| 1 | An acorn squash (Cucurbita pepo ssp. ovifera) fruit and seed transcriptome as a resource for the study of fruit traits in Cucurbita显示文摘Acorn squash(Cucurbita pepo)is an iconic fall vegetable in the United States,known for its unique fruit shape and also prized for its culinary properties.Little is known about the metabolism that underlies the development of fruit quality attributes such as color,sweetness,texture and nutritional qualities in acorn squash,or any other winter squash grown worldwide.To provide insight into winter squash fruit and seed development and add to the genomic resources in the Cucurbita genus,RNA sequencing was used to generate an acorn squash fruit and seed transcriptome from the cultivar Sweet REBA at critical points throughout fruit development.141838600 high-quality paired-end Illumina reads were assembled into 55949 unigenes.85%of unigenes with predicted open reading frames had homology with previously identified genes and over 62%could be functionally annotated.Comparison with the watermelon and cucumber genomes provided confirmation that the unigenes are full-length and comprehensive,covering an average of 90%of the coding sequence of their homologs and 72%of the cucumber and watermelon exomes.Key candidate genes associated with carotenoid and carbohydrate metabolism were identified toward a resource for winter squash fruit quality trait dissection.This transcriptome represents a major advance in C.pepo genomics,providing significant new sequence information and revealing the repertoire of genes expressed throughout winter squash fruit and seed development.Future studies on the genetic basis of fruit quality and future breeding efforts will be enhanced by tools and insights developed from this resource. | Lindsay E Wyatt Susan R Strickler Lukas A Mueller Michael Mazourek | 2015 | Horticulture Research2015，2，1： | 11 |
| 2 | Acinar cell injury induced by inadequate unfolded protein response in acute pancreatitis显示文摘Acute pancreatitis (AP) is an inflammatory disorder of pancreatic tissue initiated in injured acinar cells. Severe AP remains a significant challenge due to the lack of effective treatment. The widely-accepted autodigestion theory of AP is now facing challenges, since inhibiting protease activation has negligible effectiveness for AP treatment despite numerous efforts. Furthermore, accumulating evidence supports a new concept that malfunction of a self-protective mechanism, the unfolded protein response(UPR), is the driving force behind the pathogenesis of AP. The UPR is induced by endoplasmic reticulum(ER) stress, a disturbance frequently found in acinar cells, to prevent the aggravation of ER stress that can otherwise lead to cell injury. In addition, the UPR's signaling pathways control NFκB activation and autophagy flux, and these dysregulations cause acinar cell inflammatory injury in AP, but with poorly understood mechanisms. We therefore summarize the protective role of the UPR in AP, propose mechanistic models of how inadequate UPR could promote NFκB's pro-inflammatory activity and impair autophagy's protective function in acinar cells, and discuss its relevance to current AP treatment. | 3 | Comparative analysis of Cucurbita pepo metabolism throughout fruit development in acorn squash and oilseed pumpkin显示文摘Both the fruit mesocarp and the seeds of winter squash can be used for consumption,although the focus of breeding efforts varies by cultivar.Cultivars bred for fruit consumption are selected for fruit mesocarp quality traits such as carotenoid content,percent dry matter,and percent soluble solids,while these traits are essentially ignored in oilseed pumpkins.To compare fruit development in these two types of squash,we sequenced the fruit transcriptome of two cultivars bred for different purposes:an acorn squash,‘Sweet REBA’,and an oilseed pumpkin,‘Lady Godiva’.Putative metabolic pathways were developed for carotenoid,starch,and sucrose synthesis in winter squash fruit and squash homologs were identified for each of the structural genes in the pathways.Gene expression,especially of known rate-limiting and branch point genes,corresponded with metabolite accumulation both across development and between the two cultivars.Thus,developmental regulation of metabolite genes is an important factor in winter squash fruit quality. | Lindsay E Wyatt Susan R Strickler Lukas A Mueller Michael Mazourek | 2016 | Horticulture Research2016，3，1： | 7 |
| 4 | Acute pancreatitis in aging animals:Loss of pancreatitis-associated protein protection?显示文摘AIM:To investigate the effect of age on severity of acute pancreatitis(AP) using biochemical markers,histology and expression of the protective pancreatitisassociated proteins(PAPs).METHODS:AP was induced via intraductal injection of 4% sodium taurocholate in young and old rats.Sera and pancreata were assayed at 24 h for the parameters listed above;we also employed a novel molecular technique to assess bacterial infiltration using polymerase chain reaction to measure bacterial genomic ribosomal RNA.RESULTS:At 24 h after induction of AP,the pancreata of older animals had less edema(mean ± SE histologic score of young vs old:3.11 ± 0.16 vs 2.50 ±-0.11,P < 0.05),decreased local inflammatory response(histologic score of stromal infiltrate:3.11 ± 0.27 vs 2.00 ± 0.17,P < 0.05) and increased bacterial infiltration(174% ± 52% increase from sham vs 377% ± 4%,P < 0.05).A decreased expression of PAP1 and PAP2 was demonstrated by Western blotting analysis and immunohistochemical staining.There were no differences in serum amylase and lipase activity,or tissue myeloperoxidase or monocyte chemotactic protein-1 levels.However,in the most-aged group,serum C-reactive protein levels were higher(young vs old:0.249 ± 0.04 mg/dL vs 2.45 ± 0.68 mg/dL,P < 0.05).CONCLUSION:In older animals,there is depressed PAP expression related to a blunted inflammatory response in AP which is associated with worsened bacterial infiltration and higher C-reactive protein level;this may explain the more aggressive clinical course. | Sophia Fu Albert Stanek Cathy M Mueller Nefertti A Brown Chongmin Huan Martin H Bluth Michael E Zenilman | 2012 | World Journal of Gastroenterology2012，18，26： | 5 |
| 5 | Cachexia and pancreatic cancer: Are there treatment options?显示文摘Cachexia is frequently described in patients with pancreatic ductal adenocarcinoma(PDAC)and is associated with reduced survival and quality of life.Unfortunately,the therapeutic options of this multi-factorial and complex syndrome are limited.This is due to the fact that,despite extensive preclinical and clinical research,the underlying pathological mechanisms leading to PDAC-associated cachexia are still not fully understood.Furthermore,there is still a lack of consensus on the definition of cachexia,which complicates the standardization of diagnosis and treatment as well as the analysis of the current literature.In order to provide an efficient therapy for cachexia,an early and reliable diagnosis and consistent monitoring is required,which can be challenging especially in obese patients.Although many substances have been tested in clinical and preclinical settings,so far none of them have been proven to have a long-term effect in ameliorating cancer-associated cachexia.However,recent studies have demonstrated that multidimensional therapeutic modalities are able to alleviate pancreatic cancerassociated cachexia and ultimately improve patients’outcome.In this current review,we propose a stepwise and pragmatic approach to facilitate and standardize the treatment of cachexia in pancreatic cancer patients.This strategy consists of nutritional,dietary,pharmacological,physical and psychological methods. | Tara C Mueller Marc A Burmeister Jeannine Bachmann Marc E Martignoni | 2014 | World Journal of Gastroenterology2014，20，28： | 3 |
