| 1 | Dementia and osteoporosis in a geriatric population: Is there a common link?显示文摘AIM To determine the existence of a common pathological link between dementia and osteoporosis through reviewing the current evidence base. METHODS This paper reviews the current literature on osteoporosis and dementia in order to ascertain evidence of a common predisposing aetiology. A literature search of Ovid MEDLINE(1950 to June 2016) was conducted. The keywords 'osteoporosis', 'osteoporotic fracture', 'dementia' and 'Alzheimer's disease'(AD) were used to determine the theoretical links with the most significant evidence base behind them. The key links were found to be vitamins D and K, calcium, thyroid disease, statins, alcohol and sex steroids. These subjects were then searched in combination with the previous terms and the resulting papers manually examined. Theoretical, in vitro and in vivo research were all used to inform this review which focuses on the most well developed theoretical common causes for dementia(predominantly Alzheimer's type) and osteoporosis.RESULTS Dementia and osteoporosis are multifaceted disease processes with similar epidemiology and a marked increase in prevalence in elderly populations. The existence of a common link between the two has been suggested despite a lack of clear pathological overlap in our current understanding. Research to date has tended to be fragmented and relatively weak in nature with multiple confounding factors reflecting the difficulties of in vivo experimentation in the population of interest. Despite exploration of various possible mechanisms in search for a link between the two pathologies, this paper found that it is possible that these associations are coincidental due to the nature of the evidence available. One finding in this review is that prior investigation into common aetiologies has found raised amyloid beta peptide levels in osteoporotic bone tissue, with a hypothesis that amyloid beta disorders are systemic disorders resulting in differing tissue manifestations. However, our findings were that the most compelling evidence of a common yet independent aetiology lies in the APOE4 allele, which is a well-established risk for AD but also carries an independent association with fracture risk. The mechanism behind this is thought to be the reduced plasma vitamin K levels in individuals exhibiting the APOE4 allele which may be amplified by the nutritional deficiencies associated with dementia, which are known to include vitamins K and D. The vitamin theory postulates that malnutrition and reduced exposure to sunlight in patients with AD leads to vitamin deficiencies. CONCLUSION Robust evidence remains to be produced regarding potential links and regarding the exact aetiology of these diseases and remains relevant given the burden of dementia and osteoporosis in our ageing population. | 2 | NLRP3 inflammasome activation determines the fibrogenic potential of PM_(2.5) air pollution particles in the lung显示文摘Airborne fine particulate matter(PM_(2.5))is known to cause respiratory inflammation such as chronic obstructive pulmonary disease and lung fibrosis.NLRP3 inflammasome activation has been implicated in these diseases;however,due to the complexity in PM_(2.5)compositions,it is difficult to differentiate the roles of the components in triggering this pathway.We collected eight real-life PM_(2.5)samples for a comparative analysis of their effects on NLRP3 inflammasome activation and lung fibrosis.In vitro assays showed that although the PM_(2.5)particles did not induce significant cytotoxicity at the dose range of 12.5to 100μg/m L,they induced potent TNF-αand IL-1βproduction in PMA differentiated THP-1 human macrophages and TGF-β1 production in BEAS-2B human bronchial epithelial cells.At the dose of 100μg/m L,PM_(2.5)induced NLRP3 inflammasome activation by inducing lysosomal damage and cathepsin B release,leading to IL-1βproduction.This was confirmed by using NLRP3-and ASC-deficient cells as well as a cathepsin B inhibitor,ca-074 ME.Administration of PM_(2.5)via oropharyngeal aspiration at 2 mg/kg induced significant TGF-β1 production in the bronchoalveolar lavage fluid and collagen deposition in the lung at 21days post-exposure,suggesting PM_(2.5)has the potential to induce pulmonary fibrosis.The ranking of in vitro IL-1βproduction correlates well with the in vivo total cell count,TGF-β1 production,and collagen deposition.In summary,we demonstrate that the PM_(2.5)is capable of inducing NLRP3 inflammasome activation,which triggers a series of cellular responses in the lung to induce fibrosis. | Wei Cao Xiang Wang Jiulong Li Ming Yan Chong Hyun Chang Joshua Kim Jinhong Jiang Yu-Pei Liao Shannon Tseng Sydney Kusumoputro Candice Lau Marissa Huang Pengli Han Pengju Lu Tian Xia | 2022 | Journal of Environmental Sciences2022,34,1: | 3 |

| 4 | Alpha-adrenoceptor antagonism by Crassostrea gigas oyster extract inhibits noradrenalineinduced vascular contraction in Wistar rats显示文摘OBJECTIVE: Crassostrea gigas oyster extract has been reported to have antioxidant, antihypertensive and lipid-lowering properties that may be useful for treating cardiovascular diseases. This study aimed to evaluate the effect of C. gigas oyster extract on cardiovascular function in tissues from healthy rats. METHODS: Single-cell microelectrode and isolated thoracic aortic organ bath studies were performed on tissues from 8-week-old healthy Wistar rats, using varying concentrations of C. gigas oyster extract. To elucidate a mechanism of action for the oyster's vasoactive properties, concentration response curves were carried out in the presence of a calcium channel inhibitior(verapamil), a nitric oxide synthase inhibitor(N G-nitro-L-arginine methyl ester), a potassium channel inhibitor(4-aminopyridine), in addition to the α-adrenoceptor inhibitor prazosin. RESULTS: Oyster solution at 7 500 mg/m L inhibited noradrenaline-induced contraction in isolated aortic rings. Cardiac electrophysiology results showed that neither concentration of oyster solution was able to significantly reduce action potential duration at all phases of repolarisation in left ventricular papillary muscles from healthy animals. CONCLUSION: When administered to healthy vascular tissue, C. gigas oyster extract inhibits contraction induced by noradrenaline. | 7 | Dynamic Patterns of Gene Expression Additivity and Regulatory Variation throughout Maize Development显示文摘Gene expression variation is a key component underlying phenotypic variation and heterosis. Transcriptome profiling was performed on 23 different tissues or developmental stages of two maize inbreds, B73 and Mo17, as well as their hybrid. The obtained large-scale datasets provided opportunities to monitor the developmental dynamics of differential expression, additivity for gene expression, and regulatory variation. The transcriptome can be divided into .30 000 genes that are expressed in at least one tissue of one in bred and an additional ~10 000 “silent” genes that are not expressed in any tissue of any genotype, 90% of which are non-syntenic relative to other grasses. Many (.74%) of the expressed genes exhibit differential expression in at least one tissue. However, the majority of genes with differential expression do not exhibit consistent differential expression in different tissues. These genes often exhibit tissue-specific differential expression with equivalent expression in other tissues, and in many cases they switch the directionality of differential expression in different tissues. This suggests widespread variation for tissue-specific regulation of gene expression between the two maize inbreds B73 and Mo17. Nearly 5000 genes are expressed in only one parent in at least one tissue (single parent expression) and 97% of these genes are expressed at mid-parent levels or higher in the hybrid, providing extensive opportunities for hybrid complementation in heterosis. In general, additive expression patterns are much more common than non-additive patterns, and this trend is more pronounced for genes with strong differential expression or single pare nt expressi on. There is relatively little evidence for non-additive expression patterns that are maintained in multiple tissues. The analysis of allele-specific expression allowed classification of cis. and trans-regulatory variation. Genes with c/s-regulatory variation often exhibit additive expression and tend to have more consistent regulatory variation throughout development. In contrast, genes with trans-reguiatory variation are enriched for non-additive patterns and often show tissue-specific differential expression. Taken together, this study provides a deeper understatiding of regulatory variation and the degree of additive gene expression throughout maize development. | 8 | Multiscale patterning of a metallic glass using sacrificial imprint lithography显示文摘Bulk metallic glasses(BMGs)have been developed as a means to achieve durable multiscale,nanotextured surfaces with desirable properties dictated by topography for a multitude of applications.One barrier to this achievement is the lack of a bridging technique between macroscale thermoplastic forming and nanoimprint lithography,which arises from the difficulty and cost of generating controlled nanostructures on complex geometries using conventional top-down approaches.This difficulty is compounded by the necessary destruction of any resulting reentrant structures during rigid demolding.We have developed a generalized method to overcome this limitation by sacrificial template imprinting using zinc oxide(ZnO)nanostructures.It is established that such structures can be grown inexpensively and quickly with tunable morphologies on a wide variety of substrates out of solution,which we exploit to generate the nanoscale portion of the multiscale pattern through this bottom-up approach.In this way,we achieve metallic structures that simultaneously demonstrate features from the macroscale down to the nanoscale,requiring only the top-down fabrication of macro/microstructured molds.Upon detachment of the formed part from the multiscale molds,the ZnO remains embedded in the surface and can be removed by etching in mild conditions to both regenerate the mold and render the surface of the BMGs nanoporous.The ability to pattern metallic surfaces in a single step on length scales from centimeters down to nanometers is a critical step toward fabricating devices with complex shapes that rely on multiscale topography for their intended functions,such as biomedical and electrochemical applications. | Jonathan P.Singer Candice I.Pelligra Noga Kornblum Youngwoo Choo Manesh Gopinadhan Punnathat Bordeenithikasem Jittisa Ketkaew Seng Fatt Liew Hui Cao Jan Schroers Chinedum O.Osuji | 2015 | Microsystems & Nanoengineering2015,1,1: | 2 |
