| 1 | Higher PD-1 expression concurrent with exhausted CD8+ T cells in patients with de novo acute myeloid leukemia显示文摘Objective: To investigate the association between the T cell inhibitory receptor programmed death 1(PD-1)and T cell exhaustion status in T cells from patients with de novo acute myeloid leukemia(AML) and AML in complete remission(CR).Methods:Surface expression of PD-1 and the exhaustion and immunosenescence markers CD244 and CD57 on CD3+,CD4+ and CD8+ T cells from peripheral blood samples from 20 newly diagnosed,untreated AML patients and 10 cases with AML in CR was analyzed by flow cytometry.Twenty-three healthy individuals served as control.Results:A significantly higher percentage of PD-1+ cells were found for CD3+ T cells in the de novo AML group compared with healthy controls.In addition,an increased level of PD-1+ CD8+ T cells,but not PD-1+ CD4+,was found for CD3+ T cells in the de novo AML and AML-CR samples.A higher percentage of CD244+ CD4+,CD244+ CD8+,CD57+ CD4+ and CD57+ CD8+ T cells was found in CD3+ T cells in samples from those with de novo AML compared with those from healthy controls.Strong increased PD-1+ CD244+ and PD-1+ CD57+ coexpression was found for CD4+ and CD8+ T cells in the de novo AML group compared with healthy controls.Conclusions:We characterized the major T cell defects,including co-expression of PD-1 and CD244,CD57-exhausted T cells in patients with de novo AML,and found a particular influence on CD8+ T cells,suggesting a poor anti-leukemia immune response in these patients. | Jiaxiong Tan Shaohua Chen Yuhong Lu Danlin Yao Ling Xu Yikai Zhang Lijian Yang Jie Chen Jing Lai Zhi Yu Kanger Zhu Yangqiu Li | 2017 | Chinese Journal of Cancer Research2017,29,5: | 10 |
| 2 | Improvement potential analysis for integrated fractionating and heat exchange processes in delayed coking units显示文摘A novel diagram-based representation approach is developed to analyze the thermodynamic efficiency and identify quickly the promising energy-use improvement for integrated fractionating and heat exchange processes in delayed coking units. For considering temperature dependence of heat capacity and integrating fractionating and heat exchange processes, an advanced energy level composite curve is constructed by using the simulation results and a stepwise procedure. More accurate results of exergy analysis are obtained and the interaction between different components of the integrated system can be properly revealed in an integrated figure. Then the exergy calculation is performed to validate the performance of processes and to define the targets for improvement. The avoidable exergy destruction is also analyzed by applying the concepts of avoidable and unavoidable exergy destructions for the integrated system. In a case study for a Chinese refinery, the results reveal that the heat exchange between gas oil and deethanization gasoline is the most inefficient process with the highest retrofitting potential, and the lowest exergy efficiency of component in the integration system is only29.4%. | 6 | Human myoblast genome therapy显示文摘Human Myoblast Genome Therapy (HMGT) is a platform technology of cell transplantation, nuclear transfer, and tissue engineering. Unlike stem cells, myoblasts are differentiated, immature cells destined to become muscles. Myoblasts cultured from satellite cells of adult muscle biopsies survive, develop, and function to revitalize degenerative muscles upon transplantation. Injection injury activates regeneration of host myofibers that fuse with the engrafted myoblasts, sharing their nuclei in a common gene pool of the syncytium. Thus, through nuclear transfer and complementation, the normal human genome can be transferred into muscles of patients with genetic disorders to achieve phenotype repair or disease prevention. Myoblasts are safe and efficient gene transfer vehicles endogenous to muscles that constitute 50% of body weight. Results of over 280 HMGT procedures on Duchenne Muscular Dystrophy (DMD) subjects in the past 15 years demonstrated absolute safety. Myoblast-injected DMD muscles showed improved histology. Strength increase at 18 months post-operatively averaged 123%. In another application of HMGT on ischemic cardiomyopathy, the first human myoblast transfer into porcine myocardium revealed that it was safe and effective. Clinical trials on approximately 220 severe cardiomyopathy patients in 15 countries showed a <10% mortality. Most subjects received autologous cells implanted on the epicardial surface during coronory artery bypass graft, or injected on the endomyocardial surface percutaneously through guiding catheters. Significant increases in left ventricular ejection fraction, wall thickness, and wall motion have been reported, with reduction in perfusion defective areas, angina, and shortness of breath. As a new modality of treatment for disease in the skeletal muscle or myocardium, HMGT emerged as safe and effective. Large randomized multi-center trials are under way to confirm these preliminary results. | 10 | Dielectric properties of saline soil based on a modified Dobson dielectric model显示文摘Soil salinization is a major concern for agricultural development in arid areas. In this paper, a modified Dobson dielectric model was applied to simulate the dielectric constant of saline soil in the Ugan-Kuqa river delta oasis of Xinjiang Uygur autonomous region, northwestern China. The model performance was examined through analyzing the influences of its parameters on the soil dielectric constant and the relationship between radar backscattering coefficient and the dielectric constant of saline soil. The results of the study indicate that:(1) The real part of the soil dielectric constant is affected by soil water content at low radar frequencies; the imaginary part is closely related with both the soil water content and soil salt content.(2) The soil water and salt contents are related with the coefficient of dialectical loss, which is consistent with the natural conditions of saline soil in arid areas and provides valuable references for the study of soil dielectric properties.(3) The changes of soil water content and soil salt content have instant influences on the dielectric constant of saline soil. Subsequently, the radar backscattering coefficient is affected to respond to the dielectric constant of saline soil. The radar backscattering coefficient is most responsible to the radar's cross polarization pattern with a correlation coefficient of R2=0.75. | 20 | Distribution and gas-bearing properties of Permian igneous rocks in Sichuan Basin, SW China显示文摘Based on the analysis of outcrop, seismic, logging and drilling data, combined with exploration practice, the characteristics,distribution, reservoir performance and gas-bearing properties of Permian igneous rocks in Sichuan Basin are studied. The study shows that central volcanic eruptive facies are developed in Sichuan Basin, and their lithological assemblages and distribution characteristics show obvious differences. The igneous rocks are mainly distributed in three regions: the southwestern part of the basin has dominantly largescale overflow facies basalts; the central and western part of the basin, Jianyang-Santai area, develop intrusive rocks, volcanic lavas(basalts)and pyroclastic rocks; and the eastern part of Sichuan, Dazhou-Liangping area, only develop diabase and basalts. Five aspects of understandings are achieved:(1) The Upper Permian igneous rocks can be divided into intrusive rocks and extrusive rocks, with the extrusive rocks as the main body. The chemical compositions of the extrusive rocks are characterized by both alkaline basalt and tholeiitic basalt, and belong to the subalkaline type of transitional basalt magma eruption.(2) There are obvious rhythmic structures vertically among overflow facies basalt, and the single rhythmic layer consists of, from bottom up, pyroclastic rocks(undeveloped), gray and dark gray porphyritic basalts(unstable), dark gray and purple microcrystalline-cryptocrystalline basalts, dark greyish green porous and amygdaloid basalts; the central volcanic eruption shows the rhythm and the vertical sequence of volcanic clastic rocks(agglomerates and breccias), volcanic lava, tuffaceous lava from bottom to top.(3) The pore types of basalt and pyroclastic rocks are diverse, mainly dissolution pore and de-vitrification micropore, but their physical properties are different. Basalt is characterized by ultra-low pore permeability, small reservoir thickness, and reservoirs are distributed in the upper and middle parts of the cycle, with poor lateral comparability. Volcanic clastic rocks are medium to high porous reservoirs(Well YT1: porosity: 8.66%?16.48%, average 13.76%) with large thickness and good reservoir quality.(4) Natural gas in basalts in southwestern basin mainly comes from Middle Permian, and natural gas in volcanic clastic rocks in central and western basin comes from Cambrian Qiongzhusi Formation.(5) Analysis of igneous reservoir-forming conditions in different areas shows that there are relatively insufficient gas sources and great differences in preservation conditions in southwestern basin.Reservoirs are poorly developed and gas-bearing is complex.
