| 1 | Effects of modified Wenjing decoction on microcirculation in reproductive organs in rats with symptom patterns of cold coagulation and blood stasis显示文摘OBJECTIVE: To investigate the relationship between symptom patterns of cold coagulation and blood stasis(CCBS) and microcirculation disturbance. In addition, we determined the efficacy of modified Wenjing decoction(WJD) for the treatment of CCBS.METHODS: CCBS was induced in rats with an icewater bath treatment. The ovarian function, microvascular and circulatory status of reproductive organs, and function of local microvascular endothelial cells(VECs) and vascular smooth muscle cells(VSMCs) were evaluated.RESULTS: Ovarian dysfunction was observed in the rats with CCBS. It was characterized by the presence of an estrous cycle disorder and a decrease in reproductive hormone levels. Microvascular circulation disorders were associated with an imbalance in vasoconstriction, relaxation substances, nitric oxide, abnormal blood flow in whole blood, and decreased blood flow in the auricle and uterus. VECs were damaged, and VSMCs contracted and proliferated in ovarian and uterine tissues.CONCLUSION: Our findings suggest that the dysfunctional reproductive organs observed in gynecological CCBS may be closely related to the microcirculation disturbance of local tissues, microvascular contraction, and vascular remodeling. | 3 | Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART. | Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu | 2021 | Cell Research2021,31,8: | 2 |

| 9 | Feeding Effect of Growing-Finishing Pigs under Fine Division of Feeding Stages显示文摘In order to study the feeding effect of fine division of feeding stages on the growing-finishing pigs,96 healthy Duroc × Landrace ×Yorkshire hybrid commercial pigs with similar age in days,size,and body weight about 15 kg,half boars and half sows,randomly divided into3 treatments( each treatment repeated 4 times,and each repetition 8 pigs). According to the body weight division standard,three stages,four stages,and five stages of feeding were adopted separately. The daily ration of with different nutrition levels was used,body weight and feed consumption were measured,and effects of different feeding methods on body weight increase,feed utilization,and economic benefits of growing-finishing pigs. The results showed that compared with the three-stage feeding,the body weight of five-stage and four-stage feeding increased by 4. 18%( P < 0. 01) and 2. 41%( P < 0. 05),respectively,and the body weight of five-stage feeding increased by 1. 74%( P < 0. 05) compared with that of four-stage feeding; the daily feed consumption of five-stage and four-stage feeding increased 3. 10%( P < 0. 01) and 1. 70%(P <0. 01),respectively,and the daily feed consumption of five-stage feeding was 1. 37% higher than that of four-stage feeding. The feed cost of five-stage feeding and four-stage feeding increased 3. 32%( P < 0. 01) and 1. 61%( P < 0. 05) separately,and the feed cost of five-stage feeding was 1. 69% higher than that of four-stage feeding( P < 0. 01). The income from average body weight increase of five-stage and fourstage feeding increased by 5. 47%( P < 0. 01) and 3. 59%( P < 0. 01) respectively,and the benefit of average body weight increase of fivestage feeding was 1. 81% higher than that of four-stage feeding( P > 0. 05). | 11 | Extraction Process of Total Flavonoids from Eucommiae Cortex and Its in vitro Antioxidant Activity显示文摘[Objectives]To optimize the extraction process of total flavonoids from Eucommiae Cortex,establish the extraction and content determination methods of its medicinal materials,and study its in vitro antioxidant activity.[Methods]The total flavonoids from Eucommiae Cortex were extracted by reflux extraction method,and the effects of extraction method,extraction solvent concentration,extraction volume,and extraction time on the total flavonoids content of the medicinal materials were explored through the single factor experiment.Orthogonal experiment was carried out to optimize the extraction process conditions,and the optimal extraction process for total flavonoids from Eucommiae Cortex was screened out.The total antioxidant capacity index was used to determine the free radical scavenging ability of the total flavonoids from Eucommiae Cortex.[Results]The optimal extraction process of total flavonoids from Eucommiae Cortex was 50%ethanol,1∶40 solid-to-liquid-ratio,and 1.5 h reflux extraction time.Through the antioxidant experiment in vitro,it is found that the total flavonoids from Eucommiae Cortex have a higher scavenging ability for the total antioxidant capacity,and there is a certain positive correlation with the mass concentration of total flavonoids.[Conclusions]This method can effectively determine the total flavonoids content of Eucommiae Cortex medicinal material,and provide a certain scientific basis for the quality standard research of the medicinal material.The total flavonoids from Eucommiae Cortex have good in vitro antioxidant activity.And the method for extracting total flavonoids from Eucommiae Cortex medicinal material in this experiment has good repeatability,high stability and feasibility. | Yunli TANG Xiumei MA Huazhen SU Cheng PENG Xiaofang XIE Hongxia CHEN | 2021 | Medicinal Plant2021,12,4: | 0 |
