| 1 | Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. | 3 | Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE. | Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu | 2019 | Cellular & Molecular Immunology2019,16,3: | 15 |
| 4 | Pravastatin alleviates lipopolysaccharide-induced placental TLR4 over-activation and promotes uterine arteriole remodeling without impairing rat fetal development显示文摘Preeclampsia is associated with over-activation of the innate immune system in the placenta,in which toll-like receptor 4(TLR4) plays an essential part.With their potent anti-inflammatory effects,statins have been suggested as potential prevention or treatment of preeclampsia,although evidence remains inadequate.Herewith,we investigated whether pravastatin could ameliorate preeclampsia-like phenotypes in a previously established lipopolysaccharide(LPS)-induced rat preeclampsia model,through targeting the TLR4/NF-κB pathway.The results showed that pravastatin reduced the blood pressure [maximum decline on gestational day(GD) 12,(101.33±2.49) mmHg vs.(118.3±1.37) mmHg,P<0.05] and urine protein level [maximum decline on GD9,(3,726.23± 1,572.86) μg vs.(1,991.03 ±609.37)μg,P< 0.05],which were elevated following LPS administration.Pravastatin also significantly reduced the rate of fetal growth restriction in LPS-treated rats(34.10% vs.8.99%,P<0.05).Further pathological analyses suggested a restoration of normal spiral artery remodeling in preeclampsia rats by pravastatin treatment.These effects of pravastatin were associated with decreased TLR4/NF-κB protein levels in the placenta and IL-6/MCP-1 levels in serum.Additionally,no obvious abnormalities in fetal liver,brain,and kidney were found after administration of pravastatin.These results provide supportive evidence for use of pravastatin in preventing preeclampsia. | Muyi Yang Zhenyu Diao Zhiyin Wang Guijun Yan Guangfeng Zhao Mingming Zheng Anyi Dai Yimin Dai Yali Hu | 2018 | The Journal of Biomedical Research2018,32,4: | 5 |
| 5 | Study on Pretreatment during Simultaneous Determination of Sulfonamides and Chloramphenicols in Honey显示文摘[Objective]This study was conducted to establish a new UPLC-MS method for simultaneous detection of sulfonamides and chloramphenicols in honey.[Method]In this experiment,0.2%formic acid-acetonitrile was used as an extractant to simultaneously extract chloramphenicols and sulfonamides from drugs.The extract was then loaded and extracted with an efficient separation column Oasis~?PRiME HLB.After nitrogen blow-concentration and dissolution with mobile phase,the extract was loaded on UPLC and detected by MS.[Result]There was a good linear relation in the range of 0.1-10ng/ml for chloramphenicols,with a correlation coefficient of 0.991,while sulfonamides had a good linear relation in the range of 0.5-50ng/ml.The lowest detection limit of chloramphenicols and sulfonamides were 0.1and 0.5μg/kg,respectively.The recoveries were in the range of 82.50%-101.2%,with RSDvalues in the range of 3.2%-4.0%.[Conclusion]This method is simple and fast with low detection limit,high recovery and good reproducibility,and could be used for simultaneous detection of residual chloramphenicols and sulfonamides in honey. | Guangfeng ZHONG Xiaoling LIU Chengmei WANG | 2017 | Agricultural Biotechnology2017,6,2: | 2 |
| 6 | Acclimatory responses to high-salt stress in Chlamydomonas (Chlorophyta, Chlorophyceae) from Antarctica显示文摘Antarctic ice microalga can survive and thrive in channels or pores containing high salinity in Antarctic ice layer. In this study, it was found that cell membrane permeability of green microalga Chlamydomonas sp. L4 from Antarctic sea ice was high in cells treated with hypersalinity due to the induction of active oxygen and radicals. However, increased super oxide dismutase (SOD) scavenged harmful free radicals effectively to keep cell membrane integrity. Also, the analysis of membrane fatty acids demonstrated the content of saturated fatty acids and monounsaturated fatty acids increased and polyunsaturated fatty acids decreased under the high-salt treatment for 14 d, which effectively reduced the membrane fluidity and minimized the injury to cell membrane. The morpho- logical changes showed that hypersalinity induced the increase of cell volume and the consumption of starch granules. However, because of the increase in detoxification of vacuoles, electron-dense deposits and SOD activity under high-salt stress, the complete noninterference thylakoids, mitochondria and cell nucleus maintained cellular fundamental metabolism. Global-expression profiling of proteins showed eight protein spots disappeared, 18 protein spots decreased and 18 protein spots were enhanced after the high-salt shock obviously (P <0.05). One new peptide (pI 6.90; MW 51 kDa) was primarily confirmed as the processor of light reaction center protein CP43 in photosystem II, which increased photosynthesis ability of Chlamydomonas sp. | 7 | Efficacy compared between entecavir and adefovir dipivoxil on HBeAg-positive nucleos(t)ide-naive patients with chronic hepatitis B at week 12 and week 48显示文摘Objective:To evaluate the efficacy of entecavir and adefovir dipivoxil on HBeAg-positive nucleos(t)ide-naive patients with chronic hepatitis B with the method of Meta analysis.Methods:We searched PUBMED,EMBASE,CNKI (China National Knowledge Infrastructure),the Cochrane Central Register of Controlled Trials and the Cochrane Database of Systematic Reviews with reference to all data documented before May 2010.The dosage of entecavir and adefovir dipivoxil was 0.5 mg/d and 10 mg/d,respectively.Heterogeneity was examined by Chi-square test,the relative risk calculated and forest plot drawn.Rates of undetected serum HBV DNA,serum alanine aminotransferase (ALT) normalization,HBeAg clearance,HBeAg seroconversion and adverse effect occurrence were analyzed.Results:Six articles were included,which fit well in with this study.Meta analysis showed that the rate of undetected serum HBV DNA(P=0.000 2 at week 12,P=0.002 at week 48)and that of serum ALT normalization(P=0.04 at week 12,P=0.008 at week 48)in the entecavir group were higher than those in the adefovir dipivoxil group.However,no statistic significance existed between the two groups in the rate of HBeAg clearance (P=0.17),the rate of HBeAg seroconversion(P=0.53)or the rate of adverse effect occurrence(P=0.92)at week 48.Conclusion:Entecavir was superior to adefovir dipivoxil in decreasing serum HBV DNA and normalizing serum ALT in the HBeAg-positive nucleos(t)ide-naive patients with chronic hepatitis B. | Zhao Pan Xu Dongping Wang Xi Duan Guangfeng Huang Lili | 2010 | Journal of Medical Colleges of PLA(China)2010,25,5: | 2 |
