| 1 | Evolution of transanal total mesorectal excision for rectal cancer:From top to bottom显示文摘The gold standard for curative treatment of locally advanced rectal cancer involves radical resection with a total mesorectal excision(TME). TME is the most effective treatment strategy to reduce local recurrence and improve survival outcomes regardless of the surgical platform used. However, there are associated morbidities, functional consequences, and quality of life(QoL) issues associated with TME; these risks must be considered during the modern-day multidisciplinary treatment for rectal cancer. This has led to the development of new surgical techniques to improve patient, oncologic, and QoL outcomes. In this work, we review the evolution of TME to the transanal total mesorectal excision(TaTME) through more traditional minimally invasive platforms. The review the development, safety and feasibility, proposed benefits and risks of the procedure, implementation and education models, and future direction for research and implementation of the TaTME in colorectal surgery. | 4 | Assessing the benthic quality status of the Bohai Bay(China)with proposed modifications of M-AMBI显示文摘Multivariate AZTI's Marine Biotic Index(M-AMBI) was designed to indicate the ecological status of European coastal areas.Based upon samples collected from 2009 to 2012 in the Bohai Bay,we have tested the response of variations of M-AMBI,using biomass(M-BAMBI) in the calculations,with different transformations of the raw data.The results showed that the ecological quality of most areas in the study indicated by M-AMBI was from moderate to bad status with the worse status in the coastal areas,especially around the estuaries,harbors and outfalls,and better status in the offshore areas except the area close to oil platforms or disposal sites.Despite large variations in nature of the input data,all variations of M-AMBI gave similar spatial and temporal distribution patterns of the ecological status within the bay,and showed high correlation between them.The agreement of new ecological status obtained from all M-AMBI variations,which were calculated according to linear regression,was almost perfect.The benthic quality,assessed using different input data,could be related to human pressures in the bay,such as water discharges,land reclamation,dredged sediment and drilling cuts disposal sites.It seems that M-BAMBI were more effective than M-NABMI(M-AMBI calculated using abundance data) in indicating human pressures of the Bay.Finally,indices calculated with more severe transformations,such as presence/absence data,could not indicate the higher density of human pressures in the coastal areas of the north part of our study area,but those calculated using mild transformation(i.e.,square root) did. | CAI Wenqian BORJA Angel LIN Kuixuan ZHU Yanzhong ZHOU Juan LIU Lusan | 2015 | Acta Oceanologica Sinica2015，34，10： | 5 |

| 6 | Preclinical evaluation of azathioprine plus buthionine sulfoximine in the treatment of human hepatocarcinoma and colon carcinoma显示文摘AIM: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tumor in vivo.METHODS: Different hepatoma and colon carcinoma cell lines (HepG2, HuH7, Chang liver, LoVo, RKO, SW-48, SW-480) were grown in minimal essencial medium supplemented with 10% fetal bovine serum and 1% antibiotic/antimycotic solution and maintained in a humidified 37 ℃ incubator with 5% CO2. These cells were pretreated with BSO for 24 h and then with AZA for different times. We examined the effects of this combination on some proteins and on cellular death. We also studied the eff icacy and the safety of AZA (6 mg/kg per day) and BSO (90 mg/kg per day) in HepG2 tumor growth in vivo using athymic mice. We measured safety by serological markers such as aminotransferases and creatine kinase.RESULTS: The in vitro studies revealed a new mechanism of action for the AZA plus BSO combination in the cancer cells compared with other thiopurines (6-mercaptopurine, 6-methylmercaptopurine, 6-thioguanine and 6-methylthioguanine) in combination with BSO. The cytotoxic effect of AZA plus BSO in HepG2 cells resulted from necroptosis induction in a mitochondrial-dependent manner. From kinetic studies we suggest that glutathione (GSH) depletion stimulates c-Jun amino-terminal kinase and Bax translocation in HepG2 cells with subsequent deregulation of mitochondria (cytochrome c release, loss of membrane potential), and proteolysis activation leading to loss of membrane integrity, release of lactate dehydrogenase and DNA degradation. Some of this biochemical and cellular changes could be reversed by N-acetylcysteine (a GSH replenisher).
