1m^6A Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2显示文摘N6-methyladenosine (m6A),catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14,is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However,the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here,we examined the function of Mettl3,the key component of the complex,in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neu-ronal development and skewed the differentiation of aNSCs more toward glial lineage,but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecip-itation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically,m6A was present on the transcripts of histone methyltransferase Ezh2,and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. 3Large-scale blow spinning of heat-resistant nanofibrous air filters显示文摘Particulate matter(PM)pollution has become a serious problem worldwide and various kinds of nanofibrous filters aiming to solve the problem have been developed.It is urgent to remove PM from high-temperature pollution sources,such as industrial emissions,coal furnaces,and automobile exhaust gases.However,filtration at pollution sources remains challenging because most existing air filters are not resistant to high temperature.Herein,heat-resistant polyimide(PI)nanofibrous air filters are fabricated via a simple and scalable solution blow-spinning method.These air filters show excellent thermal stability at high temperature up to 420℃.They exhibit a filtration efficiency as high as 99.73%at ambient temperature and over 97%at 300℃.In addition,a field test shows that the filters remove>97%of PM from the car exhaust fumes.Hence,the blow-spun PI nanofibrous membranes combined with the facile preparation strategy have great potential in high temperature air filtration fields and other similar applications such as water purification and protein separation.Ziwei Li Jianan Song Yuanzheng Long Chao Jia Zhenglian Liu Lei Li Cheng Yang Junchen Liu Sen Lin Haiyang Wang Yibo Liu Minghao Fang Hui Wu 2020Nano Research2020,13,3:6

6Joint formation mechanism of high depth-to-width ratio friction stir welding显示文摘High depth-to-width ratio friction stir welding is an attractive method for the joining demands of alum inum profiles, which is sparked with its extrem ely low heat input and high mechanical performance. In this study, the joint form ation mechanism was studied by a num erical model of plastic flow combined with experim ental approaches. A fluid-solid-interaction algorithm was proposed to establish the coupling model, and the material to be welded was treated as non-New tonian fluid. The thread structure and the milling facets on tool pin prom oted drastic turbulence of material. The thread structure converged the plasticized material by its inclined plane, and then drove the attached material to refill the welds. The milling facets brought about the periodic dynamic material flow. The thread structure and the milling facets increased the strain rate greatly under the extremely low heat input, which avoided the welding defects. The condition of the peak tem perature of 648 K and the strain rate of 151 s^-1 attributed to the lowest coarsening degree of precipitate. The tensile strength of the joint reached 265 MPa, equivalent to 86% of base material. 13Ten-Hour Stable Noninvasive Brain-Computer Interface Realized by Semidry Hydrogel-Based Electrodes显示文摘Noninvasive brain computer interface(BCI)has been extensively studied from many aspects in the past decade.In order to broaden the practical applications of BCI technique,it is essential to develop electrodes for electroencephalogram(EEG)collection with advanced characteristics such as high conductivity,long term efectiveness,and biocompatibility.In this study,we devcloped a silver-nanowire/PVA hydrogc/mclamine sponge(AgPHMS)semidry EEG electrode for long lasting monitoring of EEG signal.Benefiting from the water storage capacity of PVA hydrogel,the clectrolyte solution can be continuously released to the scalpelctrode interface during used.The electrolyte solution can infiltrate the stratum corneum and reduce the scalp-electrode impedance to 10k0-15k0.The flexible structure enabls the electrode with mechanical stability,increases the wearing comfort,and reduces the scalp-electrode gap to reduce contact impedance.As a result,a long-term BCI application based on measurements of motion-onset visual evoked potentials(mVEPs)shows that the 3-hour BCI accuracy of the new electrode(77%to 100%)is approximately the same as that of conventional eletrodes supported by a conductive gel during the first hour.Furthermore,the BCI system based on the new electrode can retain low contact impedance for 10 hours on scalp,which greatly improved the ability of BCI technique.Junchen Liu Sen Lin Wenzheng Li Yanzhen Zhao Dingkun Liu Zhaofeng He Dong Wang Ming Lei Bo Hong Hui Wu 2022Research2022,,2:1

15A large-area AgNW-modified textile with high-performance electromagnetic interference shielding显示文摘Manufacturing a flexible,light,large-area,and high-efficiency electromagnetic shielding materials in a straightforward and cost-effective manner presently remains a significant challenge.In this work,we propose a conductive network design and verify its electromagnetic interference(EMI)shielding effectiveness(SE)by simulation.Using the structure and parameters obtained by simulation,we prepare a flexible EMI shielding material using silver nanowires(AgNWs)/polyvinyl butyral(PVB)ethanol solution and textile substructure via a facile immersing method.In the frequency range of 5-18 GHz,the AgNWs/PVB textile with 1.4 mm thickness achieves an EMI SE of 59 dB,which exceeds the requirements for commercial applications.Due to the low density of 56 mg/cm^(3),specific shielding effectiveness(SSE)of this material reaches 1053 dB m^(3)/g.It is found that the AgNWs/PVB textile is more resistant to washing with water and oxidation than AgNWs textile without a PVB protector.As a result,the conductivity of AgNWs/PVB textile exhibits no change after washing with water and varies slightly after being kept in hot air.We find that a signal monitor is unable to detect a signal emitted by a mobile phone from a jacket lined with AgNWs/PVB textile.AgNWs/PVB textile with these properties can be mass-produced as high-efficiency EMI shielding material for commercial applications.Junchen Liu Sen Lin Kai Huang Chao Jia Qingmin Wang Ziwei Li Jianan Song Zhenglian Liu Haiyang Wang Ming Lei Hui Wu 2020npj Flexible Electronics2020,4,1:0
16Relationship of Microchannels and Plaque Erosion in Patients with ST-Segment Elevation Myocardial Infarction: An Optical Coherence Tomography Study显示文摘Objective:Microchannels are associated with the progression of atherosclerotic vulnerable plaques.However,in patients with culprit optical coherence tomography(OCT)-defined plaque erosion,the knowledge of microchannels and culprit lesion vulnerability is limited.The aim of this study was to investigate culprit lesion characteristics in patients with ST-segment elevated myocardial infarction(STEMI)caused by plaque erosion with and without microchannels using OCT.Methods:In all,348 STEMI patients with plaque erosion who underwent OCT of the culprit lesion at the 2nd Affiliated Hospital of Harbin Medical University(Harbin,China)from August 2014 to December 2017 were included and divided into the microchannel group(n=116,33.3%)and no-microchannel group(n=232,66.7%).The clinical characteristics and OCT-derived plaque features were compared between both groups.Results:Among the 348 STEMI patients with plaque erosion,culprit lesions with microchannels had higher incidence of lipid plaque(59.5%vs.45.3%,P=0.012);calcification(41.4%vs.24.6%,P=0.002);spotty calcification(30.2%vs.18.1%,P=0.014);macrophages accumulation(72.4%vs.45.7%,P<0.001);and cholesterol crystals(32.8%vs.14.2%,P<0.001)than those without microchannels.In addition,minimal lumen area was smaller((1.9±0.9)mm2 vs.(2.8±2.3)mm2,P<0.001)and lumen area stenosis was greater((71.3%±13.4%)vs.(65.3%±19.3%),P=0.001)in the microchannel group than in the no-microchannel group.Conclusion:In patients with STEMI caused by plaque erosion,one-third manifested typical microchannel characteristics,and those with microchannels were associated with more severe luminal stenosis and more vulnerable plaque features than those without microchannels.Senqing Jiang Junchen Guo Yanwei Yin Chao Fang Jifei Wang Yidan Wang Fangmeng Lei Sibo Sun Xueying Pei Ruyi Jia Shaotao Zhang Lulu Li Yini Wang Lei Xing Huai Yu Huimin Liu Maoen Xu Xuefeng Ren Lijia Ma Guo Wei Jingbo Hou Jiannan Dai Bo Yu 2022Cardiology Discovery2022,2,2:0
17Investigating the ENSO prediction skills of the Beijing Climate Center climate prediction system version 2显示文摘The El Niño-Southern Oscillation(ENSO)ensemble prediction skills of the Beijing Climate Center(BCC)climate prediction system version 2(BCC-CPS2)are examined for the period from 1991 to 2018.The upper-limit ENSO predictability of this system is quantified by measuring its“potential”predictability using information-based metrics,whereas the actual prediction skill is evaluated using deterministic and probabilistic skill measures.Results show that:(1)In general,the current operational BCC model achieves an effective 10-month lead predictability for ENSO.Moreover,prediction skills are up to 10–11 months for the warm and cold ENSO phases,while the normal phase has a prediction skill of just 6 months.(2)Similar to previous results of the intermediate coupled models,the relative entropy(RE)with a dominating ENSO signal component can more effectively quantify correlation-based prediction skills compared to the predictive information(PI)and the predictive power(PP).(3)An evaluation of the signal-dependent feature of the prediction skill scores suggests the relationship between the“Spring predictability barrier(SPB)”of ENSO prediction and the weak ENSO signal phase during boreal spring and early summer.Yanjie Cheng Youmin Tang Tongwen Wu Xiaoge Xin Xiangwen Liu Jianglong Li Xiaoyun Liang Qiaoping Li Junchen Yao Jinghui Yan 2022Acta Oceanologica Sinica2022,41,5:0
18Computational prediction and functional analysis of arsenic-binding proteins in human cells显示文摘Background:Arsenic has a broad anti-cancer ability against hematologic malignancies and solid tumors.To systematically understand the biological functions of arsenic,we need to identify arsenic-binding proteins in human cells.However,due to lack of effective theoretical tools and experimental methods,only a few arsenic-binding proteins have been identified.Methods:Based on the crystal structure of ArsM,we generated a single mutation free energy profile for arsenic binding using free energy perturbation methods.Multiple validations provide an indication that our computational model has the ability to predict arsenic-binding proteins with desirable accuracy.We subsequently apply this computational model to scan the entire human genome to identify all the potential arsenic-binding proteins.Results:The computationally predicted arsenic-binding proteins show a wide range of biological functions,especially in the signaling transduction pathways.In the signaling transduction pathways,arsenic directly binds to the key factors(e.g.,Notch receptors,Notch ligands,Wnt family proteins,TGF-beta,and their interacting proteins)and results in significant inhibitions on their enzymatic activities,further having a crucial impact on the related signaling pathways.Conclusions:Arsenic has a significant impact on signaling transduction in cells.Arsenic binding to proteins can lead to dysfunctions of the target proteins,having crucial impacts on both signaling pathway and gene transcription.We hope that the computationally predicted arsenic-binding proteins and the functional analysis can provide a novel insight into the biological functions of arsenic,revealing a mechanism for the broad anti-cancer of arsenic.Shichao Pang Junchen Yang Yilei Zhao Yixue Li Jingfang Wang 2019Quantitative Biology2019,7,3:0
19Themis suppresses the effector function of CD8^(+)T cells in acute viral infection显示文摘CD8^(+)T cells play a central role in antiviral immune responses.Upon infection,naive CD8^(+)T cells differentiate into effector cells to eliminate virus-infected cells,and some of these effector cells further differentiate into memory cells to provide long-term protection after infection is resolved.Although extensively investigated,the underlying mechanisms of CD+T-cell differentiation remain incompletely understood.Themis is a T-cell-specific protein that plays critical roles in T-cell development.Recent studies using Themis T-cell conditional knockout mice also demonstrated that Themis is required to promote mature CD8^(+)T-cell homeostasis,cytokine responsiveness,and antibacterial responses.In this study,we used LCMV Armstrong infection as a probe to explore the role of Themis in viral infection.We found that preexisting CD8^(+)T-cell homeostasis defects and cytokine hyporesponsiveness do not impair viral clearance in Themis T-cell conditional knockout mice.Further analyses showed that in the primary immune response,Themis deficiency promoted the differentiation of CD8^(+)effector cells and increased their TNF and IFNy production.Moreover,Themis deficiency impaired memory precursor cell(MPEC)differentiation but promoted short-lived effector cell(SLEC)differentiation.Themis deficiency also enhanced effector cytokine production in memory CD8^(+)T cells while impairing central memory CD8^(+)T-cell formation.Mechanistically,we found that Themis mediates PD-1 expression and its signaling in effector CD8^(+)T cells,which explains the elevated cytokine production in these cells when Themis is disrupted.Jian Tang Xian Jia Jian Li Junchen Dong Jiayu Wang Wanyun Li Yuzhen Zhu Yanyan Hu Bowen Hou Chunjie Lin Yu Cong Tong Ren Changsheng Yan Hongying Yang Qian Lai Haiping Zheng Yuzhou Bao Namrata Gautam Hong-Rui Wang Bing Xu Xiao Lei Chen Qing Li Nicholas R.J.Gascoigne Guo Fu 2023Cellular & Molecular Immunology2023,20,5:0
20A new member of Sphenopsida,Neolobatannularia gen. nov. from Late Triassic of western Liaoning,China显示文摘A new genus Neolobatannularia Sun et Li gen. nov. with the type species Neolobatannularia liaoningensis Sun et Li sp. nov. from the Upper Triassic Yangcaogou Formation of Beipiao, Liaoning, China, is described in this paper. The new taxon is attributed to Equisetales of Sphenopsida based on its branch with internodes, node and leaves borne in whorls on nodes. It is a new member of the Mesozoic Sphenopsida. Although the new taxon shares some morphological characters with the genus Lobatannularia Kawasaki, a genus widely distributed in the Permian Cathaysia flora in East Asia, and the genus Lobatannulariopsis Yang from the Late Triassic of southwestern China, its unique branching pattern is markedly distinguished from the two known genera.
