| 1 | Comparison of the effects of bevacizumab and ranibizumab injection on corneal angiogenesis in an alkali burn induced model显示文摘·AIM:To investigate the effects of bevacizumab and ranibizumab on corneal neovascularization in an alkali burn-induced model of corneal angiogenesis. ·METHODS:Fifteen Wistar-Albino rats were divided randomly into 3 groups after chemical cauterization of the cornea. The first group received a single dose of 0.1mL saline solution as a control group whereas second and third groups received a single dose of 2.5mg bevacizumab or 1mg ranibizumab by subconjunctival injection, respectively. After three weeks, the rat corneas were evaluated by biomicroscopy and corneal photographs were taken. The percentage of neovascularization area, length of the longest new vessel, corneal oedema and corneal opacity scores were assessed. ·RESULTS:The analysis of digital photographs showed that the percentage of neovascularization area to the total corneal area, the length of the longest new vessel, corneal oedema and opacity scores were significantly lower in both study groups compared to the control group (P <0.05). Additionally, the percentage of corneal neovascularization area, the length of the longest new vessel and corneal opacity score were less with bevacizumab than ranibizumab. ·CONCLUSION:Subconjunctival bevacizumab and ranibizumab treatments may be effective methods in reducing corneal neovascularization. | 2 | 吸烟,血清抗氧化维生素水平和老年性黄斑变性之间的关系(英文)显示文摘目的:研究老年性黄斑变性(AMD)等级、血清抗氧化维生素水平(维生素A、C和E)和吸烟的关系。方法:对84例患者行黄斑彩色眼底成像,根据AMD损伤率和损伤程度将其分为五组(等级I^V)。应用高效液相色谱法(HPLC)测量血清抗氧化维生素水平。根据吸烟状况分不吸烟者、已戒烟者和吸烟者三组,并统计吸烟者每年的吸烟总量。结果:对照组中,维生素A、E、C水平分别为0.874±0.326mg/L,10.739±4.874mg/L和1.737±0.447mg/L,AMD组中分别为0.880±0.305mg/L,9.487±6.060mg/L和1.870±2.191mg/L,无显著差异(P>0.05)。AMD不同等级分组间维生素A(P=0.881)和E(P=0.293)水平差异无统计学意义,维生素C水平(P=0.044)随AMD程度的加深而增加。根据吸烟状况,AMD组与对照组之间无显著差异。根据每年的吸烟总量,两组之间有显著差异(P=0.02)。AMD越严重,年吸烟总量越高(P=0.007)。结论:研究结果显示AMD与维生素A、E血清水平无关,但和维生素C的血清水平相关,并与吸烟量有关。 | Sezen Akkaya Cakir Ozlem Sahin Haluk Kazokoglu Feyza Tekeli Goncagul Haklar Yelda Ozkurt | 2014 | 国际眼科杂志2014,14,5: | 5 |
| 3 | Total oxidative stress, paraoxonase and arylesterase levels at patients with pseudoexfoliation syndrome and pseudoexfoliative glaucoma显示文摘AIM: To investigate the oxidative stress status of the aqueous humor and serum of patients with pseudoexfoliation(PEX) syndrome and pseudoexfoliative glaucoma(PEG) and to measure paraoxonase(PON) and arylesterase(ARE) levels.·METHODS: A total of 78 patients were enrolled in the study, with 26 patients in each separate group. The patients were divided into three groups: the first group entailed PEX syndrome patients, while the second group consisted of patients with PEG and the third group involved patients with no additional systemic diseases,other than the diagnosis of cataract as control. Total oxidative stress(TOS), total antioxidant capacity(TAC),PON, and ARE levels in aqueous humor and serum were measured.·RESULTS: TAC, PON and arylesterase levels in aqueous humor and serum of the PEX syndrome and PEG patients were significantly decreased compared with control group(P <0.05). TOS values were higher in patients with PEX syndrome and PEG than controls(P <0.05). TAC, PON and ARE levels of aqueous humor did not differ significantly between the PEX syndrome and PEG groups·CONCLUSION: These findings are potentially of significance and add to the growing body of evidence for oxidative stress in PEX syndrome and PEG. | 4 | Discriminating performance of macular ganglion cellinner plexiform layer thicknesses at different stages of glaucoma显示文摘AIM: To determine the discriminating performance of the macular ganglion cell-inner plexiform layer(GCIPL) parameters between all the consecutive stages of glaucoma(from healthy to moderate-to-severe glaucoma), and to compare it with the discriminating performances of the peripapillary retinal nerve fiber layer(RNFL) parameters and optic nerve head(ONH) parameters.METHODS: Totally 147 eyes(40 healthy, 40 glaucoma suspects, 40 early glaucoma, and 27 moderate-to-severe glaucoma) of 133 subjects were included. Optical coherence tomography(OCT) was obtained using Cirrus HD-OCT 5000. The diagnostic performances of GC-IPL, RNFL, and ONH parameters were evaluated by determining the area under the curve(AUC) of the receiver operating characteristics. RESULTS: All GC-IPL parameters discriminated glaucoma suspect patients from subjects with healthy eyes and moderate-to-severe glaucoma from early glaucoma patients(P<0.017, for all). Also, minimum, inferotemporal and inferonasal GC-IPL parameters discriminated early glaucoma patients from glaucoma suspects, whereas no RNFL or ONH parameter could discriminate between the two. The best parameters to discriminate glaucoma suspects from subjects with healthy eyes were superonasal GC-IPL, superior RNFL and average c/d ratio(AUC=0.746, 0.810 and 0.746, respectively). Discriminating performances of all the parameters for early glaucoma vs glaucoma suspect comparison were lower than that of the other consecutive group comparisons, with the bestGC-IPL parameters being minimum and inferotemporal(AUC=0.669 and 0.662, respectively). Moreover, minimum GC-IPL, average RNFL, and rim area(AUC=0.900, 0.858, 0.768, respectively) were the best parameters for discriminating moderate-to-severe glaucoma patients from early glaucoma patients.CONCLUSION: GC-IPL parameters can discriminate glaucoma suspect patients from subjects with healthy eyes, and also all the consecutive stages of glaucoma from each other(from glaucoma suspect to moderate-tosevere glaucoma). | 5 | Hepatocellular carcinoma xenograft supports HCV replication:A mouse model for evaluating antivirals显示文摘AIM: To develop a hepatocellular carcinoma (HCC) xenograft model for studying hepatitis C virus (HCV) replication in a mice, and antiviral treatment.METHODS: We developed a stable S3-green fluorescence protein (GFP) cell line that replicated the GFP-tagged HCV sub-genomic RNA derived from a highly efficient JFH1 virus. S3-GFP replicon cell line was injected subcutaneously into γ-irradiated SCID mice. We showed that the S3-GFP replicon cell line formed human HCC xenografts in SCID mice. Cells were isolated from subcutaneous tumors and then serially passaged multiple times in SCID mice by culturing in growth medium supplemented with G-418. The mouse-adapted S3-GFP replicon cells were implanted subcutaneously and also into the liver of SCID mice via intrasplenic infusion to study the replication of HCV in the HCC xenografts. The tumor model was validated for antiviral testing after intraperitoneal injection of interferon-α (IFN-α). RESULTS: A highly tumorigenic S3-GFP replicon cell line was developed that formed subcutaneous tumors within 2 wk and diffuse liver metastasis within 4 wk in SCID mice. Replication of HCV in the subcutaneous and liver tumors was confirmed by cell colony assay, detection of the viral RNA by ribonuclease protection assay and real-time quantitative reverse transcription polymerase chain reaction. High-level replication of HCV sub-genomic RNA in the tumor could be visualized by GFP expression using fluorescence microscopy. IFN-α cleared HCV RNA replication in the subcutaneous tumors within 2 wk and 4 wk in the liver tumor model. CONCLUSION: A non-infectious mouse model allows us to study replication of HCV in subcutaneous and metastatic liver tumors.
