1Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. 3Vaccination of poultry successfully eliminated human infection with H7N9 virus in China显示文摘The H7 N9 viruses that emerged in China in 2013 were nonpathogenic in chickens but mutated to a highly pathogenic form in early 2017 and caused severe disease outbreaks in chickens. The H7 N9 influenza viruses have caused five waves of human infection, with almost half of the total number of human cases(766 of 1,567) being reported in the fifth wave, raising concerns that even more human infections could occur in the sixth wave. In September 2017, an H5/H7 bivalent inactivated vaccine for chickens was introduced, and the H7 N9 virus isolation rate in poultry dropped by 93.3% after vaccination. More importantly,only three H7 N9 human cases were reported between October 1, 2017 and September 30, 2018, indicating that vaccination of poultry successfully eliminated human infection with H7 N9 virus. 4Effect of Root Architecture on Structural Stability and Erodibility of Topsoils during Concentrated Flow in Hilly Loess Plateau显示文摘Traditional vegetation techniques for the control of concentrated flow erosion are widely recognized, whereas only a few studies have experimentally investigated the impacts of belowground roots on the erodibility of topsoils in semi-arid areas. To quantify the effects of root architectures on soil erodibility and its relevant structural properties, simulated flow experiments were conducted at six-week intervals from 18 July to 20 October in 2012 in the hilly Loess Plateau. Five treatments were: 1) bare(control), 2) purple alfalfa(Medicago sativa), representing tap roots(T), 3) switchgrass(Panicum virgatum), representing fibrous roots(F), 4) purple alfalfa and switchgrass, representing both tap and fibrous roots(T + F), and 5) natural recovery(N). For each treatment, soil structural properties and root characteristics were measured at an interval of six weeks. Soil anti-scouribility was calculated. Results showed that grass planting slightly reduced soil bulk density, but increased soil aggregate content by 19.1%, 10.6%, 28.5%, and 41.2% in the treatments T, F, T + F, and N, respectively. Soil shear strength(cohesion and angle of internal friction(φ)) significantly increased after the grass was planted. As roots grew, soil cohesion increased by 115.2%–135.5%, while soil disintegration rate decreased by 39.0%–58.1% in the 21 th week compared with the recorded value in the 9th week. Meanwhile, root density and root surface area density increased by 64.0%–104.7% and 75.9%–157.1%, respectively. No significant differences in soil anti-scouribility were observed between the treatments of T and F or of T + F and N, but the treatments of T + F and N performed more effectively than T or F treatment alone in retarding concentrated flow. Soil aggregation and root surface-area density explained the observed soil anti-scouribility during concentrated flow well for the different treatments. 5Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 7Distribution and drug resistance of pathogenic bacteria isolated from cancer hospital in 2013显示文摘Objective: To understand distribution and drug resistance of pathogenic bacteria from a specialized cancer hospital in 2013 in order to provide a basis for rational clinical antimicrobial agents. Methods: Pathogenic bacteria identification and drug sensitivity tests were performed with a VITEK 2 compact automatic identification system and data were analyzed using WHONET5.6 software.Results: Of the 1,378 strains tested, 980 were Gram-negative bacilli, accounting for 71.1%, in which Klebsiella pneumonia, Escherichia coli and Pseudomonas aeruginosa were the dominant strains. We found 328 Gram-positive coccus, accounting for 23.8%, in which the amount of Staphylococcus aureus was the highest. We identified 46 fungi, accounting for 4.1%. According to the departmental distribution within the hospital, the surgical departments isolated the major strains, accounting for 49.7%. According to disease types, lung cancer, intestinal cancer and esophagus cancer were the top three, accounting for 20.9%, 17.3% and 14.2%, respectively. 8Water in the Mogao Grottoes,China:where it comes from and how it is driven显示文摘The Dunhuang Mogao Grottoes in China was designated as a world heritage site by UNESCO in 1987 and is famous for its cultural relics. Water is the most active factor that harms the relics in the caves as it damages the grotto murals and painted sculptures. Thus, determining the water sources and driving forces of water movement is a key issue for protecting these cultural relics. These issues have troubled relics protectors for a long time. In this study, the authors chose a representative cave in the Mogao Grottoes and, by completely sealing the cave to make a closed system, measured the water vapor from the surrounding rock. This was accomplished by installing a condensation–dehumidification temperature–humidity control system for the collection of water vapor. The results show that there is continuous evaporation from the deep surrounding rock into the cave. The daily evaporation capacity is determined to be 1.02 g/(d?m2). The water sources and driving forces of water movement were further analyzed according to the character of the water evaporation and by monitoring the temperature and humidity of the surrounding rock. It was found that the water vapor in the cave derives from phreatic water. Moreover, the yearly fluctuation of temperature in the surrounding rock and geothermal forces are the basic powers responsible for driving phreatic evaporation. Under the action of the yearly temperature fluctuations, decomposition and combination of bound water acts as a 'pump' that drives phreatic water migration and evaporation. When the temperature rises, bound water decomposes and evaporates; and when it falls, the rock absorbs moisture. This causes the phreatic water to move from deep regions to shallow ones. 9Fine Mapping of RppP25, a Southern Rust Resistance Gene in Maize显示文摘Southern rust (Puccinia polysora Underw.) is a major disease that can cause severe yield losses in maize (Zea mays L.). In our previous study, a major gene RppP25 that confers resistance to southern rust was identified in inbred line P25. Here, we report the fine mapping and candidate gene analysis of RppP25 from the near-isogenic line F939, which harbors RppP25 in the genetic background of the susceptible inbred line F349. The inheritance of resistance to southern rust was investigated in the BC1F1 and BC3F1 populations, which were derived from a cross between F939 and F349 (as the recurrent parent). The 1:1 segregation ratio of resistance to susceptible plants in these two populations indicated that the resistance is controlled by a single dominant gene. Ten markers, including three simple sequence repeat (SSR) markers and seven insertion/deletion (InDel) markers, were developed in the RppP25 region. RppP25 was delimited to an interval between P091 and M271, with an estimated length of 40 kb based on the physical map of B73. In this region, a candidate gene was identified that was predicted to encode a putative nucleotide-binding site leucine-rich repeat (NBS-LRR) protein. 10Recent Changes in Precipitation Extremes in the Heihe River Basin,Northwest China显示文摘Changes in rainfall extremes pose a serious and additional threat to water resources planning and management,natural and artificial oasis stability,and sustainable development in the fragile ecosystems of arid inland river basins. In this study,the trend and temporal variation of extreme precipitation are analyzed using daily precipitation datasets at 11 stations over the arid inland Heihe River basin in Northwest China from 1960 to 2011. Eight indices of extreme precipitation are studied.The results show statistically significant and large-magnitude increasing and decreasing trends for most indices,primarily in the Qilian Mountains and eastern Hexi Corridor. More frequent and intense rainfall extremes have occurred in the southern part of the desert area than in the northern portion. In general,the temporal variation in precipitation extremes has changed throughout the basin. Wet day precipitation and heavy precipitation days show statistically significant linear increasing trends and step changes in the Qilian Mountains and Hexi Corridor. Consecutive dry days have decreased obviously in the region in most years after approximately the late 1980 s,but meanwhile very long dry spells have increased,especially in the Hexi Corridor. The probability density function indicates that very long wet spells have increased in the Qilian Mountains. The East Asian summer monsoon index and western Pacific subtropical high intensity index possess strong and significant negative and positive correlations with rainfall extremes,respectively. 11Application of the oblique ionogram as vertical ionogram显示文摘The vertical ionogram can provide the important ionospheric parameters, such as critical frequency, virtual height and electron density, for ionospheric research. The oblique ionosonde has the ability to detect the ionosphere over sea and other terrain where it is not practical to deploy vertical sounder and provide more ionograms with less transmitting and receiving devices. Therefore, the conversion of the oblique ionogram to vertical ionogram for obtaining the important ionospheric parameters is a very useful inversion technology. The experimental comparison between oblique and vertical detections was carried out in the equatorial ionospheric anomaly (EIA) region of south China on 25 and 26 August 2010. The oblique detecting path was from Wuhan to Shenzhen and the VI ionosonde was located in the midpoint of the oblique path. 12Reaction thermodynamics and kinetics on in situ Al_2O_3/Cu composites显示文摘The preferred internal oxidation of aluminum in Cu Al alloy was used to obtain in situ Al 2O 3/Cu composites. The reinforcement particles were mainly γ Al 2O 3, some θ Al 2O 3 and a little α Al 2O 3. Thermodynamics analyses show that the chemical reactions are 3Cu 2O+2Al=6Cu+Al 2O 3 or 3CuO+2Al=3Cu+Al 2O 3. A related equilibrium diagram was drawn. 13Diagnostic value of negative enrichment and immune fluorescence in situ hybridization for intraperitoneal free cancer cells of gastric cancer显示文摘Objective:To explore the intraperitoneal free cancer cell(IFCC)detection value of negative enrichment and immune fluorescence in situ hybridization(NEimFISH)on chromosomes(CEN)8/17.Methods:To verify the reliability of NEimFISH,29 gastric cancer tumors,their adjacent tissues and greater omental tissues were tested.Our study then included 105 gastric cancer patients for IFCC.We defined patients as IFCC-positive if a signal was detected,regardless of the detailed cancer cell numbers.A comparison of clinicopathological features was conducted among IFCC groups.We also compared the diagnosis value and peritoneal recurrence predictive value among different detection methods.The comparison of IFCC number was also conducted among different groups.Results:A cutoff of 2.5 positive cells could distinguish all benign tissue samples and 97%of malignant tissue samples in our study.Compared to intestinal gastric cancer,patients with diffuse gastric cancer tended to have more IFCCs(6 vs.4,P=0.002).The IFCC counts were often higher in the lymphovascular invasion positive group than negative group(3 vs.1,P=0.022).All IFCC samples that were considered positive using conventional cytology were also found to be positive using NEimFISH.When compared to conventional cytology and paraffin pathology,NEimFISH had a higher IFCC positive rate(68.9%)and higher one-year peritoneal recurrence predictive value with area under the curve(AUC)of 0.922.Conclusions:Gastric cancer could be effectively diagnosed by NEimFISH.The IFCC number found using NEimFISH on CEN8/17 is closely associated with Lauren type and vascular invasion of cancer.NEimFISH is a reliable detection modality with a higher positive detection rate,higher one-year peritoneal recurrence predictive value and quantitative features for IFCC of gastric cancer.Anqiang Wang Zhongwu Li Qian Wang Yali Bai Xin Ji Tao Fu Ke Ji Yanwen Xue Tingxu Han Xiaojiang Wu Ji Zhang Yingjie Yang Guobin Xu Zhaode Bu Jiafu Ji 2019Chinese Journal of Cancer Research2019,31,6:3
14Influenza A virus use of BinCARD1 to facilitate the binding of viral NP to importinα7 is counteracted by TBK1-p62 axis-mediated autophagy显示文摘As a major component of the viral ribonucleoprotein(vRNP)complex in influenza A virus(IAV),nucleoprotein(NP)interacts with isoforms of importinαfamily members,leading to the import of itself and vRNP complex into the nucleus,a process pivotal in the replication cycle of IAV.In this study,we found that BinCARD1,an isoform of Bcl10-interacting protein with CARD(BinCARD),was leveraged by IAV for efficient viral replication.BinCARD1 promoted the nuclear import of the vRNP complex and newly synthesized NP and thus enhanced vRNP complex activity.Moreover,we found that BinCARD1 interacted with NP to promote NP binding to importinα7,an adaptor in the host nuclear import pathway.However,we also found that BinCARD1 promoted RIG-I-mediated innate immune signaling by mediating Lys63-linked polyubiquitination of TRAF3,and that TBK1 appeared to degrade BinCARD1.We showed that BinCARD1 was polyubiquitinated at residue K103 through a Lys63 linkage,which was recognized by the TBK1-p62 axis for autophagic degradation.Overall,our data demonstrate that IAV leverages BinCARD1 as an important host factor that promotes viral replication,and two mechanisms in the host defense system are triggered—innate immune signaling and autophagic degradation—to mitigate the promoting effect of BinCARD1 on the life cycle of IAV.Xuyuan Wang Li Jiang Guangwen Wang Wenjun Shi Yuzhen Hu Bo Wang Xianying Zeng Guobin Tian Guohua Deng Jianzhong Shi Liling Liu Chengjun Li Hualan Chen 2022Cellular & Molecular Immunology2022,19,10:3
15Reverse Microemulsion Synthesis and Characterization of Pd-Ag Bimetallic Alloy Catalysts Supported on Al_2O_3 for Acetylene Hydrogenation显示文摘Pd-Ag bimetallic alloy nanoparticles were synthesized by the reverse microemulsion method,and then deposited on Al2O3 to form the supported catalyst.The nanoparticles of Pd-Ag and Pd-Ag/Al2O3 samples were characterized by UV/Vis,HRTEM,EDX,XRD,and XPS.The test results indicated that Pd-Ag bimetallic alloy nanoparticles with a size of about 2 nm and a face-centered cubic(fcc) structure were formed in the measured area of microemulsion.The growth of nanoparticles was effectively limited within the droplet of micoremulsion.TEM image exhibited that the Pd-Ag alloy nanoparticles were well-dispersed on the Al2O3 support.The catalytic performance of various catalysts for selective hydrogenation of acetylene showed that a higher acetylene conversion and selectivity to ethylene upon acetylene hydrogenation was achieved on a nano-sized Pd-Ag bimetallic catalyst with a Pd/Ag alloy supported molar ratio of 1:1.5.Wei Guobin Dai Wei Li Qian Cao Weiliang Zhang Jingchang 2012China Petroleum Processing & Petrochemical Technology2012,14,3:3
16H7N9 virus infection triggers lethal cytokine storm by activating gasdermin E-mediated pyroptosis of lung alveolar epithelial cells显示文摘The H7N9 influenza virus emerged in China in 2013,causing more than 1560 human infections,39% of which were fatal.A ‘cytokine storm’ in the lungs of H7N9 patients has been linked to a poor prognosis and death;however,the underlying mechanism that triggers the cytokine storm is unknown.Here,we found that efficient replication of the H7N9 virus in mouse lungs activates gasdermin E(GSDME)-mediated pyroptosis in alveolar epithelial cells,and that the released cytosolic contents then trigger a cytokine storm.Knockout of Gsdme switched the manner of death of A549 and human primary alveolar epithelial cells from pyroptosis to apoptosis upon H7N9 virus infection,and Gsdme knockout mice survived H7N9 virus lethal infection.Our findings reveal that GSDME activation is a key and unique mechanism for the pulmonary cytokine storm and lethal outcome of H7N9 virus infection and thus opens a new door for the development of antivirals against the H7N9 virus.Xiaopeng Wan Jiqing Li Yupeng Wang Xiaofei Yu Xijun He Jianzhong Shi Guohua Deng Xianying Zeng Guobin Tian Yanbing Li Yongping Jiang Yuntao Guan Chengjun Li Feng Shao Hualan Chen 2022National Science Review2022,9,1:3

18A chromosome-scale genome assembly of Artemisia argyi reveals unbiased subgenome evolution and key contributions of gene duplication to volatile terpenoid diversity显示文摘Artemisia argyi Le´vl.et Vant.,a perennial Artemisia herb with an intense fragrance,is widely used in traditional medicine in China and many other Asian countries.Here,we present a chromosome-scale genome assembly of A.argyi comprising 3.89 Gb assembled into 17 pseudochromosomes.Phylogenetic and comparative genomic analyses revealed that A.argyi underwent a recent lineage-specificwhole-genomeduplication(WGD)event after divergence fromArtemisia annua,resulting in two subgenomes.Wedeciphered the diploid ancestral genome of A.argyi,and unbiased subgenome evolution was observed.The recent WGD led to a large number of duplicated genes in the A.argyi genome.Expansion of the terpene synthase(TPS)gene family through various types of gene duplication may have greatly contributed to the diversity of volatile terpenoids in A.argyi.In particular,we identified a typical germacrene D synthase gene cluster within the expanded TPS gene family.The entire biosynthetic pathways of germacrenes,(+)-borneol,and(+)-camphor were elucidated in A.argyi.In addition,partial deletion of the amorpha-4,11-diene synthase(ADS)gene and loss of function of ADS homologs may have resulted in the lack of artemisinin production in A.argyi.Our study provides newinsights into the genome evolution of Artemisia and lays a foundation for further improvement of the quality of this important medicinal plant.Hongyu Chen Miaoxian Guo Shuting Dong Xinling Wu Guobin Zhang Liu He Yuannian Jiao Shilin Chen Li Li Hongmei Luo 2023Plant Communications2023,4,3:2
19In-situ Synthesis and Formation Mechanism ofTiN-Al_2O_3/Al Composite显示文摘The synthesis of TiN and Al2O3 by in situ injection of reactive nitrogen gas into molten Al alloys has been evaluated over the temperature range from 1000 to 1600℃. It is shown that TiN and Al2O3 can be formed in melt with nitrogen and surplus oxygen (in vacuum room of the induction furnace) as the reactive gases over 1000℃. Up to 2.1 wt pct Al2O3 and 5.2 wt pct of TiN in situ phases in an Al alloy has been formed in a range of size from 0.8 to 5 μm. 20Molecular cloning and expression patterns of the cholesterol side chain cleavage enzyme(CYP11A1) gene during the reproductive cycle in goose(Anas cygnoides)显示文摘Background: CYP11A1, a gene belonging to the family 11 of cytochrome P450, encodes a crucial steroidogenic enzyme that catalyzes the initial step in the production of all classes of steroids. Many studies show that CYP11A1 plays a role in ovary function. However, the role of CYP11A1 in goose reproductive cycle remains largely unknown.Results: In this study, full-length CYP11A1 c DNA of Zhedong goose was obtained using reverse transcription polymerase chain reaction(RT-PCR) and rapid amplification of c DNA ends(RACE). The c DNA consisted of a 96-base pair(bp) 5′untranslated region(UTR), a 179-bp 3′UTR and a 1509-bp open reading frame. The open reading frame encodes a putative 503 amino acid protein that shares high homology with CYP11A1 of other birds. The amino acid sequence possesses conserved domains of the P450 superfamily, which include the steroid-binding domain and the heme-binding region. Real-time quantitative polymerase chain reaction(q PCR) analysis revealed CYP11A1 mR NA was expressed ubiquitously in every Zhedong goose tissue analyzed, including the heart, liver, glandular stomach,lung, spleen, kidney, intestinum tenue, intestinum crassum, cerebrum, cerebellum, muscle, oviduct, pituitary,hypothalamus and ovary.. The relatively low levels of CYP11A1 m RNA were detected in pituitary, ovary and oviduct tissues at ovulation when compared with levels at oviposition. Interestingly, higher expression was observed in ovary and oviduct tissues during brooding.
