| 2 | Mechanism of inhibiting type I interferon induction by hepatitis B virus X protein显示文摘Hepatitis B virus(HBV)is regarded as a stealth virus,invading and replicating efficiently in human liver undetected by host innate antiviral immunity.Here,we show that type I interferon(IFN)induction but not its downstream signaling is blocked by HBV replication in HepG2.2.15 cells.This effect may be partially due to HBV X protein(HBx),which impairs IFNβpromoter activation by both Sendai virus(SeV)and components implicated in signaling by viral sensors.As a deubiquitinating enzyme(DUB),HBx cleaves Lys63-linked polyubiquitin chains from many proteins except TANK-binding kinase 1(TBK1).It binds and deconjugates retinoic acid-inducible gene I(RIG I)and TNF receptor-associated factor 3(TRAF3),causing their dissociation from the downstream adaptor CARDIF or TBK1 kinase.In addition to RIG I and TRAF3,HBx also interacts with CARDIF,TRIF,NEMO,TBK1,inhibitor of kappa light polypeptide gene enhancer in B-cells,kinase epsilon(IKKi)and interferon regulatory factor 3(IRF3).Our data indicate that multiple points of signaling pathways can be targeted by HBx to negatively regulate production of type I IFN. | Junyi Jiang Hong Tang | 2010 | Protein & Cell2010,1,12: | 13 |

| 5 | Brilliant blue G attenuates lipopolysaccharide-mediated microglial activation and inflammation显示文摘Previous studies have confirmed that oxidized adenosine triphosphate, a P2X7 receptor antagonist, attenuates lipopolysaccharide-mediated microglial activation and inflammatory expression following neuronal damage in rat brain. NaCl and temperature may affect the potency of oxidized adenosine triphosphate. Brilliant blue G is a derivative of a widely used food additive and has little toxicity. This study explored the effects of brilliant blue G, a selective P2X7 receptor antagonist, on microglial activation and inflammation. Results demonstrated that brilliant blue G inhibited the release of cyclooxygenase-2 and interleukin-6 in BV2 cells. Immunofluorescence displayed that brilliant blue G could suppress lipopolysaccharide-induced microglial activation. This study used RNA interference to block P2X7 receptor expression and found that small interfering RNA also suppressed the release of cyclooxygenase-2 and interleukin-6 in BV2 cells. | 7 | stAPAminer:Mining Spatial Patterns of Alternative Polyadenylation for Spatially Resolved Transcriptomic Studies显示文摘Alternative polyadenylation(APA)contributes to transcriptome complexity and gene expression regulation and has been implicated in various cellular processes and diseases.Singlecell RNA sequencing(scRNA-seq)has enabled the profiling of APA at the single-cell level;however,the spatial information of cells is not preserved in scRNA-seq.Alternatively,spatial transcriptomics(ST)technologies provide opportunities to decipher the spatial context of the transcriptomic landscape.Pioneering studies have revealed potential spatially variable genes and/or splice isoforms;however,the pattern of APA usage in spatial contexts remains unappreciated.In this study,we developed a toolkit called stAPAminer for mining spatial patterns of APA from spatially barcoded ST data.APA sites were identified and quantified from the ST data.In particular,an imputation model based on the k-nearest neighbors algorithm was designed to recover APA signals,and then APA genes with spatial patterns of APA usage variation were identified.By analyzing wellestablished ST data of the mouse olfactory bulb(MOB),we presented a detailed view of spatial APA usage across morphological layers of the MOB.We compiled a comprehensive list of genes with spatial APA dynamics and obtained several major spatial expression patterns that represent spatial APA dynamics in different morphological layers.By extending this analysis to two additional replicates of the MOB ST data,we observed that the spatial APA patterns of several genes were reproducible among replicates.stAPAminer employs the power of ST to explore the transcriptional atlas of spatial APA patterns with spatial resolution.This toolkit is available at http://gffzz188fe103f8f1460asnpq99wb9oko96669.ffgz.tsg.suse.edu.cn/BMILAB/stAPAminer and http://gffzz77e3413bc06540edsnpq99wb9oko96669.ffgz.tsg.suse.edu.cn/biocode/tools/BT007320. | Guoli Ji Qi Tang Sheng Zhu Junyi Zhu Pengchao Ye Shuting Xia Xiaohui Wu | 2023 | Genomics, Proteomics & Bioinformatics2023,21,3: | 1 |

| 12 | A chorismate mutase from Radopholus similis plays an essential role in pathogenicity显示文摘In the process of infecting plants, plant parasitic nematodes release a series of proteins that play an essential role in the successful infection and pathogenesis of plant cells and tissues through stylets or body walls. In this study,based on transcriptome data, a chorismate mutase gene of Radopholus similis(RsCM) was identified and cloned,which is a single copy gene specifically expressed in the oesophageal gland and highly expressed in juveniles and females. Transient expression of RsCM in tobacco leaves showed that it was localised in the cytoplasm and nucleus of tobacco leaf cells, which inhibited the pattern-triggered immunity(PTI) induced by flg22, including callose deposition and defence gene expression, and cell death induced by immune elicitors BAX, but could not inhibit cell death induced by immune elicitors Gpa2/RBP-1. The RNA interference(RNAi) transgenic tomato of RsCM obviously inhibited the infection, pathogenicity, and reproduction of R. similis. However, the resistance of the overexpression transgenic tomato of RsCM to R. similis infection was significantly reduced, and the expression levels of two salicylic acid(SA) pathway genes(PR1 and PR5) in roots infected by the nematode were significantly down-regulated,which indicated that RsCM might be involved in the inhibition of SA pathway. | 15 | Measurements of the effective mass transfer areas for the gas–liquid rotating packed bed显示文摘Rotating packed bed(RPB) is one of the most effective gas–liquid mass transfer enhancement reactors, its effective specific mass transfer area(ae) is critical to understand the mass transfer process. By using the NaOH–CO_(2) chemical absorption method, the aevalues of three RPB reactors with different rotor sizes were measured under different operation conditions. The results showed that the high gravity factor and liquid flow rate were major affecting factors, while the gas flow rate exhibited minor influence.The radius of packing is the dominant equipment factor to affect aevalue. The results indicated that the contact area depends on the dispersion of the liquid phase, thus the centrifugal force of rotating packed bed greatly influenced the aevalue. Moreover, the measured ae/ap(effective specific mass transfer area/specific surface area of packing) values were fitted with dimensionless correlation formulas. | 20 | Red-emitting salicylaldehyde Schiff base with AIE behaviour and large Stokes shift显示文摘Three salicylaldehyde Schiff base(SSB), iso-PBP, PBP and EPB, were facilely synthesized and exhibited aggregation-induced emission. The introduction of C= N-N = C moiety in these SSB dyes largely extend the conjugation system and push their emission to yellow to red spectral region. These SSB dyes were negligibly fluorescent in dilute THF solution. In THF/water mixtures with high water fractions, they displayed strong yellow to red fluorescence(up to 617nm) and large Stokes shifts(up to 152 nm). Single crystal analysis on EBP showed the longer emission of in aggregated state was attributed to the molecular packing effect as compared with that in dilute solution.
