2Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 3Retrieval and Analysis of Coal Fire Temperature in Wuda Coalfield, Inner Mongolia, China显示文摘Coal fire burning around the world is an environmental catastrophe characterized by the emission of noxious gases, particulate matter, and condensation by-products. In this study, coal fire temperature is retrieved based on Landsat 5 TM images and Generalized Single-Channel Algorithm (GSCA), in Wuda coalfield, Inner Mongolia, China. Then coal fire zones are extracted by Jenks′ natural breaks and threshold methods based on temperature images. Changes of coal fire zones are analyzed from 1989 to 2008. The results are summarized as follows: 1) The coal fire temperature retrieval method based on Landsat 5 TM and the GSCA model is effective and feasible, because the temperature error is relatively small (from –2.9℃ to +2.6℃) between the measured temperature and the retrieved temperature. 2) The accuracy is relatively high to extract coal fire zones through the Jenks′ natural breaks and threshold methods, because 83.56% of surveyed area is located in the coal fire zones extracted in 2005. 3) The coal fire area increased 9.81 × 10 5 m 2 from 1989 to 2005, and the annual growth is about 6.1 × 10 4 m 2 , with an annual increasing rate of 2.48%. 4Mining the key regulatory genes of chicken inosine 5'-monophosphate metabolism based on time series microarray data显示文摘IMP(inosine 5'-monophosphate) is a compound that enhances the flavor of poultry meat.IMP has become a new breeding trait to improve poultry meat quality.We tried to identify several potential regulatory genes,and construct their predicted regulatory relationships.Time series gene expression profiles of thigh muscle tissues of Rugao chicken,a famous indigenous breed in China,were performed for analysis of genes that are co-expressed or correlated with the concentration of IMP.We found 15 crucial co-expression genes,which are Hspa2,Pten,Gabpa,Bpi,Mkl1,Srf,Cd34,Hspa4,Etv6,Bmpr2,Gde1,Igfbp5,Cd28,Pecam1 and Gja1,that may directly or indirectly regulate IMP metabolism.Eventually,we computed the correlation coefficient between 19 IMP Genes and 15 CGs(15co-expression genes),and we identified and constructed a predicted regulation network.In conclusion,variation of IMP concentration was primarily connected with the muscle development process.During this process,15 CGs were identified that may have significant influence on IMP metabolism.In particular,Bmpr2,Pten and co-expression genes correlated with Entpd8 might play important roles in regulating IMP de novo synthesis,decomposition and salvage synthesis.Teng Ma Lu Xu Hongzhi Wang Jing Chen Lu Liu Guobin Chang Guohong Chen 2015Journal of Animal Science and Biotechnology2015,6,3:6

12Exponential isothermal amplification coupled MALDI-TOF MS for microRNAs detection显示文摘Micro RNAs(mi RNAs) have attracted significant attention in biomedical research and clinical diagnosis.However, due to their inherent characteristics of low abundance and the high complexity of corresponding biological matrices, simultaneous detection of multiple mi RNAs at low abundance is still a challenge.In this work, a method coupling exponential amplification reaction(EXPAR) with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS) is developed for label-free and simultaneous detection of multiple mi RNAs. The assay can be performed under isothermal conditions in a single reaction tube, and finished in less than 30 min. It exhibits good quantification ability and with attomolar-level sensitivity for mi RNAs detection. It also shows high specificity to distinguish mi RNAs at single-nucleotide resolution. We used the method to detect the mi RNA-21, let-7a, mi RNA-100, and mi RNA-125b in samples of spiked human serum and breast cancer cells(i.e., MCF-7, MDA-MB-231 and SK-BR-3). 15Ratiometric fluorescence immunoassay of SARS-CoV-2 nucleocapsid protein via Si-FITC nanoprobe-based inner filter effect显示文摘The global pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)virus has necessitated rapid,easy-to-use,and accurate diagnostic methods to monitor the virus infection.Herein,a ratiometric fluorescence enzyme-linked immunosorbent assay(ELISA)was developed using Si-fluorescein isothiocyanate nanoparticles(FITC NPs)for detecting SARSCoV-2 nucleocapsid(N)protein.Si-FITC NPs were prepared by a one-pot hydrothermal method using 3-aminopropyl triethoxysilane(APTES)-FITC as the Si source.This method did not need post-modification and avoided the reduction in quantum yield and stability.The p-nitrophenyl(pNP)produced by the alkaline phosphatase(ALP)-mediated hydrolysis of pnitrophenyl phosphate(pNPP)could quench Si fluorescence in Si-FITC NPs via the inner filter effect.In ELISA,an immunocomplex was formed by the recognition of capture antibody/N protein/reporter antibody.ALP-linked secondary antibody bound to the reporter antibody and induced pNPP hydrolysis to specifically quench Si fluorescence in Si-FITC NPs.The change in fluorescence intensity ratio could be used for detecting N protein,with a wide linearity range(0.01-10.0 and 50-300 ng/mL)and low detection limit(0.002 ng/mL).The concentration of spiked SARS-CoV-2 N protein could be determined accurately in human serum.Moreover,this proposed method can accurately distinguish coronavirus disease 2019(COVID-19)and non-COVID-19 patient samples.Therefore,this simple,sensitive,and accurate method can be applied for the early diagnosis of SARS-CoV-2 virus infection.Guobin Mao Yang Yang Shijie Cao Silu Ye Yifang Li Wei Zhao Hongwei An Yingxia Liu Junbiao Dai Yingxin Ma 2023Nano Research2023,16,4:0

17Phase Characterization and Formation Behavior in 6 wt%Si High-silicon Austenitic Stainless Steel during Isothermal Aging显示文摘The precipitation behavior of different phases in a high-silicon stainless steel(6 wt%Si)during aging at 600–1050℃for24 h was investigated.The morphology,crystal structure and composition of various precipitates were detailly characterized using optical microscopy,scanning electron microscopy and transmission electron microscopy.Four phases were mainly identified:χ-phase,M_6C carbides,σphase and a new type of fcc-phase.During aging at 600–900℃,the main precipitate was(Cr,Mo and Si)-richχ-phase which was directly precipitated fromγmatrix.Theχ-phase was calibrated as bcc structure with a lattice parameter of 8.90?.The peak temperature for the precipitation ofχ-phase was 800℃,and it was dissolved when aging at temperatures above 1000℃.Theσ-phase was observed only at 700℃and grew next toχ-phase.Above 700℃,a new fcc-phase was found to be precipitated along withχ-phase,with a space group of Fd3c and a lattice parameter of 12.56?.The M_6C carbides started to be precipitated at 700℃in the vicinity ofχ-phase.And its amount basically increased with the increasing of temperature.An orientation relationship between M_6C/γwas found:[100]c//[100]γ,(001)c//(001)γ,i.e.,the cube-on-cube relationship.Sihan Chen Tian Liang Yangtao Zhou Weiwei Xing Chengwu Zheng Yingche Ma JinMing Wu Guobin Li Kui Liu 2021Acta Metallurgica Sinica(English Letters)2021,34,5:0

19Multiferroic oxide BFCNT/BFCO heterojunction black silicon photovoltaic devices显示文摘Multiferroics are being studied increasingly in applications of photovoltaic devices for the carrier separation driven by polarization and magnetization.In this work,textured black silicon photovoltaic devices are fabricated with Bi_(6)Fe_(1.6)Co_(0.2)Ni_(0.2)Ti_(3)O_(18)/Bi_(2)FeCrO_(6)(BFCNT/BFCO)multiferroic heterojunction as an absorber and graphene as an anode.The structural and optical analyses showed that the bandgap of Aurivillius-typed BFCNT and double perovskite BFCO are 1.62±0.04 eV and 1.74±0.04 eV respectively,meeting the requirements for the active layer in solar cells.Under the simulated AM 1.5 G illumination,the black silicon photovoltaic devices delivered a photoconversion efficiency(η)of 3.9%with open-circuit voltage(Voc),short-circuit current density(Jsc),and fill factor(FF)of 0.75 V,10.8 mA cm^(-2),and 48.3%,respectively.Analyses of modulation of an applied electric and magnetic field on the photovoltaic properties revealed that both polarization and magnetization of multiferroics play an important role in tuning the built-in electric field and the transport mechanisms of charge carriers,thus providing a new idea for the design of future high-performance multiferroic oxide photovoltaic devices.Kaixin Guo Xu Wang Rongfen Zhang Zhao Fu Liangyu Zhang Guobin Ma Chaoyong Deng 2021Light(Science & Applications)2021,10,11:0
20Inducing Fe 3d Electron Delocalization and Spin‑State Transition of FeN_(4) Species Boosts Oxygen Reduction Reaction for Wearable Zinc–Air Battery显示文摘Transition metal-nitrogen-carbon materials(M-N-Cs),particularly Fe-N-Cs,have been found to be electroactive for accelerating oxygen reduction reaction(ORR)kinetics.Although substantial efforts have been devoted to design Fe-N-Cs with increased active species content,surface area,and electronic conductivity,their performance is still far from satisfactory.Hitherto,there is limited research about regulation on the electronic spin states of Fe centers for Fe-N-Cs electrocatalysts to improve their catalytic performance.Here,we introduce Ti_(3)C_(2) MXene with sulfur terminals to regulate the electronic configuration of FeN_(4) species and dramatically enhance catalytic activity toward ORR.The MXene with sulfur terminals induce the spin-state transition of FeN_(4) species and Fe 3d electron delocalization with d band center upshift,enabling the Fe(II)ions to bind oxygen in the end-on adsorption mode favorable to initiate the reduction of oxygen and boosting oxygen-containing groups adsorption on FeN_(4) species and ORR kinetics.The resulting FeN_(4)-Ti_(3)C_(2)Sx exhibits comparable catalytic performance to those of commercial Pt-C.The developed wearable ZABs using FeN_(4)-Ti_(3)C_(2)Sx also exhibit fast kinetics and excellent stability.This study confirms that regulation of the electronic structure of active species via coupling with their support can be a major contributor to enhance their catalytic activity.Shengmei Chen Xiongyi Liang Sixia Hu Xinliang Li Guobin Zhang Shuyun Wang Longtao Ma Chi‑Man Lawrence Wu Chunyi Zhi Juan Antonio Zapien 2023Nano-Micro Letters2023,15,4:0
