1Early treadmill exercise increases macrophage migration inhibitory factor expression after cerebral ischemia/reperfusion显示文摘The neuroprotective function of macrophage migration inhibitory factor(MIF) in ischemic stroke was rarely evaluated.This study aimed to investigate the effects of early treadmill exercise on recovery from ischemic stroke and to determine whether these effects are associated with the expression levels of MIF and brain-derived neurotrophic factor(BDNF) in the ischemic area.A total of 40 male Sprague-Dawley rats were randomly assigned to the ischemia and exercise group [middle cerebral artery occlusion(MCAO)-Ex,n = 10),ischemia and sedentary group(MCAO-St,n = 10),sham-surgery and exercise group(Sham-Ex,n= 10),or sham-surgery and sedentary group(Sham-St,n = 10).The MCAO-Ex and MCAO-St groups were subjected to MCAO for 60 minutes,whereas the Sham-Ex and Sham-St groups were subjected to an identical operation without MCAO.Rats in the MCAO-Ex and Sham-Ex groups then ran on a treadmill for 30 minutes once a day for 5 consecutive days.After reperfusion,the hanging time tested by the wire hang test was longer and the relative fractional anisotropy determined by MRI was higher in the peri-infarct region of the MCAO-Ex group compared with the MCAO-St group.The expression levels of MIF and BDNF in the peri-infarct region were upregulated in the MCAO-Ex group.Increased MIF and BDNF levels were positively correlated with relative fractional anisotropy changes in the peri-infarct region.There was no significant difference in the levels of MIF and BDNF in the peri-infarct region between the Sham-Ex and Sham-St groups.Our study demonstrated that early exercise(initiated 48 hours after the MCAO) could improve motor and neuronal recovery after ischemic stroke.Furthermore,the increased levels of MIF and BDNF in the peri-infarct region(penumbra) may be one of the mechanisms of enhanced neurological function recovery.All experiments were approved by the Institutional Animal Care and Use Committee in Asan Medical Center in South Korea(2016-12-126).Min Cheol Chang Chae Ri Park Seung Hwa Rhie Woo Hyun Shim Dae Yul Kim 2019Neural Regeneration Research2019,14,7:7

3MRI of magnetically labeled mesenchymal stem cells in hepatic failure model显示文摘AIM:To track intravascularly transplanted mesenchymal stem cells (MSCs) labeled with superparamagnetic iron oxide (SPIO) by using magnetic resonance imaging (MRI) in an experimental rabbit model of hepatic failure.METHODS:Human MSCs labeled with FDA-approved SPIO particles (Feridex) were transplanted via the mes-enteric vein into rabbits (n=16) with carbon tetrachloride-induced hepatic failure.Magnetic resonance (MR) examinations were performed with a 3.0 T clinical scanner immediately before and 2 h and 1,3,and 7 d after transplantation.Signal intensity (SI) changes on T2weighted MRI were measured,and correlation between MR findings and histomorphologic findings was also investigated.RESULTS:SI on T2-weighted MRI decreased significantly in the liver 2 h after injection of human MSCs and returned gradually to the levels found before injection in 7 d.Changes in SI in the liver at 2 h,1,3,and 7 d were 41.87% ± 9.63%,10.42% ± 4.3%,5.12% ± 1.9%,3.75% ± 1.2%,respectively (P < 0.001).Histologic analyses confirmed the presence of MSCs in the liver,localized mainly in the sinusoids in early period (2 h and 1 d) and concentrated to the border zone in late period (3 and 7 d).The number of iron-positive cells in the liver at 2 h and on 1,3 and 7 d after transplantation was 29.2 ± 4.8,10.1 ± 3.7,6.7 ± 2.2,and 5.8 ± 2.1,respectively (P=0.013).CONCLUSION:Intravascularly injected SPIO-labeled MSCs in an experimental rabbit model of hepatic failure can be detected and followed with MRI.Kyu Ri Son Se Young Chung Hyo-Cheol Kim Hoe Suk Kim Seung Hong Choi Jeong Min Lee Woo Kyung Moon 2010World Journal of Gastroenterology2010,16,44:4
4Effects of traditional Chinese herbal medicine San-HuangXie-Xin-Tang on gastrointestinal motility in mice显示文摘AIM: To investigate the effects of San-Huang-Xie-XinTang(SHXXT), a herbal product used in traditional Chinese medicine, on gastrointestinal(GI) motility in mice.METHODS: The in vivo effects of SHXXT on GI motility were investigated by measuring the intestinal transit rates(ITRs) using Evans blue in normal mice and in mice with experimentally induced GI motility dysfunction(GMD).RESULTS: In normal ICR mice, ITRs were significantly and dose-dependently increased by SHXXT(0.1-1 g/kg). GMD was induced by injecting acetic acid or streptozotocin intraperitoneally. 8Cancer-associated fibroblast-derived secreted phosphoprotein 1 contributes to resistance of hepatocellular carcinoma to sorafenib and lenvatinib显示文摘Background:Cancer-associated fibroblasts(CAFs)play an important role in the induction of chemo-resistance.This study aimed to clarify the mechanism underlying CAF-mediated resistance to two tyrosine kinase inhibitors(TKIs),sorafenib and lenvatinib,and to identify a novel therapeutic target for overcoming TKI resistance in hepatocellular carcinoma(HCC).Methods:We performed a systematic integrative analysis of publicly available gene expression datasets and whole-transcriptome sequencing data from 9 pairs of CAFs and para-cancer fibroblasts isolated from human HCC and para-tumor tissues,respectively,to identify key molecules that might induce resistance to TKIs.We then performed in vitro and in vivo experiments to validate selected targets and related mechanisms.The associations of plasma secreted phosphoprotein 1(SPP1)expression levels before sorafenib/lenvatinib treatment with progression-free survival(PFS)and overall survival(OS)of 54 patients with advanced HCC were evaluated using Kaplan-Meier and Cox regression analysis.Results:Bioinformatic analysis identified CAF-derived SPP1 as a candidate molecule driving TKI resistance.SPP1 inhibitors reversed CAF-induced TKI resistance in vitro and in vivo.CAF-derived SPP1 activated rapidly accelerated fibrosarcoma(RAF)/mitogen-activated protein kinase(MAPK)and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)through the integrin-protein kinase C-alpha(PKCα)signaling pathway and promoted epithelial-to-mesenchymal transition(EMT).A high plasma SPP1 level before TKI treatment was identified as an independent predictor of poor PFS(P=0.026)and OS(P=0.047)in patients with advanced HCC after TKI treatment.Conclusions:CAF-derived SPP1 enhances TKI resistance in HCC via bypass activation of oncogenic signals and EMT promotion.Its inhibition represents a promising therapeutic strategy against TKI resistance inHCC.Moreover,plasma SPP1 level before TKI treatment represents a potential biomarker for treatment response prediction.JungWoo Eun Jung Hwan Yoon Hye Ri Ahn Seokhwi Kim Young Bae Kim Su Bin Lim Won Park TaeWook Kang Geum Ok Baek Moon Gyeong Yoon Ju A Son Ji HyangWeon Soon Sun Kim Hyo Jung Cho Jae Youn Cheong 2023Cancer Communications2023,43,4:2

12Recombinant human fibroblast growth factor-2 promotes nerve regeneration and functional recovery after mental nerve crush injury显示文摘Several studies have shown that fibroblast growth factor-2(FGF2) can directly affect axon regeneration after peripheral nerve damage. In this study, we performed sensory tests and histological analyses to study the effect of recombinant human FGF-2(rh FGF2) treatment on damaged mental nerves. The mental nerves of 6-week-old male Sprague-Dawley rats were crush-injured for 1 minute and then treated with 10 or 50 μg/m L rh FGF2 or PBS in crush injury area with a mini Osmotic pump. Sensory test using von Frey filaments at 1 week revealed the presence of sensory degeneration based on decreased gap score and increased difference score. However, at 2 weeks, the gap score and difference score were significantly rebounded in the mental nerve crush group treated with 10 μg/m L rh FGF2. Interestingly, treatment with 10 μg/m L rh FGF had a more obviously positive effect on the gap score than treatment with 50 μg/m L rh FGF2. In addition, retrograde neuronal tracing with Dil revealed a significant increase in nerve regeneration in the trigeminal ganglion at 2 and 4 weeks in the rh FGF2 groups(10 μg/m L and 50 μg/m L) than in the PBS group. The 10 μg/m L rh FGF2 group also showed an obviously robust regeneration in axon density in the mental nerve at 4 weeks. 14Downregulated miR-18b-5p triggers apoptosis by inhibition of calcium signaling and neuronal cell differentiation in transgenic SOD1(G93A)mice and SOD1(G17S and G86S)ALS patients显示文摘Background:MicroRNAs(miRNAs)are endogenous non-coding RNAS that regulate gene expression at the post-transcriptional level and are key modulators in neurodegenerative diseases.Overexpressed miRNAs play an important role in amyotrophic lateral sclerosis(ALS);however,the pathogenic mechanisms of deregulated miRNAS are still unclear.Methods:We aimed to assess the dysfunction of RNAS or miRNAs in fALS(SOD1 mutations).We compared the RNA-seq of subcellular fractions in NSC-34 WT(hSOD1)and MT(hSOD1(G93A))cells to find altered RNAs or miRNAs.We identified that Hif1a and Mef2c were upregulated,and Mctp1 and Rarb were downregulated in the cytoplasm of NSC-34 MT cells.Results:SOD1 mutations decreased the level of miR-18b-5p.Induced Hif1a which is the target for miR-18b increased Mef2c expression as a transcription factor.Mef2c upregulated miR-206 as a transcription factor.Inhibition of Mctp1 and Rarb,which are targets of miR-206,induced intracellular Ca^2+ levels and reduced cell differentiation,respectively.The miR-18b-5p pathway was also observed in G93A Tg mice,fALS(G86S)patient,and iPSC-derived motor neurons from fALS(G17S)patient.Conclusions:Our data indicate that SOD1 mutation decreases miR-18b-5p,which sequentially regulates Hif1a,Mef2c,miR-206,Mctp1 and Rarb in fALS-linked SOD1 mutation.These results provide new insights into the downregulation of miR-18b-5p-dependent pathogenic mechanisms of ALS.Ki Yoon Kim Yu Ri Kim Kyung Won Choi Mijung Lee Somyung Lee Wooseok Im Je-Young Shin Jin Young Kim Yoon Ho Hong Manho Kim Jong-ll Kim Jung-Joon Sung 2020Translational Neurodegeneration2020,9,3:2

20Analysis of effects of operating parameters on rate of penetration in drilling process with air down-the-hole hammer显示文摘Air down-the-hole(DTH)hammer drilling has long been recognized to have the potential of drilling faster than conventional rotary drill,especially in some hard rocks such as granite,sandstone,limestone,dolomite,etc.with the same weight on bit(WOB)and rotations per minute(RPM).So,it has been widely used in many drilling fields including mineral resource exploration drilling,oil and gas drilling and geothermal drilling.In order to reduce drilling cost by selecting optimal drilling parameters,rate of penetration(ROP)should be estimated accurately and the effects of different factors on ROP should be analyzed.In this research,ANN model with several multi-layer perception back propagation(BP)networks for predicting ROP of air DTH hammer drilling was developed using controllable parameters such as impact energy,impact frequency,WOB,RPM and bit operating time for the formations with a certain drillability index of rock.Several BP neural networks with the different neurons in hidden layers were developed and compared for selecting optimal architecture of ANN.The effects of the drilling parameters such as impact energy,impacting frequency,WOB,RPM and bit operating time on the ROP of air DTH hammer drilling were investigated by trained ANN.From the analyses,the optimum range of drilling parameters for providing high ROP were determined and analyzed for a formation with a certain drillability index of rock.The methodology proposed in this study can be used in many mathematical problems for optimization of drilling process with air DTH hammer.HO Yinchol PAK Kumdol PENG Jianming RI Jaemyong KIM Yongnam CHOE Cholho 2021Global Geology2021,24,1:1
