1IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6
2Numerical Simulation of 2D Supersonic Magnetohydrodynamic Channel and Study on Hall Effect显示文摘In this research effort,numerical simulation of two-dimensional magnetohydrodynamic(MHD) channel is performed and Hall effect is studied.The computational model consists of the Navier-Stokes(N-S) equations coupled with electrical-magnetic source terms,Maxwell equations and the generalized Ohm's law.Boundary conditions for the electrical potential equation considering Hall effect are derived.To start with,the MHD channel with single-pair electrodes is studied and flow of the electric current is in accordance with physical principle.Then the MHD channel with five-pair electrodes is numerically simulated.The results show that the electrical current concentrates on the downstream of the anode and the upstream of the cathode due to Hall effect,and the flow field becomes asymmetrical.At the current value of the magnetic interaction parameter,the electrical-magnetic force affects the flow remarkably,decreasing the outlet Mach number and increasing the outlet pressure;what's more,the flow structure in the channel becomes extremely complex.Performances of MHD channels with continual electrodes and segmented electrodes are compared.The results show that performance of the MHD channel with segmented electrodes is better than that with continual electrodes with the increase of Hall parameter.ZHENG Xiaomei LU Haoyu XU Dajun CAI Guobiao 2011Chinese Journal of Aeronautics2011,24,2:3
3Optimal Control of Slurry Pressure during Shield Tunnelling Based on Random Forest and Particle Swarm Optimization显示文摘The control of slurry pressure aiming to be consistent with the external water and earth pressure during shield tunnelling has great significance for face stability,especially in urban areas or underwater where the surrounding environment is very sensitive to the fluctuation of slurry pressure.In this study,an optimal control method for slurry pressure during shield tunnelling is developed,which is composed of an identifier and a controller.The established identifier based on the random forest(RF)can describe the complex non-linear relationship between slurry pressure and its influencing factors.The proposed controller based on particle swarm optimization(PSO)can optimize the key factor to precisely control the slurry pressure at the normal state of advancement.A data set from Tsinghua Yuan Tunnel in China was used to train the RF model and several performance measures like R2,RMSE,etc.,were employed to evaluate.Then,the hybrid RF-PSO control method is adopted to optimize the control of slurry pressure.The good agreement between optimized slurry pressure and expected values demonstrates a high identifying and control precision.Weiping Luo Dajun Yuan Dalong Jin Ping Lu Jian Chen 2021Computer Modeling in Engineering & Sciences2021,,7:3

5A Block Parallel l_0-Norm Penalized Shrinkage and Widely Linear Affine Projection Algorithm for Adaptive Filter显示文摘To improve the identification capability of AP algorithm in time-varying sparse system, we propose a block parallel l_0-SWL-DCD-AP algorithm in this paper. In the proposed algorithm, we first introduce the l_0-norm constraint to promote its application for sparse system. Second, we use the shrinkage denoising method to improve its track ability. Third, we adopt the widely linear processing to take advantage of the non-circular properties of communication signals. Last, to reduce the high computational complexity and make it easy to implemented, we utilize the dichotomous coordinate descent(DCD) iterations and the parallel processing to deal with the tapweight update in the proposed algorithm. To verify the convergence condition of the proposed algorithm, we also analyze its steadystate behavior. Several simulation are done and results show that the proposed algorithm can achieve a faster convergence speed and a lower steady-state misalignment than similar APA-type algorithm. 13Kaiso mainly locates in the nucleus in vivo and binds to methylated,but not hydroxymethylated DNA显示文摘Objective:Kaiso is upregulated in many cancers and proposed to bind with both methylated- and unmethylated-DNA in the nucleus as a transcriptional repressor.The objective is to define its subcellular localization in vivo and exact binding DNA sequences in cells.Methods:Compartmentalization of exogenous Kaiso in cells was tracked with enhanced green fluorescence protein(EGFP) tag.The endogenous Kaiso expression in gastric carcinoma tissue was examined with immunohistochemical staining.Kaiso-DNA binding was tested using electrophoretic mobility shift assay(EMSA) and chromatin immunoprecipitation assay(ChIP).Results:Kaiso mainly localized in the nucleus of cancer and stromal cells in vivo,but remained in the cytoplasm of cultured cells.Most importantly,nuclear Kaiso can bind with the methylated-CGCGcontaining sequence in the CDKN2 A promoter,but not with the hydroxymethylated-CGCG sequence in HCT116 cells.Conclusions:Kaiso locates mainly in the nucleus in vivo where it binds with the methylated-CGCG sequences,but does not bind with the hydroxymethylated-CGCG sequences.Sisi Qin Baozhen Zhang Wei Tian Liankun Gu Zheming Lu Dajun Deng 2015Chinese Journal of Cancer Research2015,27,2:1
14Differentiation and adaptation epigenetic networks:Translational research in gastric carcinogenesis显示文摘There are several kinds of epigenetic networks in the human body including the cell differentiation epigenetic network(DiEN) and the host adaptation epigenetic network(AdEN).DiEN networks are static and cell/tissue-specific.AdEN networks are variable and dependent upon environmental factors.DiEN and AdEN alterations can respectively serve as biomarkers for different kinds of diseases.Cancer is a consequence of accumulated pathophysiological adaptations of tissue stem cells to exposure of environmental carcinogens.Cancer cells are de-differentiated cells that obtain the capacity of unrestricted proliferation,local invasion,and distant migration/metastasis.Both DiEN and AdEN changes can be observed in cancer tissues.Alterations of DNA methylation are the most stable epigenetic modifications and can be sensitively detected in a small cell population.These advantages make DNA methylation the optimal biomarkers for detection of initiated cells in precancerous lesions and metastasis stem cells in cancer tissues.It has been proven that p16 methylation can be used as a diagnostic biomarker to determine malignant potential of epithelium dysplasia in many organs including the stomach.In a large-scale validation study on the DNA methylome of gastric carcinomas(GC),the methylation status of more than 90 CpG islands has been analyzed by DHPLC.Furthermore,GFRA1 demethylation and methylation of SRF and ZNF382 are frequent events during gastric carcinogenesis and consistently correlate to GC metastasis and overall survival of GC patients from China,Japan,and Korea,respectively.In a population study,it has been demonstrated that gradual increasing of plasma miR-211 and other miRNA levels may be an early risk predictor for GC development.DENG DaJun LU ZheMing 2013Chinese Science Bulletin2013,58,1:1

19Hydraulic fracturing pressure of concentric double-layered cylinder in cohesive soil显示文摘This study aims to investigate hydrofracturing in double-layered soil through theoretical and experimental analysis,as multilayered soils where the difference in mechanical properties exists are generally encountered in practical engineering.First,an analytical solution for fracturing pressure in two different concentric regions of soil was presented based on the cavity expansion theory.Then,several triaxial hydraulic fracturing tests were carried out to validate the analytical solution.The comparison between the experimental and analytical results indicates the remarkable accuracy of the derived formula,and the following conclusions were also obtained.First,there is a linear relationship between the fracturing pressure and confining pressure in concentric double-layered cohesive soil.Second,when the internal-layer soil is softer than the external-layer soil,the presence of internal soil on the fracturing pressure approximately brings the weakening effect,and the greater strength distinction between the two layers,the greater the weakening effect.Third,when the internal-layer soil is harder than the external-layer soil,the existence of the internal-layer soil has a strengthening effect on the fracturing pressure regardless of the proportion of internal-layer soil.Moreover,the influence of strength distinction between the two layers on the fracturing pressure is significant when the proportion of internal-layer soil is less than half,while it’s limited when the proportion is more than half.The proposed solution is potentially useful for geotechnical problems involving aspects of cohesive soil layering in a composite formation.Dajun YUAN Weiping LUO Dalong JIN Ping LU 2021Frontiers of Structural and Civil Engineering2021,15,4:0
20A Symbol-Based Passband Doppler Tracking and Compensation Algorithm for Underwater Acoustic DSSS Communications显示文摘In this paper,we consider the problem of Doppler tracking and compensation for a direct sequence spread spectrum(DSSS)signal in underwater acoustic(UWA)communication.Since the dynamic property of the UWA channel and the long duration of DSSS signals result in significant Doppler spread that severely distorts the propagated signal,Doppler tracking and compensation are required.Based on the ultra-wideband property of UWA signal,the Doppler spread not only results in the frequency shift,but also changes the signal duration.Therefore,the accurate estimation of the signal expansion/compression in the time domain can reflect the Doppler spread.Accordingly,we present a Doppler tracking and compensation algorithm for a DSSS signal operating on the correlation output of passband signal at a symbol-by-symbol basis.Note that the carrier frequency of UWA communication is around several kilohertz,and thus the time delay estimation can be performed on the passband to improve the accuracy.Furthermore,the prior information of Doppler limit is used to refine the resolution of delay estimation and achieve sequential estimation.To compensate the correlation magnitude distortion induced by the velocity variation,the local reference signal is selected adaptively based on the filtered Doppler factor.Simulation results demonstrate that the proposed passband Doppler tracking algorithm achieves a superior performance compared with the conventional receiver.Dajun Sun Xiaoping Hong Hongyu Cui Lu Liu 2020Journal of Communications and Information Networks2020,5,2:0
