1DDIT4 promotes gastric cancer proliferation and tumorigenesis through the p53 and MAPK pathways显示文摘Background:Gastric cancer(GC)is one of the most common malignancies worldwide,particularly in China.DNA damage-inducible transcript 4(DDIT4)is a mammalian target of rapamycin inhibitor and is induced by various cellular stresses;however,its critical role in GC remains poorly understood.The present study aimed to investigate the poten-tial relationship and the underlying mechanism between DDIT4 and GC development.Methods:We used western blotting,real-time polymerase chain reaction,and immunohistochemical or immunoflu-orescence to determine DDIT4 expression in GC cells and tissues.High-content screening,cell counting kit-8 assays,colony formation,and in vivo tumorigenesis assays were performed to evaluate cell proliferation.Flow cytometry was used to investigate cell apoptosis and cell cycle distribution.Results:DDIT4 was upregulated in GC cells and tissue.Furthermore,downregulating DDIT4 in GC cells inhibited proliferation both in vitro and in vivo and increased 5-fluorouracil-induced apoptosis and cell cycle arrest.In contrast,ectopic expression of DDIT4 in normal gastric epithelial cells promoted proliferation and attenuated chemosensitivity.Further analysis indicated that the mitogen-activated protein kinase and p53 signaling pathways were involved in the suppression of proliferation,and increased chemosensitivity upon DDIT4 downregulation.Conclusion:DDIT4 promotes GC proliferation and tumorigenesis,providing new insights into the role of DDIT4 in the tumorigenesis of human GC.Feng Du Lina Sun Yi Chu Tingyu Li Chao Lei Xin Wang Mingzuo Jiang Yali Min Yuanyuan Lu Xiaodi Zhao Yongzhan Nie Daiming Fan 2018Cancer Communications2018,38,1:10
2Modeling CADASIL vascular pathologies with patient-derived induced pluripotent stem cells显示文摘Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy(CADASIL)is a rare hereditary cerebrovascular disease caused by a NOTCH3 mutation.However,the underlying cellular and molecular mechanisms remain unidentified.Here,we generated non-integrative induced pluripotent stem cells(iPSCs)from fibroblasts of a CADASIL patient harboring a heterozygous NOTCH3 mutation(c.3226C>T,p.R1076C).Vascular smooth muscle cells(VSMCs)differentiated from CADASIL-specific iPSCs showed gene expression changes associated with disease phenotypes,including activation of the NOTCH and NF-kB signaling pathway,cytoskeleton disorganization,and excessive cell proliferation.In comparison,these abnormalities were not observed in vascular endothelial cells(VECs)derived from the patients iPSCs.Importantly,the abnormal upregulation of NF-kB target genes in CADASIL VSMCs was diminished by a NOTCH pathway inhibitor,providing a potential therapeutic strategy for CADASIL.Overall,using this iPSCbased disease model,our study identified clues for studying the pathogenic mechanisms of CADASIL and developing treatment strategies for this disease.Chen Ling Zunpeng Liu Moshi Song Weiqi Zhang Si Wang Xiaoqian Liu Shuai Ma Shuhui Sun Lina Fu Qun Chu Juan Carlos Izpisua Belmonte Zhaoxia Wang Jing Qu Yun Yuan Guang-Hui Liu 2019Protein & Cell2019,10,4:7

5Decomposition of acid dissolved titanium slag from Australia by sodium hydroxide显示文摘The kinetics of the decomposition of acid dissolved titanium slag with a sodium hydroxide system under atmospheric pressure was studied. The effect of reaction temperature, particle size and NaOH-to-slag mass ratio on titanium extraction was investigated. The results show that temperature and particle size have significant influence on titanium extraction. The experimental data of titanium extraction show that the shrinking core model with chemical reaction controlled process is most applicable for the decomposition of slag, with an apparent activation energy of 62.4 kJ·mol-1. Approximately 85 wt.%-90 wt.% of the titanium can be extracted from the slag under the optimal conditions. 6Primed macrophages directly and specifically reject allografts显示文摘Monocytes and macrophages have long been associated with acute and chronic allograft rejection;this is mediated by their abilities to promote inflammation,kill target cells via antibody-dependent cytotoxicity and modulate adaptive immunity.Our present study showed that allogeneic antigen-primed macrophages acutely rejected skin grafts with specificity after adoptive transfer into MHC-matched immunodeficient mice.The ability of primed macrophages to reject allografts essentially requires the help of CD4^(+) T cells and does not require the help of CD8^(+) T cells.Moreover,the primed,perforin-deficient macrophages rejected the skin grafts in a significantly delayed pattern compared with WT macrophages,indicating that the perforin pathway of the primed macrophages is likely involved in the rejection process.Thus,primed macrophages are endowed with adaptive immunitylike features,such as specificity,with the help of CD4^(+) T cells during the immune response to allografts.The present study challenges our traditional views of macrophage functions and highlights the biological functions of macrophages beyond innate immunity in mammals.Zhulang Chu Chenming Sun Lina Sun Chang Feng Fan Yang Yanan Xu Yong Zhao 2020Cellular & Molecular Immunology2020,17,3:4
7A complete genome assembly of Glaciecola mesophila sp. nov. sequenced by using BIGIS-4 sequencer system显示文摘Using a pyrosequencing-based custom-made sequencer BIGIS-4, we sequenced a Gram-negative bacterium Glaciecola mesophila sp. nov. (Gmn) isolated from marine invertebrate specimens. We generated 152043 sequencing reads with a mean high-quality length of 406 bp, and assembled them using the BIGIS-4 post-processing module. No systematic low-quality data was detected beyond expected homopolymer-derived errors. The assembled Gmn genome is 5144318 bp in length and harbors 4303 annotated genes. A large number of metabolic genes correspond to various nutrients from surface marine invertebrates. 9Axial gradient excitation accelerates volumetric imaging of two-photon microscopy显示文摘Two-photon excitation fluorescence microscopy(TPM),owing to its capacity for subcellular resolution,intrinsic optical sectioning,and superior penetration depth in turbid samples,has revolutionized biomedical research.However,its layer-by-layer scanning to form a three-dimensional image inherently limits the volumetric imaging speed and increases phototoxicity significantly.In this study,we develop a gradient excitation technique to accelerate TPM volumetric imaging.The axial positions of the fluorophores can be decoded from the intensity ratio of the paired images obtained by sequentially exciting the specimen with two axially elongated two-photon beams of complementary gradient intensities.We achieved a 0.63μm axial localization precision and demonstrate the flexibility of the gradient TPM on various sparsely labeled samples,including bead phantoms,mouse brain tissues,and live macrophages.Compared with traditional TPM,our technique improves volumetric imaging speed(by at least sixfold),decreases photobleaching(i.e.,less than 2.07±2.89%in 25 min),and minimizes photodamages.Yufeng Gao Xianyuan Xia Lina Liu Ting Wu Tingai Chen Jia Yu Zhili Xu Liang Wang Fei Yan Zhuo Du Jun Chu Yang Zhan Bo Peng Hui Li Wei Zheng 2022Photonics Research2022,10,3:3
10Synthesis of Core-Shell HZSM-5@SBA-15 Composite and Its Performance in the Conversion of Methanol to Aromatics显示文摘A core-shell HZSM-5@SBA-15 composite was prepared by the acidic sol-gel coating method using Pluronic P123 as the template. Its structure, properties, and catalytic performance in the conversion of methanol to aromatics(MTA) were compared with HZSM-5@SiO_2. The silica shells of these two samples can both reduce the acid sites of HZSM-5, and consequently can decrease the formation of coke on the catalyst. However, in the ordered mesoporous SBA-15 shell the aromatics and the coke precursors could diffuse more effectively than in the amorphous SiO_2 shell; therefore, in the MTA reaction catalyzed by HZSM-5@SBA-15 composite a higher BTX selectivity and a longer catalyst cycle length were observed. 18Barley leaves mediated biosynthesis of Au nanomaterials as a potential contrast agent for computed tomography imaging显示文摘Over the past few years, environment-friendly green synthetic methods for metal nanoparticles(NPs) have received considerable attention. The use of plant extracts is considered as potential for the biosynthesis of metal NPs due to its easier obtainment, cheap accessibility to industrial scale-up and high biosecurity. Herein, with the aqueous leaf extract of barley leaves as the template, we reported a rapid, single step, cost-effective and eco-friendly synthetic route to prepare new gold nanoparticles(BL-Au NPs).Significantly, except for the key role as the reducing, barley leaves was further utilized as the capping agent that ensure the stability of the BL-Au NPs. The characterization results of TEM, FTIR and UV-vis indicated that the gold nanoparticles coated with barley leaves extract were successfully synthesized. In vitro and zebra fish toxicity and stability assays demonstrated its good biocompatibility and high colloidal stability. The CT images in vitro demonstrated that the constructed BL-Au NPs possessed superiority in X-ray attenuation ability compared to clinically commonly used iodinated small molecular contrast agent. And BL-Au NPs can be used as contrast agent for effective CT imaging of zebra fish model. 19Role of Intracellular Distribution of Feline and Bovine SAMHD1 Proteins in Lentiviral Restriction显示文摘Human SAMHD1(h SAM)restricts lentiviruses at the reverse transcription step through its d NTP triphosphohydrolase(d NTPase)activity.Besides humans,several mammalian species such as cats and cows that carry their own lentiviruses also express SAMHD1.However,the intracellular distribution of feline and bovine SAMHD1(f SAM and b SAM)and its significance in their lentiviral restriction function is not known.Here,we demonstrated that f SAM and b SAM were both predominantly localized to the nucleus and nuclear localization signal(11KRPR14)-deleted f SAM and b SAM relocalized to the cytoplasm.Both cytoplasmic f SAM and b SAM retained the antiviral function against different lentiviruses and cytoplasmic f SAM could restrict Vpx-encoding SIV and HIV-2 more efficiently than its wild-type(WT)protein as cytoplasmic h SAM.Further investigation revealed that cytoplasmic f SAM was resistant to Vpx-induced degradation like cytoplasmic h SAM,while cytoplasmic b SAM was not,but they all demonstrated the same in vitro d NTPase activity and all could interact with Vpx as their WT proteins,indicating that cytoplasmic h SAM and f SAM can suppress more SIV and HIV-2 by being less sensitive to Vpx-mediated degradation.Our results suggested that f SAM-and b SAM-mediated lentiviral restriction does not require their nuclear localization and that f SAM shares more common features with h SAM.These findings may provide insights for the establishment of alternative animal models to study SAMHD1 in vivo.Chu Wang Lina Meng Jialin Wang Kaikai Zhang Sizhu Duan Pengyu Ren Yingzhe Wei Xinyu Fu Bin Yu Jiaxin Wu Xianghui Yu 2021Virologica Sinica2021,36,5:0
20Unveiling the Contribution of Lactic Acid to the Passivation Behavior of Ti-6Al-4V Fabricated by Laser Powder Bed Fusion in Hank’s Solution显示文摘In actual physiological environments,bacteria can activate the immune system and release lactic acid.However,the detailed contribution of lactic acid to the passivation behavior of titanium(Ti)alloys is still unclear.The current work investigated the in vitro passivation behavior of Ti-6Al-4V(TC4)alloys fabricated by laser powder bed fusion in Hank's solution with and without adding lactic acid.Electrochemical methods,inductively coupled plasma atomic emission spectrometer,and X-ray photoelectron spectroscopy were jointly used.Adding lactic acid decreases the corrosion resistance of samples by degrading the formed passive film.The film formed in the(lactic acid)-containing solution exhibits a higher level of oxygen vacancies and a lower thickness,attributed to the suppressed formation of Ti^(4+)transformed from Ti^(3+)and Ti^(2+).Moreover,the presence of lactic acid would increase the open circuit potential,relieve the ions release,and hinder the deposition of calcium phosphates within 24 h immersion.Yu-Hang Chu Liang-Yu Chen Bo-Yuan Qin Wenbin Gao Fanmin Shang Hong-Yu Yang Lina Zhang Peng Qin Lai-Chang Zhang 2024Acta Metallurgica Sinica(English Letters)2024,37,1:0
