1A comprehensive review of integrative pharmacology-based investigation:A paradigm shift in traditional Chinese medicine显示文摘Over the past decade,traditional Chinese medicine(TCM) has widely embraced systems biology and its various data integration approaches to promote its modernization.Thus,integrative pharmacology-based traditional Chinese medicine(TCMIP) was proposed as a paradigm shift in TCM.This review focuses on the presentation of this novel concept and the main research contents,methodologies and applications of TCMIP.First,TCMIP is an interdisciplinary science that can establish qualitative and quantitative pharmacokinetics-pharmacodynamics(PK-PD) correlations through the integration of knowledge from multiple disciplines and techniques and from different PK-PD processes in vivo.Then,the main research contents of TCMIP are introduced as follows:chemical and ADME/PK profiles of TCM formulas;confirming the three forms of active substances and the three action modes;establishing the qualitative PK-PD correlation;and building the quantitative PK-PD correlations,etc.After that,we summarize the existing data resources,computational models and experimental methods of TCMIP and highlight the urgent establishment of mathematical modeling and experimental methods.Finally,we further discuss the applications of TCMIP for the improvement of TCM quality control,clarification of the molecular mechanisms underlying the actions of TCMs and discovery of potential new drugs,especially TCM-related combination drug disco very.Haiyu Xu Yanqiong Zhang Ping Wang Junhong Zhang Hong Chen Luoqi Zhang Xia Du Chunhui Zhao Dan Wu Feng Liu Hongjun Yang Changxiao Liu 2021Acta Pharmaceutica Sinica B2021,11,6:18

3Heavy Oil and Oil Sands:Global Distribution and Resource Assessment显示文摘Global recoverable resources of heavy oil and oil sands have been assessed by CNPC using a geology-based assessment method combined with the traditional volumetric method, spatial interpolation method, parametric-probability method etc. The most favourable areas for exploration have been selected in accordance with a comprehensive scoring system. The results show:(1) For geological resources, CNPC estimate 991.18 billion tonnes of heavy oil and 501.26 billion tonnes of oil sands globally, of which technically recoverable resources of heavy oil and oil sands comprise 126.74 billion tonnes and 64.13 billion tonnes respectively. More than 80% of the resources occur within Tertiary and Cretaceous reservoirs distributed across 69 heavy oil basins and 32 oil sands basins. 99% of recoverable resources of heavy oil and oil sands occur within foreland basins, passive continental-margin basins and cratonic basins.(2) Since residual hydrocarbon resources remain following large-scale hydrocarbon migration and destruction, heavy oil and oil sands are characterized most commonly by late hydrocarbon accumulation, the same basin types and source-reservoir conditions as for conventional hydrocarbon resources, shallow burial depth and stratabound reservoirs.(3) Three accumulation models are recognised, depending on basin type: degradation along slope; destruction by uplift; and migration along faults.(4) In addition to mature exploration regions such as Canada and Venezuela, the Volga-Ural Basin and the Pre-Caspian Basin are less well-explored and have good potential for oil-sand discoveries, and it is predicted that the Middle East will be an important region for heavy oil development.LIU Zuodong WANG Hongjun Graham BLACKBOURN MA Feng HE Zhengjun WEN Zhixing WANG Zhaoming YANG Zi LUAN Tiansi WU Zhenzhen 2019Acta Geologica Sinica(English Edition)2019,93,1:9
4Effect of Ginkgo biloba leaf extract on cerebral cortex amino acid levels in cerebral ischemia model rats显示文摘OBJECTIVE: To investigate the effect of Ginkgo biloba leaf extract on amino acid levels in the cerebral cortex of cerebral ischemia model rats induced by middle cerebral artery occlusion(MCAO).METHODS: A rat model of cerebral ischemia was established by MCAO. Male rats were divided into a negative control group(Control), a sham-operated group(Sham), an ischemic group(MCAO), and an ischemic group treated with Ginkgo biloba leaf extract(MCAO_D). All groups were divided into two subgroups with occlusion times of 12 and 24 h, respectively. The levels of 18 endogenous amino acids in the cerebral cortex were quantified by triple quadrupole-liquid chromatography-mass spectrometry.RESULTS: Compared with the MCAO group, behavioral performance, neurological deficit score, and cerebral infarct volume were significantly improved in the MCAO_D group(P < 0.05, P < 0.01). Compared with the sham group, the levels of 17 amino acids in the cerebral cortex were markedly changed in the MCAO group. The levels of Alanine(Ala), Isoleucine(Ile), Glutamic acid(Glu), Serine(Ser), Valine(Val), Phenylalanine(Phe), Proline(Pro),Threonine(Thr), Lysine(Lys), Tyrosine(Tyr), Hydroxyproline(Hyp), Arginine(Arg), Leucine(Leu),Tryptophan(Trp), and Glycine(Gly) were increased(P < 0.001, P < 0.05), while levels of Gln and Tau were decreased(P < 0.001, P < 0.05). Compared with the MCAO group, Ginkgo biloba extract treatment in the MCAO_D group significantly down-regulated the levels of 11 amino acids, especially those of Arg, Thr, and Ser in 12 or 24 h.CONCLUSION: Injection of Ginkgo biloba leaf extract has a therapeutic effect on model rats with MCAO-induced cerebral ischemia by acting on amino acids in the cerebral cortex. 5Novel SNPs in IL-17F and IL-17A genes associated with somatic cell count in Chinese Holstein and Inner-Mongolia Sanhe cattle显示文摘Background: Bovine mastitis is the most common and costly disease of lactating cattle worldwide. Apart from milk somatic cell count(SCC) and somatic cell score(SCS), serum cytokines such as interleukin-17(IL-17) and interleukin-4(IL-4) may also be potential indicators for bovine mastitis. The present study was designed to investigate the effects of single nucleotide polymorphisms(SNPs) in bovine IL-17 F and IL-17 A genes on SCC, SCS and serum cytokines in Chinese Holstein and Inner-Mongolia Sanhe cattle, and to compare the m RNA expression variations of the cows with different genotypes.Results: A total of 464 lactating cows(337 Holstein and 127 Inner-Mongolia Sanhe cattle) were screened for SNPs identification and the data were analyzed using fixed effects of herd, parity, season and year of calving by general linear model procedure. The results revealed that SNP g.24392436 C > T in IL-17 F and SNP g.24345410 A > G in IL-17 A showed significant effects on SCC and IL-4 in Holstein(n = 337) and on IL-17 and IL-4 in Sanhe cattle(n = 127). 6Methylation of RAR-β2, RASSF1A, and CDKN2A Genes Induced by Nickel Subsulfide and Nickel-carcinogenesis in Rats显示文摘Objective To investigate the expression variation of RAR‐β2, RASSF1A, and CDKN2A gene in the process of nickel‐induced carcinogenesis. Methods Nickel subsulfide (Ni 3 S 2 ) at dose of 10 mg was given to Wistar rats by intramuscular injection. The mRNA expression of the three genes in induced tumors and their lung metastasis were examined by Real‐time PCR. The methylation status of the 5' region of these genes were detected by Quantitative Real‐time methylation specific PCR. Results The mRNA expressions of the three genes both in muscle and lung tumor were decreased distinctly in comparison with normal tissue. But hypermethylation was found only in muscle tumor. 9Elemental composition and risk assessment of heavy metals in the PM10 fractions of road dust and roadside soil显示文摘A total of 64 dust samples were analyzed to determine the size distribution and elemental composition of the PM10 fraction, including42 road dust(RD), 12 roadside soil(RSD), and 10 park road dust(PRD) samples. The mass of dust smaller than 20μm was dominated by particles sized 2.5-16 μm, which accounted for 85%, 88%, and 87% of the RD, PRD, and RSD, respectively. Additionally, crustal elements accounted for 30.14%, 36.35%, and 37.14% of the PMio fractions of the RD, RSD, and PRD, respectively. The most abundant trace elements in RD, RSD, and PRD were Zn, Mn, and Cu (range, 277 to 874 mg/kg). Moreover, the /geo values revealed all dusts were contaminated with Pb, Zn, Cu, Sb, Sn, and Cd. Health risk assessment showed that Mn, Ni, Cu, Zn, Cd, Sb, and Pb in the PM10 fraction of three types of dusts posed non-cancer risks to children but posed no non-cancer risk to adults. 11Improvement Effect of Bioaugmentation with Phenol Degrading Bacteria on Lurgi Coal Gasification Wastewater Treatment显示文摘The improvement effect of bioaugmentation with phenol degrading bacteria( PDB) on pollutants removal and chemicals consumption was investigated in a full-scale Lurgi coal gasification wastewater( LCGW)treatment plant. Bioaugmentation with PDB applied in biological contact oxidation tank( BCOT) was carried out in summer to prevent the limitation of low temperature on the bacteria activity. After augmentation,the removal of COD and total phenol(TPh) was significantly enhanced,with efficiencies from 78.5% and 80% to 82.3% and 86.6% in BCOT,respectively. The improvement could also be detected in further processes,including anoxic-oxic,coagulation sedimentation and biological aerated filter,with COD and TPh removal efficiencies increment from 70.1%,24. 7% and 53. 4% to 73. 9%,29. 1% and 55. 9%,from 67. 1%,20% and 25% to 72.5%,25% and 32%, respectively. In addition, chemicals used for denitrification and coagulation sedimentation showed considerable reduction after bioaugmentation,with methanol,coagulant,coagulant aid and bleaching dosage from 100. 0,100. 0,80. 0 and 60. 0 mg/L to 85. 0,70. 6,57. 8 and 45.7 mg/L,respectively. 12Distribution and potential of global oil and gas resources显示文摘Using conventional and unconventional oil and gas resource evaluation methods with play as a unit, this study evaluates the oil and gas geology and resource potential of conventional oil and gas resources and seven types of unconventional resources in the global major oil and gas basins(excluding China). For the first time, resource evaluation data with independent intellectual property rights has been obtained. According to evaluation and calculation, the global recoverable conventional oil resources are 5 350.0×108 t, the recoverable condensate oil resources are 496.2×108 t, and the recoverable natural gas resources are 588.4×1012 m3. The remaining oil and gas 2 P recoverable reserves are 4 212.6×108 t, the reserve growth of oil and gas fields are 1 531.7×108 t. The undiscovered oil and gas recoverable resources are 3 065.5×108 t. The global unconventional oil recoverable resources are 4 209.4×108 t and the unconventional natural gas recoverable resources are 195.4×1012 m3. 14Scattered seeding of CAR T cells in solid tumors augments anticancer efficacy显示文摘Chimeric antigen receptor T cell(CAR T) therapy was a milestone in the treatment of relapsed and refractory B cell malignancies. However, beneficial effects of CAR T cells have not been obtained in solid tumors yet. Herein, we implement a porous microneedle patch that accommodates CAR T cells and allows in situ penetration-mediated seeding of CAR T cells when implanted in the tumor bed or in the post-surgical resection cavity. CAR T cells loaded in the pores of the microneedle tips were readily escorted to the tumor in an evenly scattered manner without losing their activity. Such microneedle-mediated local delivery enhanced infiltration and immunostimulation of CAR T cells as compared to direct intratumoral injection. 15IFN-stimulated P2Y13 protects mice from viral infection by suppressing the cAMP/EPACl signaling pathway显示文摘Among the most important sensors of extracellular danger signals, purinergic receptors have been demonstrated to play crucial roles in host defense against infection. However, the function of P2 receptors in viral infection has been little explored. Here we demonstrated that P2Y13 and its ligand ADP play an important role in protecting hosts from viral infections. First, we demonstrate that P2Y13, as a typical interferon-stimulated gene, is induced together with extracellular ADP during viral infection. Most importantly, extracellular ADP restricts the replication of different kinds of viruses, including vesicular stomatitis virus, Newcastle disease virus, herpes simplex virus 1, and murine leukemia virus. This kind of protection is dependent on P2Y13 but not P2Y1 or P2Y12, which are also considered as receptors for ADP. Furthermore, cyclic adenosine monophosphate and EPAC1 are downregu-lated by extracellular ADP through the P2Y13-coupled Gi alpha subunit. Accordingly, inhibition or deletion of EPAC1 significantly eliminates ADP/P2Y13-mediated antiviral activities. 18Mapping of wheat stripe rust resistance gene Yr041133 by BSR-Seq analysis显示文摘Puccinia striiformis Westend. f. sp. tritici(Pst) pathotype CYR34 is widely virulent and prevalent in China.Here, we report identification of a strpie rust resistance(Yr) gene, designated Yr041133, in winter wheat line 041133. This line produced a hypersensitive reaction to CYR34 and conferred resistance to 13 other pathotypes. Resistance to CYR34 in line 041133 was controlled by a single dominant gene. Bulked segregant RNA sequencing(BSR-Seq) was performed on a pair of RNA bulks generated by pooling resistant and susceptible recombinant inbred lines. Yr041133 was mapped to a 1.7 c M genetic interval on the chromosome arm 7 BL that corresponded to a 0.8 Mb physical interval(608.9–609.7 Mb) in the Chinese Spring reference genome.
