SKIP Confers Osmotic Tolerance during Salt Stress by Controlling Alternative Gene Splicing in Arabidopsis显示文摘译解位于下面的机制种回答到不能生活的应力为改进植物压力抵抗是关键的。 Jinlin Feng dingjing Li Zhaoxu Gao Yaru Lu Junya Yu Qian Zheng Shuning Yan Wenjiao Zhang Hang He Ligeng Ma Zhengge Zhu 2015Molecular Plant2015,8,7:10
A triple-RBD-based mucosal vaccine provides broad protectionagainst SARS-CoV-2 variants of concern显示文摘The rapid mutation and spread of SARS-CoV-2 variants urge the development of effective mucosal vaccines to provide broadspectrum protection against the initial infection and thereby curb the transmission potential. Here,we designed a chimeric tripleRBD immunogen,3Ro-NC,harboring one Delta RBD and two Omicron RBDs within a novel protein scaffold.3Ro-NC elicits potent and broad RBD-specific neutralizing immunity against SARS-CoV-2 variants of concern.Notably, intranasal immunization with 3RoNC plus the mucosal adjuvant KFD(3Ro-NC+KFDi.n)elicits coordinated mucosal IgA and higher neutralizing antibody specificity(closer antigenic distance)against the Omicron variant. In Omicron-challenged human ACE2 transgenic mice,3Ro-NC+KFDi.n immunization significantly reduces the tissue pathology in the lung and lowers the viral RNA copy numbers in both the lung(85.7-fold)and the nasal turbinate(13.6-fold).Nasal virologic control is highly correlated with RBD-specific secretory IgA antibodies. Our data show that 3Ro-NC plus KFD is a promising mucosal vaccine candidate for protection against SARS-CoV-2 Omicron infection, pathology and transmission potential.Jingyi Yang Mei-Qin Liu Lin Liu Xian Li Mengxin Xu Haofeng Lin Shuning Liu Yunqi Hu Bei Li Bowen Liu Min Li Ying Sun Yao-Qing Chen Zheng-Li Shi Huimin Yan 2022Cellular & Molecular Immunology2022,19,11:3
Rapid isolation and immune profiling of SARS-CoV-2 specific memory B cell in convalescent COVID-19 patients via LIBRA-seq显示文摘B cell response plays a critical role against SARS-CoV-2 infection. However, little is known about the diversity and frequency of the paired SARS-CoV-2 antigen-specific BCR repertoire after SARS-CoV-2 infection. Here,we performed single-cell RNA sequencing and VDJ sequencing using the memory and plasma B cells isolated from five convalescent COVID-19 patients, and analyzed the spectrum and transcriptional heterogeneity of antibody immune responses.Via linking BCR to antigen specificity through sequencing(LIBRA-seq),we identified a distinct activated memory B cell subgroup(CD11c^(high) CD95^(high))had a higher proportion of SARS-CoV-2 antigen-labeled cells compared with memory B cells. Our results revealed the diversity of paired BCR repertoire and the non-stochastic pairing of SARS-CoV-2 antigen-specific immunoglobulin heavy and light chains after SARS-CoV-2 infection. The public antibody clonotypes were shared by distinct convalescent individuals. Moreover, several antibodies isolated by LIBRA-seq showed high binding affinity against SARS-CoV-2 receptor-binding domain(RBD)or nucleoprotein(NP)via ELISA assay. Two RBD-reactive antibodies C14646P3S and C2767P3S isolated by LIBRA-seq exhibited high neutralizing activities against both pseudotyped and authentic SARS-CoV-2 viruses in vitro. Our study provides fundamental insights into B cell response following SARS-CoV-2 infection at the single-cell level.Bing He Shuning Liu Yuanyuan Wang Mengxin Xu Wei Cai Jia Liu Wendi Bai Shupei Ye Yong Ma Hengrui Hu Huicui Meng Tao Sun Yanling Li Huanle Luo Mang Shi Xiangjun Du Wenjing Zhao Shoudeng Chen Jingyi Yang Haipeng Zhu Yusheng Jie Yuedong Yang Deyin Guo Qiao Wang Yuwen Liu Huimin Yan Manli Wang Yao-Qing Chen 2021Signal Transduction and Targeted Therapy2021,6,6:2
Downregulation of MANCR inhibits cancer cell proliferation in mantle cell lymphoma possibly by interacting with RUNX2显示文摘The mitotically associated lncRNA(MANCR)participates in breast cancer cell proliferation, while its involvement in other cancers is still unknown. In this study,we therefore studied the role of MANCR in mantle cell lymphoma(MCL).We found that serum MANCR and Runt-related transcription factor 2(RUNX2)were upregulated in MCL patients when compared with those in healthy controls.A positive correlation between serum MANCR and RUNX2 was found in MCL patients but not in controls.Upregulation of serum MANCR distinguished MCL patients from controls.MANCR overexpression promoted RUNX2 expression in MCL cells, while RUNX2 overexpression failed to significantly change the expression levels of MANCR.MANCR overexpression promoted the proliferation of MCL cells, while MANCR silencing inhibited the proliferation of MCL cells. In addition,RUNX2 overexpression attenuated the inhibitory effects of MANCR silencing on cell proliferation. However,MANCR overexpression and silencing had no significant effects on cell migration and invasion.Further bioinformatics analysis showed that MANCR may sponge miR-218 to upregulate RUNX2.Therefore,we conclude that downregulation of MANCR may inhibit cancer cell proliferation in MCL possibly by interacting with RUNX2.Shujuan Wen Min Zeng Yan Li Xin Hu Shan Li Xiao Liang Lin Zhu Shune Yang 2019Acta Biochimica et Biophysica Sinica2019,51,11:1
Exome sequencing identifies rare mutations of LDLR and QTRT1 conferring risk for early-onset coronary artery disease in Chinese显示文摘Despite the advances made over the past decades, the known loci for coronary artery disease(CAD)still only explain<20%of the genetic variation in risk[1,2].The known loci with the strongest effects usually confer a 20%-37%increased CAD risk and the most loci modulate risk by≤10%[1].While most of the current data are from European populations, the use of trans-ethnic analyses could be helpful to identify additional loci.Genetic inheritance may impose a high burden on early-onset coronary artery disease(EOCAD).Kang Yao Yuxiang Dai Juan Shen Yi Wang Huanjie Yang Runda Wu Qijun Liao Hongyi Wu Xiaodong Fang Shalaimaiti Shali Lili Xu Meng Hao Chenhao Lin Zhonghan Sun Yilian Liu Mengxin Li Zhen Wang Qiang Gao Shuning Zhang Chenguang Li Wei Gao Lei Ge Yunzeng Zou Aijun Sun Juying Qian Li Jin Shangyu Hong Yan Zheng Junbo Ge 2022National Science Review2022,9,8:1
Catalytic ozonation in advanced treatment of kitchen wastewater:multi-scale simulation and pilot-scale study显示文摘Catalytic ozonation is regarded as a promising technology in the advanced treatment of refractory organic wastewater.Packed-bed reactors are widely used in practical applications due to simple structures, installation and operation. However, mass transfer of packed-bed reactors is relatively restrained and amplified deviations usually occurred in scale-up application.Herein,a multi-scale packed-bed model of catalytic ozonation was established to guide pilot tests.First,a laboratory-scale test was conducted to obtain kinetic parameters needed for modeling. Then,a multi-scale packed-bed model was developed to research the effects of water distribution structure, catalyst particle size, and hydraulic retention time(HRT)on catalytic ozonation. It was found that the performance of packed bed reactor was increased with evenly distributed water inlet,HRT of 60 min,and catalyst diameter of about 3-7 mm.Last,an optimized reactor was manufactured and a pilot-scale test was conducted to treat kitchen wastewater using catalytic ozonation process. In the pilot-scale test with an ozone dosage of 50 mg/L and HRT of 60 min,the packed-bed reactor filled with catalysts I was able to reduce chemical oxygen demand(COD)from 117 to 59 mg/L.The performance of the catalytic ozonation process in the packed-bed reactor for the advanced treatment of actual kitchen wastewater was investigated via both multi-scale simulation and pilot-scale tests in this study, which provided a practical method for optimizing the reactors of treating refractory organic wastewater.Zuoyong Zhou Ni Yan Mengxi Yin Tengfei Ren Shuning Chen Kechao Lu Xiaoxin Cao Xia Huang Xiaoyuan Zhang 2023Frontiers of Environmental Science & Engineering2023,17,12:0
A neutralizing-protective supersite of human monoclonal antibodies for yellow fever virus显示文摘The yellow fever virus(YFV)is a life-threatening human pathogen.Owing to the lack of available therapeutics,non-vaccinated individuals are at risk. Here,we isolated eight human monoclonal antibodies that neutralize YFV infection.Five recognized overlapping epitopes and exhibited potent neutralizing activity.Two(YD6 and YD73)were ultra-potent and conferred complete protection against the lethal challenge of YFV as both prophylactics and therapeutics in a mouse model.Crystal structures revealed that YD6 engaged the YFV envelope protein in both pre-and post-fusion states, suggesting viral inhibition by a“double-lock”mechanism.The recognition determinants for YD6 and YD73 are clustered at the premembrane(prM)-binding site.Notably, antibodies targeting this site were present in minute traces in YFV-infected individuals but contributed significantly to neutralization, suggesting a vulnerable supersite of YFV.We provide two promising candidates for immunotherapy against YFV,and the supersite represents an ideal target for epitope-based vaccine design.Yan Li Zhihai Chen Lili Wu Lianpan Dai Jianxun Qi Yan Chai Shihua Li Qihui Wang Zhou Tong Sufang Ma Xiaomin Duan Shuning Ren Rui Song Mifang Liang Wenjun Liu Jinghua Yan George F.Gao 2022The Innovation2022,3,6:0
The impact of venetoclax based regimens in the preemptive of measurable residual disease in acute myeloid leukemia显示文摘To The Editor:The role of measurable residual disease(MRD)in prognosis and treatment in acute myeloid leukemia(AML)is evolving.Studies have demonstrated the correlation between MRD and risks of relapse in adult AML:persistently positive MRD after induction is associated with a high risk of relapse,1,2 and these patients should consider allogeneic transplantation(allo-Hematopoietic Stem Cell Transplantation(HSCT))and clinical trial, even in favorable-risk groups. However, because of the financial issue or lack of suitable transplant donors, many of the patients could not receive allo-HSCT,so how to prolong the relapse-free survival of these patients remains a challenge.Platzbecker et al treated MRD-positive patients with azacytidine(AZA),and found pre-emptive therapy with AZA can prevent or substantially delay hematological relapse in MRD-positive patients with MDS(myelodysplastic syndrome)or AML who are at a high risk of relapse.Qiuyun Fang Xiaoyuan Gong Yan Li Benfa Gong Yuntao Liu Kaiqi Liu Guangji Zhang Shuning Wei Dong Lin Bingcheng Liu Ying Wang Hui Wei Yingchang Mi Jianxiang Wang 2022Blood Science2022,4,1:0
