| 1 | Successful embryo transfer in Tianzhu white yak using standard protocol显示文摘The present study was carried out to investigate the efficiency of superovulation, oestrus synchroni-zation, and embryo recovery in Tianzhu white yaks and also to confirm the pregnancy rate of black yaks, to which embryos collected from white yaks were transplanted. Forty-seven yaks were selected from different experiment groups, including 10 Tianzhu white female yaks (donor, group A) and 37 black female yaks (recipient, groups B and C). Superovulation of the donor was induced by the application procedure of CIDR-B + FSH + PG. Oestrus synchronization of recipients was induced using two meth-ods: group B was given the same treatment as group A, except that the follicle-stimulating hormone (FSH) injection was not administered, whereas group C was injected with cloprostenol only once when corpus luteum (corpora lutea) was (were) palpated. The results showed that the oestrous rates in group A were higher (80%) than those in group B (60%) and group C (44.5%). As for the efficiency of su-perovulation, it was indicated that the mean numbers (±SD) of total corpora lutea, follicles, viable (transferable), and degenerated embryos were 4.75 ± 2.19, 1.13 ± 0.83, 2.50 ± 1.31, and 1.38 ± 0.92, re-spectively. The mean embryo recovery rates were 55.6%. All together, 18 viable embryos of Tianzhu white yak were obtained and 12 of them were transplanted to 10 recipients. | 2 | Proteasomal deubiquitinase UCH37 inhibits degradation of β-catenin and promotes cell proliferation and motility显示文摘The ubiquitin–proteasome system degrades most cellular proteins in eukaryotes.UCH37,also known as UCH-L5,is a deubiquitinase binding to Rpn13,a receptor for ubiquitinated substrates in the 26 S proteasome.But,it remains unclear how UCH37 influences the proteasomal degradation of the ubiquitinated substrates.Because deletion of UCH37 is embryonically lethal in mice,this study aims to investigate the role of UCH37 in proteasomal degradation by constructing the UCH37-deficient cell lines using CRISPR/Cas9 technology.Our results demonstrated that deletion of UCH37 decreased the levels of proteasomal Rpn13,implying that UCH37 might facilitate incorporation of Rpn13 into the proteasome.Meanwhile,deletion of UCH37 decreased the levels of β-catenin and the early endosomal protein Rab8.β-Catenin interacts with TCF/LEF to control transcription,and is involved in development,tissue homeostasis and tumorigenesis.We further found that deletion of UCH37 increased the levels of the ubiquitinated β-catenin and accelerated the hydrogen peroxide-stimulated degradation of β-catenin.Deletion of UCH37 also down-regulated the transcription of c-Myc,a downstream effector of β-catenin,and inhibited cell proliferation and motility.These results raise the possibility that UCH37 maintains the homeostasis of proteasomal degradation reciprocally by assisting the recruitment of the ubiquitin receptor Rpn13 into the proteasome and by reversing ubiquitination of certain critical substrates of the 26 S proteasome. | Zijian Li Luming Zhou Tianxia Jiang Libin Fan Xiaoying Liu Xiaobo Qiu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,3: | 4 |
| 3 | Targeting histones for degradation in cancer cells as a novel strategy in cancer treatment显示文摘The anticancer therapies with the joint treatment of a histone deacetylase(HDAC) inhibitor and a DNA-damaging approach are actively under clinical investigations, but the underlying mechanism is unclear. Histone homeostasis is critical to genome stability, transcriptional accuracy, DNA repair process, senescence, and survival. We have previously demonstrated that the HDAC inhibitor, trichostatin A(TSA), could promote the degradation of the core histones induced by γ-radiation or the DNAalkylating agent methyl methanesulfonate(MMS) in non-cancer cells, including mouse spermatocyte and embryonic fibroblast cell lines. In this study, we found that the joint treatment by TSA and MMS induced the death of the cultured cancer cells with an additive effect, but induced degradation of the core histones synergistically in these cells. We then analyzed various combinations of other HDAC inhibitors, including suberoylanilide hydroxamic acid and valproate sodium, with MMS or other DNAdamaging agents, including etoposide and camptothecin. Most of these combined treatments induced cell death additively, but all the tested combinations induced degradation of the core histones synergistically. Meanwhile, we showed that cell cycle arrest might not be a primary consequence for the joint treatment of TSA and MMS. Given that clinic treatments of cancers jointly with an HDAC inhibitor and a DNA-damaging approach often show synergistic effects, histone degradation might more accurately underlie the synergistic effects of these joint treatments in clinic applications than other parameters, such as cell death and cell cycle arrest. | 8 | lncRNA LINC00960 promotes apoptosis by sponging ubiquitin ligase Nrdp1-targeting miR-183-5p显示文摘The ubiquitin ligase Nrdp1/RNF41 promotes the ubiquitin-dependent degradation of multiple important substrates,including BRUCE/BIRC6,a giant ubiquitin-conjugating enzyme inhibiting both apoptosis and autophagy.miR-183-5p is associated with various malignancies potentially by targeting dozens of genes.Here,we show that the lncRNA LINC00960 binds to the Nrdp1-targeting miR-183-5p and promotes apoptosis.Compared to other known miR-183-5p targets,Nrdp1 mRNA is among the few with top scores to complement miR-183-5p.miR-183-5p binds to the 3′UTR of Nrdp1 mRNA and downregulates Nrdp1 at both the mRNA and protein levels.The miR-183-5p mimics inhibit DNA damage-induced apoptosis probably by upregulating BRUCE level,whereas the miR-183-5p inhibitor suppresses the effects of miR-183-5p.LINC00960 is the noncoding RNA with the highest score to complement miR-183-5p.LINC00960 overexpression reduces,but its knockdown increases,the level of miR-183-5p,whereas LINC00960 overexpression increases,but its knockdown decreases,the level of Nrdp1 and apoptosis.Importantly,the expression of LINC00960,which is associated with multiple types of tumors,positively correlates with that of Nrdp1 in several tumors but inversely correlates with that of miR-183-5p in multiple human tumor cell lines,as analysed by quantitative PCR.Thus,miR-183-5p downregulates Nrdp1 expression and inhibits apoptosis,whereas LINC00960 upregulates Nrdp1 and promotes apoptosis by inhibiting miR-183-5p.These results may provide new ideas for the prevention,diagnosis and treatment of apoptosis-related diseases,such as tumors and neurodegenerative diseases. | Hao Yang Tianxia Jiang Libin Fan Xiaobo Qiu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,1: | 1 |
| 9 | Investigation of the mesospheric temperature over Fort Collins region显示文摘The nightly mean mesospheric temperature profiles between 80 and 107 km,observed by Na lidar,over Fort Collins,Colorado(41°N,105°W)from 1990 to 2010,are employed to research the temporal and spatial variations and mesopause.We find that the maximum mean temperature is in summer months above 95 km,but reverse below 95 km,and there is a cooler region below 185 K around 97 km in August.The largest seasonal variation is 39.2 K at 81 km,and the minimum is 6.5 K at 96.5 km.The maximum standard derivation in spring and autumn months are larger than other seasons above 105 km,but the temperatures in March,June and September are lower than the other months between 82 km and 100 km where winter is the largest season.Moreover,the seasonal variations of the temperature are about 36,8 and 21 K at 85,95 and 105 km,respectively.winter is colder and summer is warmer above 97.5 km,but reverse below 92 km.The mesopause height is around 102 km in winter,but 84 km in summer,and the mean speed of decreasing or increasing of the mesopause height is about 5 km/month in spring and autumn months which are about 90 km.The lasting time of the mesopause in winter is near 6 months,longer than other seasons,and the mesopause temperature is about 165 K in cool summer,and 185 K in warm winter. | FAN Xin WENG LiBin ZHANG JianBin FANG HanXian XIE YanQiong | 2014 | Science China(Technological Sciences)2014,57,8: | 0 |

[Method] Taking Tobacco Mosaic Virus(TMV) and Cucumber Mosaic Virus(CMV) of sweet(hot) pepper as the sources of virus,an affordable indoor virus inoculation and identification technology was developed in the paper. [Result] The suitable inoculation concentration of CMV was five to ten times,and the best seedling age for inoculation was five to six leaves. Suitable inoculation concentration of TMV was 20 to 30 times,and the best seedling age for inoculation was three to six leaves. Single inoculation technology was mainly used for indoor virus inoculation and identification of sweet(hot) pepper,and complex inoculation technology could also be adopted with first inoculation of CMV and late inoculation of TMV. For mixed inoculation technology,CMV∶TMV should be 1∶1. Complex inoculation and mix inoculation should base on the technology of single inoculation.
