2Comparison of novel and standard diagnostic tools for the detection of Schistosoma mekongi infection in Lao People’s Democratic Republic and Cambodia显示文摘Background:Given the restricted distribution of Schistosoma mekongi in one province in Lao People’s Democratic Republic(Lao PDR)and two provinces in Cambodia,together with progress of the national control programmes aimed at reducing morbidity and infection prevalence,the elimination of schistosomiasis mekongi seems feasible.However,sensitive diagnostic tools will be required to determine whether elimination has been achieved.We compared several standard and novel diagnostic tools in S.mekongi-endemic areas.Methods:The prevalence and infection intensity of S.mekongi were evaluated in 377 study participants from four villages in the endemic areas in Lao PDR and Cambodia using Kato-Katz stool examination,antibody detection based on an enzyme-linked immunosorbent assay(ELISA)and schistosome circulating antigen detection by lateral-flow tests.Two highly sensitive test systems for the detection of cathodic and anodic circulating antigens(CCA,CAA)in urine and serum were utilized.Results:Stool microscopy revealed an overall prevalence of S.mekongi of 6.4%(one case in Cambodia and 23 cases in Lao PDR),while that of Opisthorchis viverrini,hookworm,Trichuris trichiura,Ascaris lumbricoides and Taenia spp.were 50.4%,28.1%,3.5%,0.3%and 1.9%,respectively.In the urine samples,the tests for CCA and CAA detected S.mekongi infections in 21.0%and 38.7%of the study participants,respectively.In the serum samples,the CAA assay revealed a prevalence of 32.4%,while a combination of the CAA assay in serum and in urine revealed a prevalence of 43.2%.There was a difference between the two study locations with a higher prevalence reached in the samples from Lao PDR.Conclusions:The CCA,CAA and ELISA results showed substantially higher prevalence estimates for S.mekongi compared to Kato-Katz thick smears.Active schistosomiasis mekongi in Lao PDR and Cambodia might thus have been considerably underestimated previously.Hence,sustained control efforts are still needed to break transmission of S.mekongi.The pivotal role of highly sensitive diagnostic assays in areas targeting elimination cannot be overemphasised.Youthanavanh Vonghachack Somphou Sayasone Virak Khieu Robert Bergquist Govert Jvan Dam Pytsje THoekstra Paul L.A.M.Corstjens Beatrice Nickel Hanspeter Marti Jürg Utzinger Sinuon Muth Peter Odermatt 2017Infectious Diseases of Poverty2017,6,1:4

5Understanding and tuning blue-to-near-infrared photon cutting by the Tm^(3+)/Yb^(3+) couple显示文摘Lanthanide-based photon-cutting phosphors absorb high-energy photons and‘cut’them into multiple smaller excitation quanta.These quanta are subsequently emitted,resulting in photon-conversion efficiencies exceeding unity.The photon-cutting process relies on energy transfer between optically active lanthanide ions doped in the phosphor.However,it is not always easy to determine,let alone predict,which energy-transfer mechanisms are operative in a particular phosphor.This makes the identification and design of new promising photon-cutting phosphors difficult.Here we unravel the possibility of using the Tm^(3+)/Yb^(3+) lanthanide couple for photon cutting.We compare the performance of this couple in four different host materials.Cooperative energy transfer from Tm^(3+)to Yb^(3+) would enable blue-to-near-infrared conversion with 200% efficiency.However,we identify phonon-assisted cross-relaxation as the dominant Tm^(3+)-to-Yb^(3+) energy-transfer mechanism in YBO_(3),YAG,and Y_(2)O_(3).In NaYF_(4),in contrast,the low maximum phonon energy renders phonon-assisted cross-relaxation impossible,making the desired cooperative mechanism the dominant energy-transfer pathway.Our work demonstrates that previous claims of high photon-cutting efficiencies obtained with the Tm^(3+)/Yb^(3+) couple must be interpreted with care.Nevertheless,the Tm^(3+)/Yb^(3+) couple is potentially promising,but the host material-more specifically,its maximum phonon energy-has a critical effect on the energy-transfer mechanisms and thereby on the photon-cutting performance.Dechao Yu Ting Yu Arnoldus Jvan Bunningen Qinyuan Zhang Andries Meijerink Freddy T.Rabouw 2020Light(Science & Applications)2020,9,1:2
6Post-hepatectomy liver regeneration in the context of bile acid homeostasis and the gut-liver signaling axis显示文摘Background:Liver regeneration following partial hepatectomy(PHx)is a complicated process involving multiple organs and several types of signaling networks.The bile acid-activated metabolic pathways occupy an auxiliary yet important chapter in the entire biochemical story.PHx is characterized by rapid but transient bile acid overload in the liver,which constitutes the first wave of proliferative signaling in the remnant hepatocytes.Bile acids trigger hepatocyte proliferation through activation of several nuclear receptors.Following biliary passage into the intestines,enterocytes reabsorb the bile acids,which results in the activation of farnesoid X receptor(FXR),the consequent excretion of fibroblast growth factor(FGF)19/FGF15,and its release into the enterohepatic circulation.FGF19/FGF15 subsequently binds to its cognate receptor,fibroblast growth factor receptor 4(FGFR4)complexed withβ-klotho,on the hepatocyte membrane,which initiates the second wave of proliferative signaling.Because some bile acids are toxic,the remnant hepatocytes must resolve the potentially detrimental state of bile acid excess.Therefore,the hepatocytes orchestrate a bile acid detoxification and elimination response as a protective mechanism in concurrence with the proliferative signaling.The response in part results in the excretion of(biotransformed)bile acids into the canalicular system,causing the bile acids to end up in the intestines.Relevance for patients:Recently,FXR agonists have been shown to promote regeneration via the gut-liver axis.This type of pharmacological intervention may prove beneficial for patients with hepatobiliary tumors undergoing PHx.In light of these developments,the review provides an in-depth account of the pathways that underlie post-PHx liver regeneration in the context of bile acid homeostasis in the liver and the gut-liver signaling axis.Lianne de Haan Sarah Jvan der Lely Anne-Loes K.Warps Quincy Hofsink Pim B.Olthof Mark J.de Keijzer Daniel A.Lionarons Lionel Mendes-Dias Bote G.Bruinsma Korkut Uygun Hartmut Jaeschke Geoffrey C.Farrell Narci Teoh Rowan Fvan Golen Tiangang Li Michal Heger 2018Journal of Clinical & Translational Research2018,4,1:2
