1Barometric altimetry system as virtual constellation applied in CAPS显示文摘This work describes the barometric altimetry as virtual constellation applied to the Chinese Area Positioning System (CAPS), which uses the transponders of communication satellites to transfer navigation messages to users. Barometric altimetry depends on the relationship of air pressure varying with altitude in the Earth’s atmosphere. Once the air pressure at a location is measured the site altitude can be found. This method is able to enhance and improve the availability of three-dimensional positioning. The difficulty is that the relation between barometric pressure and altitude is variable in different areas and under various weather conditions. Hence, in order to obtain higher accuracy, we need to acquire the real-time air pressure corresponding to an altimetric region’s reference height. On the other hand, the altimetry method will be applied to satellite navigation system, but the greatest difficulty lies in how to get the real-time air pressure value at the reference height in the broad areas overlaid by satellite navigation. We propose an innovational method to solve this problem. It is to collect the real-time air pressures and temperatures of the 1860 known-altitude weather observatories over China and around via satellite communication and to carry out time extrapolation forecast uniformly. To reduce data quantity, we first partition the data and encode them and then broadcast these information via navigation message to CAPS users’ receivers. Upon the interpolations being done in receivers, the reference air pressure and temperature at the receiver’s nearby place is derived. Lastly, combing with the receiver-observed real air pressure and temperature, the site’s altitude can be determined. The work is presented in the following aspects: the calculation principle, formulae, data collection, encoding, prediction, interpolation method, navigation message transmission together with errors causes and analyses. 33-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years.Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators 2021Journal of Clinical and Translational Hepatology2021,9,3:7

10Photodynamic therapy-triggered on-demand drug release from ROS-responsive core-cross-linked micelles toward synergistic anti-cancer treatment显示文摘Polymeric micelles have demonstrated wide utility for chemodrug delivery,which however,still suffer from shortcomings such as undesired drug loading,disassembly upon dilution,pre-leakage of drug cargoes during systemic circulation,and lack of cancer-selective drug release.Herein,a poly(ethylene glycol)(PEG)-polyphosphoester-based,reactive oxygen species (ROS)-responsive,core-cross-linked (CCL) micellar system was developed to encapsulate both chemodrug (doxorubicin,Dox) and photosensitizer (chlorin e6,Ce6).The hydrophobic core of the micelles was cross-linked via a thioketal (TK)-containing linker,which notably enhanced the drug loading and micelle stability.In tumor cells,far-red light irradiation of Ce6 generated ROS to cleave the TK linkers and disrupt the micelle cores.As such,micelles were destabilized and Dox release was promoted,which thereafter imparted synergistic anti-cancer effect with ROS-mediated photodynamic therapy.This study provides an effective approach to realize the precise control over drug loading,formulation stability,and cancer-selective drug release using polymeric micelles,and would render promising utilities for the programmed anti-cancer combination therapy.Yongjuan Li Jian Hu Xun Liu Yong Liu Shixian Lv Juanjuan Dang Yong Ji Jinlin He Lichen Yin 2019Nano Research2019,12,5:5
11Effect of catalyst confinement and pore size on Fischer-Tropsch synthesis over cobalt supported on carbon nanotubes显示文摘This paper studies the impact of structure of cobalt catalysts supported on carbon nanotubes(CNT) on the activity and product selectivity of Fischer-Tropsch synthesis(FTS) reaction.Three types of CNT with average pore sizes of 5,11,and 17 nm were used as the supports.The catalysts were prepared by selectively impregnating cobalt nanoparticles either inside or outside CNT.The TPR results indicated that the catalyst with Co particles inside CNT was easier to be reduced than those outside CNT,and the reducibility of cobalt oxide particles inside the CNT decreased with the cobalt oxide particle size increasing.The activity of the catalyst with Co inside CNT was higher than that of catalysts with Co particles outside CNT.Smaller CNT pore size also appears to enhance the catalyst reduction and FTS activity due to the little interaction between cobalt oxide with carbon and the enhanced electron shift on the non-planar carbon tube surface.XIE Wei ZHANG YuHua LIEW KongYong LI JinLin 2012Science China Chemistry2012,55,9:4
12Viral and Antibody Kinetics of COVID-19 Patients with Different Disease Severities in Acute and Convalescent Phases:A 6-Month Follow-Up Study显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has spread rapidly around the world,posing a major threat to human health and the economy.Currently,long-term data on viral shedding and the serum antibody responses in COVID-19 patients are still limited.Herein,we report the clinical features,viral RNA loads,and serum antibody levels in a cohort of 112 COVID-19 patients admitted to the Honghu People’s Hospital,Hubei Province,China.Overall,5.36%(6/112)of patients showed persistent viral RNA shedding(>45 days).The peak viral load was higher in the severe disease group than in the mild group(median cycle threshold value,36.4 versus 31.5;P=0.002).For most patients the disappearance of IgM antibodies occurred approximately 4–6 weeks after symptoms onset,while IgG persisted for over 194 days after the onset of symptoms,although patients showed a 46%reduction in antibodies titres against SARS-CoV-2 nucleocapsid protein compared with the acute phase.We also studied18 asymptomatic individuals with RT-qPCR confirmed SARS-CoV-2 infection together with 17 symptomatic patients,and the asymptomatic individuals were the close contacts of these symptomatic cases.Delayed IgG seroconversion and lower IgM seropositive rates were observed in asymptomatic individuals.These data indicate that higher viral loads and stronger antibody responses are related to more severe disease status in patients with SARS-CoV-2 infection,and the antibodies persisted in the recovered patient for more than 6 months so that the vaccine may provide protection against SARS-CoV-2 infection.Xiaoyong Zhang Suwen Lu Hui Li Yi Wang Zhen Lu Zhihong Liu Qingtao Lai Yali Ji Xuan Huang Yongyin Li Jian Sun Yingsong Wu Xiaoning Xu Jinlin Hou 2020Virologica Sinica2020,35,6:4
13Nickel catalysts supported on MgO with different specific surface area for carbon dioxide reforming of methane显示文摘In this paper, three kinds of MgO with different specific surface area were prepared, and their effects on the catalytic performance of nickel catalysts for the carbon dioxide reforming of methane were investigated. The results showed that MgO support with the higher specific surface area led to the higher dispersion of the active metal, which resulted in the higher initial activity. On the other hand, the specific surface area of MgO materials might not be the dominant factor for the basicity of support to chemisorb and activate CO2, which was another important factor for the performance of catalysts. 14Prediction and systematic study of protein-protein interaction networks of Leptospira interrogans显示文摘Leptospira interrogans serovar Lai is a pathogenic bacterium that causes a spirochetal zoonosis in humans and some animals. With its complete genome sequence available, it is possible to analyze protein-protein interactions from a whole- genome standpoint. Here we combine four recently developed computational approaches (gene fusion method, gene neighbor method, phylogenetic profiles method, and operon method) to predict protein-pro- tein interaction networks of Leptospira interrogans strain Lai. Through comprehensive analysis on in- teractions among proteins of motility and chemotaxis system, signal transduction, lipopolysaccaride bio- synthesis and a series of proteins related to adhesion and invasion, we provided information for further studying on its pathogenic mechanism. In addition, we also assigned 203 previously uncharacterized proteins with possible functions based on the known functions of its interacting partners. 15Blast-induced rock damage control in Fangchenggang nuclear power station, China显示文摘In the process of blasting excavation, stress wave propagation and gas expansion can basically induce damage to surrounding rocks, which is detrimental to rock mass integrity and engineering safety. In this case, evaluation and control of blast-induced effects are essential to the safety of nearby buildings and integrity of bedrock in blasting field. In Fangchenggang nuclear power station of China, the drill-andblast method was employed for bedrock excavation. In order to reduce the blast-induced damage zone, the wave propagation and associated damage to rock mass should be carefully investigated. In this paper, the wave propagation regressively obtained from field monitoring data was presented based on empirical formula(e.g. Sadovsky formula). The relationship between the peak particle velocity(PPV) at a distance of 30 m away from the charge hole and charge per delay in blast design was derived. Meanwhile,the acoustic tests before and after blasting were conducted to determine the damage depth of rock mass.The charge per delay in blast design was then calibrated based on the blast-induced wave propagation regularity. The results showed that a satisfactory effect was achieved on blast-induced damage control of rock mass. 16Facile preparation of pH-responsive PEGylated prodrugs for activated intracellular drug delivery显示文摘PEGylated prodrug,covalent attaching polyethylene glycol(PEG) polymer chains to therapeutic drugs,is one of the most promising techniques to improve the water-solubility,stability,and therapeutic effect of drugs.In this study,three PEGylated acid-sensitive prodrugs DOX-PEG-DOX with different molecular weights,were prepared via Schiff-base reaction between aldehyde-modified PEG and the amino groups of doxorubicin(DOX).This kind of amphiphilic polymeric prodrug could be self-assemble into nanoparticles in aqueous solution.The average particle size and morphologies of the prodrug nanoparticles under different pH conditions were observed by dynamic light scattering(DLS) and transmission electron microscopy(TEM),re s pectively.It turned out that the nanoparticles could be kept stable in the physiological environment,but degraded in acidic medium.Subsequently,we also investigated in vitro drug release behavior and found that the prodrug had acid-sensitive property.The cytotoxicity and intracellular uptake assays revealed that the prodrugs could rapidly internalized by HeLa or HepG2 cells to release DOX and effectively inhibited the proliferation of the tumor cells,which have the potential for use in cancer therapy.Yue Song Dian Li Jinlin He Mingzu Zhang Peihong Ni 2019Chinese Chemical Letters2019,30,12:3
1796-Week Treatment of Tenofovir Amibufenamide and Tenofovir Disoproxil Fumarate in Chronic Hepatitis B Patients显示文摘Background and Aims:Tenofovir amibufenamide(TMF)is a novel phosphoramidated prodrug of tenofovir with nonin-ferior efficacy and better bone and renal safety to tenofovir disoproxil fumarate(TDF)in 48 weeks of treatment.Here,we update 96-week comparison results.Methods:Patients with chronic hepatitis B were assigned(2:1)to receive either 25 mg TMF or 300 mg TDF with matching placebo for 96 weeks.The virological suppression was defined as HBV DNA levels<20 IU/mL at week 96.Safety was evaluated thoroughly with focusing on bone,renal,and metabolic pa-rameters.Results:Virological suppression rates at week 96 were similar between TMF and TDF group in both HBeAg-positive and HBeAg-negative populations.Noninferior efficacy was maintained in the pooled population,while it was first achieved in patients with HBV DNA≥7 or 8 log10 IU/mL at baseline.Non-indexed estimated glomerular filtration rate for renal safety assessment was adopted,while a smaller decline of which was seen in the TMF group than in the TDF group(p=0.01).For bone mineral density,patients receiv-ing TMF displayed significantly lower reduction levels in the densities of spine,hip,and femur neck at week 96 than those receiving TDF.In addition,the lipid parameters were stable after week 48 in all groups while weight change still showed the opposite trend.Conclusions:TMF maintained similar efficacy at week 96 compared with TDF with continued superior bone and renal safety profiles(NCT03903796).Zhihong Liu Qinglong Jin Yuexin Zhang Guozhong Gong Guicheng Wu Lvfeng Yao Xiaofeng Wen Zhiliang Gao Yan Huang Daokun Yang Enqiang Chen Qing Mao Shide Lin Jia Shang Huanyu Gong Lihua Zhong Huafa Yin Fengmei Wang Peng Hu Qiong Wu Chao Pan Wen Jia Chuan Li Chang’an Sun Junqi Niu Jinlin Hou TMF Study Group 2023Journal of Clinical and Translational Hepatology2023,11,3:3
18Catalytic performance of iron oxide loaded on electron-rich surfaces of carbon nitride显示文摘Carbon nitride(CN) in CN encapsulated Ni/Al_2O_3(denoted as CN/Ni/Al_2O_3) catalyst was evidenced previously as a material in electron-rich state and possessed H2-dissociative adsorption activity due to the electron doping effect from underlying nickel. In this report, iron oxide loaded on the CN/Ni/Al_2O_3 was synthesized and investigated by Fischer-Tropsch(F-T) synthesis to test the special effect of electron-rich support on the catalytic activity of iron oxide. The Fe/CN/Al_2O_3 and CN/Ni/Al_2O_3 samples were accordingly synthesized for comparison. In Fe/CN/Ni/Al_2O_3, the iron oxide was reduced to magnetite by syngas as evidenced by the in-situ XPS measurements and XRD pattern of used catalyst. Compared with Fe/CN/Al_2O_3, more light hydrocarbons over Fe/CN/Ni/Al_2O_3 were observed. It should be understood by the interaction between iron oxide and support mainly due to the effect of electron-rich state and thus enhanced H_2 adsorption ability. In addition, such a novel support facilitated the CO conversion and retarded the water-gas shift reaction and CO2 formation. 19p53 deficiency-induced Smad1 upregulation suppresses tumorigenesis and causes chemoresistance in colorectal cancers显示文摘The DNA damage response helps to maintain genome integrity,suppress tumorigenesis,and mediate the effects of radiotherapy and chemotherapy.Our previous studies have shown that Smad1 is upregulated and activated by Atm in DNA damage response,which can further bind to p53 and promote p53 stabilization.Herewe report another aspect of the interplay between p53 and Smad1.Comparison of rectal tumor against paired paraneoplastic specimens and analysis of>500 colorectal tumors revealed that Smad1 was upregulated in tumor samples,which was attributable to p53 defects.Using MEFs as a model,we found that knockdown of the elevated Smad1 in p532/2 MEFs promoted cell proliferation,E1A/Ras-induced cell transformation,and tumorigenesis.Mechanistic studies suggest that elevated Smad1 and momentary activation inhibit cell proliferation by upregulating p57Kip2 and enhancing Atm–Chk2 activation.Surprisingly,elevated Smad1 appears to have a negative effect on chemotherapy,as colorectal tumors,primary cancer cells,and cell lines with Smad1 knockdown all showed an increase in chemosensitivity,which could be attributable to elevated p57Kip2.These findings underscore the significance of Smad1–p53 interaction in tumor suppression and reveal an unexpected role for Smad1 in chemoresistance of colorectal cancers.Xinsen Ruan Qiao Zuo Hao Jia Jenny Chau Jinlin Lin Junping Ao Xuechun Xia Huijuan Liu Samy LHabib Chuangang Fu Baojie Li 2015Journal of Molecular Cell Biology2015,7,2:2
20Analysis and application of large-scale protein-protein interaction data sets显示文摘Protein-protein interactions play key roles in cells. Lots of experimental approaches and in silico methods have been developed to identify and predict large-scale pro- tein-protein interactions. However, compared with the tradi- tionally experimental results, the high-throughput pro- tein-protein interaction data often contain the false positives in high probability. In order to fully utilize the large-scale data, it is necessary to develop bioinformatic methods for systematically evaluating those data in order to further im- prove the data reliability and mine biological information. This review summarizes the methodologies of analysis and application of high-throughput protein-protein interaction data, including the evaluation methods, the relationship be- tween protein-protein interaction data and other protein biological information, and their applications in biological study.
