| 4 | Impact of organic interlayer anions on the CO2 adsorption performance of Mg-Al layered double hydroxides derived mixed oxides显示文摘Herein we report a systematical investigation on the promoting effect of the carbon chain length of the intercalated carboxylic anions on the CO_2 capture performance of Mg-Al layer double hydroxides(LDHs).A series of organo-LDHs were successfully synthesized via co-precipitation and calcination-rehydration methods. All as-prepared samples were characterized by many techniques including XRD, ATR-FTIR, BET,and TGA. The XRD and ATR-FTIR studies indicated that organic anions were successfully intercalated into LDHs. The influence of some important parameters such as calcination temperature, adsorption temperature, and coating with(Li-Na-K)NO_3 molten salt was investigated. The results exhibited that when the number of carbon is greater than 10, the CO_2 capture capacity steadily increased with the increase in carbon number. After coating with 55 mol%(Li-Na-K)NO_3 molten salt, the CO_2 uptake of LDH-C16 sample with high Mg/Al ratios can be increased up to 3.25 mmol/g. The CO_2 adsorption/desorption cycling stability was also studied using temperature swing adsorption, which showed a stable CO_2 capture performance even after 22 cycles. | 6 | Effects of chlortetracycline,Cu and their combination on the performance and microbial community dynamics in swine manure anaerobic digestion显示文摘Swine manure was typical for the combined pollution of heavy metals and antibiotics. The effects of widely used veterinary antibiotic chlortetracycline(CTC), Cu and their combination on swine manure anaerobic digestion performance and microbial community have never been investigated. Thus, four 2 L anaerobic digestion reactors were established including reactor A(control), B(CTC spiked by 0.5 g/kg dry weight, dw), C(Cu spiked by 5 g/kg dw) and D(combination of CTC, 0.5 g/kg dw, and Cu, 5 g/kg dw), and dynamics of bacterial and archaeal community structure was investigated using high throughput sequencing method. Results showed that addition of CTC and Cu separately could increase the total biogas production by21.6% and 15.8%, respectively, while combination of CTC and Cu severely inhibited anaerobic digestion(by 30.3%). Furthermore, corresponding to different stages and reactors, four kinds of microbes including bacteria and archaea were described in detail, and the effects of CTC, Cu and their combination mainly occurred at hydrolysis and acidification phases. The addition of Cu alone changed the dynamics of archaeal community significantly. | 7 | A review of progress in coupled ocean-atmosphere model developments for ENSO studies in China显示文摘El Nino-Southern Oscillation(ENSO) is the strongest interannual signal that is producedby basinscale processes in the tropical Pacific,with significant effects on weather and climate worldwide.In the past,extensive and intensive international efforts have been devoted to coupled model developments for ENSO studies.A hierarchy of coupled ocean-atmo sphere models has been formulated;in terms of their complexity,they can be categorized into intermediate coupled models(ICMs),hybrid coupled models(HCMs),and fully coupled general circulation models(CGCMs).ENSO modeling has made significant progress over the past decades,reaching a stage where coupled models can now be used to successfully predict ENSO events 6 months to one year in advance.Meanwhile,ENSO exhibits great diversity and complexity as observed in nature,which still cannot be adequately captured by current state-of-the-art coupled models,presenting a challenge to ENSO modeling.We primarily reviewed the long-term efforts in ENSO modeling continually and steadily made at different institutions in China;some selected representative examples are presented here to review the current status of ENSO model developments and applications,which have been actively pursued with noticeable progress being made recently.As ENSO simulations are very sensitive to model formulations and process representations etc.,dedicated efforts have been devoted to ENSO model developments and improvements.Now,different ocean-atmosphere coupled models have been available in China,which exhibit good model performances and have already had a variety of applications to climate modeling,including the Coupled Model Intercomparison Project Phase 6(CMIP6).Nevertheless,large biases and uncertainties still exist in ENSO simulations and predictions,and there are clear rooms for their improvements,which are still an active area of researches and applications.Here,model performances of ENSO simulations are assessed in terms of advantages and disadvantages with these differently formulated coupled models,pinpointing to the areas where they need to be further improved for ENSO studies.These analyses provide valuable guidance for future improvements in ENSO simulations and predictions. | ZHANG Rong-Hua YU Yongqiang SONG Zhenya REN Hong-Li TANG Youmin QIAO Fangli WU Tongwen GAO Chuan HU Junya TIAN Feng ZHU Yuchao CHEN Lin LIU Hailong LIN Pengfei WU Fanghua WANG Lin | 2020 | Journal of Oceanology and Limnology2020,38,4: | 4 |
| 8 | Optimized prime editing efficiently generates glyphosate-resistant rice plants carrying homozygous TAP-IVS mutation in EPSPS显示文摘Dear Editor,Prime editing is a novel and universal CRISPR-Cas-derived precise genome-editing technology and has great potentials for applications in basic plant research and crop molecular breeding(Anzalone et al.,2019).Although low efficiency has restrained the original prime editors(PEs)from being used as a routine tool for precise genome editing in plants,an iterative update of the PEs is removing this obstacle(Lin et al.,2021;Xu et al.,2022).Recently,the Liu group reported three optimization strategies for improving prime editing efficiency(Chen et al.,2021;Nelson et al.,2022).The first strategy is based on engineered prime editing guide RNAs(epegRNAs),which were generated by incorporating structured RNA motifs to the 3′terminus of pegRNAs.This strategy enhances pegRNA stability and prevents degradation of the 3′extension(Nelson et al.,2022).The second strategy is based on the optimized PE2 protein(PEmax),which harbors a SpCas9 variant with increased nuclease activity,an additional nuclear localization signal(NLS)sequences,and a new linker between nCas9 and reverse transcriptase(Chen et al.,2021).The third strategy is based on inhibition of DNA mismatch repair(MMR)in cells(Chen et al.,2021).In this work,we tested the optimized PEs generated with these three strategies in rice,demonstrating that the optimized PEs greatly improved prime editing efficiency in rice.We named the two optimized PEs ePE3max and ePE5max:the former is comprised of the PEmax protein,an epegRNA with evopreQ1 appended to its 3′end,and a nicking sgRNA;the latter is comprised of the ePE3max system and a dominant negative OsMLH1 variant for inhibiting MMR.Using the two optimized PEs,we efficiently generated homozygous and heterozygous T173I,A174V,and P177S(TAP-IVS)mutation in EPSPS in rice,which lays a solid foundation for rice non-transgenic glyphosate-resistance breeding. | Yuanyuan Jiang Yiping Chai Dexin Qiao Junya Wang Cuiping Xin Wei Sun Zhenghong Cao Yu Zhang Yun Zhou Xue-Chen Wang Qi-Jun Chen | 2022 | Molecular Plant2022,15,11: | 3 |

| 10 | Delaminated layered double hydroxide delivers DNA molecules as sandwich nanostructure into cells via a non-endocytic pathway显示文摘Layered double hydroxides(LDHs) are effective molecular carriers in cytological research, gene therapy,and transgenic applications. Herein, we investigated the internalization behavior of the LDH-DNA bioconjugates via a microscopic approach and analyzed the internalization pathway by dissipative particle dynamics(DPD) simulations. We experimentally found that LDH can efficiently carry DNA into the nucleus of cell in BY-2 suspension cells. Furthermore, atomic force microscopy and X-ray diffraction analysis demonstrated that the LDH-DNA bioconjugates mainly exist as a DNA-LDH-DNA sandwich complex,while the LDH-DNA-LDH sandwich complex and DNA-LDH complex cannot be excluded. The DPD simulations further indicated that only the DNA-LDH-DNA sandwich structure could penetrate the plasma membrane(PM), while PM is impermeable to the LDH-DNA-LDH sandwich complex and the DNA-LDH complex. This work provides novel perspective for understanding the membrane penetration mechanism of LDH nano-sheets and new insights into the design of novel molecular delivery systems.ó 2017 Science China Press. | 11 | DNA damage response-related immune activation signature predicts the response to immune checkpoint inhibitors: from gastrointestinal cancer analysis to pan-cancer validation显示文摘Objective: DNA damage response(DDR) deficiency has emerged as a prominent determinant of tumor immunogenicity. This study aimed to construct a DDR-related immune activation(DRIA) signature and evaluate the predictive accuracy of the DRIA signature for response to immune checkpoint inhibitor(ICI) therapy in gastrointestinal(GI) cancer.Methods: A DRIA signature was established based on two previously reported DNA damage immune response assays. Clinical and gene expression data from two published GI cancer cohorts were used to assess and validate the association between the DRIA score and response to ICI therapy. The predictive accuracy of the DRIA score was validated based on one ICI-treated melanoma and three pan-cancer published cohorts.Results: The DRIA signature includes three genes(CXCL10, IDO1, and IFI44L). In the discovery cancer cohort, DRIA-high patients with gastric cancer achieved a higher response rate to ICI therapy than DRIA-low patients(81.8% vs. 8.8%;P < 0.001), and the predictive accuracy of the DRIA score [area under the receiver operating characteristic curve(AUC) = 0.845] was superior to the predictive accuracy of PD-L1 expression, tumor mutational burden, microsatellite instability, and Epstein–Barr virus status. The validation cohort demonstrated that the DRIA score identified responders with microsatellite-stable colorectal and pancreatic adenocarcinoma who received dual PD-1 and CTLA-4 blockade with radiation therapy. Our aim is to maximize the EE which is defined as the ratio of the transmission rate to the total consumed power subject to the maximum transmit power of each remote antenna constraint and Qo S(target BER) requirement. According to the definition of EE and using the upper bound of average EE,the optimized objective function is provided. Based on this,utilizing Karush-KuhnTucker conditions and numerical calculation,a suboptimal energy efficient power allocation(PA) scheme is developed,and the closedform expression of PA coefficients is derived. The scheme may obtain the EE performance close to the existing optimal scheme. Moreover,it has relatively lower complexity than the existing scheme because only the statistic channel information and less iteration are required.
