2Xa7, a new executor R gene that confers durable and broad-spectrum resistance to bacterial blight disease in rice显示文摘Bacterial blight(BB)is a globally devastating rice disease caused by Xanthomonas oryzae pv.oryzae(Xoo).The use of disease resistance(R)genes in rice breeding is an effective and economical strategy for the control of this disease.Nevertheless,a majority of R genes lack durable resistance for long-term use under global warming conditions.Here,we report the isolation of a novel executor R gene,Xa7,that confers extremely durable,broad-spectrum,and heat-tolerant resistance to Xoo.The expression of Xa7 was induced by incompatible Xoo strains that secreted the transcription activator-like effector(TALE)AvrXa7 or PthXo3,which recognized effector binding elements(EBEs)in the Xa7 promoter.Furthermore,Xa7 induction was faster and stronger under high temperatures.Overexpression of Xa7 or co-transformation of Xa7 with avrXa7 triggered a hypersensitive response in plants.Constitutive expression of Xa7 activated a defense response in the absence of Xoo but inhibited the growth of transgenic rice plants.In addition,analysis of over 3000 rice varieties showed that the Xa7 locuswas found primarily in the indica and aus subgroups.A variation consisting of an 11-bp insertion and a base substitution(G to T)was found in EBEAvrXa7 in the tested varieties,resulting in a loss of Xa7 BB resistance.Through a decade of effort,we have identified an important BB resistance gene and characterized its distinctive interaction with Xoo strains;these findings will greatly facilitate research on the molecular mechanism of Xa7-mediated resistance and promote the use of this valuable gene in breeding.Xifeng Chen Pengcheng Liu Le Mei Xiaoling He Long Chen Hui Liu Shurong Shen Zhandong Ji Xixi Zheng Yuchen Zhang Zhenyu Gao Dali Zeng Qian Qian Bojun Ma 2021Plant Communications2021,2,3:9
3Engineering osteoarthritic cartilage model through differentiating senescent human mesenchymal stem cells for testing disease-modifying drugs显示文摘Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis(OA).In this study,we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs(DMOADs).Spe-cifically,human bone marrow-derived mesenchymal stromal cells(MSCs)were expanded in vitro up to passage 10(P10-MSCs).Following their senescent phenotype formation,P10-MSCs were subjected to pellet culture in chondrogenic medium.Results from qRT-PCR,histology,and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents,when compared to that from normal passage 4(P4)-MSCs.Interestingly,the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples,as demonstrated by RNA sequencing data and other analysis methods.Lastly,the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics.The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage.The P4-and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans.Ning Wang Yuchen He Silvia Liu Meagan J.Makarcyzk Guanghua Lei Alexander Chang Peter G Alexander Tingjun Hao Anne-Marie Padget Nuria de Pedro Tsapekos Menelaos Hang Lin 2022Science China(Life Sciences)2022,65,2:5

5Cyclin-dependent Kinase 18 Promotes Oligodendrocyte Precursor Cell Differentiation through Activating the Extracellular Signal-Regulated Kinase Signaling Pathway显示文摘The correct differentiation of oligodendrocyte precursor cells(OPCs) is essential for the myelination and remyelination processes in the central nervous system.Determining the regulatory mechanism is fundamental to the treatment of demyelinating diseases. By analyzing the RNA sequencing data of different neural cells, we found that cyclin-dependent kinase 18(CDK18) is exclusively expressed in oligodendrocytes. In vivo studies showed that the expression level of CDK18 gradually increased along with myelin formation during development and in the remyelination phase in a lysophosphatidylcholine-induced demyelination model, and was distinctively highly expressed in oligodendrocytes. In vitro overexpression and interference experiments revealed that CDK18 directly promotes the differentiation of OPCs, without affecting their proliferation or apoptosis. Mechanistically, CDK18 activated the RAS/mitogen-activated protein kinase kinase1/extracellular signal-regulated kinase pathway, thus promoting OPC differentiation. 7A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. 11A Yeast BiFC-seq Method for Genome-wide Interactome Mapping显示文摘Genome-wide physical protein±protein interaction(PPI)mapping remains a major challenge for current technologies.Here,we reported a high-efficiency BiFC-seq method,yeastenhanced green fluorescent protein-based bimolecular fluorescence complementation(y EGFPBiFC)coupled with next-generation DNA sequencing,for interactome mapping.We first applied y EGFP-BiFC method to systematically investigate an intraviral network of the Ebola virus.Two-thirds(9/14)of known interactions of EBOV were recaptured,and five novel interactions were discovered.Next,we used the BiFC-seq method to map the interactome of the tumor protein p53.We identified 97 interactors of p53,more than three-quarters of which were novel.Furthermore,in a more complex background,we screened potential interactors by pooling two BiFC libraries together and revealed a network of 229 interactions among 205 proteins.These results show that BiFC-seq is a highly sensitive,rapid,and economical method for genome-wide interactome mapping.Limin Shang Yuehui Zhang Yuchen Liu Chaozhi Jin Yanzhi Yuan Chunyan Tian Ming Ni Xiaochen Bo Li Zhang Dong Li Fuchu He Jian Wang 2022Genomics, Proteomics & Bioinformatics2022,20,4:1

14Interconnected N/P co-doped carbon nanocage as high capacitance electrode material for energy storage devices显示文摘Heteroatom doping carbon materials exhibit a huge application potential for energy storage devices(ESDs).Herein,interconnected N/P co-doped carbon nanocage(NP-CNC)was synthesized from pyrene molecules by using nano-MgO as template and melamine-phytic acid supramolecular aggregate as dopant coupled with KOH activation.The as-prepared NP-CNC possesses interconnected nanocages for electron transportation and abundant micropores for ion adsorption.Moreover,co-doped N/P species in NP-CNC provide active sites and additional pseudocapacitance.Consequently,NP-CNC as electrode material for symmetric supercapacitor exhibits a high gravimetric capacitance of 435 F·g^(-1) at 0.05 A·g^(-1),high volumetric capacitance of 274 F·cm^(-3) at 0.032 A·cm^(-3),and long cycle lifespan with 96.1%capacitance retention after 50,000 cycles.Furthermore,NP-CNC as cathode for zinc-ion hybrid supercapacitor delivers satisfactory energy and power densities of 130.6 Wh·kg^(-1)(82.3 Wh·L^(-1))and 14.4 kW·kg^(-1)(9.1 kWL^(-1)).This work paves a promising approach to the preparation of high capacitance NP-CNC for ESDs.Lei Yang Xiaojun He Yuchen Wei Honghui Bi Feng Wei Hongqiang Li Changzhou Yuan Jieshan Qiu 2022Nano Research2022,15,5:1

16Ultrasensitive solar-blind ultraviolet detection and optoelectronic neuromorphic computing using α-In_(2)Se_(3)phototransistors显示文摘Detection of solar-blind ultraviolet(SB-UV)light is important in applications like confidential communication,flame detection,and missile warning system.However,the existing SB-UV photodetectors still show low sensitivities.In this work,we demonstrate the extraordinary SB-UV detection performance of α-In_(2)Se_(3 )phototransistors.Benefiting from the coupled semiconductor and ferroelectricity property,the phototransistor has an ultraweak detectable power of 17.85 fW,an ultrahigh gain of 1.2×10^(6),a responsivity of 2.6×10^(5) A/W,a detectivity of 1.3×10^(16) Jones and an ultralow noise-equivalent-power of 4.2×10^(–20 )W/Hz1/2 for 275 nm light.Its performance exceeds most other UV detectors,even including commercial photomultiplier tubes and avalanche photodiodes.It can be also implemented as an optoelectronic synapse for neuromorphic computing.A 784×300×10 artificial neural network(ANN)based on this optoelectronic synapse is constructed and demonstrated with a high recognition accuracy and good noise-tolerance for the Fashion-MNIST dataset.These extraordinary features endow this phototransistor with the potential for constructing advanced SB-UV detectors and intelligent hardware.Yuchen Cai Jia Yang Feng Wang Shuhui Li Yanrong Wang Xueying Zhan Fengmei Wang Ruiqing Cheng Zhenxing Wang Jun He 2023Frontiers of physics2023,18,3:1

18Fish-Eye Image Distortion Correction Based on Adaptive Partition Fitting显示文摘The acquisition of images with a fish-eye lens can cause serious image distortion because of the short focal length of the lens.As a result,it is difficult to use the obtained image information.To make use of the effective information in the image,these distorted imagesmust first be corrected into the perspective of projection images in accordance with the human eye’s observation abilities.To solve this problem,this study presents an adaptive classification fitting method for fish-eye image correction.The degree of distortion in the image is represented by the difference value of the distances fromthe distorted point and undistorted point to the center of the image.The target points selected in the image are classified by the difference value.In the areas classified by different distortion differences,different parameter curves were used for fitting and correction.The algorithm was verified through experiments.The results showed that this method has a substantial correction effect on fish-eye images taken by different fish-eye lenses.Yibin He Wenhao Xiong Hanxin Chen Yuchen Chen Qiaosen Dai Panpan Tuand Gaorui Hu 2021Computer Modeling in Engineering & Sciences2021,,1:1
19Implementation of a TPM-Based Security Enhanced Browser Password Manager显示文摘In order to enhance the security of a browser password manager, we propose an approach based on a hardware trusted platform module(TPM). Our approach encrypts users' passwords with keys generated by the TPM, which uses a master password as the credential for authorization to access the TPM. Such a hardware-based feature may provide an efficient way to protect users' passwords. Experiment and evaluation results show that our approach performs well to defend against password stealing attack and brute force attack. Attackers cannot get passwords directly from the browser, therefore they will spend incredible time to obtain passwords.
