| 1 | The calcium-dependent kinase OsCPK24 functions in cold stress responses in rice显示文摘Calcium-dependent protein kinases(CPKs)are serine/threonine protein kinases that function in plant stress responses. Although CPKs are recognized as key messengers in signal transduction, the specific roles of CPKs and the molecular mechanisms underlying their activity remain largely unknown. Here, we characterized the function of Os CPK_(24), a cytosol-localized calciumdependent protein kinase in rice. Os CPK_(24) was universally and highly expressed in rice plants and was induced by cold treatment. Whereas Os CPK_(24) knockdown plants exhibited increased sensitivity to cold compared to wild type(WT), Os CPK_(24)-overexpressing plants exhibited increased cold tolerance. Plants overexpressing Os CPK_(24) exhibited increased accumulation of proline(an osmoprotectant) and glutathione(an antioxidant) and maintained a higher GSH/GSSG(reduced glutathione to oxidized glutathione) ratio during cold stress compared to WT. In addition to these effects in response to cold stress, we observed the kinase activity of Os CPK_(24) varied under different calcium concentrations. Further,Os CPK_(24) phosphorylated Os Grx_(10), a glutathionedependent thioltransferase, at rates modulated by changes in calcium concentration. | 2 | Plant immune signaling: Advancing on two frontiers显示文摘Plants have evolved multiple defense strategies to cope with pathogens, among which plant immune signaling that relies on cell-surface localized and intracellular receptors takes fundamental roles. Exciting breakthroughs were made recently on the signaling mechanisms of pattern recognition receptors(PRRs) and intracellular nucleotide-binding site(NBS) and leucine-rich repeat(LRR)domain receptors(NLRs). This review summarizes the current view of PRRs activation, emphasizing the most recent discoveries about PRRs’ dynamic regulation and signaling mechanisms directly leading to downstream molecular events including mitogen-activated protein kinase(MAPK) activation and calcium(Ca2+) burst. Plants also have evolved intracellular NLRs to perceive the presence of specific pathogen effectors and trigger more robust immune responses. | 3 | The receptor-like cytoplasmic kinase RIPK regulates broad-spectrum ROS signaling in multiple layers of plant immune system显示文摘Production of reactive oxygen species(ROS)via the activity of respiratory burst oxidase homologs(RBOHs)plays a vital role in multiple layers of the plant immune system,including pathogen-associated molecular pattern-triggered immunity(PTI),damage-associated molecular pattern-triggered immunity(DTI),effector-triggered immunity(ETI),and systemic acquired resistance(SAR).It is generally established that RBOHD is activated by different receptor-like cytoplasmic kinases(RLCKs)in response to various immune elicitors.In this study,we showed that RPM1-INDUCED PROTEIN KINASE(RIPK),an RLCK VII subfamily member,contributes to ROS production in multiple layers of plant immune system.The ripk mutants showed reduced ROS production in response to treatment with all examined immune elicitors that trigger PTI,DTI,ETI,and SAR.We found that RIPK can directly phosphorylate the N-terminal region of RBOHD in vitro,and the levels of phosphorylated S343/S347 residues of RBOHD are sigfniciantly lower in ripk mutants compared with the wild type upon treatment with all tested immune elicitors.We further demonstrated that phosphorylation of RIPK is required for its function in regulating RBOHD-mediated ROS production.Similar to rbohd,ripk mutants showed reduced stomatal closure and impaired SAR,and were susceptible to the necrotrophic bacterium Pectobacterium carotovorum.Collectively,our results indicate that RIPK regulates broad-spectrum RBOHD-mediated ROS signaling during PTI,DTI,ETI,and SAR,leading to subsequent RBOHD-dependent immune responses. | Ping Li Lulu Zhao Fan Qi Nang Myint Phyu Sin Htwe Qiuying Li Dawei Zhang Fucheng Lin Keke Shang-Guan Yan Liang | 2021 | Molecular Plant2021,14,10: | 5 |

| 5 | Plant plasma membrane-resident receptors:Surveillance for infections and coordination for growth and development显示文摘As sessile organisms, plants are exposed to pathogen invasions and environmental fluctuations. To overcome the challenges of their surroundings, plants acquire the potential to sense endogenous and exogenous cues, resulting in their adaptability. Hence, plants have evolved a large collection of plasma membrane-resident receptors, including RECEPTOR-LIKE KINASEs(RLKs) and RECEPTOR-LIKE PROTEINs(RLPs) to perceive those signals and regulate plant growth,development, and immunity. The ability of RLKs and RLPs to recognize distinct ligands relies on diverse categories of extracellular domains evolved. Co-regulatory receptors are often required to associate with RLKs and RLPs to facilitate cellular signal transduction. RECEPTOR-LIKE CYTOPLASMIC KINASEs(RLCKs) also associate with the complex, bifurcating the signal to key signaling hubs, such as MITOGEN-ACTIVATED PROTEIN KINASE(MAPK) cascades, to regulate diverse biological processes. | 6 | Two Arabidopsis Receptor-like Cytoplasmic Kinases SZE1 and SZE2 Associate with the ZAR1-ZED1 Complex and Are Required for Effector-Triggered Immunity显示文摘Plants utilize intracellular nucleotide-binding leucine-rich repeat domain-containing receptors (NLRs) to recognize pathogen effectors and induce a robust defense response named effector-triggered immunity (ETI). The Arabidopsis NLR protein HOPZ-ACTIVATED RESISTANCE 1 (ZAR1) forms a precomplex with HOPZ-ETI-DEFICIENT 1 (ZED1),a receptor-like cytoplasmic kinase (RLCK) XII-2 subfamily member, to recognize the Pseudomonas syringae effector HopZ1 a. We previously described a dominant mutant of Arabi-dopsis ZED1, zed1-D, which displays temperature-sensitive autoimmunity in a ZAR1-dependent manner. Here, we report that the RLCKs SUPPRESSOR OF ZED1-D1 (SZE1) and SZE2 associate with the ZAR1-ZED1 complex and are required for the ZED7-D-activated autoimmune response and HopZ1a-triggered immunity. We show that SZE1 but not SZE2 has autophosphorylation activity, and that the N-terminal myristoylation of both SZE1 and SZE2 is critical for their plasma membrane localization and ZED1-D-activated autoimmunity. Furthermore, we demonstrate that SZE1 and SZE2 both interact with ZAR1 to form ja functional complex and are required for resistance against P. syringae pv. tomato DC3000 ex-pressing HopZIa. We also provide evidence that SZE1 and SZE2 interact with HopZ1a and function together with ZED1 to change the intramolecular interactions of ZAR1, leading to its activation.
