| 3 | Effect of pyrophosphate on the light scatter in KDP crystal显示文摘Pyrophosphate doped potassium dihydrogen phosphate (KDP) crystal was grown from aqueous solution by the temperature lowering method. Light scatter in KDP crystal was detected with the ultramicroscopic method. The light scatter in KDP crystal was aggravated when pyro-phosphate was doped into the growth solution, which was distributed ununiformly in prism and pyramidal sectors of KDP crystal. Different effects of pyrophosphate on prism and pyramidal sectors of KDP crystal can explain this case. | 4 | Statistical Analysis of Regularity of Pesticide Residues in Vegetables Produced in Inner Mongoli显示文摘[Objective]The paper was to study regularity of pesticide residues in vegetables produced in Inner Mongolia.[Method]Mathematical statistic analysis was carried out on 6 800 samples of veggies,fruits,leguminous vegetables,nuts and seeds produced in Inner Mongolia.[Result]The dominated vegetables in Inner Mongolia were green leafy vegetables,solanaceous vegetables and melon vegetables,and their yields accounted for 70%of the total vegetables production.Since 2003,with the rapid increase of detected vegetable numbers,the status of vegetable quality safety had entered into a new stage of sustainable steady after a rapid decline or periodic fluctuation,and the differences in safety levels were obvious,of which the range of exceeding standard rate of pesticide residues in solanaceous vegetables,melon vegetables and leguminous vegetables(about 55%of the total vegetables)was under 2%,with average values of 1.1%,1.6%and 3.1%,respectively.They belonged to stable type.The exceeding standard rate of pesticide residues in green leafy vegetables and Chinese cabbage group(about 30%of total vegetables)presented a decreasing trend year by year,wondering in the range of 7%-10%.They belonged to main risk type.The time period of exceeding standard of pesticide residues in root vegetables and cole vegetables was under predictable,and its exceeding standard rate in some years was over 5%(amplitude variation is over 15percentage points).They belonged to random risk type.The kinds of pesticides,which exceeding standard rate in vegetables were relatively intensive,presented variance in different vegetable species.70%of the pesticides belonged to intermittent over-limits and the probability was below 5%.About 20%tradition pesticides often exceeded standard and their probabilities were over 30%.The exceeding standard of organophosphorus and carbamates pesticides in vegetables presented a decreasing tend,while the risk of some new pesticides containing nitrogen element and high effective pyrethroids showed an increasing tendency.[Conclusion]The paper provided decision making services for agricultural management departments. | Fujin ZHANG Dekun HOU Jiang HE Tianyun GAO Hong LUO Songyan LANG Xinxin ZHANG Yiping YAO | 2013 | Agricultural Science & Technology2013,14,10: | 2 |

| 19 | miR-126 mitigates the osteogenic differentiation of human bone marrow-derived mesenchymal stem cells by targeting the ERK1/2 and Bcl-2 pathways显示文摘Human bone marrow mesenchymal stem cells(hBMMSCs)are a promising cell source for bone engineering owing to their high potential to differentiate into osteoblasts.The objective of the present study is to assess microRNA-126(miR-126)and examine its effects on the osteogenic differentiation of hBMMSCs.In this study,we investigate the role of miR-126 in the progression of osteogenic differentiation(OD)as well as the apoptosis and inflammation of hBMMSCs during OD induction.OD is induced in hBMMSCs,and matrix mineralization along with other ODassociated markers are evaluated by Alizarin Red S(AR)staining and quantitative PCR(qPCR).Gain-and loss-offunction studies are performed to demonstrate the role of miR-126 in the OD of hBMMSCs.Flow cytometry and qPCR-based cytokine expression studies are performed to investigate the effect of miR-126 on the apoptosis and inflammation of hBMMSCs.The results indicate that miR-126 expression is downregulated during the OD of hBMMSCs.Gain-and loss-of function assays reveal that miR-126 upregulation inhibits the differentiation of hBMMSCs into osteoblasts,whereas the downregulation of miR-126 promotes hBMMSC differentiation,as assessed by the determination of osteogenic genes and alkaline phosphatase activity.Furthermore,the miR-126 level is positively correlated with the production of inflammatory cytokines and apoptotic cell death.Additionally,our results suggest that miR-126 negatively regulates not only B-cell lymphoma 2(Bcl-2)expression but also the phosphorylation of extracellular signal-regulated protein kinase(ERK)1/2.Moreover,restoring ERK1/2 activity and upregulating Bcl-2 expression counteract the miR-126-mediated suppression of OD in hBMMSCs by promoting inflammation and apoptosis,respectively.Overall,our findings suggest a novel molecular mechanism relevant to the differentiation of hBMMSCs into osteoblasts,which can potentially facilitate bone formation by counteracting miR-126-mediated suppression of ERK1/2 activity and Bcl-2 expression. | Ying Zhang Yiping Dong Qiushi Wei Zhikun Zhuang Youwen Liu Qiang Yuan Wei He Zhenhao Jing Jitian Li Peifeng Li Leilei Zhang Zhinan Hong Ning Zhang Haibin Wang Wuyin Li | 2023 | Acta Biochimica et Biophysica Sinica2023,55,3: | 0 |
| 20 | Construction and application of gene targeting replacement vector formouse coagulation factor IX gene显示文摘Objective: To construct and apply a replacement targeting vector for mouse coagulation factor IX(mFIX) gene in embryonic stem (ES) cells. Methods: Based on the cloning and structural analysis of the genomic DNA fragment of coagulation factor IX gene from 129Sv mouse genomic DNA A phage library, PMFIXDEL plasmid was designed with positive-negative-selection (PNS) strategy, and constructed with commonmolecular cloning techniques. Structure of PMFIXDEL was identified by PCR and restriction analysis. Afterelectroporation with the linearized PMFIXDEL DNA, transfected ES cells were cultured in G418/GANC drugselection medium. The recombination efficacy of this vector was tested. Results: The main components ofPMFIXDEL were two copies of negative selection gene (HSV-tk expression cassette), a positive selectiongene (Neo expression cassette), long and short homologous fragments and plasmid backbone.
