastric mucosa, can persist for life. Infection is associated with chronic gastritis, gastric atrophy and ulceration, reduced appetite and a lower body mass index (BMI). The large majority of studies investigating levels of circulating ghrelin and ghrelin expression in the stomach in patients with H. pylori infection indicate that the bacterium has a negative impact on ghrelin production and/or secretion. Eradication of infection restores ghrelin, improves appetite and increases BMI in some studies, however, a causative relationship between H. pylori-associated serum ghrelin decline and food intake and obesity has not been established. Most studies measure total ghrelin in the circulation although the measurement of the ratio of acyl/total ghrelin gives a clearer indication that the ghrelin acylation process is altered during infection and atrophy. 2Second harmonic generation spectroscopy on hybrid plasmonic/dielectric nanoantennas显示文摘Plasmonic nanoantennas provide unprecedented opportunities to concentrate light fields in subwavelength-sized volumes.By placing a nonlinear dielectric nanoparticle in such a hot spot,one can hope to take advantage of both the field enhancement provided by nanoantennas and the large,nonlinear optical susceptibility of dielectric nanoparticles.To test this concept,we combine gold gap nanoantennas with second-order,nonlinear zinc sulfide nanoparticles,and performsecond harmonic generation(SHG)spectroscopy on the combined hybrid dielectric/plasmonic nanoantennas as well as on the individual constituents.We find that SHG from the bare gold nanoantennas,even though it should be forbidden due tosymmetry reasons,is several orders ofmagnitude larger than that of the bare zinc sulfide nanoparticles.Even stronger second harmonic signals are generated by the hybrid dielectric/plasmonic nanoantennas.Control experiments with nanoantennas containing linear lanthanumfluoride nanoparticles reveal;however,that the increasedSHG efficiency of the hybrid dielectric/plasmonic nanoantennas does not depend on the nonlinear optical susceptibility of the dielectric nanoparticles but is an effect of the modification of the dielectric environment.The combination of a hybrid dielectric/plasmonic nanoantenna,which is only resonant for the incoming pump light field,with a second nanoantenna,which is resonant for the generated second harmonic light,allows for a further increase in the efficiency of SHG.As the second nanoantenna mediates the coupling of the second harmonic light to the far field,this double-resonant approach also provides us with control over the polarization of the generated light.Heiko Linnenbank Yevgen Grynko Jens Förstner Stefan Linden 2016Light(Science & Applications)2016,5,1:7

4Therapeutic ACPA inhibits NET formation: a potential therapy for neutrophil-mediated inflammatory diseases显示文摘Excessive release of neutrophil extracellular traps(NETs)is associated with disease severity and contributes to tissue injury,followed by severe organ damage.Pharmacological or genetic inhibition of NET release reduces pathology in multiple inflammatory disease models,indicating that NETs are potential therapeutic targets.Here,we demonstrate using a preclinical basket approach that our therapeutic anti-citrullinated protein antibody(tACPA)has broad therapeutic potential.Treatment with tACPA prevents disease symptoms in various mouse models with plausible NET-mediated pathology,including inflammatory arthritis(IA),pulmonary fibrosis,inflammatory bowel disease and sepsis.We show that citrulline residues in the N-termini of histones 2A and 4 are specific targets for therapeutic intervention,whereas antibodies against other N-terminal post-translational histone modifications have no therapeutic effects.Because citrullinated histones are generated during NET release,we investigated the ability of tACPA to inhibit NET formation.tACPA suppressed NET release from human neutrophils triggered with physiologically relevant human disease-related stimuli.Moreover,tACPA diminished NET release and potentially initiated NET uptake by macrophages in vivo,which was associated with reduced tissue damage in the joints of a chronic arthritis mouse model of IA.To our knowledge,we are the first to describe an antibody with NET-inhibiting properties and thereby propose tACPA as a drug candidate for NET-mediated inflammatory diseases,as it eliminates the noxious triggers that lead to continued inflammation and tissue damage in a multidimensional manner.Renato G.S.Chirivi Jos W.Gvan Rosmalen Maarten van der Linden Maximilien Euler Gonny Schmets Galina Bogatkevich Konstantinos Kambas Jonas Hahn Quinte Braster Oliver Soehnlein Markus H.Hoffmann Helmuth H.Gvan Es Jos M.H.Raats 2021Cellular & Molecular Immunology2021,18,6:5

7Surface functionalized N-C-TiO_(2)/C nanocomposites derived from metal-organic framework in water vapour for enhanced photocatalytic H generation显示文摘Surface-functionalized nitrogen/carbon co-doped polymorphic TiO_(2) phase junction nanoparticles uniformly distributed in porous carbon matrix were synthesized by a simple one-step pyrolysis of titanium based metal-organic framework(MOF),NH_(2)-MIL^(-1)25(Ti) at 700℃ under water vapour atmosphere.Introducing water vapour during the pyrolysis of NH_(2)-MIL^(-1)25(Ti) not only functionalizes the derived porous carbon matrix with carboxyl groups but also forms additional oxygen-rich N like interstitial/intraband states lying above the valence band of TiO2 along with the self-doped carbo n,which further narrows the energy band gaps of polymorphic TiO2 nanoparticles that enhance photocatalytic charge transfer efficiency.Without co-catalyst,sample N-C-TiO2/CArW demonstrates H_(2) evolution activity of 426 μmol gcat-1h^(-1),which remarkably outperforms commercial TiO_(2)(P-25) and N-C-TiO_(2)/CAr with a 5-fold and 3-fold H_(2) generation,respectively.This study clearly shows that water vapour atmosphere during the pyrolysis increases the hydrophilicity of the Ti-MOF derived composites by functionalizing porous carbon matrix with carboxylic groups,as well as enhancing the electrical conductivity and charge transfer efficiency due to the formation of additional localized oxygen-rich N like interstitial/intraband states.This work also demonstrates that by optimizing the anatase-rutile phase composition of the TiO2 polymorphs,tuning the energy band gaps by N/C co-doping and functionalizing the porous carbon matrix in the N-C-TiO2/C nanocomposites,the photocatalytic H_(2) generation activity can be further enhanced.Mian Zahid Hussain Zhuxian Yang Bart van der Linden Zheng Huang Quanli Jia Erik Cerrato Roland A.Fischer Freek Kapteijn Yanqiu Zhu Yongde Xia 2021Journal of Energy Chemistry2021,30,6:3

10Single-port laparoscopic cholecystectomy vs standard laparoscopic cholecystectomy:A non-randomized,agematched single center trial显示文摘AIM: To compare the safety of single-port laparoscopic cholecystectomies with standard four-port cholecystectomies.METHODS: Between January 2011 and December 2012 datas were gathered from 100 consecutive patients who received a single-port cholecystectomy. Patient baseline characteristics of all 100 single-port cholecystectomies were collected(body mass index, age, etc.) in a database. This group was compared with 100 age-matched patients who underwent a conventional laparoscopic cholecystectomy in the same period. Retrospectively, per- and postoperative data were added. The two groups were compared to each other using independent t-tests and χ2-tests, P values below 0.05 were considered significantly different.RESULTS: No differences were found between both groups regarding baseline characteristics. Operating time was significantly shorter in the total single-port group(42 min vs 62 min, P < 0.05); in procedures performed by surgeons the same trend was seen(45 min vs 59 min, P < 0.05). Peroperative complications between both groups were equal(3 in the single-port group vs 5 in the multiport group; P = 0.42). Although not significant less postoperative complications were seen in the single-port group compared with the multiport group(3 vs 9; P = 0.07). No statistically significant differences were found between both groupswith regard to length of hospital stay, readmissions and mortality. CONCLUSION: Single-port laparoscopic cholecystectomy has the potential to be a safe technique with a low complication rate, short in-hospital stay and comparable operating time. Single-port cholecystectomy provides the patient an almost non-visible scar while preserving optimal quality of surgery. 14Effects of a general practitioner cooperative co-located with an emergency department on patient throughput显示文摘BACKGROUND:In 2013 a General Practitioner Cooperative(GPC) was introduced at the Emergency Department(ED) of our hospital.One of the aims of this co-located GPC was to improve throughput of the remaining patients at the ED.To determine the change in patient flow,we assessed the number of self-referrals,redirection of self-referrals to the GPC and back to the ED,as well as ward and ICU admission rates and length of stay of the remaining ED population.METHODS:We conducted a four months' pre-post comparison before and after the implementation of a co-located GPC with an urban ED in the Netherlands.RESULTS:More than half of our ED patients were self-referrals.At triage,54.5%of these selfreferrals were redirected to the GPC.After assessment at the GPC,8.5%of them were referred back to the ED.The number of patients treated at the ED declined with 20.3%after the introduction of the GPC.In the remaining ED population,there was a significant increase of highly urgent patients(P<0.001),regular admissions(P<0.001),and ICU admissions(P<0.001).Despite the decline of the number of patients at the ED,the total length of stay of patients treated at the ED increased from 14 682 hours in the two months' control period to 14 962 hours in the two months' intervention period,a total increase of 270 hours in two months(P<0.001).CONCLUSION:Introduction of a GPC led to efficient redirection of self-referrals but failed to improve throughput of the remaining patients at the ED.Michiel J.van Veelen Crispijn L.van den Brand Resi Reijnen M.Christien van der Linden 2016World Journal of Emergency Medicine2016,7,4:2

18Diabetic neuropathy: An evaluation of the use of quercetin in the cecum of rats显示文摘AIM:To investigate the effect of quercetin supplementation on the myenteric neurons and glia in the cecum of diabetic rats.METHODS:Total preparations of the muscular tunic were prepared from the ceca of twenty-four rats divided into the following groups:control(C),control supplemented with quercetin(200 mg/kg quercetin body weight)(CQ),diabetic(D)and diabetic supplemented with quercetin(DQ).Immunohistochemical double staining technique was performed with HuC/D(general population)/nitric oxide synthase(nNOS),HuC-D/S-100and VIP.Density analysis of the general neuronal population HuC/D-IR,the nNOS-IR(nitrergic subpopulation)and the enteric glial cells(S-100)was performed,and the morphometry and the reduction in varicosity population(VIP-IR)in these populations were analyzed.RESULTS:Diabetes promoted a significant reduction(25%)in the neuronal density of the HuC/D-IR(general population)and the nNOS-IR(nitrergic subpopulation)compared with the C group.Diabetes also significantly increased the areas of neurons,glial cells and VIP-IR varicosities.Supplementation with quercetin in the DQ group prevented neuronal loss in the general population and increased its area(P<0.001)and the area of nitrergic subpopulation(P<0.001),when compared to C group.Quercetin induced a VIP-IR and glial cells areas(P<0.001)in DQ group when compared to C,CQ and D groups.CONCLUSION:In diabetes,quercetin exhibited a neuroprotective effect by maintaining the density of the general neuronal population but did not affect the density of the nNOS subpopulation.Paulo Emilio Botura Ferreira Cláudia Regina Pinheiro Lopes Angela Maria Pereira Alves der Paulo Belato Alves David Robert Linden Jacqueline Nelisis Zanoni Nilza Cristina Buttow 2013World Journal of Gastroenterology2013,19,38:2
