1MicroRNAs in the prognosis and therapy of colorectal cancer: From bench to bedside显示文摘MicroRNAs(miRNAs) are small, single-stranded, noncoding RNAs that can post-transcriptionally regulate the expression of various oncogenes and tumor suppressor genes. Dysregulated expression of many miRNAs have been shown to mediate the signaling pathways critical in the multistep carcinogenesis of colorectal cancer(CRC). MiR NAs are stable and protected from RNase-mediated degradation, thereby enabling its detection in biological fluids and archival tissues for biomarker studies. This review focuses on the role and application of miRNAs in the prognosis and therapy of CRC. While stage Ⅱ CRC is potentially curable by surgical resection, a significant percentage of stage Ⅱ CRC patients do develop recurrence. MiRNA biomarkers may be used to stratify such high-risk population for adjuvant chemotherapy to provide better prognoses. Growing evidence also suggests that miRNAs are involved in the metastatic process of CRC. Certain of these miRNAs may thus be used as prognostic biomarkers to identify patients more likely to have micro-metastasis, who could be monitored more closely after surgery and/or given more aggressive adjuvant chemotherapy. Intrinsic and acquired resistance to chemotherapy severely hinders successful chemotherapy in CRC treatment. Predictive miRNA biomarkers for response to chemotherapy may identify patients who will benefit the most from a particular regimen and also spare the patients from unnecessary side effects. Selection of patients to receive the new targeted therapy is becoming possible with the use of predictive miRNA biomarkers. 4Compressive Stress Enhances Invasive Phenotype of Cancer Cells via Piezo1 Activation显示文摘In this study,we hypothesized that Piezo 1 channels mediate the compression-enhanced invasive phenotype of cancer cells via a caveolae-dependent mechanism.To test this hypothesis,we examined in vitro cultured human breast cancer cells for their ability to invade and degrade extracellular matrix in the presence or absence of compressive stress,together with corresponding changes in Piezo1 as well as cytoskeletal remodeling and calcium signaling.Here we show that compressive stress enhanced invasion,matrix degradation,and invadopodia formation of breast cancer cells.We further identified Piezo1 as the putative mechanosensitive cellular component that transmits compression to induce calcium influx,which in turn triggers several downstream pathways.Interestingly,for the first time we observed inv-adopodia with matrix degradation ability on the apical side of the cells, similar to those commonly observed at the cell s ventral side.Furthermore,we demonstrate that Piezo1 and caveolae were both involved in mediating the compressive stress-induced cancer cell invasive phenotype as Piezo 1 and caveolae were often colocalized,and reduction of Cav-1 expression or disruption of caveolae with methyl-β-cyclodextrin led to not only reduced Piezo1 expression but also attenuation of the invasive phenotypes promoted by compressive stress.Taken together,we first observed that in breast cancer cells,simulating uncontrolled growth-induced compressive stress enhanced cancer cell invasion,matrix degradation,and invadopodia and stress fiber formation.Our study also confirmed that Piezo1 channels are highly expressed in breast cancer cells compared to normal breast cells,and is consistent with the data that compressive stress regulates cell migration of breast cancer cells but not normal breast cells.Additionally,we identified that Piezol mediated these processes and the invasive phenotypes also depended on the integrity of caveolae.These findings provide the first demonstration that compressive stress enhances matrix degradation by breast cancer cells and Piezo1 is an essential mechanosensor and transducer for such stress in breast cancer.Additionally,our data supports the model where caveolae might be the'mechanical force foci'which concentrates Piezol to facilitate force sensing and transduction in mammalian cells.Our work may have relevance to human tumors in vivo.As solid tumor experiences high compressive stress due to uncontrolled proliferation and confinement by the stiff extracellular matrix environment,this microenvironment facilitates compression-enhanced cell invasion.The identification of Piezo1’s crucial role in this process provides the first demonstration of the dependence of Piezo1 channels on the response of breast cancer cells to physiological compressive stress.The functional dependence of Piezo1 on caveolae further highlights the importance of membrane organization and composition on forcegated ion channels.Both of these findings underscore the cardinal role that Piezo1 channels play in regulating cell invasion and may inspire further development targeting Piezol as a potential cancer therapeutic target.Mingzhi Luo Kenneth KYHo Zhaowen Tong Linhong Deng Allen PLiu 2019医用生物力学2019,34,A01:1
5Uranium sequestration in sediment at an iron-rich contaminated site at Oak Ridge, Tennessee via.bioreduction followed by reoxidation显示文摘This study evaluated uranium sequestration performance in iron-rich (30 g/kg) sediment via bioreduction followed by reoxidation.Field tests (1383 days) at Oak Ridge,Tennessee demonstrated that uranium contents in sediments increased after bioreduced sediments were re-exposed to nitrate and oxygen in contaminated groundwater.Bioreduction of contaminated sediments (1200 mg/kg U) with ethanol in microcosm reduced aqueous U from 0.37 to 0.023 mg/L.Aliquots of the bioreduced sediment were reoxidized with O2,H2O2,and NaNO3,respectively,over 285 days,resulting in aqueous U of 0.024,1.58 and 14.4 mg/L at pH 6.30,6.63 and 7.62,respectively.The source-and the three reoxidized sediments showed different desorption and adsorption behaviors of U,but all fit a Freundlich model.The adsorption capacities increased sharply at pH 4.5 to 5.5,plateaued at pH 5.5 to 7.0,then decreased sharply as pH increased from 7.0 to 8.0.The O2-reoxidized sediment retained a lower desorption efficiency at pH over 6.0.The NO3--reoxidized sediment exhibited higher adsorption capacity at pH 5.5 to 6.0.The pH-dependent adsorption onto Fe(Ⅲ) oxides and formation of U coated particles and precipitates resulted in U sequestration,and bioreduction followed by reoxidation can enhance the U sequestration in sediment.Peng-Song Li Wei-Min Wu Debra H.Phillips David B.Watson Shelly Kelly Bing Li Tonia Mehlhorn Kenneth Lowe Jennifer Earles Hu-Chun Tao Tong Zhang Craig S.Criddle 2019Journal of Environmental Sciences2019,31,11:1

8Large Scale Antenna Arrays with Increasing Antennas in Limited Physical Space显示文摘Large Scale multiple input multiple output(MIMO) systems have recently emerged as a promising technology for 5G communications.While they have been shown to offer significant performance benefits in theoretical studies,the large scale MIMO transmitters will have to be deployed in the limited physical space of today's base stations(BSs).Accordingly,this paper examines effects of deploying increasing numbers of antennas in fixed physical space,by reducing the antenna spacing.We focus on the resulting performance of large-scale MIMO transmitters using low complexity closed form precoding techniques.In particular,we investigate the combined effect of reducing the distance between the antenna elements with increasing the number of elements in a fixed transmitter space.This gives rise to two contradicting phenomena:the reduction of spatial diversity due to reducing the separation between antennas and the increase in transmit diversity by increasing the number of elements.To quantify this tradeoff,we investigate densely deployed uniform antenna arrays modelled by detailed electromagnetic simulation.Our results show the somewhat surprising result that,by reducing the separations between the antennas to significantly less than the transmit wavelength to fit more antennas,the resulting system performance improves.Christos Masouros CHEN Jianling Kenneth Tong Mathini Sellathurai Tharmalingam Ratnaraiah WANG Junhong 2014China Communications2014,11,11:1
9Luminescent gold nanocluster-based sensing platform for accurate H_(2)S detection in vitro and in vivo with improved anti-interference显示文摘Gold nanoclusters(Au NCs)are promising luminescent nanomaterials due to their outstanding optical properties.However,their relatively low quantum yields and environment-dependent photoluminescence properties have limited their biological applications.To address these problems,we developed a novel strategy to prepare chitosan oligosaccharide lactate(Chi)-functionalized Au NCs(Au NCs@Chi),which exhibited emission with enhanced quantum yield and elongated emission lifetime as compared to the Au NCs,as well as exhibited environment-independent photoluminescence properties.In addition,utilizing the free amino groups of Chi onto Au NCs@Chi,we designed a FRET-based sensing platform for the detection of hydrogen sulfide(H_(2)S).The Au NCs and the specific H_(2)S-sensitive merocyanine compound were respectively employed as an energy donor and acceptor in the platform.The addition of H_(2)S induced changes in the emission profile and luminescence lifetime of the platform with high sensitivity and selectivity.Utilization of the platform was demonstrated to detect exogenous and endogenous H_(2)S in vitro and in vivo through wavelength-ratiometric and time-resolved luminescence imaging(TLI).Compared to previously reported luminescent molecules,the platform was less affected by experimental conditions and showed minimized autofluorescence interference and improved accuracy of detection.Qi Yu Pengli Gao Kenneth Yin Zhang Xiao Tong Huiran Yang Shujuan Liu Jing Du Qiang Zhao Wei Huang 2017Light(Science & Applications)2017,6,1:1

18Membrane-based treatment of shale oil and gas wastewater:The current state of knowledge显示文摘Shale oil and gas exploitation not only consumes substantial amounts of freshwater but also generates large quantities of hazardous wastewater. Tremendous research efforts have been invested in developing membrane-based technologies for the treatment of shale oil and gas wastewater. Despite their success at the laboratory scale, membrane processes have not been implemented at full scale in the oil and gas fields. In this article, we analyze the growing demands of wastewater treatment in shale oil and gas production, and then critically review the current stage of membrane technologies applied to the treatment of shale oil and gas wastewater. We focus on the unique niche of those technologies due to their advantages and limitations, and use mechanical vapor compression as the benchmark for comparison. We also highlight the importance of pretreatment as a key component of integrated treatment trains, in order to improve the performance of downstream membrane processes and water product quality. We emphasize the lack of sufficient efforts to scale up existing membrane technologies, and suggest that a stronger collaboration between academia and industry is of paramount importance to translate membrane technologies developed in the laboratory to the practical applications by the shale oil and gas industry.Tiezheng Tong Kenneth H. 20The Hong Kong consensus recommendations on the diagnosis and management of pancreatic cystic lesions显示文摘Background:The finding of pancreatic cystic lesions(PCL)on incidental imaging is becoming increasingly common.International studies report a prevalence of 2.2-44.7%depending on the population,imaging modality and indication for imaging,and the prevalence increases with age.Patients with PCL are at risk of developing pancreatic cancer,a disease with a poor prognosis.This publication summarizes recommendations for the diagnosis and management of PCL and post-operative pancreatic exocrine insufficiency(PEI)from a group of local specialists.Methods:Clinical evidence was consolidated from narrative reviews and consensus statements formulated during two online meetings in March 2022.The expert panel included gastroenterologists,hepatobiliary surgeons,oncologists,radiologists,and endocrinologists.Results:Patients with PCL require careful investigation and follow-up due to the risk of malignant transformation of these lesions.They should undergo clinical investigation and pancreas-specific imaging to classify lesions and understand the risk profile of the patient.Where indicated,patients should undergo pancreatectomy to excise PCL.Following pancreatectomy,patients are at risk of PEI,leading to gastrointestinal dysfunction and malnutrition.Therefore,such patients should be monitored for symptoms of PEI,and promptly treated with pancreatic enzyme replacement therapy(PERT).Patients with poor response to PERT may require increases in dose,addition of a proton pump inhibitor,and/or further investigation,including tests for pancreatic function.Patients are also at risk of new-onset diabetes mellitus after pancreatectomy;they should be screened and treated with insulin if indicated.Conclusions:These statements are an accurate summary of our approach to the diagnosis and management of patients with PCL and will be of assistance to clinicians treating these patients in a similar clinical landscape.Tan-To Cheung Yuk Tong Lee Raymond Shing-Yan Tang Wong Hoi She Kai Chi Cheng Chin Cheung Cheung Keith Wan Hang Chiu Kenneth Siu Ho Chok Wing Sun Chow Tak Wing Lai Wai-Kay Seto Thomas Yau 2023Hepatobiliary Surgery and Nutrition2023,12,5:0
