| 1 | Glial cell-derived neurotrophic factor mRNA expression in a rat model of spinal cord injury following bone marrow stromal cell transplantation显示文摘BACKGROUND: Several animal experiments utilizing bone marrow stromal cell (BMSC) transplantation for the treatment of spinal cord injury have proposed a hypothesis that BMSC transplantation effects are associated with increased glial cell-derived neurotrophic factor (GDNF) expression. OBJECTIVE: To confirm the effects of BMSC transplantation on GDNF mRNA expression in rats with spinal cord injury by reverse transcription-polymerase chain reaction (RT-PCR). DESIGN, TIME AND SETTING: The present molecular, cell biology experiment was performed at the Key Laboratory of Children's Congenital Malformation, Ministry of Health of China & Department of Developmental Biology, Basic Medical College, China Medical University between March 2006 and May 2007. MATERIALS: Sixty healthy Wistar rats aged 2-4-months and of either gender were included in this study. Spinal cord injury was induced in all rats by hemisection of T9 on the left side. RT-PCR kits were purchased from TaKaRa Company, China. Type 9600 RCR amplifier was provided by PerkinElmer Company, USA. METHODS: Three rats were selected for BMSC culture and subsequent transplantation (after three passages). Of the remaining 57 rats, nine were selected for sham-operation (sham-operated group), where only the T9 spinal cord was exposed without hemisection. A total of 48 rats were randomly and evenly divided into BMSC transplantation and model groups. In the BMSC transplantation group, following spinal cord injury induction, each rat was administered a BMSC suspension through two injection sites selected on the gray and white matter boundary caudally and cephalically, seperately and near to injury site in the spinal cord. The model group received an equal volume of PBS through the identical injection sites. MAIN OUTCOME MEASURES: At 24 and 72 hours, as well as at 7 days, following spinal cord injury, the spinal cord at the T 9 segment was removed. Eight rats were allocated to each time point in the BMSC transplantation and model groups, with three rats allocated to the sham-operated group. GDNF mRNA expression was semiquantitatively analyzed by RT-PCR. RESULTS: The sham-operated group exhibited extremely low GDNF mRNA expression. GDNF mRNA expression significantly increased at 24 hours after spinal cord injury, reached a peak level at 72 hours, and slowly decreased thereafter. However, it remained higher than normal levels at 7 days (P < 0.05). At all time points following spinal cord injury, GDNF mRNA expression was significantly greater in the BMSC transplantation group than in the model group (P < 0.05). | 2 | Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. | 3 | Experimental investigation of ghost imaging of reflective objects with different surface roughness显示文摘We present an experimental demonstration of ghost imaging of reflective objects with different surface roughness.The influence of the surface roughness, the transverse size of the test detector, and the reflective angle on the signal-to-noise ratio(SNR) is analyzed by measuring the second-order correlation of the light field based on classical statistical optics. It is shown that the SNR decreases with an increment of the surface roughness and the detector's transverse size or a decrease of the reflective angle. | 18 | Lynch syndrome pre-screening and comprehensive characterization in a multi-center large cohort of Chinese patients with colorectal cancer显示文摘Objective:Lynch syndrome(LS)pre-screening methods remain under-investigated in colorectal cancers(CRCs)in Asia.Here,we aimed to systematically investigate LS pre-screening and comprehensively characterize LS CRCs.Methods:Microsatellite instability(MSI)and germline variants of DNA mismatch repair(MMR)genes were examined in 406 deficient MMR(dMMR)and 250 proficient MMR CRCs.The genetic differences between LS and sporadic CRCs were studied with whole exome sequencing analysis.Results:The incidence of dMMR in Chinese patients with CRCs was 13.8%.Consistency analysis between MMR immunohistochemistry(IHC)and MSI testing showed the kappa value was 0.758.With next-generation sequencing(NGS),germline variants were detected in 154 CRCs.Finally,88 patients with CRC were identified as having LS by Sanger sequencing.Among them,we discovered 21 previously unreported pathogenic germline variants of MMR genes.Chinese patients with LS,compared with sporadic CRCs,tended to be early-onset,right-sided,early-stage and mucinous.Overall,the performance of MMR IHC and MSI testing for LS pre-screening was comparable:the area under the ROC curve for dMMR,MSI-H,and MSI-H/L was 0.725,0.750,and 0.745,respectively.dMMR_MSI-H LS and sporadic CRCs showed substantial differences in somatic genetic characteristics,including different variant frequencies of APC,CREBBP,and KRAS,as well as different enriched pathways of VEGF,Notch,TGFβR,mTOR,ErbB,and Rac protein signal transduction.Conclusions:MMR IHC and MSI testing were effective methods for LS pre-screening.The revealed clinical and somatic genetic characteristics in LS CRCs may have the potential to improve the performance of LS pre-screening in combination with dMMR/MSI. | Yan Li Lihong Fan Jianming Zheng Xiu Nie Yu Sun Qin Feng Shenyi Lian Wenqi Bai Weijing Cai Yanan Yang Bo Su Yanfeng Xi Dongmei Lin | 2022 | Cancer Biology & Medicine2022,19,8: | 0 |
