4Cold acclimation improves photosynthesis by regulating the ascorbate–glutathione cycle in chloroplasts of Kandelia obovata显示文摘As the most northerly mangrove species in China, Kandelia obovata may undergo extreme cold event stress. Enhancing the cold tolerance of this species is crucial to its successful afforestation. This study aimed to determine the resistance of K. obovata seedlings to low temperature stress by cold acclimation and to explain the mechanisms for alleviating cold injury. To understand these mechanisms, seedlings that were acclimatized and not acclimatized were exposed to 5℃/- 2℃(day/night)for 48 h.Results showed that low temperature stress reduced leaf photosynthesis of non-acclimatized seedlings by inducing oxidative stress and structural damage to chloroplasts. These phenomena were shown by increasing levels of malondialdehyde (MDA), O2-and H2O2, as well as decreasing enzyme activities in the ascorbate–glutathione (AsA-GSH) cycle. However, cold-acclimatized seedlings had improved photosynthetic rates and efficiency of photosystem II (PSII) under low temperature stress. Compared with non-acclimatized seedlings, leaves of coldacclimatized seedlings under low temperature stress for 48 h exhibited higher anti-oxidative enzyme activities, lower levels of O2^- and H2O2, less damage to chloroplast structure, and removed 33.7% of MDA at low temperature stress for 48 h. 5SCM-198 protects endometrial stromal cells from oxidative damage through Bax/Bcl-2 and ERK signaling pathways显示文摘In creasi ng amounts of evidence dem on strated that accumulative reactive oxyge n species (ROS) and apoptosis of human endometrial stromal cells (ESCs) are closely associated with endometrial dysfunction induced by oxidative stress, which plays an important role in the pathological process of multiple gyn ecological and reproduction-related diseases. SCM-198, an alkaloid active component of Leonurus japonicas Houtt, has been reported to have anti-oxidative activity. However, the specific mechanisms of SCM-198 in the prevention of endometrial damage remain unknown. In the present study, we assessed the effect of SCM-198 on hydrogen peroxide (H2O2Hnduced oxidative injury in ESCs. ESCs were pretreated with SCM-198 for 4 h and then challenged with H2O2.Morphology ch a nges, apoptosis rate, and intracellular ROS producti on were measured to assess the level of oxidative injury. Flow cytometry and western blot analysis were performed to detect the expression levels of Bax, Bcl-2, active-caspase-3, and mitogen-activated protein kinases pathways. Classic inflammation cytokines were measured by real-time polymerase chain reactions. Our results showed that SCM-198 attenuated apoptosis and ROS generation of ESCs induced by H2O2. H2O2 induced the apparent apoptotic characteristics, including fragmentation of DNA, upregulation of Bax/Bcl2, activation of caspase-3, and secretion of inflammation cytokines, which were all ameliorated by SCM-198. Furthermore, H2O2-induced apoptosis-related ERK1/2 pathway activation was restrained by SCM-198 pretreatme nt. 6Image universal steganalysis based on best wavelet packet decomposition显示文摘Based on the best wavelet packet decomposition of images, a new universal steganalysis method with high detection correct ratio is proposed. First, the best wavelet packet decomposition of image based on the Shannon entropy information cost function is made. Second, high order absolute characteristic function moments of histogram extracted from the coefficient subbands obtained by best wavelet packet decomposition are regarded as features. Finally, these features are processed and a back-propagation (BP) neural network is designed to classify original and stego images. Three different steganalysis algorithms for three different cases of background and application condition are presented. To validate the performance of the proposed method, a series of experiments are made for six kinds of typical steganography methods, i.e. LSB, LTSB, PMK, Jsteg, F5 and JPHide. Results show that, the average detection accuracy of the proposed method exceeds at least 6.4% and up to 15.4% and has a better universal performance than its closest competitors. 7Laboratory Characterizations on 2007 Cases of Monoclonal Gammopathies in East China显示文摘Monoclonal gammopathies are characterized by the presence of monoclonal immunoglobulin in patients with or without evidence of multiple myeloma (MM),macroglobulinemia,amyloidosis (AL),or a related plasma cell proliferative disorder. This study aims to evaluate laboratory diagnostic characters of monoclonal gammopathies and investigates the correlation between monoclonal gammopathies and transforming growth factor β1 (TGFβ1). Immunofixation electrophoresis (IFE),serum protein electrophoresis (SPE),nephelometry and urine light chain ELISA were used for laboratory identification of monoclonal immunoglobulins. Plasma TGFβ1 was detected with double-antibodies ELISA. Lightcycler was used for single nucleotide polymorphism (SNP) analysis. Totally 2,007 cases of monoclonal immunoglobulin (M protein) were identified in 10,682 samples. The isotypes of M protein were IgG type 47.1%,IgA 23.0%,IgM 8.7%,IgD 5.3%,free light chain κ 6.1%,λ 9.8%. In reference to IFE,the coherency of diagnosis was serum light chain ratio (κ/λ) 94.4%,quantitation of Igs 83%,light chain quantitation 80.9%,and urine light chain ratio (κ/λ) 58.0%. Plasma TGFβ1 was elevated significantly compared to normal control. The allelic frequency of codon 10 (C > T) was neither associated with the existence of the M protein nor with the M protein isotype. Monoclonal gammopathies can be identified with the combination of IFE,SPE,Igs quantitaion and urine light chain determination. 9Fine Mapping of RppP25, a Southern Rust Resistance Gene in Maize显示文摘Southern rust (Puccinia polysora Underw.) is a major disease that can cause severe yield losses in maize (Zea mays L.). In our previous study, a major gene RppP25 that confers resistance to southern rust was identified in inbred line P25. Here, we report the fine mapping and candidate gene analysis of RppP25 from the near-isogenic line F939, which harbors RppP25 in the genetic background of the susceptible inbred line F349. The inheritance of resistance to southern rust was investigated in the BC1F1 and BC3F1 populations, which were derived from a cross between F939 and F349 (as the recurrent parent). The 1:1 segregation ratio of resistance to susceptible plants in these two populations indicated that the resistance is controlled by a single dominant gene. Ten markers, including three simple sequence repeat (SSR) markers and seven insertion/deletion (InDel) markers, were developed in the RppP25 region. RppP25 was delimited to an interval between P091 and M271, with an estimated length of 40 kb based on the physical map of B73. In this region, a candidate gene was identified that was predicted to encode a putative nucleotide-binding site leucine-rich repeat (NBS-LRR) protein. 10Redox condition during Ediacaran–Cambrian transition in the Lower Yangtze deep water basin, South China: constraints from iron speciation and δ^(13) Corg in the Diben section, Zhejiang显示文摘Cherts of the Piyuancun Formation and black mudstones of the Hetang Formation in Diben Town, Kaihua County, Zhejiang were deposited in a deep water basin of the Lower Yangtze area during the Ediacaran–Cambrian transition(E–C transition), offering an opportunity to reconstruct the ancient redox condition of the deep water basin. Total 53 samples were collected and analyzed for iron speciation, including pyrite iron(FePy), carbonate associated iron(FeCarb), iron oxide and hydroxide(FeOx),magnetite iron(FeMag) and total iron(FeT) as well as d13 Corgto gain a deeper understanding of specific redox condition during the period. The results show that(1) most samples have pyrite concentration \1 %, with the FePy/FeHR ratio ranging from \0.01 to 0.81(mostly \0.5), and the FeHR/FeT ratio between 0.39 and 0.93(mostly [0.5).(2) The E–C boundary in the Diben section occurred at the maximum negative excursion of organic carbon isotope within the Piyuancun Formation, which can be correlated with other sections by using d13 Corg. We thus concluded that the relatively high content of highly reactive iron and low content of pyrite iron in the Diben section indicate a minor occurrence of H2 S and a predominance of anoxic and ferruginous environment, which is similar to other reported sections such as the Longbizui and Yanwutan sections, western Hunan. In addition, oxygen concentration in sea water showed an increasing trend in the interval corresponding to the bottom and upper layer of the Hetang Formation as indicated by the FeHR/FeT ratio, which decreased from 0.7 to 0.38, however, it was not great enough to oxidize the whole deep water. Therefore, during the E–C transition deep water basin in South China was most likely an anoxic and ferruginous enrironment, which prevented metazoon from the evolution in the deep water basin. 11Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19.Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang 2021Signal Transduction and Targeted Therapy2021,6,4:4
12Effects of compound microbial inoculant treated wastewater irrigation on soil nutrients and enzyme activities显示文摘Wastewater treated by compound microbial inoculant(CMI)in agricultural irrigation can enrich soil fertility and decompose the possible pollutants.In this study,a greenhouse experiment using tomato as the model crop was performed to investigate the effects of treated wastewater irrigation on soil nutrients and enzymes.For this purpose,certain parameters were measured,including soil total nitrogen(N),nitrate N,total phosphorus(P),available potassium(K)and the activities of the enzymes urease,acid phosphatase and catalase in soils irrigated with fresh water,wastewater and CMI-treated wastewater under three amount of irrigation water.The results showed that irrigation with both treated and untreated wastewater significantly increased soil total N,total P,and available K,however the treated wastewater showed higher effects on soil enrichment,especially on available K.The activity of soil urease and acid phosphatase reached highest with treated wastewater irrigation,whereas wastewater irrigation increased the activity of catalase obviously.Soil enzyme and nutrient with fresh water irrigation decreased with increasing water amount;the content of soil urease,nitrate-nitrogen,total N and total P in treated wastewater and wastewater irrigation rose with increasing water amount,but the highest activity of acid phosphatase and the lowest activity of catalase were found in medium irrigation water amount.Under the condition of tomato cultivation,total N,nitrate N and total P were closely correlated with soil urease and catalase;there were significant positive correlation among soil urease,catalase,total N,nitrate N and total P;there existed significantly positive correlation between acid phosphatase and all measured soil nutrient indexes.The results suggested that irrigation with CMI-treated wastewater is a security and effective strategy to agricultural land management.Wang Chunfang Shao Xiaohou Xu Huilian Chang Tingting Wang Weina 2016International Journal of Agricultural and Biological Engineering2016,9,6:4
13Dynamic Analysis of Supply and Demand Coupling of Ecosystem Services in Loess Hilly Region:A Case Study of Lanzhou,China显示文摘The relationship between the supply and demand for ecosystem services(ESs)is a key issue for the rational allocation of natural resources and optimisation of sustainable development capacity.This paper investigateed the dynamic evolution features of supply and demand of four ESs in Lanzhou of China,namely,water supply,food supply,carbon fixation and soil retention services.The crosssectional data of 2005 and 2017 were used for calculating ESs value and its supply and demand through ArcGIS software,InVEST model,elastic coefficient model and coupling coordination model.Results showed that:1)from 2005 to 2017,the supply of water supply services increased,the demand of soil retention services decreased,and the supply and demand of food supply and carbon fixation services increased.The high-value areas of service supply were mainly distributed in the rocky mountain areas in the southeast and northwest with high vegetation coverage,while the high-value areas of demand were mainly distributed in the urban areas and surrounding areas with high population density.2)There were five different types of coupling relations.Water supply service was dominated by a negative coupling type D,which means that the decrease in demand for ESs has had a positive response on the supply of ESs.Negative coupling type C was the main type of food supply and carbon fixation services,which means that the increase in demand for ESs has had a negative response on the supply of ESs.All three services were supplemented by a positive coupling type A,which means that the increase in demand for ESs has had a positive response on the supply of ESs.Soil retention service generally exhibits a positive coupling type B,which means that the decrease in demand for ESs has had a negative response on the supply of ESs.3)Over the past 12 yr,the coordination degree of supply and demand of water supply,food supply and soil retention services decreased,and the coordination degree of carbon fixation service increased.Various types of ES had a low degree of coupling and coordination,showing different characteristics of temporal and spatial evolution.The areas with imbalanced ESs supply and demand were mainly distributed in urban areas dominated by construction land.The research results are valuable to the optimisation of urban and rural ecological environments and the sustainable development of territory space under the framework of ecological civilisation,including similar ecologically vulnerable areas in other developing countries.LI Pengjie LIU Chunfang LIU Licheng WANG Weiting 2021Chinese Geographical Science2021,31,2:3
14Expressions of cyclooxygenase-2 and matrix metalloproteinase-9 in cervical carcinoma and their clinical significance显示文摘Objective: To investigate the expressions of cyclooxygenase (COX)-2 and matrix metalloproteinase (MMP)-9 in cervical carcinoma and their clinical significance. Methods: Immunohistochemistry SP method was used to detect the expres- sions of COX-2 and MMP-9 in 72 cases of invasive carcinoma of cervix (ICC) and 16 cases of normal cervical epithelium remote from tumor (NCE). The relationships between the expressions of COX-2, MMP-9 in ICC and some characteristics relating to clinical pathology of cervical carcinoma such as histological grading, lymph node metastasis, stromal invasion and FIGO stage were analyzed statistically. Results: The rates of the positive expressions of COX-2 and MMP-9 in ICC were significantly higher than those in NCE. COX-2: 88.9% (64/72) in group ICC and 12.5% (2/16) in group NCE, P = 0.000; MMP-9: 94.4% (68/72) in group ICC and 43.8% (7/16) in group NCE, P = 0.000. The expression of COX-2 was positively correlated with lymph node metastasis (r = 0.296, P = 0.012) and stromal invasion (r = 0.257, P = 0.029). The expression of MMP-9 was positively correlated with FIGO stage (r = 0.329, P = 0.005) and histological grading (r = 0.351, P = 0.003). The expression of COX-2 was positively correlated with the expression of MMP-9 in ICC (r = 0.297, P = 0.011). Conclusion: The overexpressions of COX-2 and MMP-9 were closely related to the invasion and growth of cervical carcinoma. The tissue with the overexpression of COX-2 had strong invasion ability. COX-2 and MMP-9 had synergistic effect on proliferation, invasion and metastasis of cancer cells. 17Preparation and Characterization of Natural Rubber/Silica Nanocomposites using Rule of Similarity in Latex显示文摘Rule of similarity and latex compounding techniques were combined for the first time to prepare natural rubber/nanosilica(NR/SiO2) nanocomposite with core-shell nanosilica-poly(methyl methacrylate)(SiO2-PMMA) particles and PMMA-modified natural rubber matrix(NR-PMMA).The microstructure of SiO2 and nanocomposites with different SiO2 contents was characterized by fourier transform infrared spectroscopy(FTIR);the morphology of nanocomposites was investigated with scanning electron microscopy(SEM);the tensile strength was characterized by tensile testing machine and the thermal stability of composites was studied by thermal gravimetric analysis.Results showed that PMMA chains have successfully grafted onto the surface of SiO2,and the core-shell SiO2-PMMA nanoparticles and NR-PMMA latex have been perfectly incorporated.SiO2-PMMA nanoparticles are evenly distributed over the NR matrix with an average size in the range of 60-100 nm at the low content(SiO2 ≤ 3 wt%),while aggregations are apparently observed when 5 wt% SiO2 is loaded.In addition,NR/SiO2 composities possess a considerable improvement in ageing resistance compared with the pure NR.The tensile strength of composite increases from 6.99 to 12.72 MPa,reaching the highest value at a 0.5 wt% SiO2 loading,and then the figure decreases gradually because of the aggregation of SiO2 nanoparticles.It is anticipated that the reported process is to provide a simple and economic way for preparing NR composites.罗勇悦 FENG Chunfang WANG Qinghuang YI Zhifeng QIU Quanfang Kong LX 彭政 2013Journal of Wuhan University of Technology(Materials Science)2013,28,5:1
18Association between time in range and cancer mortality among patients with type 2 diabetes:a prospective cohort study显示文摘Background:Little was known about the association among time in range(TIR),time above range(TAR),time below range(TBR),and cancer mortality among patients with type 2 diabetes.We aimed to investigate the association among TIR,TAR,TBR,and the risk of cancer mortality among patients with type 2 diabetes.Methods:A total of 6225 patients with type 2 diabetes were prospectively recruited in Shanghai,China.TIR was measured with continuous glucose monitoring at baseline and was defined as the average percentage of time in the target glucose range during a 24 h period.Cox proportion hazard regression analysis was used to determine the association between TIR and the risk of cancer mortality.Results:During a mean follow-up of 7.10 years,we confirmed 237 death events related to cancer.The multivariable-adjusted hazard ratio(HR)for cancer mortality was 1.32(95%confidence interval[CI]:1.01-1.75)in patients with TIR≤70%compared with those with TIR>70%.When TIR was considered as a continuous variable,the multivariable-adjusted HR for cancer mortality associated with each 10%decrease in TIR was 1.07(95%CI:1.02-1.14).In the site-specific analysis,a significant association between TIR as a continuous variable and the risk of hepatocellular cancer was found(HR:1.24;95%CI:1.09-1.41).However,no relationship between hemoglobin A1c and cancer mortality was observed(HR:1.04;95%CI:0.97-1.10).Conclusions:The present study found an inverse association of TIR with the risk of cancer mortality among patients with type 2 diabetes.New evidence of TIR was added into the clinical practice that TIR may be an optimal target of glycemic control among patients with type 2 diabetes.Yun Shen Chunfang Wang Yaxin Wang Jingyi Lu Lei Chen Lei Zhang Wei Lu Wei Zhu Gang Hu Tian Xia Jian Zhou 2022Chinese Medical Journal2022,,3:1
19Biological effects of extract from newborn porcine liver on hepatocytes,hepatic stellate cells,and hepatoma cell line显示文摘Objective: Porcine liver extract has been shown to be effective in the clinical treatment of severe hepatitis. The aim of the present study was to study its antifibrotic as well as immune regulatory effect in vitro. Methods: Hepatocytes, hepatic stellate cells (HSCs), hepatoma cell line (HepG2) and human peripheral blood mononuclear cells (PMNCs) were studied with respect to proliferation, extracellular matrix production and apoptotic activities by proliferation assay, radioimmunoassay, gene transfection, reporter gene analysis and flow cytometry, respectively. Results: A strong stimulatory proliferation effect was observed in hepatocytes, and an inhibitory effect was found in HSCs. Hyaluronic acid (HA) production and reporter gene activities driven by various α1(I) procollagen gene promoters in HSC-T6 were significantly decreased after treatment with the extract. Fluo-Anexin V binding apoptotic HepG2 cells were more prominent in the presence of 60 μg/ml extract. More CD4+/CD69+ positive T lymphocytes existed in the presence of the extract. Conclusion: Porcine liver extract is effective for antifibrogenesis via hepatocyteregeneration, HSC and hepatoma cell inhibition in vitro. The elevation of active T lymphocytes is helpful for immune surveillance.
