1Actively priming autophagic cell death with novel transferrin receptor-targeted nanomedicine for synergistic chemotherapy against breast cancer显示文摘Recently, considerable attention in the field of cancer therapy has been focused on the mammalian rapamycin target(m TOR), inhibition of which could result in autophagic cell death(ACD). Though novel combination chemotherapy of autophagy inducers with chemotherapeutic agents is extensively investigated, nanomedicine-based combination therapy for ACD remains in infancy. In attempt to actively trigger ACD for synergistic chemotherapy, here we incorporated autophagy inducer rapamycin(RAP) into 7 pep-modified PEG-DSPE polymer micelles(7 pep-M-RAP) to specifically target and efficiently priming ACD of MCF-7 human breast cancer cells with high expression of transferrin receptor(Tf R). Cytotoxic paclitaxel(PTX)-loaded micelle(7 pep-M-PTX) was regarded as chemotherapeutic drug model. We discovered that with superior intracellular uptake in vitro and more tumor accumulation of micelles in vivo, 7 pep-M-RAP exhibited excellent autophagy induction and synergistic antitumor efficacy with 7 pep-M-PTX. Mechanism study further revealed that 7 pep-M-RAP and 7 pep-MPTX used in combination provided enhanced efficacy through induction of both apoptosis-and mitochondria-associated autophagic cell death. 2Phospholipase C Delta 3 inhibits apoptosis and promotes proliferation,migration,and invasion of thyroid cancer cells via Hippo pathway显示文摘In recent decades,the incidence of thyroid cancer(TC)has rapidly increased,leading us to explore the complex underlying mechanisms.We identified the gene Phospholipase C Delta 3(PLCD3)as a potential oncogene in TC by conducting the whole transcriptome sequencing.Our study is to understand the oncogenic role of PLCD3 in TC.We verified the overexpression of PLCD3 in TC from The Cancer Genome Atlas,Gene Expression Omnibus databases,and a locally validated cohort.Clinical correlation analysis showed that PLCD3 expression was related to histological type,T stage,lymph node metastasis(LNM),and disease stage.The high expression of PLCD3 could be a distinguishing factor for TC and its LNM.The biological function was examined using small interfering RNA-transfected TC cell lines.Silenced PLCD3 could inhibit colony formation,migration,and invasion ability and promote apoptosis of TC cell lines.PLCD3 silencing reversed the epithelial-mesenchymal transition but induced the apoptotic progress.Further exploration revealed that PLCD3 might be associated with critical genes of the Hippo pathway.The expressions of RHOA,YAP1/TAZ,and their downstream targets were decreased significantly when PLCD3 was down-regulated.YAP1 overexpression rescued the tumor-suppressive effect caused by PLCD3 silencing.This study demonstrates that PLCD3 is an oncogene that supports tumorigenesis and progression in TC,and PLCD3 may be a potential target gene for TC treatment.Lizhi Lin Jialiang Wen Bangyi Lin Hao Chen Adheesh Bhandari Yufeng Qi Danni Zheng Ouchen Wang 2021Acta Biochimica et Biophysica Sinica2021,53,4:4
3Genome-wide analysis identified 17 new loci influencing intraocular pressure in Chinese population显示文摘Intraocular pressure(IOP) is a major risk factor for glaucoma. Genetic determinants of intraocular pressure can provide critical insights into the genetic architecture of glaucoma and, as a result, open new avenues for therapeutic intervention. We performed a genome-wide association study and replication analysis of 8,552 Chinese participants. In the genome-wide association study, we identified 51 loci that surpassed the significance of P<9×10^(-7), and we formally replicated these loci. A combined discovery and replication meta-analysis identified 21 genome-wide loci that surpassed the genome-wide significance of P<5×10^(-8), including 4 previously reported loci: rs145063132(7 p21.2, ETV1/DGKB), rs548030386(7 q31.2, ST7 near CAV1/CAV2), rs7047871(9 p24.2, GLIS3), and rs2472494(9 q31.1, ABCA1/SLC44 A1). Of the 17 newly identified loci, five were reported to have ocular related phenotypes: PTCH2(rs7525308 in 1 p34.1), LRIF1/DRAM2(rs1282146 in 1 p13.3), COLEC11(rs201143466 in 2 p25.3),SPTBN1(rs4514918 in 2 p16.2), and CRK(rs11078446 in 17 p13.3). 4Electromagnetic and microwave absorption properties of Er-Ho-Fe alloys显示文摘The Er_xHo_(2-x)Fe_(17)(x = 0.0, 0.1,0.2, 0.3, 0.4) powders were prepared by arc melting and high energy ball milling method. The influence of the Er substitution on phase structure, morphology, saturation magnetization,electromagnetic parameters was investigated by X-ray diffraction(XRD),scanning electron microscopy(SEM), vibrating sample magnetometer(VSM) and vector network analyzer(VNA),respectively. The results show that the saturation magnetization increases and the average particle size increases with the increase of Er content. The minimum absorption peak frequency shifts towards a lower frequency region with the increase of Er content. The Er_(0.3)Ho_(1.7)Fe_(17) powder can achieve the minimum RL of-24.07 dB at 6.96 GHz with a thickness of 2.0 mm and the minimum RL is less than-20 dB at the thickness range from 2.0 to 3.0 mm. The minimum RL of Er_(0.3)Ho_(1.7)Fe_(17) is-37.26 dB at5.68 GHz and the frequency bandwidth of R<-10 dB reaches about 1.2 GHz with a thickness of 2.4 mm.And the microwave absorbing properties of the composite with different weight ratios of Er_(0.3)Ho_(1.7)Fe_(17)/graphene were researched. The microwave absorbing peaks of the composite shift to lower frequency with the increase of graphene content. 7Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. 9Visual information quantification for object recognition and retrieval显示文摘The rapid development of computer vision has led to an increasing amount of 3 D data,such as multiple views and point clouds,which are widely used in 3 D object recognition and retrieval.Intuitively,the quality of 3 D data is the most crucial factor that directly affects the performance of 3 D applications.However,how to evaluate the 3 D data quality,especially the multi-view data quality,is still an open question.To tackle this issue,we propose an entropy-based multi-view information quantification model(MV-Info model)to quantitatively evaluate the multi-view data information.Our proposed MV-Info model consists of hierarchical data module,feature generation module,and quantitative calculation module.Besides,it considers the information entropy theory for more reasonable quantification results.In our method,how much information we can observe from a group of views can be quantified,which can be used to support 3 D recognition and retrieval.We also designed a series of experiments to evaluate the effectiveness of the proposed model.The experimental results demonstrate the rationality and validity of the proposed model.CHENG JiaLiang BIE Lin ZHAO XiBin GAO Yue 2021Science China(Technological Sciences)2021,64,12:1

11Preliminary study on the safety and efficacy of a new polymyxin B-immoblized resin column in treatment of LPS-induced sepsis beagles显示文摘Background:This study aims to assess the safety and efficacy of direct hemoperfu-sion using a new polymyxin B-immobilized resin column(disposable endotoxin ad-sorber,KCEA)in an endotoxin/lipopolysaccharide(LPS)-induced sepsis model.Methods:Eighteen beagles were randomized into 1 intervention group(KCEA group,n=6)and 2 control groups(sham group and model group,n=6 each).Sepsis was in-duced by continuous intravenous application of 0.5 mg/kg body weight of endotoxin for 60 min.An extracorporeal hemoperfusion device made with KCEA for endotoxin adsorption was used.Model group beagles received standard treatment with fluids and vasoactive drugs,KCEA group beagles received standard treatment and direct hemoperfusion of KCEA for 2 h,and sham group beagles were treated with standard treatment and direct hemoperfusion of a sham column for 2 h.Results:Good blood compatibility of KCEA was confirmed by assessing clinical pa-rameters.Blood endotoxin peak levels in the KCEA group were significantly lower,resulting in a significant suppression of IL-6,TNF-αand procalcitonin,which improved mean arterial pressure and significantly lowered vasopressor demand,thereby pro-tecting organ function and improving survival time and rate.In the KCEA group,MAP was significantly higher over 6 h than those recorded both in the sham group and model group.The 7-day survival rates of the KCEA,sham and model groups were 50%,0%and 0%,respectively.Conclusion:KCEA hemoadsorption was effective at detoxifying circulatory endotoxin and inflammatory mediators and contributed to the decreased mortality rate in the sepsis beagles.Yonggui Li Zhenggen Yang Jialiang Hu Zhennan Lin 2022Animal Models and Experimental Medicine2022,5,2:1
12Anti-PD-L1 antibody enhances curative effect of cryoablation via antibody-dependent cell-mediated cytotoxicity mediating PD-L1^(high)CD11b^(+)cells elimination in hepatocellular carcinoma显示文摘Cryoablation(CRA)and microwave ablation(MWA)are two main local treatments for hepatocellular carcinoma(HCC).However,which one is more curative and suitable for combining with immunotherapy is still controversial.Herein,CRA induced higher tumoral PD-L1 expression and more T cells infiltration,but less PD-L1^(high)CD11b^(+)myeloid cells infiltration than MWA in HCC.Furthermore,CRA had better curative effect than MWA for anti-PD-L1 combination therapy in mouse models.Mechanistically,anti-PD-L1 antibody facilitated infiltration of CD8^(+)T cells by enhancing the secretion of CXCL9 from cDC1 cells after CRA therapy.On the other hand,anti-PD-L1 antibody promoted the infiltration of NK cells to eliminate PD-L1^(high)CD11b^(+)myeloid cells by antibody-dependent cell-mediated cytotoxicity(ADCC)effect after CRA therapy.Both aspects relieved the immunosuppressive microenvironment after CRA therapy.Notably,the wild-type PD-L1 Avelumab(Bavencio),compared to the mutant PD-L1 atezolizumab(Tecentriq),was better at inducing the ADCC effect to target PD-L1^(high)CD11b^(+)myeloid cells.Collectively,our study uncovered the novel insights that CRA showed superior curative effect than MWA in combining with anti-PD-L1 antibody by strengthening CTL/NK cell immune responses,which provided a strong rationale for combining CRA and PD-L1 blockade in the clinical treatment for HCC.Jizhou Tan Ting Liu Wenzhe Fan Jialiang Wei Bowen Zhu Yafang Liu Lingwei Liu Xiaokai Zhang Songling Chen Haibiao Lin Yuanqing Zhang Jiaping Li 2023Acta Pharmaceutica Sinica B2023,13,2:0
13Integrative analysis of gene expression profiles reveals specific signaling pathways associated with pancreatic duct adenocarcinoma显示文摘Background:Pancreatic duct adenocarcinoma(PDAC)remains a major health problem because conventional can-cer treatments are relatively ineffective against it.Microarray studies have linked many genes to pancreatic cancer,but the available data have not been extensively mined for potential insights into PDAC.This study attempted to identify PDAC-associated genes and signaling pathways based on six microarray-based profiles of gene expression in pancre-atic cancer deposited in the gene expression omnibus database.Methods:Pathway network methods were used to analyze core pathways in six publicly available pancreatic cancer gene(GSE71989,GSE15471,GSE16515,GSE32676,GSE41368 and GSE28735)expression profiles.Genes potentially linked to PDAC were assessed for potential impact on survival time based on data in The Cancer Genome Atlas and International Cancer Genome Consortium databases,and the expression of one candidate gene(CKS2)and its association with survival was examined in 102 patients with PDAC from our hospital.Effects of CKS2 knockdown were explored in the PDAC cell lines BxPC-3 and CFPAC-1.Results:The KEGG signaling pathway called“pathway in cancer”may play an important role in pancreatic cancer development and progression.Five genes(BIRC5,CKS2,ITGA3,ITGA6 and RALA)in this pathway were significantly associated with survival time in patients with PDAC.CKS2 was overexpressed in PDAC samples from our hospital,and higher CKS2 expression in these patients was associated with shorter survival time.CKS2 knockdown substantially inhibited PDAC cell proliferation in vitro.Conclusions:Analysis integrating existing microarray datasets allowed identification of the“pathway in cancer”as an important signaling pathway in PDAC.This integrative approach may be powerful for identifying genes and pathways involved in cancer.Jun Li Wenle Tan Linna Peng Jialiang Zhang Xudong Huang Qionghua Cui Jian Zheng Wen Tan Chen Wu Dongxin Lin 2018Cancer Communications2018,38,1:0

16Prediction of network public opinion based on bald eagle algorithm optimized radial basis function neural network显示文摘Purpose-In the new era of highly developed Internet information,the prediction of the development trend of network public opinion has a very important reference significance for monitoring and control of public opinion by relevant government departments.Design/methodology/approach-Aiming at the complex and nonlinear characteristics of the network public opinion,considering the accuracy and stability of the applicable model,a network public opinion prediction model based on the bald eagle algorithm optimized radial basis function neural network(BES-RBF)is proposed.Empirical research is conducted with Baidu indexes such as“COVID-19”,“Winter Olympic Games”,“The 100th Anniversary of the Founding of the Party”and“Aerospace”as samples of network public opinion.Findings-The experimental results show that the model proposed in this paper can better describe the development trend of different network public opinion information,has good stability in predictive performance and can provide a good decision-making reference for government public opinion control departments.Originality/value-A method for optimizing the central value,weight,width and other parameters of the radial basis function neural network with the bald eagle algorithm is given,and it is applied to network public opinion trend prediction.The example verifies that the prediction algorithm has higher accuracy and better stability.Jialiang Xie Shanli Zhang Ling Lin 2022International Journal of Intelligent Computing and Cybernetics2022,15,2:0
17Efficient heterogeneous integration of InP/Si and GaSb/Si templates with ultra-smooth surfaces显示文摘Heterogeneous integration of InP and GaS b on Si substrates holds a huge potential interest in near-infrared and mid-infrared optoelectronic devices.In this study,2-inch 180-nm-thick InP and 185-nmthick GaS b thin layers were successfully transferred onto the Si substrates to form high-quality and ultrasmooth InP/Si and GaS b/Si templates using molecular beam epitaxy(MBE)and the ion-slicing technique together with selective chemical etching.The relocation of the implantation-introduced damage in the sacrificial layer enables the transfer of relatively defect-free InP and GaS b thin films.The sacrificial layers were completely etched off by selective chemical etching,leaving ultra-smooth epitaxial surfaces with a roughness of 0.2 nm for the InP/Si template and 0.9 nm for the GaS b/Si template,respectively.Thus,the chemical mechanical polishing(CMP)process was not required to smooth the surface which usually introduces particles and chemical contaminations on the transferred templates.Furthermore,the donor substrate is not consumed and can be recycled to reduce the cost,which provides a paradigm for the sustainable and economic development of the Si integration platform.Tingting JIN Jiajie LIN Tiangui YOU Xiaolei ZHANG Hao LIANG Yifan ZHU Jialiang SUN Hangning SHI Chaodan CHI Min ZHOU Robert KUDRAWIEC Shumin WANG Xin OU 2022Science China(Information Sciences)2022,65,8:0
18Supramolecular Self-assembly Behaviors of Asymmetric Diblock Copolymer Blends with Hydrogen Bonding Interactions between Shorter Blocks Modelled by Yukawa Potentials显示文摘We employed the extended self-consistent field theory to investigate the supramolecular self-assembly behaviors of asymmetric diblock copolymer blends(AB/B’C)with hydrogen bonding interactions between shorter B and B’blocks.The hydrogen bonding interactions are described by Yukawa potentials,where the hydrogen bonding donors and acceptors were modelled as two blocks smeared with opposite screened charges.The hierarchical microstructures with parallelly packed lamellae-in-lamellae(Lam)and 4.8.8 Archimedean tilting pattern(4.8.8)were observed at lower and higher hydrogen bonding density(θ),respectively.The hierarchy of Lam and 4.8.8 were demonstrated by the one-and two-dimensional density profiles and the underlying order of the large-length-scale and small-length-scale microstructures were also clarified.It was found that the 4.8.8 is favorable to the stronger hydrogen bonding density or interactions.Asθincreases,the microphase transition from Lam to 4.8.8 occurs atθ=0.34,which is mainly attributed to the optimization of the electrostatic energy and conformational entropy with sacrificing the interfacial energy.This work can provide a new strategy to understand the supramolecular self-assembly as well as the mechanism behind the formation of complex hierarchical microstructures.Xu Zhang Jialiang Chen Lin Xu Tianxi Liu 2021Chinese Journal of Polymer Science2021,39,11:0
