1Super enhancer inhibitors suppress MYC driven transcriptional amplification and tumor progression in osteosarcoma显示文摘Osteosarcoma is the most common primary bone sarcoma that mostly occurs in young adults. The causes of osteosarcoma are heterogeneous and still not fully understood. Identification of novel, important oncogenic factors in osteosarcoma and development of better, effective therapeutic approaches are in urgent need for better treatment of osteosarcoma patients. In this study, we uncovered that the oncogene MYC is significantly upregulated in metastastic osteosarcoma samples. In addition, high MYC expression is associated with poor survival of osteosarcoma patients. Analysis of MYC targets in osteosarcoma revealed that most of the osteosarcoma super enhancer genes are bound by MYC. Treatment of osteosarcoma cells with super enhancer inhibitors THZ1 and JQ1 effectively suppresses the proliferation, migration, and invasion of osteosarcoma cells. Mechanistically,THZ1 treatment suppresses a large group of super enhancer containing MYC target genes including CDK6 and TGFB2. 2Indole methylation protects diketopiperazine configuration in the maremycin biosynthetic pathway显示文摘The maremycin biosynthetic gene cluster has been identified in Streptomyces sp. B9173. Comparative metabolic profiling with knockout mutant strains led to the identification of new products correlated to the maremycin biosynthesis, in particular the'demethyl'-maremycins with an unexpected D-tryptophan unit. 4Deep-red emitting Mg2Ti04:Mn^(4+)phosphor ceramics for plant lighting显示文摘In this study,deep-red emitting Mg2T i04:Mn4 phosphor ceramics were synthesized by the high temperature solid-state reaction method.The ceramics can be excited by the 465 nm blue light and had a narrow emission with a full width at half maximum(FWMH)value of 31 nm.The peak wavelength was located at 658 nm,which matched the demanded wavelength for photosynthesis.The crystal field strength(Dq)and the Racah parameters(B and C)were estimated by the Tanabe-Sugano diagram.The thermal conductivity of the Mg2Ti(0.999)O4:0.001Mn^(4+)ceramic was 7.535 W/(m·K)at room temperature,which was one order of magnitude higher than that of the traditional packaging method using the silicone gel.A set of phosphor converted LEDs were fabricated by mounting the phosphor ceramics onto the 460 nm blue LED chips and the CIE coordinates can move from the blue region to the purple light region with the thickness of the ceramic increasing.These results indicated that the Mg2Ti04:Mn^(4+)phosphor ceramic was suitable for plant lighting when combined with a blue LED chip.Zixin WANG Hui LIN Dawei ZHANG Yiming SHEN Yang LI Ruijin HONG Chunxian TAO Zhaoxia HAN Lei CHEN Shengming ZHOU 2021Journal of Advanced Ceramics2021,10,1:2

7Microbial diversity and structure in the gastrointestinal tracts of two stranded short-finned pilot whales(Globicephala macrorhynchus)and a pygmy sperm whale(Kogia breviceps)显示文摘Information on the gut microbiome composition of different mammals could provide novel insights into the evolution of mammals and succession of microbial communities in different hosts.However,there is limited information on the gut microbiome composition of marine mammals,especially cetaceans because of sampling constraints.In this study,we investigated the diversity and composition of microbial communities in the stomach,midgut,and hindgut of 2 stranded short-finned pilot whales(Globicephala macrorhynchus)and hindgut of a stranded pygmy sperm whale(Kogia breviceps)by using 16S rRNA gene amplicon sequencing technology.On the basis of the 50 most abundant operational taxonomic units,principal coordinate analysis,and non-metric multidimensional scaling analysis,we confirmed that the gut microbial communities of the 3 whales were different.Our results revealed that the gut microbiome of 1 stranded short-finned pilot whale GM16 was dominated by Firmicutes(mainly Clostridium)and Fusobacteria;whereas that of the other pilot whale GM19 was composed of Gammaproteobacteria and Bacteroidetes(mainly Vibrio and Bacteroides,respectively),probably caused by intestinal disease and antibiotic treatment.The gut microbiome of the pygmy sperm whale was dominated by Firmicutes and Bacteroidetes.Moreover,different gastrointestinal tract regions harbored different microbial community structures.To our knowledge,this is the first report of the gut microbiome of short-finned pilot whales,and our findings will expand our current knowledge on microbial diversity and composition in the gastrointestinal tract of cetaceans.Shijie BAI Peijun ZHANG Mingli LIN Wenzhi LIN Zixin YANG Songhai LI 2021Integrative Zoology2021,16,3:1
8A near-complete genome assembly of the allotetrapolyploid Cenchrus fungigraminus(JUJUNCAO)provides insights into its evolution and C4 photosynthesis显示文摘JUJUNCAO(Cenchrus fungigraminus;2n=4x=28)is a Cenchrus grass with the highest biomass production among cultivated plants,and it can be used for mushroom cultivation,animal feed,and biofuel production.Here,we report a nearly complete genome assembly of JUJUNCAO and reveal that JUJUNCAO is an allopolyploid that originated2.7 million years ago(mya).Its genome consists of two subgenomes,and subgenome A shares high collinear synteny with pearl millet.We also investigated the genome evolution of JUJUNCAO and suggest that the ancestral karyotype of Cenchrus split into the A and B ancestral karyotypes of JUJUNCAO.Comparative transcriptome and DNA methylome analyses revealed functional divergence of homeologous gene pairs between the two subgenomes,which was a further indication of asymmetric DNA methylation.The three types of centromeric repeat in the JUJUNCAO genome(CEN137,CEN148,and CEN156)may have evolved independently within each subgenome,with some introgressions of CEN156 from the B to the A subgenome.We investigated the photosynthetic characteristics of JUJUNCAO,revealing its typical C4 Kranz anatomy and high photosynthetic efficiency.NADP-ME and PEPCK appear to cooperate in the major C4 decarboxylation reaction of JUJUNCAO,which is different from other C4 photosynthetic subtypes and may contribute to its high photosynthetic efficiency and biomass yield.Taken together,our results provide insights into the highly efficient photosynthetic mechanism of JUJUNCAO and provide a valuable reference genome for future genetic and evolutionary studies,as well as genetic improvement of Cenchrus grasses.Huakun Zheng Baiyu Wang Xiuting Hua Ruiting Gao Yuhao Wang Zixin Zhang Yixing Zhang Jing Mei Yongji Huang Yumin Huang Hui Lin Xingtan Zhang Dongmei Lin Siren Lan Zhongjian Liu Guodong Lu Zonghua Wang Ray Ming Jisen Zhang Zhanxi Lin 2023Plant Communications2023,4,5:1

10Characterization of the tunicamycin gene cluster unveiling unique steps involved in its biosynthesis显示文摘Tunicamycin,a potent reversible translocase I inhibitor,is produced by several Actinomycetes species.The tunicamycin structure is highly unusual,and contains an 11-carbon dialdose sugar and anα,β-1″,11′-glycosidic linkage.Here we report the identification of a gene cluster essential for tunicamycin biosynthesis by high-throughput heterologous expression(HHE)strategy combined with a bioassay.Introduction of the genes into heterologous non-producing Streptomyces hosts results in production of tunicamycin by these strains,demonstrating the role of the genes for the biosynthesis of tunicamycins.Gene disruption experiments coupled with bioinformatic analysis revealed that the tunicamycin gene cluster is minimally composed of 12 genes(tunA–tunL).Amongst these is a putative radical SAM enzyme(Tun B)with a potentially unique role in biosynthetic carbon-carbon bond formation.Hence,a seven-step novel pathway is proposed for tunicamycin biosynthesis.Moreover,two gene clusters for the potential biosynthesis of tunicamycin-like antibiotics were also identified in Streptomyces clavuligerus ATCC 27064 and Actinosynnema mirums DSM 43827.These data provide clarification of the novel mechanisms for tunicamycin biosynthesis,and for the generation of new-designer tunicamycin analogs with selective/enhanced bioactivity via combinatorial biosynthesis strategies.Wenqing Chen Dongjing Qu Lipeng Zhai Meifeng Tao Yemin Wang Shuangjun Lin Neil P.J.Price Zixin Deng 2010Protein & Cell2010,1,12:1
11Enhancing tylosin production by combinatorial overexpression of efflux,SAM biosynthesis,and regulatory genes in hyperproducing Streptomyces xinghaiensis strain显示文摘Tylosin is a 16-membered macrolide antibiotic widely used in veterinary medicine to control infections caused by Gram-positive pathogens and mycoplasmas.To improve the fermentation titer of tylosin in the hyperproducing Streptomyces xinghaiensis strain TL01,we sequenced its whole genome and identified the biosynthetic gene cluster therein.Overexpression of the tylosin efflux gene tlrC,the cluster-situated S-adenosyl methionine(SAM)synthetase gene metK_(cs),the SAM biosynthetic genes adoK_(cs)-metFcs,or the pathway-specific activator gene tylR enhanced tylosin production by 18%,12%,11%,and 11%in the respective engineered strains TLPH08-2,TLPH09,TLPH10,and TLPH12.Co-overexpression of metK_(cs)and adoK_(cs)-metFcs as two transcripts increased tylosin production by 22%in the resultant strain TLPH11 compared to that in TL01.Furthermore,combinational overexpression of tlrC,metK_(cs),adoK_(cs)-metFcs,and tylR as four transcripts increased tylosin production by 23%(10.93g/L)in the resultant strain TLPH17 compared to that in TL01.However,a negligible additive effect was displayed upon combinational overexpression in TLPH17 as suggested by the limited increment of fermentation titer compared to that in TLPH08-2.Transcription analyses indicated that the expression of tlrC and three SAM biosynthetic genes in TLPH17 was considerably lower than that of TLPH08-2 and TLPH11.Based on this observation,the five genes were rearranged into one or two operons to coordinate their overexpression,yielding two engineered strains TLPH23 and TLPH24,and leading to further enhancement of tylosin production over TLPH17.In particular,the production of TLPH23 reached 11.35 g/L.These findings indicated that the combinatorial strategy is a promising approach for enhancing tylosin production in high-yielding industrial strains.Penghui Dai Yuyao Qin Luyuan Li Haidi Li Lihuo Lv Danying Xu Yuqing Song Tingting Huang Shuangjun Lin Zixin Deng Meifeng Tao 2023Synthetic and Systems Biotechnology2023,8,3:0
12A hierarchical bilayer architecture for complex tissue regeneration显示文摘Engineering a complete,physiologically functional,periodontal complex structure remains a great clinical challenge due to the highly hierarchical architecture of the periodontium and coordinated regulation of multiple growth factors required to induce stem cell multilineage differentiation.Using biomimetic self-assembly and microstamping techniques,we construct a hierarchical bilayer architecture consisting of intrafibrillarly mineralized collagen resembling bone and cementum,and unmineralized parallel-aligned fibrils mimicking periodontal ligament.The prepared biphasic scaffold possesses unique micro/nano structure,differential mechanical properties,and growth factor-rich microenvironment between the two phases,realizing a perfect simulation of natural periodontal hard/soft tissue interface.The interconnected porous hard compartment with a Young’s modulus of 1409.00±160.83 MPa could induce cross-arrangement and osteogenic differentiation of stem cells in vitro,whereas the micropatterned soft compartment with a Young’s modulus of 42.62±4.58 MPa containing abundant endogenous growth factors,could guide parallel arrangement and fibrogenic differentiation of stem cells in vitro.After implantation in critical-sized complete periodontal tissue defect,the biomimetic bilayer architecture potently reconstructs native periodontium with the insertion of periodontal ligament fibers into newly formed cementum and alveolar bone by recruiting host mesenchymal stem cells and activating the transforming growth factor beta 1/Smad3 signaling pathway.Taken together,integration of self-assembly and microstamping strategies could successfully fabricate a hierarchical bilayer architecture,which exhibits great potential for recruiting and regulating host stem cells to promote synergistic regeneration of hard/soft tissues.Min Yu Dan Luo Jing Qiao Jiusi Guo Danqing He Shanshan Jin Lin Tang Yu Wang Xin Shi Jing Mao Shengjie Cui Yu Fu Zixin Li Dawei Liu Ting Zhang Chi Zhang Zhou Li Yongsheng Zhou Yan Liu 2022Bioactive Materials2022,7,4:0

15Design and comparative analysis of self-propelling drill bit applied to deep-sea stratum drilling robot显示文摘Robotic subsea stratum drilling robot is a method for new subsea stratigraphic geological investigation and resource exploration.Resistance at the front end is the main source of resistance to the robot’s motion in the strata.Since there is no continuous and strong downward drilling force as in conventional drilling rigs,robot movement relies heavily on the drill bit to reduce the drilling resistance.In this study we propose a self-propelling drill bit that can discharge soil debris to provide propulsive force and reduce the resistance.The key parameter of the drill bit design,the spiral blade lead angle,was determined by theoretical analysis of the drill bit’s soil discharging effect.To verify the structural advantages of the self-propelling drill bit in reducing resistance,a comparative analysis with a conventional conical drill bit was conducted.The drilling process of both bits was simulated using finite element simulation at various rotation speeds,the penetration force and torque data of both drill bits were obtained,and tests prepared accordingly in subsea soil were conducted.The simulations and tests verified that the penetration force of the self-propelling drill bit was lower than that of the conventional conical drill bit.The self-propelling drill bit can reduce the resistance effectively,and may play an important role in the stratum movement of drilling robots.Peihao ZHANG Xingshuang LIN Hao WANG Jiawang CHEN Zhenwei TIAN Zixin WENG Ziqiang REN Peng ZHOU 2023Journal of Zhejiang University-Science A(Applied Physics & Engineering)2023,24,10:0
16Surveillance and examination of microbial contamination in ice cream in China显示文摘This study investigated the microbial contamination of ice cream in China.A total of 2887 ice cream samples were collected from different regions of China.Contamination by the aerobic plate count(APC),coliforms,and three foodborne pathogens,Listeria monocytogenes(L.monocytogenes),Staphylococcus aureus(S.aureus),and Salmonella spp.,was detected in the samples.L.monocytogenes isolates were further analyzed for antibiotic susceptibility and multi-locus sequence typing(MLST).The results showed that APC was more than 10^(5) colony forming units(CFU)/g in 6.10%(176/2887)and coliform was more than 10^(2) CFU/g in 15.69%(453/2887)of all samples.The positive rates of S.aureus,L.monocytogenes,and Salmonella spp.were 0.66%,0.62%,and 0.10%,respectively.Among these,S.aureus contamination was more than 102 CFU/g in two samples,and L.monocytogenes in the positive sample was in the range of 0.3-240 most probable number(MPN)/g.with a median of 4.3 MPN/g.The hygienic status of the packaged samples was much better than that of the bulk samples(P<0.05).Catering samples were more frequently and heavily contaminated than samples from retail and wholesale outlets(P<0.05).No significant differences were observed in samples bought from urban and rural areas(P>0.05).For 18 L.monocytogenes strains isolated from ice cream,the resistance rate of nine antibiotics was 5.56%(1/18).By MLST,the L.monocytogenes strains were classified into nine sequence types(STs),of which ST8 was the most common(six isolates).These results indicate that a potential health risk to the public may be caused by ice cream,particularly in susceptiblepopulations.Lin Yan Xiaoyan Pei Junjie Miao Ying Li Shuran Yang Zixin Peng Xiaorong Yang Lingling Mei Qingwen Yang Huan Ren Dajin Yang Hongmei Shi 2022Food Quality and Safety2022,6,3:0

18Characterization of endogenous nucleic acids that bind to NgAgo in Natronobacterium gregoryi sp2 cells显示文摘As nucleic acid-guided endonucleases,some prokaryotic Argonautes have been used as programmable nucleases.Natronobacterium gregoryi Argonaute(NgAgo)has also been proposed for gene editing,but this remains very controversial.Until now,the endogenous nucleic acids that bind to NgAgo in Natronobacterium gregoryi sp2(N.gregoryi sp2)have not been characterized.We expressed the conserved PIWI domain of NgAgo and used it to induce anti-PIWI antibody.We also cultured the N.gregoryi sp2 strain and performed immunoprecipitation,chromatin immunoprecipitation(ChIP),and RNA immunoprecipitation(RIP)assays.The nucleic acids that endogenously bound NgAgo in N.gregoryi sp2 cells were sequenced and analyzed.The results showed that NgAgo endogenously bound RNA rather than DNA.NgAgo-associated RNAs were mainly transcripts of genes that encoded tRNA,transcriptional regulators,RNA polymerases,and RNA-binding proteins.NgAgo mainly binds to the transcripts inside genes or in their upstream sequences.Interestingly,the top enriched motif of peaks was the same as that of miR-1289,suggesting that NgAgo may regulate gene expression post-transcriptionally.GO enrichment analysis showed that the peak-associated genes were enriched in transmembrane transport processes.These results revealed that NgAgo binds RNA and may function in post-transcriptional regulation in vivo.LIXU JIANG LIN NING CHUNCHAO PU ZIXIN WANG BIFANG HE JIAN HUANG 2022BIOCELL2022,46,2:0
19An Investigation of Bubble Motion in the Fluidic Oscillator显示文摘Enhanced gas-liquid mass transfer is significant for the desulfurization and denitration of ship exhaust gases.As a fluid device,the special structure of the fluidic oscillator generates self-excited oscillations that can effectively enhance the mass transfer process of gas-liquid.But there are few studies on the internal gas-liquid flow.The transportation of individual bubbles in the fluidic oscillator was investigated by a high-speed camera and digital image analysis.The results show that the bubble experienced a significant deceleration process in the chamber region of the fluidic oscillator.In addition,the maximum bubble offset increased with the diameter of the initial bubble.The trajectory of the bubble showed zigzag movement due to the deflecting oscillation of the fluidic oscillator.At the same time,the deformation of the bubble was intensified by the deflecting oscillation.The deformation ratio of the bubble increased with the increase of Reynolds number.By studying the transport process of a single bubble in the fluid oscillator,it is considered that the fluid oscillator has the potential to be a new bubble generator.BIE Haiyan XUE Licheng LI Yulong LI Yunxia LIN Zixin AN Weizhong 2023Journal of Ocean University of China2023,22,2:0
