| 3 | Actively priming autophagic cell death with novel transferrin receptor-targeted nanomedicine for synergistic chemotherapy against breast cancer显示文摘Recently, considerable attention in the field of cancer therapy has been focused on the mammalian rapamycin target(m TOR), inhibition of which could result in autophagic cell death(ACD). Though novel combination chemotherapy of autophagy inducers with chemotherapeutic agents is extensively investigated, nanomedicine-based combination therapy for ACD remains in infancy. In attempt to actively trigger ACD for synergistic chemotherapy, here we incorporated autophagy inducer rapamycin(RAP) into 7 pep-modified PEG-DSPE polymer micelles(7 pep-M-RAP) to specifically target and efficiently priming ACD of MCF-7 human breast cancer cells with high expression of transferrin receptor(Tf R). Cytotoxic paclitaxel(PTX)-loaded micelle(7 pep-M-PTX) was regarded as chemotherapeutic drug model. We discovered that with superior intracellular uptake in vitro and more tumor accumulation of micelles in vivo, 7 pep-M-RAP exhibited excellent autophagy induction and synergistic antitumor efficacy with 7 pep-M-PTX. Mechanism study further revealed that 7 pep-M-RAP and 7 pep-MPTX used in combination provided enhanced efficacy through induction of both apoptosis-and mitochondria-associated autophagic cell death. | 7 | Cartilage-targeting peptide-modified dual-drug delivery nanoplatform with NIR laser response for osteoarthritis therapy显示文摘Cartilage-targeting delivery of therapeutic agents is still an effective strategy for osteoarthritis(OA)therapy.Recently,scavenging for reactive oxygen species(ROS)and activating autophagy have been increasingly reported to treat OA effectively.In this study,we designed,for the first time,a dual-drug delivery system based on metal organic framework(MOF)-decorated mesoporous polydopamine(MPDA)which composed of rapamycin(Rap)loaded into the mesopores and bilirubin(Br)loaded onto the shell of MOF.The collagen II-targeting peptide(WYRGRL)was then conjugated on the surface of above nanocarrier to develop a cartilage-targeting dual-drug delivery nanoplatform(RB@MPMW).Our results indicated the sequential release of two agents from RB@MPMW could be achieved via near-infrared(NIR)laser irritation.Briefly,the rapid release of Br from the MOF shell exhibited excellent ROS scavenging ability and anti-apoptosis effects,however responsively reduced autophagy activity,to a certain extent.Meanwhile,following the NIR irradiation,Rap was rapidly released from MPDA core and further enhanced autophagy activation and chondrocyte protection.RB@MPMW continuously phosphorylated AMPK and further rescued mitochondrial energy metabolism of chondrocytes following IL-1βstimulation via activating SIRT1-PGC-1αsignaling pathway.Additionally,the cartilage-targeting property of peptide-modified nanocarrier could be monitored via Magnetic Resonance(MR)and IVIS imaging.More significantly,RB@MPMW effectively delayed cartilage degeneration in ACLT rat model.Overall,our findings indicated that the as-prepared dual-drug delivery nanoplatform exerted potent anti-inflammation and anti-apoptotic effects,rescued energy metabolism of chondrocytes in vitro and prevented cartilage degeneration in vivo,which thereby showed positive performance for OA therapy. | Song Xue Xiaojun Zhou Weilin Sang Cong Wang Haiming Lu Yiming Xu Yiming Zhong Libo Zhu Chuanglong He Jinzhong Ma | 2021 | Bioactive Materials2021,6,8: | 5 |
| 8 | Simultaneous extraction of DNA and RNA from Escherichia coli BL 21 based on silica-coated magnetic nanoparticles显示文摘The extraction of nucleic acid is recognized as one of the most essential steps in molecular biology for initiating other downstream applications such as sequencing, amplification, hybridization, and cloning. Many commercial kits and methods are currently available that allow the extraction of only one type of nucleic acids-DNA or RNA. However, in parallel clinical detection of several diseases, a method for simultaneous extraction of both DNA and RNA from the same source is needed in such cases. In this study, a method for simultaneous extraction of DNA and RNA from bacteria based on magnetic nanoparticles(MNPs) was described. Lysis buffers were prepared to help the nucleic acid released and adsorbed to MNPs. Then, two washing buffers were used to remove the contamination of proteins and carbohydrates. The nucleic acids were finally eluted by Deoxyribonuclease(DNase) and Ribonucleases(RNase) free water. Different factors which might affect the purification of the nucleic acid were investigated, and the quantity and quality parameters of the nucleic acid were also recorded. The DNA and RNA extracted from bacteria were then respectively subjected to polymerase chain reaction(PCR) and reverse transcription PCR(RT-PCR) to further confirm its quality. | 10 | Size-tunable synthesis of gold nanorods using pyrogallol as a reducing agent显示文摘A novel and facile seed-mediated method for the preparation of monodispersed gold nanorods(GNRs) is presented by introducing pyrogallol as a reductant. Fast Fourier transformation of high-resolution transmission electron microscopy reveals that the synthesized GNRs are single crystalline. The longitudinal surface plasmon resonance of GNRs can be finely tuned by varying silver ion concentrations or seed amounts. Also, both thick(diameter >30 nm) and thin(diameter <10 nm) GNRs with exceptional monodispersity can be well prepared by this method. | 11 | Plant plasma membrane-resident receptors:Surveillance for infections and coordination for growth and development显示文摘As sessile organisms, plants are exposed to pathogen invasions and environmental fluctuations. To overcome the challenges of their surroundings, plants acquire the potential to sense endogenous and exogenous cues, resulting in their adaptability. Hence, plants have evolved a large collection of plasma membrane-resident receptors, including RECEPTOR-LIKE KINASEs(RLKs) and RECEPTOR-LIKE PROTEINs(RLPs) to perceive those signals and regulate plant growth,development, and immunity. The ability of RLKs and RLPs to recognize distinct ligands relies on diverse categories of extracellular domains evolved. Co-regulatory receptors are often required to associate with RLKs and RLPs to facilitate cellular signal transduction. RECEPTOR-LIKE CYTOPLASMIC KINASEs(RLCKs) also associate with the complex, bifurcating the signal to key signaling hubs, such as MITOGEN-ACTIVATED PROTEIN KINASE(MAPK) cascades, to regulate diverse biological processes.
