1Tom20 senses iron-activated ROS signaling to promote melanoma cell pyroptosis显示文摘Iron has been shown to trigger oxidative stress by elevating reactive oxygen species (ROS) and to participate in different modes of cell death,such as ferroptosis,apoptosis and necroptosis. However,whether iron-elevated ROS is also linked to pyroptosis has not been reported. Here,we demonstrate that iron-activated ROS can induce pyroptosis via a Tom20-Bax-caspase-GSDME pathway. In melanoma cells,iron enhanced ROS signaling initiated by CCCP,causing the oxidation and oligomerization of the mitochondrial outer membrane protein Tom20. Bax is recruited to mitochondria by oxidized Tom20,which facilitates cytochrome c release to cytosol to activate caspase-3;eventually triggering pyroptotic death by inducing GSDME cleavage. Therefore,ROS acts as a causative factor and Tom20 senses ROS signaling for iron-driven pyroptotic death of melanoma cells. Since iron activates ROS for GSDME-dependent pyroptosis induction and melanoma cells specifically express a high level of GSDME,iron may be a potential candidate for melanoma therapy. Based on the functional mechanism of iron shown above,we further demonstrate that iron supplementation at a dosage used in iron-deficient patients is sufficient to maximize the anti-tumor effect of clinical ROS-inducing drugs to inhibit xenograft tumor growth and metastasis of melanoma cells through GSDME-dependent pyroptosis. Moreover,no obvious side effects are observed in the normal tissues and organs of mice during the combined treatment of clinical drugs and iron. 3Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. 4Osteoimmunomodulatory effects of biomaterial modification strategies on macrophage polarization and bone regeneration显示文摘Biomaterials as bone substitutes are always considered as foreign bodies that can trigger host immune responses.Traditional designing principles have been always aimed at minimizing the immune reactions by fabricating inert biomaterials.However,clinical evidence revealed that those methods still have limitations and many of which were only feasible in the laboratory.Currently,osteoimmunology,the very pioneering concept is drawing more and more attention-it does not simply regard the immune response as an obstacle during bone healing but emphasizes the intimate relationship of the immune and skeletal system,which includes diverse cells,cytokines,and signaling pathways.Properties of biomaterials like topography,wettability,surface charge,the release of cytokines,mediators,ions and other bioactive molecules can impose effects on immune responses to interfere with the skeletal system.Based on the bone formation mechanisms,the designing methods of the biomaterials change from immune evasive to immune reprogramming.Here,we discuss the osteoimmunomodulatory effects of the new modification strategies—adjusting properties of bone biomaterials to induce a favorable osteoimmune environment.Such strategies showed potential to benefit the development of bone materials and lay a solid foundation for the future clinical application.Yajuan Xie Cheng Hu Yi Feng Danfeng Li Tingting Ai Yulei Huang Xiaodan Chen Lijia Huang Jiali Tan 2020Regenerative Biomaterials2020,7,3:22

93D Geometry-Based UAV-MIMO Channel Modeling and Simulation显示文摘A more general narrowband regular-shaped geometry-based statistical model(RS-GBSM) combined with the line of sight(LoS) and single bounce(SB) rays for unmanned aerial vehicle(UAV) multiple-input multiple-output(MIMO) channel is proposed in this paper. The channel characteristics, including space-time correlation function(STCF), Doppler power spectral density(DPSD), level crossing rate(LCR) and average fade duration(AFD), are derived based on the single sphere reference model for a non-isotropic environment. The corresponding sum-of-sinusoids(SoS) simulation models including both the deterministic model and statistical model with finite scatterers are also proposed for practicable implementation. The simulation results illustrate that the simulation models well reproduce the channel characteristics of the single sphere reference model with sufficient simulation scatterers. And the statistical model has a better approximation of the reference model in comparison with the deterministic one when the simulation trials of the stochastic model are sufficient. 11Differentially expressed proteins of gamma-ray irradiated mouse intestinal epithelial cells by two-dimensional electrophoresis and MALDI-TOF mass spectrometry显示文摘AIM: To identify the differentially expressed proteins involved in ionizing radiation in mice and to explore new ways for studying radiation-related proteins.METHODS: Bal B/c mice grouped as sham-irradiation, 3h and 72 h irradiation were exposed to 9.0Gy single dose of γ-irradiation. Intestinal epithelia were isolated from mice,and total proteins were extracted with urea containing solution. A series of methods were used, including twodimensional electrophoresis, PDQuest 2-DE software analysis, peptide mass fingerprinting based on matrixassisted laser desorption/ionization time of flight masss pectrometry (MALDI-TOF-MS) and SWISS-PROT database searching, to separate and identify the differential proteins.Western blotting and RT-PCR were used to validate the differentially expressed proteins.RESULTS: Mouse intestine was severely damaged by 9.0 Gy γ-irradiation. Image analysis of two-dimensional gels revealed that averages of 638±39, 566±32 and 591±29 protein spots were detected in 3 groups, respectively, and the majority of these protein spots were matched. About 360 protein spots were matched between normal group and 3 h irradiation group, and the correlation coefficient was 0.78 by correlation analysis of gels. Also 312 protein spots matched between normal group and 72 h irradiation group, and 282 protein spots between 3 h and 72 h irradiation groups. Twenty-eight differential protein spots were isolated from gels, digested with trypsin, and measured with MALDI-TOF-MS. A total of 25 spots yielded good spectra, and 19 spots matched known proteins after database searching. These proteins were mainly involved in anti-oxidation, metabolism, signal transduction,and protein post-translational processes. Western-blotting confirmed that enolase was up-regulated by γ-irradiation. Up-regulation of peroxiredoxin I was verified by applying RTPCR technique, but no change occurred in QSVC72.CONCLUSION: These differentially expressed proteins might play important roles when mouse intestine was severely injured by γ-irradiation. 12Preparation of Li_4Ti_5O_(12) submicrospheres and their application as anode materials of rechargeable lithium-ion batteries显示文摘We report on the preparation of spinel Li4Ti5O12 submicrospheres and their application as anode materials of rechargeable lithium-ion batteries. The spinel Li4Ti5O12 submicrospheres are synthesized with three steps of the hydrolysis of TiCl4 to form rutile TiO2, the hydrothermal treatment of rutile TiO2 with LiOH to prepare an intermediate phase of LiTi2O4+δ, and the calcinations of LiTi2O4+δ to obtain spinel Li4Ti5O12. The as-prepared products are investigated by X-ray powder diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The diameters of Li4Ti5O12 submicrospheres with novel hierarchical microstructures are about 200–300 nm with the assembly of 20–30 nm nanoparticles. The electrochemical properties of Li4Ti5O12 submicrospheres are measured by galvanostatical discharge/charge test and cyclic voltammetry (CV). The as-prepared Li4Ti5O12 display excellent discharge/charge rate and cycling capability. A high discharge capacity of 174.3 mAh/g is obtained in the first discharge at 1 C rate. Meanwhile, there is only tiny capacity fading with nearly 100% columbic efficiency in the sequential 5–50 cycles. Moreover, lithium-ion diffusion coefficient in Li4Ti5O12 is calculated to be 1.03 × 10-7 cm2/s. 13H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak显示文摘AIM To investigate the protective efficacy ofH2 strain attenuated live hepatitis A vaccines(H2-strain vaccines)in hepatitis A(HA)outbreaks.METHODS With the permission of theirparents,5551 pre-school and grade 1-3 primaryschool children were inoculated with 1 dose(106.5TCID50)of H2-strain vaccines in anonrandomized,controlled trial conducted inFucheng County,Hebei Province in May 1997.Another 6485 children in the same grades andcompatible in gender and age were enrolled ascontrols.Epidemiological and serological surveywas conducted to evaluate the protectiveefficacy of the vaccines.ELISA was used todetect serum IgM anti-HAV.RESULTS HA outbreak started in early May1998,peaked in the middle of the same month,and lasted about 80 days.Overall 302 HA caseswere found,192(53.58%)were 5-9 years old.One vaccinee and 25 control cases were found tohave hepatitis A,which account for 0.28%(1/356)and 5.92%(25/422)of all vaccinees andcontrols in the 14 villages,respectively.The protective efficacy of vaccines was 95.27%(95% Cl:85.83%-104.72%).In subjects testedfor anti-HAV IgM from 13 villages,1(0.40%)overt and 11(4.06%)asymptomatic HAV caseswere found in 271 vaccinees,but 21(6.69%)ofovert and asymptomatic ones were found in 314controls.CONCLUSION H2 strain vaccines were excellent in preventing overt hepatitis A, but not so effective in preventing asymptomatic hepatitis A virus infection. 15Composite solid electrolyte of Na3PS4-PEO for all-solid-state SnS2/Na batteries with excellent interfacial compatibility between electrolyte and Na metal显示文摘High ionic conductivity and superior interfacial stability of solid electrolytes at the electrodes are crucial factors for high-performance all-solid-state sodium batteries. Herein, a composite solid electrolyte Na3PS4-polyethylene oxide is synthesized by the solution-phase reaction method with an improved ionic conductivity up to 9.4 × 10-5 S/cm at room temperature. Moreover, polyethylene oxide polymer layer is wrapped homogeneously on the surface of Na3PS4 particles, which could effectively avoid the direct contact between Na3PS4 electrolyte and sodium metal, thus alleviate their side reactions. 18Leveraging Auxiliary Knowledge for Web Service Clustering显示文摘By grouping Web services that share similar functionalities, Web service clustering can greatly enhance Web service discovery and selection. Most existing clustering techniques are designed to handle long text documents. However, the descriptions of most publicly available Web services are in the form of short text, which impairs the quality of service clustering due to the sparseness of useful information. Towards this issue, we propose a new service clustering approach based on transfer learning from auxiliary long text data obtained from Wikipedia.To handle the inconsistencies in semantics and topics between service descriptions and auxiliary data, we introduce a novel topic model – Tag aided dual Author topical model(TD-ATM), which jointly learns two sets of topics on the two data sets and automatically couples the topic parameters to avoid the potential inconsistencies between these two data sets. 20Epidermal growth factor receptor mutation analysis in cytological specimens and responsiveness to gefitinib in advanced non-small cell lung cancer patients显示文摘Background: Epidermal growth factor receptor(EGFR) mutation is the key predictor of EGFR tyrosine kinase inhibitors(TKIs) efficacy in non-small cell lung cancer(NSCLC). We conducted this study to verify the feasibility of EGFR mutation analysis in cytological specimens and investigate the responsiveness to gefitinib treatment in patients carrying EGFR mutations.Methods: A total of 210 cytological specimens were collected for EGFR mutation detection by both direct sequencing and amplification refractory mutation system(ARMS). We analyzed EGFR mutation status by both methods and evaluated the responsiveness to gefitinib treatment in patients harboring EGFR mutations by overall response rate(ORR), disease control rate(DCR) and progression free survival(PFS).Results: Of all patients, EGFR mutation rate was 28.6%(60/210) by direct sequencing and 45.2%(95/210) by ARMS(P<0.001) respectively. Among the EGFR wild type patients tested by direct sequencing, 26.7% of them were positive by ARMS. For the 72 EGFR mutation positive patients treated with gefitinib, the ORR, DCR and median PFS were 69.4%, 90.2% and 9.3 months respectively. The patients whose EGFR mutation status was negative by direct sequencing but positive by ARMS had lower ORR(48.0% vs. 80.9%, P=0.004) and shorter median PFS(7.4 vs. 10.5 months, P=0.009) as compared with that of EGFR mutation positive patients by both detection methods. Conclusions: Our study verified the feasibility of EGFR analysis in cytological specimens in advanced NSCLC. ARMS is more sensitive than direct sequencing in EGFR mutation detection. EGFR Mutation status tested on cytological samples is applicable for predicting the response to gefitinib.
