1Roles of Chemokine Receptor 4 (CXCR4) and Chemokine Ligand 12 (CXCL12) in Metastasis of Hepatocellular Carcinoma Cells显示文摘Chemokines are involved in human hepatocellular carcinoma (HCC) carcinogenesis. However, the exact mechanism of chemokines in HCC carcinogenesis remains unknown. Here we investigated the roles of chemokine receptor 4 (CXCR4) and chemokine ligand 12 (CXCL12) in the metastasis of HCC. We found that the expression levels of CXCR4 mRNA in HCC tissues, MHCC97 cells, and HUVEC cells were 2.52 ± 1.13, 2.34 ± 1.16 and 1.63 ± 1.26, respectively and that the CXCR4 protein levels were 1.38 ± 0.13, 1.96 ± 0.32 and 1.86 ± 0.21, respectively. In contrast, CXCR4 was not detected in normal hepatic tissues. In 78 HCC patients, we also found that the concentration of CXCL12 in cancerous ascitic fluid was 783-8,364 pg/ml and that CXCL12 mRNA level in HCC metastasis portal lymph nodes was 1.21 ± 0.87 but undetectable in normal hepatic tissues. Finally we discovered that recombinant human CXCL12 could induce MHCC97 cells and HUVEC cells to migrate with chemotactic indexes (CI) of 3.9 ± 1.1 and 4.1 ± 1.6, respectively. Cancerous ascitic fluid could also induce the migration of MHCC97 cells with a CI of 1.9 ± 0.8. 2Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 3Microbiota transplantation reveals beneficial impact of berberine on hepatotoxicity by improving gut homeostasis显示文摘Berberine has been shown to reduce acute liver injury although the underlying mechanism is not fully understood. Because of the anatomic connection, the liver is constantly exposed to gut-derived bacterial products and metabolites. In this study, we showed that berberine has beneficial effects on both hepatotoxicity and intestinal damage in a rat model of chronic or acute liver injury. Microbiota transplantation from the rats with chronic hepatotoxicity could aggravate acute hepatotoxicity in mice treated with diethylnitrosamine(DEN). In rat models with gut homeostasis disruption induced by penicillin or dextran sulfate sodium(DSS), their fecal microbiota could also cause an enhanced hepatotoxicity of recipient mice. When treated with berberine,the DSS-induced enteric dysbacteriosis could be mitigated and their fecal bacteria were able to reduce acute hepatotoxicity in recipient mice. This study indicates that berberine could improve intestinal dysbacteriosis, which reduces the hepatotoxicity caused by pathological or pharmacological intervention. 5Application of moderate nitrogen levels alleviates yield loss and grain quality deterioration caused by post-silking heat stress in fresh waxy maize显示文摘High temperature(HT)during grain filling is one of the most important environmental factors limiting maize yield and grain quality.Nitrogen(N)fertilizer is essential for maintaining normal plant growth and defense against environmental stresses.The effects of three N rates and two temperature regimes on the grain yield and quality of fresh waxy maize were studied using the hybrids Suyunuo 5(SYN5)and Yunuo 7(YN7)as materials.N application rates were 1.5,4.5,and 7.5 g plant-1,representing low,moderate,and high N levels(LN,MN,and HN,respectively).Mean day/night temperatures during the grain filling of spring-and summer-sown plants were 27.6/21.0°C and 28.6/20.0°C for ambient temperature(AT)and 35/21.0°C and 35/20.0°C for HT,respectively.On average,HT reduced kernel number,weight,yield,and moisture content by 29.8%,17.9%,38.7%,and 3.3%,respectively.Kernel number,weight,yield,moisture,and starch contents were highest under MN among the three N rates under both temperature regimes.HT reduced grain starch content at all N levels.HT increased grain protein content,which gradually increased with N rate.Mean starch granule size under MN was larger(10.9μm)than that under LN and HN(both 10.4μm)at AT.However,the mean size of starch granules was higher under LN(11.7μm)and lower under MN(11.2μm)at HT.Iodine binding capacity(IBC)was lowest under MN and highest under HN among the three N levels under both temperature regimes.In general,IBC at all N rates was increased by HT.Peak viscosity(PV)was gradually reduced with increasing N rate at AT.In comparison with LN,PV was increased by MN and decreased by HN at HT.Retrogradation percentage gradually increased with N rate at AT,but was lowest under MN among the three N rates at HT.LN+AT and MN+HT produced grain with high pasting viscosity and low retrogradation tendency.MN application could alleviate the negative effects of HT on the grain yield and quality of fresh waxy maize.Jue Wang Pengxiao Fu Weiping Lu Dalei Lu 2020The Crop Journal2020,8,6:5
6The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 7SCStore: Managing Scientific Computing Packages for Hybrid System with Containers显示文摘Managing software packages in a scientific computing environment is a challenging task, especially in the case of heterogeneous systems. It is error prone when installing and updating software packages in a sophisticated computing environment. Testing and performance evaluation in an on-the-fly manner is also a troublesome task for a production system. In this paper, we discuss a package management scheme based on containers. The newly developed method can ease the maintenance complexity and reduce human mistakes. We can benefit from the self-containing and isolation features of container technologies for maintaining the software packages among intricately connected clusters. 8Facile shape and size-controlled growth of uniform magnetite and hematite nanocrystals with tunable properties显示文摘Monodispersed magnetite Fe3O4 and hematite α-Fe2O3 nanocrystals have been grown in co-solvents of alcohol and water. Either the shape or the size of the nanocrystals could be easily controlled. Both the phases and nanostructures have been characterized by powder X-ray diffraction patterns and electron microscopy. The magnetic and catalytic properties of these products were investigated and compared with each other. The obtained results clearly demonstrate that these iron oxide nanocrystals are soft ferromagnetic at room temperature and α-Fe2O3 has a more effective catalytic property on the thermal decomposition of ammonium perchlorate than Fe3O4. Based on the experimental data, it is proposed that the magnetic and catalytic properties of these nanocrystals are dependent not only on the size and shape, but also on the surface structure of the nanocrystals. 9Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research. 10A proposed direct measurement of cross section at Gamow window for key reaction 19F(p,α) 16O in Asymptotic Giant Branch stars with a planned accelerator in CJPL显示文摘In 2014, the National Natural Science Foundation of China(NSFC) approved the Jinping Underground Nuclear Astrophysics laboratory(JUNA) project, which aims at direct cross-section measurements of four key stellar nuclear reactions right down to the Gamow windows. In order to solve the observed fluorine overabundances in Asymptotic Giant Branch(AGB) stars, measuring the key ^(19)F( p,α)^(16)O reaction at effective burning energies(i.e., at Gamow window) is established as one of the scientific research sub-projects. 11Microstructure and Mechanical Behavior of FeNiCoCr and FeNiCoCrMn High-Entropy Alloys Fabricated by Powder Metallurgy显示文摘The present work reports a systematic investigation on phase evolution,microstructure and microstructure-property relationship of two typical face-centered cubic(FCC)structured high-entropy alloys(HEAs),FeNiCoCr and FeNiCoCrMn,prepared via mechanical alloying(MA)followed by spark plasma sintering(SPS).Following 50 h of MA,the two HEAs consisted of a mixture of FCC and body-centered cubic phases.Following SPS,the bulk FeNiCoCr alloy showed a primary FCC phase with a small amount of Cr_(23)C_(6) and Cr_(2)O_(3) contaminants,while the bulk FeNiCoCrMn alloy was composed of a primary FCC phase with some(Cr,Mn)23C6 and MnCr_(2)O_(4) contaminants.The average grain size of the primary FCC phase in the bulk FeNiCoCr alloy was~416 nm,while that of the primary FCC phase in the bulk FeNiCoCrMn alloy was~547 nm.The yield strength,compressive strength and strain-to-failure of the bulk FeNiCoCr alloy are 1525 MPa,1987 MPa and 24.4%,respectively,whereas those of the bulk FeNiCoCrMn alloy are 1329 MPa,1761 MPa and 21.9%,respectively.It suggests that the bulk FeNiCoCrMn exhibited lower strength and plasticity in comparison with the bulk FeNiCoCr alloy.Clearly,the smaller grain size of the primary FCC phase in the FeNiCoCr alloy is mainly responsible for the better mechanical performance.Chenliang Chu Weiping Chen Zhen Chen Zhenfei Jiang Hao Wang Zhiqiang Fu 2021Acta Metallurgica Sinica(English Letters)2021,34,4:3
12Catalytic performance of iron oxide loaded on electron-rich surfaces of carbon nitride显示文摘Carbon nitride(CN) in CN encapsulated Ni/Al_2O_3(denoted as CN/Ni/Al_2O_3) catalyst was evidenced previously as a material in electron-rich state and possessed H2-dissociative adsorption activity due to the electron doping effect from underlying nickel. In this report, iron oxide loaded on the CN/Ni/Al_2O_3 was synthesized and investigated by Fischer-Tropsch(F-T) synthesis to test the special effect of electron-rich support on the catalytic activity of iron oxide. The Fe/CN/Al_2O_3 and CN/Ni/Al_2O_3 samples were accordingly synthesized for comparison. In Fe/CN/Ni/Al_2O_3, the iron oxide was reduced to magnetite by syngas as evidenced by the in-situ XPS measurements and XRD pattern of used catalyst. Compared with Fe/CN/Al_2O_3, more light hydrocarbons over Fe/CN/Ni/Al_2O_3 were observed. It should be understood by the interaction between iron oxide and support mainly due to the effect of electron-rich state and thus enhanced H_2 adsorption ability. In addition, such a novel support facilitated the CO conversion and retarded the water-gas shift reaction and CO2 formation. 16Amorphous NiO_(n)coupled with trace PtO_(x)toward superior electrocatalytic overall water splitting in alkaline seawater media显示文摘Developing corrosion resistance bifunctional electrocatalysts with high activity and stability toward both hydrogen evolution reaction(HER)and oxygen evolution reaction(OER),especially electrolysis in seawater,is of prime significance but still pressingly challenging.Herein,in-situ introduced PtO_(x)on the derivative amorphous NiO_(n)is prepared via heat treatment of Ni ZIFL nanosheets on nickel foam under low temperature(PtO_(x)-NiO_(n)/NF).The synthesized PtO_(x)-NiO_(n)/NF possesses suprahydrophilic and aerophilic surface,then in favor of intimate contact between the electrode and electrolyte and release of the generated gas bubbles during the electrocatalysis.As a result,the in-situ PtO_(x)-NiO_(n)/NF electrode presents outstanding bifunctional activity,which only requires extremely low overpotentials of 32 and 240 mV to reach a current density of 10 mA·cm^(-2)for HER and OER,respectively,which exceeds most of the electrocatalysts previously developed and even suppresses commercial Pt/C and RuO_(2)electrodes.As for two-electrode cell organized by PtO_(x)-NiO_(n)/NF,the voltages down to 1.57 and 1.58 V are necessary to drive 10 mA·cm^(-2)with remarkable durability in 1 M KOH and alkaline seawater,respectively,along with remarkable stability.Moreover,a low cell voltage of 1.88 V is needed to achieve 1,000 mA·cm^(-2)toward water-splitting under industrial conditions.This study provides a new idea for designing in-situ amorphous metal oxide bifunctional electrocatalyst with strong Pt–support interaction for overall water splitting.Wenli Yu Hongru Liu Ying Zhao Yunlei Fu Weiping Xiao Bin Dong Zexing Wu Yongming Chai Lei Wang 2023Nano Research2023,16,5:2
