| 3 | Antitumor effect and mechanism of Gecko on human esophageal carcinoma cell lines in vitro and xenografted sarcoma 180 in Kunming mice显示文摘AIM: To investigate the anti-tumor effect of Chinese medicine Gecko on human esophageal carcinoma cell lines and xenografted sarcoma 180 in Kunming mice and its mechanism. METHODS: The serum pharmacological method was used in vitro . The growth rates of the human esophageal carcinoma cells (EC9706 or EC1) were measured by a modifi ed 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The transplanted tumor model of the mouse S180 sarcoma was established. Fifty mice were randomly divided into fi ve groups (n = 10). Three Gecko groups were treated respectively with oral administration of Gecko powder at a daily dose of 13.5 g/kg, 9 g/kg, and 4.5 g/kg. The negative group (NS group) was treated with oral administration of an equal volume of saline and the positive group (CTX group) was treated with 100 mg/kg Cytoxan by intraperitoneal injection at the fi rst day. After 2 wk of treatment, the anti-tumor activity was evaluated by tumor tissue weighing. The impact on immune organ was detected based on the thymus index, spleen index, phagocytic rate and phagocytic index. The protein expression of vascular endothelingrowth factor (VEGF) and basic fibroblast growth factor (bFGF) were detected by immunohistochemistry. The cell apoptotic rate was detected by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) assay. RESULTS: The A value in each group treated with Gecko after 72 h was reduced signif icantly in EC9706 and in EC1. The tumor weight in each group of Gecko was decreased signifi cantly (1.087 ± 0.249 vs 2.167 ± 0.592; 1.021 ± 0.288 vs 2.167 ± 0.592; 1.234 ± 0.331 vs 2.167 ± 0.592; P < 0.01, respectively). However, the thymus index and Spleen index of mice in Gecko groups had no significant difference compared with the NS group. The immunoreactive score of VEGF and bFGF protein expression of each Gecko group by immunohistochemical staining were lowered signifi cantly. The apoptosis index (AI) of each group was increased progressively with increase of dose of Gecko by TUNEL. | 4 | Effects of large dose of dexamethasone on inflammatory mediators and pancreatic cell apoptosis of rats with severe acute pancreatitis显示文摘AIM: To investigate the influence of high dose of dexamethasone on inflammatory mediators and apoptosis of rats with severe acute pancreatitis (SAP). METHODS: SAP rats were randomly assigned to the model group and treatment group while the normal rats were assigned to the sham operation group. The mortality,ascite volumes,ascites/body weight ratio and pancreas pathological changes of all rats were observed at 3,6 and 12 h after operation. Their contents of amylase and endotoxin in plasma and contents of tumor necrosis factor (TNF-α),phospholipase A2 (PLA2) and IL-6 in serum were also determined. The microarray sections of their pancreatic tissues were prepared,terminal transferase dUTP nick end labeling (TUNEL) staining was performed and apoptotic indexes were calculated. RESULTS: There was no marked difference between treatment group and model group in survival. The contents of amylase and endotoxin in plasma and contents of TNF-α,PLA2 and IL-6 in serum,ascite volumes,ascites/body weight ratio and pancreas pathological scores were all lower in treatment group than in model group to different extents at different time points P < 0.05,58.3 (26.4) ng/L vs 77.535 (42.157) ng/L in TNF-α content,8.00 (2.00) points vs 9.00 (2.00) points in pathological score of pancreas respectively; P < 0.01,0.042 (0.018) EU/mL vs 0.056 (0.0195) EU/mL in endotoxin content,7791 (1863) U/L vs 9195 (1298) U/L in plasma amylase content,1.53 (0.79) vs 2.38 (1.10) in ascites/body weight ratio,8.00 (1.00) points vs 11.00 (1.50) points in pathological score of pancreas; P < 0.001,3.36 (1.56) ng/L vs 5.65 (1.08) ng/L in IL-6 content,4.50 (2.00) vs 7.20 (2.00),4.20 (1.60) vs 6.40 (2.30),3.40 (2.70) vs 7.90 (1.70) in ascite volumes,respectively. The apoptotic indexes of pancreas head and pancreas tail were all higher in treatment group than in model group at 6 h P < 0.01,0.00 (2.00)% vs 0.00 (0.00)%,0.20 (1.80) vs 0.00 (0.00) in apoptosis indexes,respectively. The proliferation of the cells wasexamined by cell counter, flow cytometry (FCM) andmicrocytotoxicity assay (MTT).RESULTS: The growth of human primary gastric cancer cellsin Met Hcy+ was suppressed, manifested by the decrease oftotal cell counts [1.46±0.42 ( x 109@L-1) in Met-Hcy+ vs .64±0.44 ( x l09@L-1) in Met+ Hcy, P<0.01], the decline inthe percentage of G0G1 phase cells (0.69±0.24 in Met-Hey+vs 0.80±0.18 in Met+ Hcy, P<0.01) and the increase of Scells(0.24±0.20inMet-Hcy+ vs 0.17 ± 0.16 in Met+ Hcy-, P< 0.01); however, gastric mucusal cells grew normally. IfMet-Hcy+ medium was used in combination withchemotherapeutic drugs, the number of surviving gastriccancer cells dropped significantly.CONCLUSION: Human primary gastric cancer cells in vitroare Met-dependent; however, gastric mucosal cells have notshown the same characteristica. | 10 | Arctic Sea Ice and Eurasian Climate:A Review显示文摘The Arctic plays a fundamental role in the climate system and has shown significant climate change in recent decades,including the Arctic warming and decline of Arctic sea-ice extent and thickness. In contrast to the Arctic warming and reduction of Arctic sea ice, Europe, East Asia and North America have experienced anomalously cold conditions, with record snowfall during recent years. In this paper, we review current understanding of the sea-ice impacts on the Eurasian climate.Paleo, observational and modelling studies are covered to summarize several major themes, including: the variability of Arctic sea ice and its controls; the likely causes and apparent impacts of the Arctic sea-ice decline during the satellite era,as well as past and projected future impacts and trends; the links and feedback mechanisms between the Arctic sea ice and the Arctic Oscillation/North Atlantic Oscillation, the recent Eurasian cooling, winter atmospheric circulation, summer precipitation in East Asia, spring snowfall over Eurasia, East Asian winter monsoon, and midlatitude extreme weather; and the remote climate response(e.g., atmospheric circulation, air temperature) to changes in Arctic sea ice. | 11 | Relation between electricity structure of the crust and deformation of crustal blocks on the northeastern margin of Qinghai-Tibet Plateau显示文摘Study on the electricity structure along a magnetotelluric (MT) sounding profile on the northeastern margin of Qinghai-Tibet Plateau indicates that four crustal blocks can be de-termined from southwest to northeast, namely Bayan Har block (BH), Qin-Qi block (QQ), Hai-yuan block (HY) or the North-South seismotectonic belt and Ordos block (OD). The BH, QQ and OD blocks display a similar electricity structure of the crust. The upper crust represents a high- resistivity layer and the upper part of lower crust represents a low-resistivity layer with the resis-tivity increasing gradually with depth from the lower part of lower crust to the upper mantle. The electricity structure of the crust in these three blocks is similar to that in the complete blocks on the southern and eastern margins of the Qinghai-Tibet Plateau and belongs to normal electricity layering of the crust in slightly deformed or complete intracontinental blocks. The crust in HY block as a boundary zone has been significantly deformed, hence its electricity layering was de-stroyed and the structure was complex and the block became a recent tectonically active and great seismo-active region. The contact belts between the blocks on the northeastern margin of Qinghai-Tibet Plateau exhibit both upthrusting outward and strike-slip movement different from those on the southern and eastern margins of the plateau. | 13 | Fast magnetic reconstruction of the portal vein with allogeneic blood vessels in canines显示文摘BACKGROUND:The resection and reconstruction of large vessels,including the portal vein,are frequently needed in tumor resection.Warm ischemia before reconstruction might have deleterious effects on the function of some vital organs and therefore,how to reconstruct the vessels quickly after resection is extremely important.The present study was to introduce a new type of magnetic compression anastomosis(MCA)device to establish a quick non-suture anastomosis of the portal vein after resection in canines.METHODS:The new MCA device consists of a pair of titanium alloy and neodymium-ferrum-boron magnet(Ti-Nd Fe B)composite rings.The Nd Fe B magnetic ring as a core of the device was hermetically sealed inside the biomedical titanium alloy case.Twelve canines were divided into two groups:a MCA group in which the end-to-end anastomoses was made with a new device after resection in the portal vein and a traditional manual suture(TMS)group consisted of 6 canines.The anastomosis time,anastomotic patency and quality were investigated at week 24 postoperatively.RESULTS:The portal vein was reconstructed successfully in all of the animals and they all survived.The duration of portal vein anastomosis was significantly shorter in the MCA group than in the TMS group(8.16±1.25 vs 36.24±2.17 min,P<0.05).Portography and ultrasound showed that the blood flow was normal without angiostenosis or thrombosis in all of the canines.Hematoxylin-eosin staining and electron microscope scanning showed in contrast to the TMS group,MCA anastomotic intimal was much smoother with more regularly arranged endothelial cells at week 24 postoperatively. | Shan-Pei Wang Xiao-Peng Yan Fei Xue Ding-Hui Dong Xu-Feng Zhang Feng Ma Hao-Hua Wang Yi Lv | 2015 | Hepatobiliary & Pancreatic Diseases International2015,14,3: | 20 |
| 14 | Moisture distribution in sludges based on different testing methods显示文摘Moisture distributions in municipal sewage sludge, printing and dyeing sludge and paper mill sludge were experimentally studied based on four different methods, i.e., drying test, thermogravimetric-differential thermal analysis (TG-DTA) test, thermogravimetric-differential scanning calorimetry (TG-DSC) test and water activity test. The results indicated that the moistures in the mechanically dewatered sludges were interstitial water, surface water and bound water. The interstitial water accounted for more than 50% wet basis (wb) of the total moisture content. The bond strength of sludge moisture increased with decreasing moisture content, especially when the moisture content was lower than 50% wb. Furthermore, the comparison among the four different testing methods was presented. The drying test was advantaged by its ability to quantify free water, interstitial water, surface water and bound water; while TG-DSC test, TG-DTA test and water activity test were capable of determining the bond strength of moisture in sludge. | 15 | High-intensity focused ultrasound treatment for patients with unresectable pancreatic cancer显示文摘BACKGROUND: High-intensity focused ultrasound (HIFU) is a non-invasive method of solid tissue ablation therapy. However, only a few studies have reported the effect of HIFU for unresectable pancreatic cancer. This study aimed to evaluate the clinical benefits, survival time and complications associated with the use of HIFU ablation in patients with unresectable pancreatic cancer. METHODS: Twenty-five patients with unresectable pancreatic cancer were enrolled in our study. All patients received HIFU therapy for tumors at least once. The therapeutic effects of HIFU was evaluated in terms of Karnofsky performance status (KPS) scores, pain relief, serum CA19-9, and imaging by B-US and CT before and after the therapy. We also recorded median overall survival time and complications caused by the treatment. RESULTS: In the 25 patients, KPS scores were above 60, and increased KPS was observed in 23 patients after treatment. Pain relief occurred in 23 patients. Serum CA19-9 levels were significantly reduced one month after HIFU treatment and became negative in 5 patients. B-US revealed enhanced tumor echogenicity in 13 patients and decreased tumor blood supply in 9. Tumor necrosis was confirmed by CT in 8 patients one month after HIFU treatment. The median overall survival time was 10 months, and the 1-year survival rate was 42%. No severe complications were observed after HIFU treatment. | 17 | Isolation and Characterization of an Algicidal Bacterium Indigenous to Lake Taihu with a Red Pigment able to Lyse Microcystis aeruginosa显示文摘Objective To isolate and characterize indigenous algicidal bacteria and their algae-lysing compounds active against Microcystis aeruginosa, strains TH1, TH2, and FACHB 905. Methods The bacteria were identified using the Biolog automated microbial identification system and 16S rDNA sequence analysis. The algae-lysing compounds were isolated and purified by silica gel column chromatography and reverse-phase high performance liquid chromatography. Their structures were confirmed by Nuclear Magnetic Resonance (NMR) and Fourier Transform Infrared (FT-IR) spectroscopy. Algae-lysing activity was observed using microscopy. Results The algae-lysing bacterium LTH-2 isolated from Lake Taihu was identified as Serratia marcescens. Strain LTH-2 secreted a red pigment identified as prodigiosin (C 20 H 25 N 3 O), which showed strong lytic activity with algal strains M. aeruginosa TH1, TH2, and FACHB 905 in a concentration-dependent manner. The 50% inhibitory concentration (IC 50 ) of prodigiosin with the algal strains was 4.8 (±0.4)×10 -2 μg/mL, 8.9 (±1.1)×10-2 μg/mL, and 1.7 (±0.1)×10 -1 μg/mL in 24 h, respectively. Conclusion The bacterium LTH-2 and its pigment had strong Microcystis-lysing activity probably related to damage of cell membranes. | 18 | Effects of dietary supplementation of Artemisia argyi aqueous extract on antioxidant indexes of small intestine in broilers显示文摘The present study was conducted to investigate the effect of Artemisia argyi aqueous extract(AAE) on antioxidant indexes in the small intestine. A total of 192 Arbor Acre broiler chickens(one-day-old) were randomly divided into 4 treatments with 6 replicates of 8 chickens. These 4 diets were formulated by adding 0, 500,1,000 and 2,000 mg/kg AAE to the basal diet. The results showed as follows: 1) compared with the control, the total antioxidant capacity(T-AOC) in ileum for the 2,000 mg/kg treatment group was significantly increased at 21 days of age(P < 0.05); the T-AOC levels in jejunum and ileum were significantly increased in broilers supplemented with 500 mg/kg AAE at 42 days of age(P < 0.05), and the T-AOC levels in jejunum and ileum were significantly improved in 1,000 mg/kg treatment group(P < 0.01), 2) At 21 days of age, supplementation of 500 mg/kg AAE significantly increased the catalase(CAT) activity of small intestine, and the glutathione peroxidase(GSH-Px) activity of jejunum was improved(P < 0.01), meanwhile, the GSH-Px activity of duodenum and the total superoxide dismutase(T-SOD) activity of duodenum and jejunum were significantly higher than those of the control group(P < 0.05); supplementation of 1,000 mg/kg AAE significantly increased the CAT activity of duodenum and ileum and the GSH-Px activity of duodenum and jejunum(P < 0.05), and the ileum GSH-Px activity was significantly increased(P < 0.01); supplementation of 2,000 mg/kg AAE significantly increased the CAT activity of duodenum and ileum(P < 0.05). At 42 days of age, supplementation of 500 mg/kg AAE significantly increased the GSH-Px activity of ileum and the T-SOD activity of duodenum(P < 0.05),meanwhile, the T-SOD activity of jejunum was significantly increased(P < 0.01); supplementation of1,000 mg/kg AAE significantly increased the CAT activity of jejunum and the T-SOD activity of ileum(P < 0.01), and the GSH-Px activity of jejunum was significantly increased(P < 0.05); supplementation of2,000 mg/kg AAE significantly increased the T-SOD activity of ileum(P < 0.05), but significantly decreased the CAT activity of ileum and the GSH-Px activity of jejunum(P < 0.05). 3) The malondialdehyde(MDA) levels of 3 AAE supplementation groups were significantly decreased at 21 and 42 days of age(P < 0.05). | 20 | Protective effects and mechanisms of Baicalin and octreotide on renal injury of rats with severe acute pancreatitis显示文摘AIM: To investigate the protective effects and mechanisms of Baicalin and octreotide on renal injury of rats with severe acute pancreatitis (SAP). METHODS: One hundred and eighty SD rats were randomly assigned to the model group, Baicalin-treated group, octreotide-treated group and sham operation group. The mortality, plasma endotoxin level, contents of blood urea nitrogen (BUN), creatinine (CREA), phospholipase A2 (PLA2), nitrogen monoxide (NO), tumor necrosis factor (TNF)-α, IL-6 and endothelin-1 (ET-1) in serum, expression levels of renal Bax and Bcl-2 protein, apoptotic indexes and pathological changes of kidney were observed at 3, 6 and 12 h after operation. RESULTS: The renal pathological changes were milder in treated group than in model group. The survival at 12 h and renal apoptotic indexes at 6 h were significantly (P < 0.05) higher in treated group than in model group [66.67% vs 100%; 0.00 (0.02)% and 0.00 (0.04)% vs 0.00 (0.00)%, respectively]. The serum CREA content was markedly lower in octreotide-treated group than in model group at 3 h and 6 h (P < 0.01, 29.200 ± 5.710 μmol/L vs 38.400 ± 11.344 μmol/L; P < 0.05, 33.533 ± 10.106 μmol/L vs 45.154 ± 17.435 μmol/L, respectively). The expression level of renal Bax protein was not significantly different between model group and treated groups at all time points. The expression level of renal Bcl-2 protein was lower in Baicalin-treated group than in model group at 6 h [P < 0.001, 0.00 (0.00) grade score vs 3.00 (3.00) grade score]. The Bcl-2 expression level was lower in octreotide-treated group than in model group at 6 h and 12 h [P < 0.05, 0.00 (0.00) grade score vs 3.00 (3.00) grade score; 0.00 (0.00) grade score vs 0.00 (1.25) grade score, respectively]. The serum NO contents were lower in treated groups than in model group at 3 h and 12 h [P < 0.05, 57.50 (22.50) and 52.50 (15.00) μmol/L vs 65.00 (7.50) μmol/L; P < 0.01, 57.50 (27.50) and 45.00 (12.50) μmol/L vs 74.10 (26.15) μmol/L, respectively]. The plasma endotoxin content and serum BUN content (at 6 h and 12 h) were lower in treated groups than in model group. The contents of IL-6, ET-1, TNF-α (at 6 h) and PLA2 (at 6 h and 12 h) were lower in treated groups than in model group [P < 0.001, 3.031 (0.870) and 2.646 (1.373) pg/mL vs 5.437 (1.025) pg/mL; 2.882 (1.392) and 3.076 (1.205) pg/mL vs 6.817 (0.810) pg/mL; 2.832 (0.597) and 2.462 (1.353) pg/mL vs 5.356 (0.747) pg/mL; 16.226 (3.174) and 14.855 (5.747) pg/mL vs 25.625 (7.973) pg/mL; 18.625 (5.780) and 15.185 (1.761) pg/mL vs 24.725 (3.759) pg/mL; 65.10 (27.51) and 47.60 (16.50) pg/mL vs 92.15 (23.12) pg/mL; 67.91 ± 20.61 and 66.86 ± 22.10 U/mL, 63.13 ± 26.31 and 53.63 ± 12.28 U/mL vs 101.46 ± 14.67 and 105.33 ± 18.10 U/mL, respectively]. CONCLUSION: Both Baicalin and octreotide can protect the kidney of rats with severe acute pancreatitis. The therapeutic mechanisms of Baicalin and octreotide might be related to their inhibition of inflammatory mediators and induction of apoptosis.
