| 3 | Internal ribosome entry site-based vectors for combined gene therapy显示文摘Gene therapy appears as a promising strategy to treatincurable diseases. In particular, combined gene therapy has shown improved therapeutic efficiency. Internal ribosome entry sites(IRESs), RNA elements naturally present in the 5' untranslated regions of a few m RNAs, constitute a powerful tool to co-express several genes of interest. IRESs are translational enhancers allowing the translational machinery to start protein synthesis by internal initiation. This feature allowed the design of multi-cistronic vectors expressing several genes from a single m RNA. IRESs exhibit tissue specificity, and drive translation in stress conditions when the global cell translation is blocked, which renders them useful for gene transfer in hypoxic conditions occurring in ischemic diseases and cancer. | 4 | Reducing Uncertainties in Climate Projections with Emergent Constraints:Concepts, Examples and Prospects显示文摘Models disagree on a significant number of responses to climate change,such as climate feedback,regional changes,or the strength of equilibrium climate sensitivity.Emergent constraints aim to reduce these uncertainties by finding links between the inter-model spread in an observable predictor and climate projections.In this paper,the concepts underlying this framework are recalled with an emphasis on the statistical inference used for narrowing uncertainties,and a review of emergent constraints found in the last two decades.Potential links between highlighted predictors are explored,especially those targeting uncertainty reductions in climate sensitivity,cloud feedback,and changes of the hydrological cycle.Yet the disagreement across emergent constraints suggests that the spread in climate sensitivity can not be significantly narrowed.This calls for weighting the realism of emergent constraints by quantifying the level of physical understanding explaining the relationship.This would also permit more efficient model evaluation and better targeted model development.In the context of the upcoming CMIP6 model intercomparison a growing number of new predictors and uncertainty reductions is expected,which call for robust statistical inferences that allow cross-validation of more likely estimates. | Florent BRIENT | 2020 | Advances in Atmospheric Sciences2020,37,1: | 4 |
| 5 | Fulminant myocarditis in adults: a narrative review显示文摘Fulminant myocarditis(FM)is an uncommon syndrome characterized by sudden and severe hemodynamic compromise secondary to acute myocardial inflammation,often presenting as profound cardiogenic shock,life-threatening ventricular arrhythmias and/or electrical storm.FM may be refractory to conventional therapies and require mechanical circulatory support(MCS).The immune system has been recognized as playing a pivotal role in the pathophysiology of myocarditis,leading to an increased focus on immunosuppressive treatment strategies.Recent data have highlighted not only the fact that FM has significantly worse outcomes than non-FM,but that prognosis and management strategies of FM are heavily dependent on histological subtype,placing greater emphasis on the role of endomyocardial biopsy in diagnosis.The impact of subtype on severity and prognosis will likewise influence how aggressively the myocarditis is managed,including whether MCS is warranted.Many patients with refractory cardiogenic shock secondary to FM end up requiring MCS,with venoarterial extracorporeal membrane oxygenation demonstrating favorable survival rates,particularly when initiated prior to the development of multiorgan failure.Among the challenges facing the field are the need to more precisely identify immunopathophysiological pathways in order to develop targeted therapies,and the need to better optimize the timing and management of MCS to minimize complications and maximize outcomes. | Santiago Montero Darryl Abrams Enrico Ammirati Florent Huang Dirk WDonker Guillaume Hekimian Cosme García-García Antoni Bayes-Genis Alain Combes Matthieu Schmidt | 2022 | Journal of Geriatric Cardiology2022,19,2: | 3 |
| 6 | Complex causes and consequences of rangeland greening in South America -multiple interacting natural and anthropogenic drivers and simultaneous ecosystem degradation and recovery trends显示文摘Land-surface greening has been reported globally over the past decades.While often seen to represent ecosystem recovery,the impacts on biodiversity and society can also be negative.Greening has been widely reported from rangelands,where drivers and processes are complex due to its high environmental heterogeneity and societal dynamics.Here,we assess the complexity behind greening and assess its links to various drivers in an iconic,heterogeneous rangeland area,the IberáWetlands and surroundings,in Argentina.Time-series satellite imagery over the past 19 years showed overall net greening,but also substantial local browning both in protected and unprotected areas,linking to land use,temporal changes in surface water,fire,and weather.We found substantial woody expansion mainly in the unprotected land,with 37%contributed by tree plantations and the remaining 63%by spontaneous woody expansion,along with widespread transitions from terrestrial land to seasonal surface water.Fire occurrences tended to reduce greening with unprotected areas experiencing widespread and frequent fire.However,protected areas had more browning in unburnt areas than burned areas.Temporal variation in annual precipitation and temperature tended to nonlinearly influence fire occurrences with an interplay of human fire management,further shaping the vegetation greening,pointing to high complexity behind the observed rangeland greening involving interactions among local drivers.Our findings highlight that the observed overall greening is an outcome of multiple trends with clear negative impacts on biodiversity and the local livestock-oriented culture(notably expanding tree plantations)and spontaneous vegetation dynamics,partly involving spontaneous woody expansion.The latter has positive potential for biodiversity and ecosystem services in terms of woodland recovery,but can become negative in such a natural savanna region if expansions develop on a too broad scale,highlighting the importance of ensuring recovery of natural fire and herbivory regimes in protected areas along with sustainable rangeland management elsewhere. | Wang Li Robert Buitenwerf Renata Nicora Chequín Javier Elias Florentín Roberto Manuel Salas Julia Carolina Mata Li Wang Zheng Niu Jens-Christian Svenning | 2020 | Geography and Sustainability2020,1,4: | 2 |
| 7 | Physico-chemical and mechanical properties of Ti_3SiC_2-based materials elaborated from SiC/Ti by reactive spark plasma sintering显示文摘In this paper, the synthesis of Ti_3 SiC_2 from SiC/Ti powder using reactive spark plasma sintering(R-SPS) in the temperature range of 1300-1400 ℃ is reported. The results show that the purity of Ti_3 SiC_2 is improved up to 75 wt% when the holding time is increased from 10 to 20 min at1400 ℃.
