| 1 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |
| 2 | Noninvasive Ultrasound Deep Brain Stimulation for the Treatment of Parkinson’s Disease Model Mouse显示文摘Modulating basal ganglia circuitry is of great signifcance in the improvement of motor function in Parkinson’s disease(PD).Here,for the frst time,we demonstrate that noninvasive ultrasound deep brain stimulation(UDBS)of the subthalamic nucleus(STN)or the globus pallidus(GP)improves motor behavior in a subacute mouse model of PD induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).Immunohistochemical c-Fos protein expression confrms that there is a relatively high level of c-Fos expression in the STN-UDBS and GP-UDBS group compared with sham group(both p<0.05).Furthermore,STN-UDBS or GPUDBS signifcantly increases the latency to fall in the rotarod test on day 9(p<0.05)and decreases the time spent climbing down a vertical rod in the pole test on day 12(p<0.05).Moreover,our results reveal that STN-UDBS or GP-UDBS protects the dopamine(DA)neurons from MPTP neurotoxicity by downregulating Bax(p<0.001),upregulating Bcl-2(p<0.01),blocking cytochrome c(Cyt C)release from mitochondria(p<0.05),and reducing cleaved-caspase 3 activity(p<0.01)in the ipsilateral substantia nigra(SN).Additionally,the safety of ultrasound stimulation is characterized by hematoxylin and eosin(HE)and Nissl staining;no hemorrhage or tissue damage is detected.Tese data demonstrate that UDBS enables modulation of STN or GP neural activity and leads to neuroprotection in PD mice,potentially serving as a noninvasive strategy for the clinical treatment of PD. | Hui Zhou Lili Niu Long Meng Zhengrong Lin Junjie Zou Xiangxiang Xia Xiaowei Huang Wei Zhou Tianyuan Bian Hairong Zheng | 2019 | Research2019,,1: | 9 |

| 4 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 6 | Relationship between atmospheric pollutants and risk of death caused by cardiovascular and respiratory diseases and malignant tumors in Shenyang, China, from 2013 to 2016: an ecological research显示文摘Background:Air pollutants and their pathogenic effects differ among regions and seasons.We aimed to explore the relationship between fine particulate matter(PM2.5),sulfur dioxide(SO2),and ozone-8 hours(O3-8h)concentrations in heating and non-heating seasons and the associated death risk due to cardiovascular diseases(CDs),respiratory diseases(RDs),and malignant tumors.Methods:Data were collected in Shenyang,China,from April 2013 to March 2016.We analyzed the correlation or lagged effect of atmospheric pollutant concentration,meteorological conditions,and death risk due to disorders of the circulatory system,respiratory system,and malignant tumor in heating and non-heating seasons.We also used multivariate models to analyze the association of air pollutants during holidays with the death risk due to the evaluated diseases while considering the presence or absence of meteorological factors.Results:An increase in the daily average SO2 concentration by 10μg/m^3 increased the death risk by CDs,which reached a maximum of 2.0%(95%confidence interval[CI]:1.3%–2.7%)on lagging day 4 during the non-heating season and 0.2%(95%CI:0.1%-0.4%)on lagging day 3 during the heating season.The risk of death caused by RDs peaked on lagging day 1 by 0.8%(95%CI:0.4%–1.2%)during the heating season.An increase in O3-8h concentration by 10μg/m^3 increased the risk of RD-related death on lagging day 2 by 1.0%(95%CI:0.4%–1.7%)during the non-heating season,which was significantly higher than the 0.1%(95%CI:0–0.9%)increase during the heating season.Further,an increase in the daily average PM2.5 concentration by 10μg/m^3 increased the risk of death caused by RDs by 0.3%and 0.8%during heating and non-heating seasons,respectively,which peaked on lagging day 0.However,air pollution was not significantly associated with the risk of death caused by malignant tumors.Conclusion:Short-term exposure to PM2.5,SO2,and O3 during the non-heating season resulted in higher risks of CD-related death,followed by RD-related death. | Shu-Yue Xia De-Sheng Huang Hui Jia Yan Zhao Nan Li Ming-Qing Mao Hong Lin Yun-Xia Li Wei He Long Zhao | 2019 | Chinese Medical Journal2019,,19: | 4 |
| 7 | Assessment of estrogen disrupting potency in animal foodstuffs of China by combined biological and chemical analyses显示文摘Food has been documented as one of major routes for human exposure to environmental estrogens(EEs), but information on the occurrence of EEs in animal foodstuffs is still scarce.This study analyzed estrogenic activity in 16 types of animal foodstuffs(n = 142) collected from four cities(Wuhan, Guangzhou, Wenzhou and Yantai) of China by combined yeast estrogen screen(YES) bioassay and chemical analysis. By bioassay, all samples' extracts were found to induce estrogenic activities and the bioassay-derived 17β-estradiol equivalent(EEQbio) ranged from 8.29 to 118.32 ng/g. In addition, the samples were analyzed by liquid chromatography coupled to tandem mass spectrometry for further chemical analysis. 17β-Estradiol was found in all samples in this survey at levels of 0.44 to 15.04 ng/g.All samples had 33.1% detection rate of 17α-ethinylestradiol(EE2), and the maximum concentration was 2.80 ng/g. Bisphenol A and 4-nonylphenols were detected in 83.8% and83.1% of samples, with concentrations up to 12.56 ng/g and 35.76 ng/g, respectively.However, the concentrations of estrone, diethylstilbestrol and 4-t-octylphenol were found to be below the limit of detection. A comparison of EEQbiomeasured from the YES assay and EEQchemcalculated from chemical analysis showed good correlation(R2= 0.84). | 10 | Dynamic profile of ocular refraction in pediatric cataract patients after lens surgeries显示文摘AIM: To study the change in ocular refraction in patients with pediatric cataracts(PCs) after lens extraction. METHODS: A total of 1258 patients who were undergoing cataract extraction with/without intraocular lens(IOL)implantation were recruited during preoperative examinations between Jan 2010 and Oct 2013. Patient ages ranged from 1.5 mo to 14y. Follow-ups were conducted at1 wk, 1, and 3 mo postoperatively and every 3 mo in the first year, then 6 mo thereafter. Ocular refraction [evaluated as spherical equivalent(SE)] and yearly myopic shift(YMS)were recorded and statistically analyzed among patients with age at surgery, baseline ocular refraction, gender,postoperative time and laterality(bilateral vs unilateral). RESULTS: By Dec 31st 2015, 1172 participants had been followed for more than 2y. The median follow-up period was 3y. The critical factors affecting the ocular refraction of PC patients were baseline ocular refraction, postoperative time for both aphakic and pseudophakic eyes. YMS grew most rapidly in young childhood and early adolescence. | 12 | Source attribution for mercury deposition with an updated atmospheric mercury emission inventory in the Pearl River Delta Region, China显示文摘We used CMAQ-Hg to simulate mercury pollution and identify main sources in the Pearl River Delta (PR.D) with updated local emission inventory and latest regional and global emissions. The total anthropogenic mercury emissions in the PRD for 2014 were 11,939.6 kg. Power plants and industrial boilers were dominant sectors, responsible for 29.4 and 22.7%. We first compared model predictions and observations and the results showed a good performance. Then five scenarios with power plants (PP), municipal solid waste incineration (MSWI), industrial point sources (IP), natural sources (NAT), and boundary conditions (BCs) zeroed out separately were simulated and compared with the base case. BCs was responsible for over 30% of annual average mercury concentration and total deposition while NAT contributed around 15%. Among the anthropogenic sources, IP (22.9%) was dominant with a contribution over 20.0% and PP (18.9%) and MSWI (11.2%) ranked second and third. Results also showed that power plants were the most important emission sources in the central PRD, where the ultra-low emission for thermal power units need to be strengthened. In the northern and western PRD, cement and metal productions were priorities for mercury control. The fast growth of municipal solid waste incineration were also a key factor in the core areas. In addition, a coordinated regional mercury emission control was important for effectively controlling pollution.
