| 1 | Influence of Ginkgo Biloba extract on beta-secretase in rat hippocampal neuronal cultures following chronic hypoxic and hypoglycemic conditions显示文摘BACKGROUND: Preparation of Ginkgo leaf has been widely used to improve cognitive deficits and dementia, in particular in Alzheimer's disease patients. However, the precise mechanism of action of Ginkgo leaf remains unclear. OBJECTIVE: To explore the effect of Ginkgo Biloba extract (Egb761), Ginaton, on β-secretase expression in rat hippocampal neuronal cultures following chronic hypoxic and hypoglycemic conditions. DESIGN, TIME AND SETTNG: Completely by randomized, grouping study. The experiment was performed at the Laboratory of Molecular Imaging, Southeast University between August 2006 and August 2007. MATERIALS: A total of 128 Wistar rats aged 24 hours were selected, and hippocampal neurons were harvested for primary cultures. METHODS: On day 7, primary hippocampal neuronal cultures were treated with Egb761 (0, 25, 50, 100, 150, and 200 μg/mL) under hypoxic/hypoglycemic or hypoglycemic culture conditions for 12, 24, and 36 hours, respectively. Hippocampal neurons cultured in primary culture medium served as control. MAIN OUTCOME MEASURES: Cell viability was assayed using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT); fluorescence detection of β-secretase activity was performed; Western Blot was used to measure β-secretase expression. RESULTS: Cell viability under hypoxic/hypoglycemic or hypoglycemic culture conditions was significantly less than control cells (P < 0.05). Under hypoxic/hypoglycemic or hypoglycemic culture conditions, treatment with 25 μg/mL Egb761 did not alter cell viability. However, > 25 μg/mL Egb761 induced greater cell viability (P < 0.05). No differences were observed between hypoxic/hypoglycemic or hypoglycemic cells (P > 0.05). α-secretase activity was increased after 12 hours in hypoxic/hypoglycemic culture (P < 0.01). There were no significant differences between the 12-, 24-, or 36-hour Egb761 groups and the hypoxic/hypoglycemic groups (P > 0.05). β-secretase activity was greater after 12, 24, and 36 hours in hypoxic/hypoglycemic culture conditions, compared with control conditions (P < 0.05). β-secretase activity was significantly decreased in neurons treated with Egb761 for 12, 24, or 36 hours, compared with the hypoxic/hypoglycemic group (P < 0.05). β-secretase protein expression was significantly up-regulated in neurons cultured in hypoxic/hypoglycemic conditions for 12, 24, or 36 hours, compared to control cells (P < 0.05), and was decreased compared to neurons treated with Egb761 (P < 0.05). CONCLUSION: β-secretase expression and activity in rat neonatal hippocampal neurons were influenced by hypoxic and hypoglycemic culture. | 4 | Transformation of Coal Gangue to Sodalite and Faujasite Using Alkali-hydrothermal Method显示文摘The coal gangue as the only source of silicon and aluminum was employed to synthesize sodalite and faujasite using hydrothermal method,which directly treated the mixture of pre-treated coal gangue and NaOH solution under hydrothermal environment.X-ray powder diffraction analysis(XRD),thermogravimetry analysis(TG)and differential thermogravimetry analysis(DTG),scanning electron microscopy(SEM),high resolution transmission electron microscopy(HRTEM),N_(2)adsorption-desorption technique,X-ray photoelectron spectroscopy(XPS),etc.were used to characterize the samples.Cd^(2+) ion was used to evaluate the heavy metal ions removal performance of the samples.The experimental results show that the coal gangue,which consists of quartz,calcium feldspar,potassium feldspar and kaolinite,can transform to sodalite and faujasite under alkali-hydrothermal condition at 150 and 180℃,respectively.The as-prepared sodalite and faujasite can effectively remove the simulated Cd2+ion wastewater and actual industrial wastewater containing As^(3+),Cd^(2+),and Cr^(3+)ions,and the good heavy metal ion removal performance of the zeolites is mainly attributed to their low Si/Al ratio and high Na+content.This alkali-hydrothermal method appears to be a simple and efficient method for transformation of coal gangue to high purity zeolites. | WANG Cheng FENG Kai WANG Lipeng YU Qianru DU Fuling LI Yan YU Kaining | 2023 | Journal of Wuhan University of Technology(Materials Science)2023,38,6: | 0 |

| 6 | Anti-cancer activity of Tonglian decoction against esophageal cancer cell proliferation through regulation of the cell cycle and PI3K/Akt signaling pathway显示文摘Objective:The purpose of this study was to observe the anti-cancer activity of Tonglian decoction(TD)on esophageal cancer(EC)cells in vitro,and to elucidate the related molecular mechanisms in the cell cycle and PI3K/Akt signaling pathway.Methods:EC9706 cells were cultured in RPMI 1640 medium supplemented with 10%calf serum at 37C in a 5%CO2 incubator.The cells were treated with rat serum containing TD or the serum of rats administered Xiaoaiping as a positive control drug.Cell proliferation was assessed by methylthiazolyldiphenyl-tetrazolium bromide assays.Cell morphology was observed under a microscope.The cell cycle was examined by flow cytometry.Protein expression in the PI3K/Akt signaling pathway was measured by western blotting.Results:TD mainly inhibited cell proliferation.Concentrations of 50%cell inhibition by rat serum containing TD or Xiaoaiping were 73.6 and 153.8 mL/mL,respectively.TD also influenced cell morphology characterized by small shrunken cells.Cell colonies became small and the cell proliferation rate was slower.In cell cycle analysis,the percentage of cells in S phase was decreased significantly by TD and Xiaoaiping compared with the blank control group(P<.05).Western blotting showed that serum containing TD strongly downregulated EGFR,PI3K,Akt,p-Akt,and mTOR expression compared with the blank control group(P<.05).Conclusion:TD could inhibit EC9706 carcinoma cell proliferation by blocking the cell cycle progression in S phase.The possible mechanism was inhibition of multiple targets in the PI3K/Akt signaling pathway by TD. | Yongsen Jia Lijuan Qin Chunhua Jiang Qing Lin Fuling Tian Huijuan Cao Xin Yan | 2015 | Journal of Traditional Chinese Medical Sciences2015,2,2: | 0 |
