| 1 | Programmed death-1 upregulation is correlated with dysfunction of tumor-infiltrating CD8^(+ ) T lymphocytes in human non-small cell lung cancer显示文摘T-cell tolerance is an important mechanism for tumor escape,but the molecular pathways involved in T-cell tolerance remain poorly understood.It remains unknown whether the inhibitory immunoreceptor programmed death-1(PD-1)plays a role in conditions of human non-small cell lung cancer(NSCLC).In this study,we detected PD-1 expression on CD81 T cells from healthy control peripheral blood mononuclear cells(PBMCs)and the PBMCs of NSCLC patients as well as NSCLC tissues.Results showed that tumor-infiltrating CD81 T cells had increased PD-1 expression and impaired immune function,including reducing cytokine production capability and impairing capacity to proliferate.Blockade of the PD-1/PD-L1 pathway by the PD-L1-specific antibody partially restored cytokine production and cell proliferation.These data provide direct evidence that the PD-1/PD-L1 pathway is involved in CD81 T-cell dysfunction in NSCLC patients.Moreover,blocking this pathway provides a potential therapy target in lung cancer. | Yan Zhang Shengdong Huang Dejun Gong Yanghua Qin Qian Shen | 2010 | Cellular & Molecular Immunology2010,7,5: | 48 |
| 2 | Identification and mapping of genes for improving yield from Chinese common wild rice (O.rufipogon Griff.) using advanced backcross QTL analysis显示文摘To identify useful genes from wild rice which have been lost or weakened in cultivated rice has become more and more important for modern breeding strategy. In this study, a BC4 population derived from 94W1, an accession of common wild rice (Oryza rufipogon Griff.) from Dongxiang in Jiangxi Province of China, as the donor, and a high-yielding Indica cultivar (O. sativa L.), 'Guichao 2', as the recipient, was used to identify quantitative trait loci (QTL) associated with yield and its components. Based on the analysis for the genotype of BC4F1 population with 87 SSR markers distributed throughout the genome and investigation of the plant height, yield and yield components of BC4F2, a total of 52 QTLs, were detected. Of 7 QTLs associated with grain yield per plant, 2 QTLs on chromosome 2 and chromosome 11 for grain yield, explaining 16% and 11% of the phenotypic variance respectively, were identified. | 4 | Identification and mapping of quantitative trait loci controlling cold-tolerance of Chinese common wild rice (O. rufipogon Griff.) at booting to flowering stages显示文摘An advanced backcross population of rice was used to identify the quantitative trait locus (QTL) controlling the cold-tolerance at booting to flowering stages. | 7 | Predicting the spread of nuclear radiation from the damaged Fukushima Nuclear Power Plant显示文摘Japan suffered a M9.0 earthquake and massive tsunami on March 11, 2011, which seriously damaged the Fukushima Nuclear Power Plant and caused a nuclear crisis. The spread of nuclear radiation from the power plant through the atmosphere and ocean was predicted with a short-term climate forecasting model and an ocean circulation model under some idealized assumptions. If nuclear matter were leaked in the near-ground layer of 992 hPa, the climate model results show that the nuclear radiation would cover North America 10 days after the initial leakage, with the concentration at the forefront dramatically reduced to 10 millionths of the initial model concentration at the source. The radiation would span Europe in 15 days and cover much of the Northern Hemisphere in 30 days. If the initial leakage was assumed to occur in the layer 5000-m above the ground, the radiation would cover Europe in 10 days and cover much of the Northern Hemisphere in 15 days. Moreover, under the assumption that the nuclear matter leaked in the 10000-m layer, the radiation would affect much of China after 10 days. The ocean circulation model indicates that the nuclear material would be slowly transported northeast of Fukushima and reach 150°E in 50 days, and the nuclear debris in the ocean would be confined to a narrow band. Compared with the spread in the ocean, the area affected by leaked nuclear radiation in the atmosphere would be very large. In our retrospective analysis there was an early assumption of at least three genes at or near the locus, which caused an erroneous presumption of genetic diversity for resistance. | 12 | Status and distribution changes of the endangered Green Peafowl (Pavo muticus) in China over the past three decades (1990s-2017)显示文摘Background: Understanding the changes in population dynamics, including demographics, distribution and threats, is essential for species status assessing. The endangered Green Peafowl(Pavo muticus) has experienced sharp population declines and distribution range diminishing both in China and Southeast Asia. Field population surveys have not been conducted in China since the 1990 s, which hindered conservation planning and decision-making.Methods: With interview and line transects methods, we figured out the population and distribution changes of Green Peafowl across its historical ranges over the past three decades in China during 2014-2017.Results: The Green Peafowl once habituated in 54 counties in China. Nearly 60% of the distribution counties were lost in the past three decades, with the left 22 counties distributed in central, southern and western Yunnan, SW China. Population decrease detected in all distribution areas except for Shuangbai and Xinping county where more than 60% of the total population is located. Only about 30% of the former bird population were recorded with the same interviewing method as 20 years ago. Three birds, 1 carcass, 6 calls and 12 footprints were detected along the 865 km line transects, indic-ating extremely low encounter rate of Green Peafowl in field. Sharp decreases in flock sizes were also detected, from 820 birds per flock in the 1990 s to 3-5 birds at present. Poaching and habitat conversion are two widespread and long-lasting threats, while poisoning caused mortality in the past and hydropower construction affect regional population's survival. Large flocks of 18–27 birds were discovered in the field, which increases our confidence of population recovery of this endangered pheasant in China.Conclusions: Only interviewed bird number and counts based on line transects were presented in this study, without further population estimation due to limitation of the data sets. | 14 | Lopingian mixed floras from Linxi Formation in Soron area,Inner Mongolia显示文摘A lot of well-preserved plants from the Linxi Formation are collected in the Soron area,Inner Mongolia,consisting of 34 species of 16 genera. They are Late Permian in age. The flora is characterized by a lot of Angaran plants,such as Paracalamites,Comia,Rhachiphyllum,Iniopteris,Rufloria,etc. Some Cathaysian elements,such as Lobatannularia lingulata,L. multifolia,Fascipteris Cathaysiantus,F. densata,Taeniopteris szei,Cladophlebis liulinensis and C. permica,are also mixed within the present flora. This indicates that the Soron of Inner Mongolia was located in the sector between the Angaran and Cathysian floristic provinces in Lopingian. It is beneficial for depicting the location and the evolution of the Solenker--Xar Moron suture zone in the phytogeographic view. | 17 | CRISPR/Cas9-mediated multiple gene editing in Brassica oleracea var.capitata using the endogenous tRNA-processing system显示文摘Cabbage(Brassica oleracea var.capitata)is a biennial plant with strong self-incompatibility and an obligate requirement for prolonged vernalization by exposure to low temperatures to induce flowering.These characteristics significantly increase the difficulty of exploiting novel germplasm induced by physical or chemical mutagens.In this study,we report a CRISPR/Cas9 gene-editing system based on endogenous tRNA processing to induce high efficiency and inheritable mutagenesis in cabbage.Using the phytoene desaturase gene BoPDS,the S-receptor kinase gene BoSRK,and the male-sterility-associated gene BoMS1 as the target genes,multisite and multiple gene mutations were achieved using a construct with tandemly arrayed tRNA-sgRNA architecture to express multiple sgRNAs.The BoSRK3 gene mutation suppressed self-incompatibility completely,converting the self-incompatible line into a self-compatible line.In addition,the BoMS1 gene mutation produced a completely male-sterile mutant,which was highly cross compatible with its nonmutant isoline at the flowering stage as a result of a simultaneous BoSRK3 gene mutation,enabling the economic propagation of the male-sterile line through bee-mediated cross-pollination.Interestingly,higher site mutation efficiency was detected when a guide sequence was inserted into a location in the tandemly arrayed tRNA-sgRNA architecture that was distal from the upstream Pol III promoter.In addition,mutation sites were also detected in the paralogous genes of the BoPDS and BoSRK genes that had fully consistent sequences or base mismatches but beyond the“seed”region in the spacer sequence compared with the target sgRNAs.Collectively,our results demonstrate that the CRISPR/Cas9 system,coupled with an endogenous tRNA-processing system,is an efficient tool to improve cabbage traits. | Cunfa Ma Chenzeng Zhu Min Zheng Mengci Liu Dejun Zhang Baoli Liu Qinfei Li Jun Si Xuesong Ren Hongyuan Song | 2019 | Horticulture Research2019,6,1: | 8 |

| 19 | Effects of complexing agents on acidic electroless nickel deposition显示文摘Our previous study reported the influences of different complexing agents on electroless nickel(EN) by examining the properties of the deposits.In the present work,the effects of four common-used complexing agents on EN deposition rate and the stability of solution pH values were examined,either with an acetic pH buffer agent or absent of them.It is indicated that the pH buffering effect of them is dominative when the EN solution is lack of the pH buffer.Under this situation,the EN deposition rate increases with the concentration of complexing agents increasing.The EN deposition rate decreases with further adding the complexing agent when the solution already has enough pH buffer capability.Electrochemical impedance spectroscopy obtained during EN deposition illustrates that,in this case,the enhanced reaction resistance is the main reason for a lower deposition rate.However,the influence of polarization caused by mass transfer is not negligible at high complex ratio for sodium citrate and malic acid EN solutions. | JIN Ying YU Hongying YANG Dejun SUN Dongbai | 2010 | Rare Metals2010,29,4: | 6 |
| 20 | Synthesis of ZrAlN coatings with thermal stability at high temperature显示文摘Dry machining will result in elevated temperatures at the tool surface (800—1000℃). So, coating materials that can provide protection for cutting tools at these temperatures are of great technological interests. ZrAlN coating is proposed to possess high-temperature stable structural and mechanical properties due to the addition of the alloying element. ZrAlN coatings were grown using a dc reactive magnetron sputtering. The XRD and nano indenter were employed to investigate the effects of reaction gas partial pressure and substrate bias on structural and mechanical properties, as well as high-temperature stability. The ZrAlN coating, when deposited under optimum conditions (-37 V substrate bias and 2×10-5 Pa N2 partial pressure), showed smooth surface with thermal stable hardness. Its internal stress was relaxed from 2.2 to 0.7 GPa after anneal- ing.
