| 1 | IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE. | Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,7: | 6 |
| 2 | Coordinated transcription of ANRIL and P16 genes is silenced by P16 DNA methylation显示文摘Objective: To investigate the relationship between the transcription of ANRIL, P15, P14 and P16 at the same locus and the regulation mechanism of ANRIL.Methods: Publicly available database of Cancer Cell Line Encyclopedia(CCLE) was used in bioinformatic analyses. Methylation of Cp G islands was detected by denaturing high performance liquid chromatography(DHPLC). Gene transcript levels were determined using quantitative real-time polymerase chain reaction(q RTPCR) assays. An engineered P16-specific transcription factor and DNA methyltransferase were used to induce P16-specific DNA demethylation and methylation.Results: The expression level of ANRIL was positively and significantly correlated with that of P16 but not with that of P15 in the CCLE database. This was confirmed in human cell lines and patient colon tissue samples. In addition, ANRIL was significantly upregulated in colon cancer tissues. Transcription of ANRIL and P16 was observed only in cell lines in which the P16 alleles were unmethylated and not in cell lines with fully methylated P16 alleles.Notably, P16-specific methylation significantly decreased transcription of P16 and ANRIL in BGC823 and GES1 cells. | 9 | Differentiation and adaptation epigenetic networks:Translational research in gastric carcinogenesis显示文摘There are several kinds of epigenetic networks in the human body including the cell differentiation epigenetic network(DiEN) and the host adaptation epigenetic network(AdEN).DiEN networks are static and cell/tissue-specific.AdEN networks are variable and dependent upon environmental factors.DiEN and AdEN alterations can respectively serve as biomarkers for different kinds of diseases.Cancer is a consequence of accumulated pathophysiological adaptations of tissue stem cells to exposure of environmental carcinogens.Cancer cells are de-differentiated cells that obtain the capacity of unrestricted proliferation,local invasion,and distant migration/metastasis.Both DiEN and AdEN changes can be observed in cancer tissues.Alterations of DNA methylation are the most stable epigenetic modifications and can be sensitively detected in a small cell population.These advantages make DNA methylation the optimal biomarkers for detection of initiated cells in precancerous lesions and metastasis stem cells in cancer tissues.It has been proven that p16 methylation can be used as a diagnostic biomarker to determine malignant potential of epithelium dysplasia in many organs including the stomach.In a large-scale validation study on the DNA methylome of gastric carcinomas(GC),the methylation status of more than 90 CpG islands has been analyzed by DHPLC.Furthermore,GFRA1 demethylation and methylation of SRF and ZNF382 are frequent events during gastric carcinogenesis and consistently correlate to GC metastasis and overall survival of GC patients from China,Japan,and Korea,respectively.In a population study,it has been demonstrated that gradual increasing of plasma miR-211 and other miRNA levels may be an early risk predictor for GC development. | DENG DaJun LU ZheMing | 2013 | Chinese Science Bulletin2013,58,1: | 1 |

| 13 | Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model. | 15 | 放疗敏感性不同宫颈癌的蛋白质组学比较研究(英文)显示文摘Objective:To investigate the proteomic differences between the high-sensitivity(HS) group and low-sensitivity(LS) group of cervical cancer treated by radiotherapy and confirm the radiotherapy sensitivity associated proteins in early cer-vical cancer.Methods:The fresh carcinoma tissues were collected from 10 untreated cervical cancer patients and preserved in the-80 ℃ refrigeratory.The tissues were classified into two groups:high sensitivity group(HS) and low sensitivity group(LS),according to their response to radiotherapy.In the first part of our experiment,protein separating was performed by using two-dimensional gel electrophoresis(2-DE) with Amersham 18 cm linear pH 3-10 immobilized pH gradient(IPG) strips.The images of the gels were acquired by the scanner and then analyzed by using PD-quest7.3 software to find the differentially expression protein-spots in each group.Then the differentially expressed protein-spots was incised from the gels and digested by trypsin.The peptide mass fingerprintings(PMF) was acquired by matrix assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) and the proteins were identified by data searching in the Mascot-database.Part of dif-ferentially expression proteins were assayed by Western Blot.Results:Most of the gels were clear and successfully analyzed by PD-quest7.3 software.Most of the protein-spots concentrated on the area of 20-100Kda(Mw) and pH4-8.The average number of the protein-spots was 754 ± 64 in HS group and 777 ± 48个in LS group.The match rate was 87.6% between two groups.Five high expression proteins were found in HS group which were low expression in LS group,3 high expres-sion protein were found in LS group which were low expression in HS group.Reselts of Western Blot were in coincidence to proteomic result.Conclusion:The 2-DE gels image of HS group and LS group with early cervical cancer tissues treated by radiotherapy are successfully acquired.Some differentially expression proteins between the two groups are further confirmed by immunohistochemical assay. | Liang Zeng Hong Zhu Dajun Li Haiping Pei Yaping Deng Jun Yuan | 2009 | The Chinese-German Journal of Clinical Oncology2009,8,4: | 0 |
| 16 | FPGA based digital phase-coding quantum key distribution system显示文摘Quantum key distribution(QKD) is a technology with the potential capability to achieve information-theoretic security. Phasecoding is an important approach to develop practical QKD systems in fiber channel. In order to improve the phase-coding modulation rate, we proposed a new digital-modulation method in this paper and constructed a compact and robust prototype of QKD system using currently available components in our lab to demonstrate the effectiveness of the method. The system was deployed in laboratory environment over a 50 km fiber and continuously operated during 87 h without manual interaction. The quantum bit error rate(QBER) of the system was stable with an average value of 3.22% and the secure key generation rate is 8.91 kbps.
