| 1 | m^6A Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2显示文摘N6-methyladenosine (m6A),catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14,is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However,the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here,we examined the function of Mettl3,the key component of the complex,in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neu-ronal development and skewed the differentiation of aNSCs more toward glial lineage,but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecip-itation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically,m6A was present on the transcripts of histone methyltransferase Ezh2,and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. | 2 | Linkage between permafrost distribution and river runoff changes across the Arctic and the Tibetan Plateau显示文摘River runoff in the Arctic and the Tibetan Plateau(TP) change significantly in recent decades. However, the mechanisms of the physical processes of permafrost river runoff change remain uncertain across large scale. This study investigated the mainstreams and tributaries of main Arctic and TP rivers dominated by permafrost and assessed the linkage between hydrological regime change and permafrost. The results show that the effects of permafrost on river runoff are highly dependent on the permafrost coverage of a watershed. For the past decades, the majority of the Arctic and TP basins showed increased discharge, while all of the studied basins showed increased baseflow, with faster increasing speed than total discharge.Both total discharge and baseflow annual change rate(ΔQ and ΔBF) increased with permafrost coverage, indicating the increments of streamflow are enhanced with high permafrost coverage. Meanwhile, the annual change of precipitation showed weak connection with total discharge and baseflow change. The high permafrost coverage basins showed high annual maximum/minimum discharge ratio(Qmax/Qmin), while the Qmax/Qminchanged slightly in low permafrost cover basins. Our results highlight the importance of permafrost coverage on streamflow regime change for permafrost basins across the northern hemisphere. | 3 | Transparently curved metamaterial with broadband millimeter wave absorption显示文摘We present a conformal metamaterial with simultaneous optical transparency and broadband millimeter-wave absorption for a curved surface. By tailoring the reflection response of meta-atoms at oblique angles, it is possible to achieve excellent absorption performance from 26.5 to 40.0 GHz within a wide angular range from 0° to 60°for transverse-electric and transverse-magnetic waves. In the meantime, by employing transparent substrates,including polyvinyl chloride and polyethylene terephthalate, good optical transmittance(80.1%) and flexibility are obtained simultaneously. The reflectivity of a curved metallic surface coated with the proposed curved metamaterial is simulated and measured experimentally. | 5 | Comparison of dimension reduction-based logistic regression models for case-control genome-wide association study:principal components analysis vs.partial least squares显示文摘With recent advances in biotechnology, genome-wide association study(GWAS) has been widely used to identify genetic variants that underlie human complex diseases and traits. In case-control GWAS, typical statistical strategy is traditional logistical regression(LR) based on single-locus analysis. However, such a single-locus analysis leads to the well-known multiplicity problem, with a risk of inflating type I error and reducing power. Dimension reduction-based techniques, such as principal component-based logistic regression(PC-LR), partial least squares-based logistic regression(PLS-LR), have recently gained much attention in the analysis of high dimensional genomic data. However, the perfor?mance of these methods is still not clear, especially in GWAS. We conducted simulations and real data application to compare the type I error and power of PC-LR, PLS-LR and LR applicable to GWAS within a defined single nucleotide polymorphism(SNP) set region. We found that PC-LR and PLS can reasonably control type I error under null hypothesis.On contrast, LR, which is corrected by Bonferroni method, was more conserved in all simulation settings. In particular, we found that PC-LR and PLS-LR had comparable power and they both outperformed LR, especially when the causal SNP was in high linkage disequilibrium with genotyped ones and with a small effective size in simulation. | 9 | Computational prediction and functional analysis of arsenic-binding proteins in human cells显示文摘Background:Arsenic has a broad anti-cancer ability against hematologic malignancies and solid tumors.To systematically understand the biological functions of arsenic,we need to identify arsenic-binding proteins in human cells.However,due to lack of effective theoretical tools and experimental methods,only a few arsenic-binding proteins have been identified.Methods:Based on the crystal structure of ArsM,we generated a single mutation free energy profile for arsenic binding using free energy perturbation methods.Multiple validations provide an indication that our computational model has the ability to predict arsenic-binding proteins with desirable accuracy.We subsequently apply this computational model to scan the entire human genome to identify all the potential arsenic-binding proteins.Results:The computationally predicted arsenic-binding proteins show a wide range of biological functions,especially in the signaling transduction pathways.In the signaling transduction pathways,arsenic directly binds to the key factors(e.g.,Notch receptors,Notch ligands,Wnt family proteins,TGF-beta,and their interacting proteins)and results in significant inhibitions on their enzymatic activities,further having a crucial impact on the related signaling pathways.Conclusions:Arsenic has a significant impact on signaling transduction in cells.Arsenic binding to proteins can lead to dysfunctions of the target proteins,having crucial impacts on both signaling pathway and gene transcription.We hope that the computationally predicted arsenic-binding proteins and the functional analysis can provide a novel insight into the biological functions of arsenic,revealing a mechanism for the broad anti-cancer of arsenic. | Shichao Pang Junchen Yang Yilei Zhao Yixue Li Jingfang Wang | 2019 | Quantitative Biology2019,7,3: | 0 |

| 13 | Manipulating micro-electric field and coordination-saturated site configuration boosted activity and safety of frustrated single-atom Cu/O Lewis pair for acetylene hydrochlorination显示文摘Simultaneously boosting acetylene hydrochlorination activity and avoiding formation of explosive copper acetylide over Cu-based catalyst,which represented a promising alternative to Hg-based and noble metal catalysts,remained challenging.Herein,we fabricated a frustrated single-atom Cu/O Lewis pair catalyst(Cu/O-FLP)by coupling epoxide group(C-O-C)with atomdispersed Cu-cis-N_(2)C_(2)Cl center to address this challenge.The basic epoxy site modulated the electron-deficient state of Lewisacidic Cu center and paired with the Cu-cis-N_(2)C_(2)Cl moiety to preferentially break HCl into different electronegative Cu-Clδ-and C-O-H^(δ+)intermediates,which further induced both an extra localized electric field to polarize acetylene and a upshift of the dband center of catalyst,thereby promoting adsorption and enrichment of acetylene by enhancing the dipolar interaction between acetylene and active intermediates.Moreover,the generated Cu-Clδ-and C-O-H^(δ+)drastically reduced the energy barrier of ratelimiting step and made vinyl chloride easier to desorb from the Lewis-basic oxygen-atom site rather than traditional Lewis-acidic Cu center.These superiorities ensured a higher activity of Cu/O-FLP compared with its counterparts.Meanwhile,preferential dissociation of HCl endowed single-atom Cu with the coordination-saturated configuration,which impeded formation of explosive copper acetylide by avoiding the direct interaction between Cu and acetylene,ensuring the intrinsic safety during catalysis. | Junchen Peng Dandan Dong Zongyuan Wang Hong Yang Dongyang Qiao Qinqin Wang Wei Sun Minmin Liu Jiajun Wang Mingyuan Zhu Bin Dai Fei He Chaofeng Huang | 2023 | Nano Research2023,16,7: | 0 |
