1Accurate diagnosis of prenatal cleft lip/palate by understanding the embryology显示文摘Cleft lip with or without cleft palate(CP) is one of the most common congenital malformations. Ultrasonographers involved in the routine 20-wk ultrasound screening could encounter these malformations. The face and palate develop in a very characteristic way. For ultrasonographers involved in screening these patients it is crucial to have a thorough understanding of the embryology of the face. This could help them to make a more accurate diagnosis and save time during the ultrasound. 2Discriminating mild from critical COVID-19 by innate and adaptive immune single-cell profiling of bronchoalveolar lavages显示文摘How the innate and adaptive host immune system miscommunicate to worsen COVID-19 immunopathology has not been fully elucidated.Here,we perform single-cell deep-immune profiling of bronchoalveolar lavage(BAL)samples from 5 patients with mild and 26 with critical COVID-19 in comparison to BALs from non-COVID-19 pneumonia and normal lung.We use pseudotime inference to build T-cell and monocyte-to-macrophage trajectories and model gene expression changes along them.In mild COVID-19,CD8^(+)resident-memory(TRM)and CD4^(+)T-helper-17(T_(H17))cells undergo active(presumably antigen-driven)expansion towards the end of the trajectory,and are characterized by good effector functions,while in critical COVID-19 they remain more naïve.Vice versa,CD4^(+)T-cells with T-helper-1 characteristics(TH1-like)and CD8^(+)T-cells expressing exhaustion markers(T_(EX)-like)are enriched halfway their trajectories in mild COVID-19,where they also exhibit good effector functions,while in critical COVID-19 they show evidence of inflammation-associated stress at the end of their trajectories.Monocyte-to-macrophage trajectories show that chronic hyperinflammatory monocytes are enriched in critical COVID-19,while alveolar macrophages,otherwise characterized by anti-inflammatory and antigen-presenting characteristics,are depleted.In critical COVID-19,monocytes contribute to an ATP-purinergic signaling-inflammasome footprint that could enable COVID-19 associated fibrosis and worsen disease-severity.Finally,viral RNA-tracking reveals infected lung epithelial cells,and a significant proportion of neutrophils and macrophages that are involved in viral clearance.Els Wauters Pierre Van Mol Abhishek Dinkarnath Garg Sander Jansen Yannick Van Herck Lore Vanderbeke Ayse Bassez Bram Boeckx Bert Malengier-Devlies Anna Timmerman Thomas Van Brussel Tina Van Buyten Rogier Schepers Elisabeth Heylen Dieter Dauwe Christophe Dooms Jan Gunst Greet Hermans Philippe Meersseman Dries Testelmans Jonas Yserbyt Sabine Tejpar Walter De Wever Patrick Matthys CONTAGIOUS collaborators Johan Neyts Joost Wauters Junbin Qian Diether Lambrechts 2021Cell Research2021,31,3:11

4RNAi mechanisms in Huntington’s disease therapy:siRNA versus shRNA显示文摘Huntington’s Disease(HD)is a genetically dominant trinucleotide repeat disorder resulting from CAG repeats within the Huntingtin(HTT)gene exceeding a normal range(>36 CAGs).Symptoms of the disease manifest in middle age and include chorea,dystonia,and cognitive decline.Typical latency from diagnosis to death is 20 years.There are currently no disease-modifying therapies available to HD patients.RNAi is a potentially curative therapy for HD.A popular line of research employs siRNA or antisense oligonucleotides(ASO)to knock down mutant Huntingtin mRNA(mHTT).Unfortunately,this modality requires repeated dosing,commonly exhibit off target effects(OTEs),and exert renal and hepatic toxicity.In contrast,a single AAV-mediated short-hairpin RNA(shRNA)dose can last years with low toxicity.In addition,we highlight research indicating that shRNA elicits fewer OTEs than siRNA when tested head-tohead.Despite this promise,shRNA therapy has been held back by difficulties controlling expression(oversaturating cells with toxic levels of RNA construct).In this review,we compare RNAi modalities for HD and propose novel methods of optimizing shRNA expression and on-target fidelity.Sebastian Aguiar Bram van der Gaag Francesco Albert Bosco Cortese 2017Translational Neurodegeneration2017,6,1:3

8UV-B light and its application potential to reduce disease and pest incidence in crops显示文摘Ultraviolet-B radiation(280–315 nm),perceived by the plant photoreceptor UVR8,is a key environmental signal that influences plant growth and development and can reduce disease and pest incidence.The positive effect of UV-B on disease resistance and incidence in various plant species supports the implementation of supplemental UV-B radiation in sustainable crop production.However,despite many studies focusing on UV-B light,there is no consensus on the best mode of application.This review aims to analyze,evaluate,and organize the different application strategies of UV-B radiation in crop production with a focus on disease resistance.We summarize the physiological effects of UV-B light on plants and discuss how plants perceive and transduce UV-B light by the UVR8 photoreceptor as well as how this perception alters plant specialized metabolite production.Next,we bring together conclusions of various studies with respect to different UV-B application methods to improve plant resistance.In general,supplemental UV-B light has a positive effect on disease resistance in many plant–pathogen combinations,mainly through the induction of the production of specialized metabolites.However,many variables(UV-B light source,plant species,dose and intensity,timing during the day,duration,background light,etc.)make it difficult to compare and draw general conclusions.We compiled the information of recent studies on UV-B light applications,including e.g.,details on the UV-B light source,experimental set-up,calculated UV-B light dose,intensity,and duration.This review provides practical insights and facilitates future research on UV-B radiation as a promising tool to reduce disease and pest incidence.Prisca Meyer Bram Van de Poel Barbara De Coninck 2021Horticulture Research2021,8,1:2
