Brazilian biosafety law and the new breeding technologies显示文摘Globally, the area of land cultivated with genetically modified(GM)crops has increased a thousand-fold over the last two decades.Although this technology has become important for food production, the regulatory frameworks that underpin these outcomes are based on a list of requirements for a risk assessment that differ from country to country. In recent years, policymakers have had the opportunity to learn from the controversies over transgenics to create effective regulatory milestones for emerging technologies, allowing them to reach their potential for a more sustainable agriculture, ensuring food security. In Brazil,Law No.11.105 of 24 March 2005 established a framework with four main organizations responsible for risk assessment and management. However, most of new breeding technologies did not exist at that time and were not considered in this law.In2016,Normative Resolution No.16 of the National Biosafety Technical Commission(CTNBio)was established to address this gap based on the evaluation of the products obtained through these techniques(termed Innovative Precision Improvement Techniques in the resolution),in a case-by-case consultation system.Briefly,if the product is designated to be a GM,the developer will have to go through the biosafety requirements and will be approved only after CTNBio risk assessment. If the product is designated not to be GM(for the purposes of the legislation),then it can be registered using the existing procedures.Currently,152 GM products are commercially approved in Brazil. In 2018,CTNBio assessed the first consultation on commercial release of plants generated using the new breeding technologies and has subsequently approved six products. It is expected that many institutions would be able to participate in Brazilian and world markets, developing and introducing new biotechnological solutions and products through a more sustainable approach and without facing public disapproval,a common issue for GM crops.Alexandre Lima NEPOMUCENO Renata FUGANTI-PAGLIARINI Maria Sueli Soares FELIPE Hugo Bruno Correa MOLINARI Edivaldo Domingues VELINI Eduardo Romano de Campos PINTO Maria Lucia Zaidan DAGLI Galdino ANDRADE FILHO Patricia Machado Bueno FERNANDES 2020Frontiers of Agricultural Science and Engineering2020,7,2:5
Clinical hematological and biochemical parameters in Swiss,BALB/c,C57BL/6 and B6D2F1 Mus musculus显示文摘Background:Animal models are widely used in scientific research in order to obtain information from a whole organism under a specific set of experimental conditions.Various lineages of mice have been used to investigate diseases and new therapeutic strategies, and, consequently, hematological and biochemical tests in these laboratory animals are essential to validate scientific studies. Our study seeks to establish reference values for hematological and biochemical parameters of four lineages of mice. Methods:We evaluated the hematological and biochemical profiles of 20 males and 20 females from the lineages Swiss(heterogeneous),BALB/c and C57BL/6(isogenic),and B6D2F1(hybrid),totaling 160 mice.Analysis were standardized using the systems pocH-100iV Diff™for 19 hematological parameters and VITROS®350 for 12 biochemical parameters. Results:Results are shown as means and standard deviation, grouped by lineage and genre.Comparing the values obtained in this study with the values from previous studies, some variations were detected, which could be explained by differences in methodologies or individual variability. Conclusion:Thus our study shows that knowledge and disclosure of the values of physiological parameters of laboratory animals is necessary, and emphasises the importance of considering variations influenced by gender, lineage and genotype in the choice of the best experimental model.Giorgio Silva-Santana Juliet Cunha Bax Débora Cristina Silva Fernandes Daniela Tendler Leibel Bacellar Cleber Hooper Alexandre Alves Souza Oliveira Dias Cristina Barbosa Silva Aline Moreira de Souza Simone Ramos Ricardo Alexandre Santos Thainara Ramos Pinto Mariana Antunes Ramão Ana Luíza Mattos-Guaraldi 2020Animal Models and Experimental Medicine2020,3,4:2
Acute cholestatic hepatitis caused by amoxicillin/ clavulanate显示文摘Amoxicillin/clavulanate is a synthetic penicillin that is currently commonly used, especially for the treatment of respiratory and cutaneous infections. In general,it is a well-tolerated oral antibiotic.However,amoxicillin/clavulanate can cause adverse effects, mainly cutaneous, gastrointestinal, hepatic and hematologic,in some cases.Presented here is a case report of a 63-yearold male patient who developed cholestatic hepatitis after recent use of amoxicillin/clavulanate.After 6 wk of prolonged use of the drug,he began to show signs of cholestatic icterus and developed severe hyperbilirubinemia(total bilirubin>300 mg/L).Diagnostic investigation was conducted by ultrasonography of the upper abdomen, serum tests for infection history, laboratory screening of autoimmune diseases, nuclear magnetic resonance(NMR)of the abdomen with bile duct-NMR and transcutaneous liver biopsy guided by ultrasound. The duration of disease was approximately 4 mo,with complete resolution of symptoms and laboratory changes at the end of that time period.Specific treatment was not instituted, only a combination of anti-emetic(metoclopramide)and cholestyramine for pruritus.Daniel Oliveira Beraldo Joanderson Fernandes Melo Alexandre Vidal Bonfim Andrei Alkmim Teixeira Ricardo Alkmim Teixeira André Loyola Duarte 2013World Journal of Gastroenterology2013,19,46:2
Biochar Increases Diuron Sorption and Reduces the Potential Contamination of Subsurface Water with Diuron in a Sandy Soil显示文摘The herbicide diuron is widely used in agricultural areas in Brazil, whereas it has high potential for subsurface water contamination due to its physicochemical characteristics. Recent studies have demonstrated the potential of biochar as a sorbent and possible pesticide leaching mitigation. The objective of this study was to investigate the long-term effect of biochar application on the kinetics of sorption and desorption of diuron in a Cerrado Haplic Plinthosol. Samples were collected in an experiment conducted in the field in a randomized block design consisting of the combination of two levels of fertilizer application (0 and 300 kg ha −1 of 5-25-15 formula of NPK fertilizers) and three doses of biochar (0, 16, and 32 Mg ha −1 ). The Freundlich isotherm accurately described the sorption of diuron in all treatments. Biochar application increased the sorption and reduced the desorption of diuron. This effect was attributed to the contribution of biochar to total organic carbon (C) and C in the humin fraction and to the increase in the reactivity of the humic acid and humin fractions, which was significantly highly correlated with the sorption coefficient ( K f ). A positive correlation between the partition coefficient of organic C and K f confirmed the importance of the soil organic compartment for the sorption of diuron. The higher diuron sorption and lower diuron desorption capacities of sandy soils after biochar application could reduce the potential risk of diuron leaching and contamination of subsurface water.Fabiano APETTER Tamara SFERREIRA Adilson PSINHORIN Larissa BLIMA Fernandes AALMEIDA Leandro PPACHECO Alexandre FSILVA 2019Pedosphere2019,29,6:1

Vitamin D deficiency, functional status, and balance in older adults with osteoarthritis显示文摘BACKGROUND Low vitamin D levels are associated with a more severe case of knee osteoarthritis(OA).However, there are few published reports concerning an association between vitamin D deficiency and functional status of individuals with OA and no reports about postural balance in this population.AIM To analyze the relationship between vitamin D deficiency and severity, functional status, and balance in elderly patients with OA.METHODS In this cross-sectional study,105 elderly patients with hip and knee OA were included. The severity was assessed by the Kellgren-Lawrence criteria. The functional status was assessed with the Lequesne index.Postural balance was assessed using a force platform, and center-of-pressure parameters(velocity at anteroposterior and mediolateral axis)were used as the balance outcomes.Serum 25(OH)vitamin D levels were measured using a chemiluminescence method.RESULTS Most of the patients(mean age:70.6±6.5 years)were female(n=78,74.3%).In the group with vitamin D deficiency,43 patients(56.6%)had severe OA,while 33 patients(43.4%)had mild or moderate OA(χ^(2) test,P=0.04).Patients with vitamin D deficiency showed a higher Lequesne index score(Mann-Whitney test,P=0.04),indicating a worse functional impairment when compared to individuals with normal vitamin D levels.Additionally, patients with vitamin D deficiency had worse postural balance according to the Mann-Whitney test(P=0.03).CONCLUSION Vitamin D deficiency is associated with worse severity, functional status, and postural balance in patients with OA.Cláudia N Montemor Marcos Tadeu P Fernandes Audrey S Marquez Regina Célia Poli-Frederico Rubens Alexandre da Silva Karen B P Fernandes 2021World Journal of Clinical Cases2021,9,31:0
Oxidative stress and labile plasmatic iron in anemic patients following blood therapy显示文摘AIM: To determine the plasmatic iron content and evaluate the oxidative stress(OS) markers in subjects receiving blood therapy.METHODS: Thirty-nine individuals with unspecified anemia receiving blood transfusions and 15 healthy subjects were included in the study. Anemic subjects were divided into three subgrou P:(1) those that received up to five blood transfusions(n = 14);(2) those that received from five to ten transfusions(n = 11); and(3) those that received more than ten transfusions(n = 14). Blood samples were collected by venous arm puncture and stored in tubes containing heparin. The plasma and cells were separated by centrifugation and subsequently used for analyses. Statistical analyses were performed using Kruskal-Wallis analysis of variance followed by Dunn's multiple comparison tests when appropriate.RESULTS: The eletrophoretic hemoglobin profiles of the subjects included in this study indicated that no patients presented with hemoglobinopathy. Labile plasmatic iron, ferritin, protein carbonyl, thiobarbituric acidreactive substances(TBARS) and dichlorofluorescein diacetate oxidation were significantly higher(P < 0.05), whereas total thiol levels were significantly lower(P < 0.05) in transfused subjects compared to controls. Additionally, the activity of catalase, superoxide dismutase and glutathione peroxidase were significantly lower in the transfused subjects(P < 0.05). Antioxidant enzyme activities and total thiol levels were positively correlated(P < 0.05), and negatively correlated with the levels of protein carbonyl and TBARS(P < 0.05). In contrast, protein carbonyl and TBARS were positively correlated(P < 0.05). Altogether, these data confirm the involvement of OS in patients following therapy with repeated blood transfusions.CONCLUSION: Our data reveal that changes in OS markers are correlated with levels of labile plasmatic iron and ferritin and the number of transfusions.Marília Sabo Fernandes Tatiana Tamborena Rissi Luisa Zuravski Juliana Mezzomo Carmen Regla Vargas Vanderlei Folmer Félix Alexandre Antunes Soares Vanusa Manfredini Mushtaq Ahmed Robson Luiz Puntel 2014World Journal of Experimental Medicine2014,4,3:0
