| 2 | The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. The ethanol sensing properties of these microcrystals are investigated by spin-coating them on a silicon substrate with Pt electrodes to fabricate a micro-structure sensor. The sensitivity is up to 8 when the sensor is exposed to 50 ppm ethanol, and the response time and recovery time are 10 s and 20 s, respectively. | 5 | Transferrin receptor and ferritin-H are developmentally regulated in oligodendrocyte lineage cells显示文摘Iron is an essential trophic element that is required for cell viability and differentiation, especially in oligodendrocytes, which consume relatively high rates of energy to produce myelin. Multiple iron metabolism proteins are expressed in the brain including transferrin receptor and ferritin-H. However, it is still unknown whether they are developmentally regulated in oligodendrocyte lineage cells for myelination. Here, using an in vitro cultured differentiation model of oligodendrocytes, we found that both transferrin receptor and ferritin-H are significantly upregulated during oligodendrocyte maturation, implying the essential role of iron in the development of oligodendrocytes. Additional different doses of Fe3+ in the cultured medium did not affect oligodendrocyte precursor cell maturation or ferritin-H expression but decreased the expression of the transferrin receptor. | 6 | Production and contribution of hydroxyl radicals between the DSA anode and water interface显示文摘Hydroxyl radicals play the key role during electrochemical oxidation and photoelectrochemical oxidation. The production and effect of hydroxyl radicals on the interface between DSA anode and water was investigated by examining the quenching effect of iso-propanol on Orange II decolorization. We observed that with an increase in electrode potential from 4 to 12 V across electrodes at pH 7.0, the contribution percentage of hydroxyl radicals increased dramatically. More OH radicals were produced in acidic and alkaline conditions than at neutral conditions. At electrode potential of 4 V, the contribution percentage of hydroxyl radicals was obviously higher at near neutral pH conditions, while removal efficiency of Orange II achieved was the lowest concurrently. | 8 | Hepatitis B virus X protein enhances hepatocarcinogenesis by depressing the targeting of NUSAP1 mRNA by miR-18b显示文摘Objective: The aim of this study was to investigate the underlying mechanism whereby HBx modulates the targeting of NUSAP1 by miR-18b to enhance hepatocarcinogenesis.Methods: We employed an integrated approach of bioinformatics analysis and molecular experiments in hepatoma cells, HBV transgenic mice, and clinical liver cancer tissues to investigate the role of HBx-regulated miR-18b in the development of liver cancer.Results: In this study, we report that the HBx-mediated tumor suppressor miR-18b modulates hepatocarcinogenesis during the host-HBV interaction. The expression levels of miR-18b were lower in clinical HBV-positive liver cancer tissues and liver tissues of HBV-transgenic mice. Interestingly, HBx inhibited miR-18b expression by inducing the methylation of CpG islands in its promoter. Accordingly, we tested the hypothesis that HBx enhanced hepatocarcinogenesis by increasing the expression of target genes of miR-18b. Moreover, we identified nucleolar spindle-associated protein 1(NUSAP1) as one of the target genes of miR-18b.NUSAP1 was expressed at high levels in liver cancer tissues. Interestingly, HBx up-regulated NUSAP1 by suppressing miR-18b.Functionally, miR-18b significantly inhibited the proliferation of hepatoma cells by depressing NUSAP1 levels in vivo and in vitro.Conclusions: Thus, we conclude that the targeting of NUSAP1 mRNA by the tumor suppressor miR-18b is controlled by HBxmodulated promoter methylation during the host-virus interaction, leading to hepatocarcinogenesis. | 11 | Genetic analysis and gene fine mapping of a rolling leaf mutant (rl_(11(t))) in rice (Oryza sativa L.)显示文摘The exploration of new genes controlling rice leaf shape is an important foundation for rice functional genomics and plant architecture improvement. In the present study, we identified a rolling leaf mutant from indica variety Yuefeng B, named rl11(t), which exhibited reduced plant height, rolling and narrow leaves. Leaves in rl11(t) mutant showed abnormal number and morphology of veins compared with those in wild type plants. In addition, rl11(t) mutant was less sensitive to the inhibitory effect of auxin than the wild type. Genetic analysis suggested that the mutant was controlled by a single recessive gene. Gene Rl11(t) was initially mapped between SSR markers RM6089 and RM124 on chromosome 4. Thirty-two new STS markers around the Rl11(t) region were developed for fine mapping. A physical map encompassing the Rl11(t) locus was constructed and the target gene was finally delimited to a 31.6 kb window between STS4-25 and STS4-26 on BAC AL606645. | 13 | Microvascular pathological features and changes in related injury factors in a rat acute blood stasis model显示文摘OBJECTIVE: To examine the microvascular pathological characteristics and changes in related injury factors in a rat model of acute blood stasis.METHODS: A total of 75 Sprague-Dawley rats were divided randomly and equally into a control group and four experimental groups assessed at different times after the induction of stasis(0, 1, 3 or 6 h after stasis)(n = 15). The acute blood stasis model was established through rat tail-vein injection of high-molecular-weight dextran. After Electrocardiograph(ECG) detection at predetermined times(0,1, 3 and 6 h after induction of stasis), the rats were sacrificed and blood and cardiac samples were harvested for analysis. Hematoxylin-eosin(HE) staining and transmission electron microscopy were used for histopathological detection; an enzyme linked immunosorbent assay(ELISA) was used to detect thromboxane B2(TXB2) and 6-keto-prostaglandin F1α(6-Keto-PGF1α) concentrations; a real-time polymerase chain reaction(PCR) reaction system was used to detect intercellular adhesion molecule1(ICAM-1) and vascular cell adhesion molecule1(VCAM-1) m RNA expression; western blotting was used to detect vascular endothelial cadherin(VE-cadherin) protein expression.RESULTS: The ST segment in the ECG showed gradual elevation after induction of stasis and continued elevation at a high level at 3 and 6 h. The HE staining showed changes in myocardial cell necrosis and tissue dissociation after the induction of stasis, along with inflammatory infiltration. Results of transmission electron microscopy showed immediate changes in blood stasis and lumen occlusion in the microvasculature, along with endothelial cell swelling. After the induction of stasis, TXB2 concentrations gradually increased while 6-Keto-PGF1αconcentrations were immediately significantly reduced. The TXB2/6-Keto-PGF1αratio was maintained at a high level. ICAM-1 m RNA expression showed an unstable elevation while VCAM-1 m RNA expression was significantly reduced after the induction of stasis. Compared with the control group, VE-cadherin protein expression increased at 0 and 3 h after the induction of stasis, while no change occurred at 1 and 6 h.CONCLUSION: The pathological manifestations of acute blood stasis are microvascular blood retention, lumen stenosis and even occlusion. The condition is also called 'blood coagulation and weep' in Traditional Chinese Medicine. | 15 | Skeletal loading regulates breast cancer-associated osteolysis in a loading intensity-dependent fashion显示文摘Osteocytes are mechanosensitive bone cells, but little is known about their effects on tumor cells in response to mechanical stimulation. We treated breast cancer cells with osteocyte-derived conditioned medium(CM) and fluid flow-treated conditioned medium(FFCM) with 0.25 Pa and 1 Pa shear stress. Notably, CM and FFCM at 0.25 Pa induced the mesenchymal-to-epithelial transition(MET), but FFCM at 1 Pa induced the epithelial-to-mesenchymal transition(EMT). This suggested that the effects of fluid flow on conditioned media depend on flow intensity. Fluorescence resonance energy transfer(FRET)-based evaluation of Src activity and vinculin molecular force showed that osteopontin was involved in EMT and MET switching. A mouse model of tumorinduced osteolysis was tested using dynamic tibia loadings of 1, 2, and 5 N. The low 1 N loading suppressed tumor-induced osteolysis, but this beneficial effect was lost and reversed with loads at 2 and 5 N, respectively. | 16 | Identification of a locus associated with genic male sterility in maize via EMS mutagenesis and bulked-segregant RNA-seq显示文摘Genic male sterility(GMS) is one of the most important resources for exploiting heterosis in crop breeding, so that identifying genomic loci regulating GMS is desirable. However, many regulatory genes controlling GMS have not yet been characterized in maize, owing partly to a lack of genetic materials. We generated a recessive male-sterile maize mutant in the Jing 724 genetic background via ethyl methanesulfonate treatment, and found the male sterility to be due to a single gene mutation. Bulk-segregant RNA sequencing of three replicates indicated that one genomic region located at the end of chromosome 4 was associated with the observed mutant phenotype. Among genes with nonsynonymous mutations,Zm00001 d053895(bHLH51) showed abolished expression in the sterile bulks and was annotated as a bHLH transcription factor orthologous to Arabidopsis AMS, suggesting an association with the male sterility of the mutant. Kompetitive Allele-Specific PCR assays further validated the exclusive correlation of male sterility with the single C-to-T mutation in the fifth exon. | 17 | Growth and metal uptake of energy sugarcane (Saccharum spp.) in different metal mine tailings with soil amendments显示文摘A pot experiment was conducted to investigate the feasibility of growing energy sugarcane(Saccharum spp.) in three different metal mine tailings(Cu, Sn and Pb/Zn tailings) amended with uncontaminated soil at different mixing ratios. The results indicated that sugarcane was highly tolerant to tailing environments. Amendments of 20% soil to Sn tailings and 30% soil to Cu tailings could increase the biomass of cane-stem for use as the raw material for bioethanol production. Heavy metals were mostly retained in roots, which indicated that sugarcane was useful for the stabilization of the tailings. Bagasse and juice, as the most valuable parts to produce bioethanol, only accounted for 0.6%– 3% and 0.6%–7% of the total metal content. | 18 | Design of depth-control planting unit with single-side gauge wheel for no-till maize precision planter显示文摘No-till planters are very popular for maize seeding in fields covered with residue in annual wheat-maize double cropping system in North China Plain.However,there is no suitable depth control mechanism for existing no-till maize planters,and as a result,it is hard to obtain consistent planting depth,uniform emergence,and good passing ability at the same time.For the above reasons,a proper planting unit with a new type of depth-control mechanism was developed in this study.The mechanism consists of a single-side gauge wheel,a parallel four-bar linkage,a pair of double-disc opener,a V-shape press wheel and a depth regulator,which can adjust the planting depth from 30 mm to 90 mm to satisfy the agronomic requirement under different field conditions.Based on analysis and calculation,the width of gauge wheel was set to 50 mm while the length of parallel four-bar linkage was set to 350 mm.Field experiment was conducted and the results indicated that the newly designed planting unit with single-side gauge wheel performed well with regard to planting depth uniformity and anti-blocking ability.The planting depth uniformity and seed spacing quality were 95.45%and 91.90%,respectively,when the average height of stubble was 22.5 cm and residue amount was 0.64 kg/m^(2)in the field.It can satisfy the requirement of no-till maize planting on the cropland with residue and stubble in North China Plain. | Zhang Rui Cui Tao Han Dandan Zhang Dongxing Li Kehong Yin Xiaowei Wang Yunxia He Xiantao Yang Li | 2016 | International Journal of Agricultural and Biological Engineering2016,9,6: | 4 |
