2Detection and identification of Vibrio parahaemolyticus by multiplex PCR and DNA-DNA hybridization on a microarray显示文摘In this paper,we developed a rapid and accurate method for the detection of Vibrio parahaemalyticus strains,using multiplex PCR and DNA—DNA hybridization.Multiplex PCR was used to simultaneously amplify three diagnostic genes(tlh,tdh and fla) that serve as molecular markers of V.parahaemolyticus.Biotinylated PCR products were hybridized to primers immobilized on a microarray,and detected by chemiluminesce with avidin—conjugated alkaline phosphatase.With this method,forty-five samples were tested.Eight known virulent strains (tlh^+/tdh^+/fla^+) and four known avirulent strains(tlh^+/tdh^-/fla^+) of the V.parahaemolyticus were successfully detected,and no non-specific hybridization and cross-hybridization reaction were found from fifteen closely-related strains(tlh^-/tdh^-/fla^+) of the Vibrio spp.In addition,all the other eighteen strains of non-Vibrio bacteria(tlh^-/tdh^-/fla^-) gave negative results.The DNA microarray successfully distinguished V. parahaemolyticus from other Vibrio spp.The results demonstrated that this was an efficient and robust method for identifying virulent strains of V.parahaemolyticus.Rongzhi Wang Jiadong Huang Wei Zhang Guangmei Lin Junwei Lian Libin Jiang Hongcong Lin Songfa Wang Shihua Wang 2011Journal of Genetics and Genomics2011,38,3:11
3PCN-Fe(Ⅲ)-PTX nanoparticles for MRI guided high efficiency chemo-photodynamic therapy in pancreatic cancer through alleviating tumor hypoxia显示文摘As nanomedicine-based clinical strategies have continued to develop,the possibility of combining chemotherapy and singlet oxygen-dependent photodynamic therapy(PDT)to treat pancreatic cancer(PaC)has emerged as a viable therapeutic modality.The efficacy of such an approach,however,is likely to be constrained by the mechanisms of drug release and tumor oxygen levels.In the present study,we developed an Fe(Ⅲ)-complexed porous coordination network(PCN)which we then used to encapsulate PTX(PCN-Fe(Ⅲ)-PTX)nanoparticles(NPs)in order to treat PaC via a combination of chemotherapy and PDT.The resultant NPs were able to release drug in response to both laser irradiation and pH changes to promote drug accumulation within tumors.Furthermore,through a Fe(Ⅲ)-based Fenton-like reaction these NPs were able to convert H2O2 in the tumor site to O2,thereby regulating local hypoxic conditions and enhancing the efficacy of PDT approaches.Also these NPs were suitable for use as a T1-MRI weighted contrast agent,making them viable for monitoring therapeutic efficacy upon treatment.Our results in both cell line and animal models of PaC suggest that these NPs represent an ideal agent for mediating effective MRI-guided chemotherapy-PDT,giving them great promise for the clinical treatment of PaC.Tao Zhang Zhenqi Jiang Libin Chen Chunshu Pan Shan Sun Chuang Liu Zihou Li Wenzhi Ren Aiguo Wu Pintong Huang 2020Nano Research2020,13,1:4
4Proteasomal deubiquitinase UCH37 inhibits degradation of β-catenin and promotes cell proliferation and motility显示文摘The ubiquitin–proteasome system degrades most cellular proteins in eukaryotes.UCH37,also known as UCH-L5,is a deubiquitinase binding to Rpn13,a receptor for ubiquitinated substrates in the 26 S proteasome.But,it remains unclear how UCH37 influences the proteasomal degradation of the ubiquitinated substrates.Because deletion of UCH37 is embryonically lethal in mice,this study aims to investigate the role of UCH37 in proteasomal degradation by constructing the UCH37-deficient cell lines using CRISPR/Cas9 technology.Our results demonstrated that deletion of UCH37 decreased the levels of proteasomal Rpn13,implying that UCH37 might facilitate incorporation of Rpn13 into the proteasome.Meanwhile,deletion of UCH37 decreased the levels of β-catenin and the early endosomal protein Rab8.β-Catenin interacts with TCF/LEF to control transcription,and is involved in development,tissue homeostasis and tumorigenesis.We further found that deletion of UCH37 increased the levels of the ubiquitinated β-catenin and accelerated the hydrogen peroxide-stimulated degradation of β-catenin.Deletion of UCH37 also down-regulated the transcription of c-Myc,a downstream effector of β-catenin,and inhibited cell proliferation and motility.These results raise the possibility that UCH37 maintains the homeostasis of proteasomal degradation reciprocally by assisting the recruitment of the ubiquitin receptor Rpn13 into the proteasome and by reversing ubiquitination of certain critical substrates of the 26 S proteasome.Zijian Li Luming Zhou Tianxia Jiang Libin Fan Xiaoying Liu Xiaobo Qiu 2019Acta Biochimica et Biophysica Sinica2019,51,3:4

6Targeting histones for degradation in cancer cells as a novel strategy in cancer treatment显示文摘The anticancer therapies with the joint treatment of a histone deacetylase(HDAC) inhibitor and a DNA-damaging approach are actively under clinical investigations, but the underlying mechanism is unclear. Histone homeostasis is critical to genome stability, transcriptional accuracy, DNA repair process, senescence, and survival. We have previously demonstrated that the HDAC inhibitor, trichostatin A(TSA), could promote the degradation of the core histones induced by γ-radiation or the DNAalkylating agent methyl methanesulfonate(MMS) in non-cancer cells, including mouse spermatocyte and embryonic fibroblast cell lines. In this study, we found that the joint treatment by TSA and MMS induced the death of the cultured cancer cells with an additive effect, but induced degradation of the core histones synergistically in these cells. We then analyzed various combinations of other HDAC inhibitors, including suberoylanilide hydroxamic acid and valproate sodium, with MMS or other DNAdamaging agents, including etoposide and camptothecin. Most of these combined treatments induced cell death additively, but all the tested combinations induced degradation of the core histones synergistically. Meanwhile, we showed that cell cycle arrest might not be a primary consequence for the joint treatment of TSA and MMS. Given that clinic treatments of cancers jointly with an HDAC inhibitor and a DNA-damaging approach often show synergistic effects, histone degradation might more accurately underlie the synergistic effects of these joint treatments in clinic applications than other parameters, such as cell death and cell cycle arrest. 7Coupled biodegradation of p-nitrophenol and p-aminophenol in bioelectrochemical system:Mechanism and microbial functional diversity显示文摘Biodegradation mechanisms and microbial functional diversity during coupled pnitrophenol(PNP)and p-aminophenol(PAP)degradation were studied in a bioelectrochemical system.PNP in the biocathode and PAP in the bioanode were almost completely removed within 28 hr and 68 hr respectively.The degradation followed the steps including hydrating hydroxyalkylation,dehydrogenating carbonylation,and hydrolating ring cleavage,etc.Metagemomic analysis based on the KEGG and egg NOG database annotations revealed the microbial composition and functional genes/enzymes related to phenol degradation in the system.The predominant bacteria genera were Lautropia,Pandoraea,Thiobacillus,Ignavibacterium,Truepera and Hyphomicrobium.The recognized biodegradation genes/enzymes related to pollutant degradation were as follows:pmo,hbd,&ppo for phenol degradation,nzba,amie,&badh for aromatic degradation,and CYP&p450 for xenobiotics degradation,etc.The co-occurrence of ARGs(antibiotic resistant genes),such as ade F,Mex J,Erm F,PDC-93 and Escherichiacolimdf A,etc.,were annotated in CARD database during the biodegradation process.The Proteobacteria&Actinobacteria phylum was the primary host of both the biodegradation genes&ARGs in this system.The microbial functional diversity ensured the effective biodegradation of the phenol pollutants in the bioelectrochemical system.Jianjun Luo Yuxi Xu Jing Wang Libin Zhang Xinbai Jiang Jinyou Shen 2021Journal of Environmental Sciences2021,33,10:2

10MXene-GaN van der Waals metal-semiconductor junctions for high performance multiple quantum well photodetectors显示文摘A MXene-GaN-MXene based multiple quantum well photodetector was prepared on patterned sapphire substrate by facile drop casting.The use of MXene electrodes improves the responsivity and reduces dark current,compared with traditional Metal-Semiconductor-Metal(MSM)photodetectors using Cr/Au electrodes.Dark current of the device using MXene-GaN van der Waals junctions is reduced by three orders of magnitude and its noise spectral intensity shows distinct improvement compared with the traditional Cr/Au–GaN–Cr/Au MSM photodetector.The improved device performance is attributed to low-defect MXene-GaN van der Waals interfaces.Thanks to the high quality MXene-GaN interfaces,it is possible to verify that the patterned substrate can locally improve both light extraction and photocurrent collection.The measured responsivity and specific detectivity reach as high as 64.6 A/W and 1.93×1012 Jones,respectively,making it a potential candidate for underwater optical detection and communication.The simple fabrication of MXene-GaN-MXene photodetectors spearheaded the way to high performance photodetection by combining the advantages of emerging 2D MXene materials with the conventional III-V materials.Lingzhi Luo Yixuan Huang Keming Cheng Abdullah Alhassan Mahdi Alqahtani Libin Tang Zhiming Wang Jiang Wu 2021Light(Science & Applications)2021,10,10:1
11A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 14Solution-processed,flexible and broadband photodetector based on CsPbBr_(3)/PbSe quantum dot heterostructures显示文摘Due to their promising applications in foldable displays,optical communication equipment and environmental monitoring systems,flexible and broadband optoelectronic devices have gained extensive attention in recent years.Here,a flexible and broadband photodetector based on CsPbBr_(3)/PbSe quantum dot(QD) heterostructures is firstly presented.The integrated QD heterostructures possess consecutive detection range from ultraviolet(UV) to long-wave length infrared(LW-IR) regions with efficient light absorption and chemical stability,in comparison with the pristine PbSe QDs.Systematic material characterizations reveal the improved exciton dissociation,carrier transport and carrier lifetime of the QD heterostructures.Flexible photodetector Ag/CsPbBr_(3)/PbSe/Ag demonstrate a high responsivity of 7.17 A/W with a specific detectivity of 8.97 × 10^(12) Jones under 25 μW/cm^(2) 365 nm illumination at 5 V.Furthermore,it could maintain 91.2 %(or 94.9 %) of its initial performance even after bending for thousands of times(or exposing in ambient air for 4 weeks).More importantly,its re s ponse time is shortened more than three orders of magnitude as that of pristine PbSe QDs-based photodetectors.Therefore,it provides a feasible and promising method for the next-generation high-performance broadband photodetectors via constructing heterostructures of various QDs.Jinming Hu Shengyi Yang Zhenheng Zhang Hailong Li Chandrasekar Perumal Veeramalai Muhammad Sulaman Muhammad Imran Saleem Yi Tang Yurong Jiang Libin Tang Bingsuo Zou 2021Journal of Materials Science & Technology2021,,9:1
15lncRNA LINC00960 promotes apoptosis by sponging ubiquitin ligase Nrdp1-targeting miR-183-5p显示文摘The ubiquitin ligase Nrdp1/RNF41 promotes the ubiquitin-dependent degradation of multiple important substrates,including BRUCE/BIRC6,a giant ubiquitin-conjugating enzyme inhibiting both apoptosis and autophagy.miR-183-5p is associated with various malignancies potentially by targeting dozens of genes.Here,we show that the lncRNA LINC00960 binds to the Nrdp1-targeting miR-183-5p and promotes apoptosis.Compared to other known miR-183-5p targets,Nrdp1 mRNA is among the few with top scores to complement miR-183-5p.miR-183-5p binds to the 3′UTR of Nrdp1 mRNA and downregulates Nrdp1 at both the mRNA and protein levels.The miR-183-5p mimics inhibit DNA damage-induced apoptosis probably by upregulating BRUCE level,whereas the miR-183-5p inhibitor suppresses the effects of miR-183-5p.LINC00960 is the noncoding RNA with the highest score to complement miR-183-5p.LINC00960 overexpression reduces,but its knockdown increases,the level of miR-183-5p,whereas LINC00960 overexpression increases,but its knockdown decreases,the level of Nrdp1 and apoptosis.Importantly,the expression of LINC00960,which is associated with multiple types of tumors,positively correlates with that of Nrdp1 in several tumors but inversely correlates with that of miR-183-5p in multiple human tumor cell lines,as analysed by quantitative PCR.Thus,miR-183-5p downregulates Nrdp1 expression and inhibits apoptosis,whereas LINC00960 upregulates Nrdp1 and promotes apoptosis by inhibiting miR-183-5p.These results may provide new ideas for the prevention,diagnosis and treatment of apoptosis-related diseases,such as tumors and neurodegenerative diseases.Hao Yang Tianxia Jiang Libin Fan Xiaobo Qiu 2023Acta Biochimica et Biophysica Sinica2023,55,1:1

18CsPbI nanorods as the interfacial layer for high-performance,all-solution-processed self-powered photodetectors显示文摘Heterojunction is regarded as a crucial step toward realizing high-performance devices,particularly,forming gradient energy band between heterojunctions benefits self-powered photodetectors.Therefore,in this paper,the synthesis of CsPbI3 nanorods(NRs)and its application as the interfacial layer in high-performance,all-solution-processed self-powered photodetectors are presented.For the bilayer photodetector ITO/ZnO(100 nm)/PbS-TBAI(150 nm)/Au,a responsivity of 3.6 A/W with a specific detectivity of 9.8×10^(12)Jones was obtained under 0.1 mW/cm^(2)white light illumination at zero bias(i.e.in self-powered mode).Meanwhile,the photocurrent was enhanced to an On/Off current ratio of 105 at zero bias with an open circuit voltage of 0.53 V for trilayer photodetector ITO/ZnO(100 nm)/PbSTBAI(150 nm)/CsPbI3(250 nm)/Au,in which the CsPbI3 NRs layer works as the interfacial layer.As a result,a specific detectivity of 4.5×10^(13)Jones with a responsivity of 11.12 A/W was obtained under0.1 mW/cm^(2) white light illumination,as well as the rising/decaying time of 0.57 s/0.41 s with excellent stability and reproducibility upto four weeks in air.The enhanced-performance is ascribed to the mismatch bandgap between PbS-TBAI/CsPbI_(3)interface,which can suppress the carrier recombination and provide efficient transport passages for charge carriers.Thus,it provides a feasible and efficient method for high-performance photodetectors.Muhammad Imran Saleem Shangyi Yang Attia Batool Muhammad Sulaman Chandrasekar Perumal Veeramalai Yurong Jiang Yi Tang Yanyan Cui Libin Tang Bingsuo Zou 2021Journal of Materials Science & Technology2021,,16:1

20Generation and application of replication-competent Venus-expressing H5N1,H7N9,and H9N2 influenza A viruses显示文摘The generation and application of replication-competent influenza A virus(IAV) expressing a reporter gene represent a valuable tool to elucidate the mechanism of viral pathogenesis and establish new countermeasures to combat the threat of influenza. Here, replication-competent IAVs with a neuraminidase(NA) segment harboring a fluorescent reporter protein, Venus, were generated in the background of H5N1, H7N9, and H9N2 influenza viruses, the three subtypes of viruses with imminent pandemic potential. All three reporter viruses maintained virion morphology, replicated with similar or slightly reduced titers relative to their parental viruses, and stably expressed the fluorescent signal for at least two passages in embryonated chicken eggs. As a proof of concept, we demonstrated that these reporter viruses,used in combination with a high-content imaging system, can serve as a convenient and rapid tool for the screening of antivirals and host factors involved in the virus life cycle.
