1p16 promoter hypermethylation:A useful serum marker for early detection of gastric cancer显示文摘AIM: To determine p16 promoter hypermethylation in gastric tumoral tissue and serum samples, its impact on p16-protein expression, and correlation with clinical and histological features. METHODS: Samples were obtained from 52 histologically confirmed cases of gastric adenocarcinoma. Gastric tissue and serum of 50 age- and sex-matched individuals with normal gastroscopy and biopsy were obtained as control samples. Methylation-specific polymerase chain reaction (MSP) was used to evaluate methylation status of p16 promoter. p16-protein expression was analyzed by immunohistochemical staining on paraffin-embedded sections. RESULTS: Methylation was detected in 44.2% (23/52) of tumoral tissues. 60.9% of them were also methylated in serum, i.e., 26.9% of all patients (14/52). Methylation was not detected in tissue and sera of control samples. p16-protein expression was decreased in 61.5% of cases (32/52), and was significantly associated with promoter hypermethylation (P < 0.001). Methylation was significantly more frequent in higher pathological grades (P < 0.05). Methylation was not associated with other clinicopathological features and environmental factors including H pylori infection and smoking. CONCLUSION: p16 promoter hypermethylation is an important event in gastric carcinogenesis. It is the principle mechanism of p16 gene silencing. It is related to malignant tumor behavior. 3Stool-based DNA testing,a new noninvasive method for colorectal cancer screening,the first report from Iran显示文摘AIM:To detect tumor-associated DNA changes in stool samples among Iranian patients with colorectal cancer(CRC)compared to healthy individuals using BAT-26,p16 hypermethylation and long DNA markers.METHODS:Stool DNA was isolated from 45 subjects including 25 CRC patients and 20 healthy individuals using a new,fast and easy extraction method.Long DNA associated with tumor was detected using polymerase chain reaction method.Microsatellite studies were performed utilizing denaturating polyacrylamide gel to determine the instability of BAT-26.Methylation status of p16 promoter was analyzed using methylation-specific PCR(MSP).RESULTS:The results showed a significant difference in existence of long DNA(16 in patients vs 1 in controls,P < 0.001)and p16(5 in patients vs none in controls,P = 0.043)in the stool samples of two groups.Long DNA was detected in 64% of CRC patients;whereas just one of the healthy individuals was positive for Long DNA.p16 methylation was found in 20% of patients and in none of healthy individuals.Instability of BAT-26 was not detected in any of stool samples.CONCLUSION:We could detect colorectal cancer related genetic alterations by analyzing stool DNA witha sensitivity of 64% and 20% and a specificity of 95% and 100% for Long DNA and p16 respectively.A non-invasive molecular stool-based DNA testing can provide a screening strategy in high-risk individuals.However,additional testing on more samples is necessary from Iranian subjects to determine the exact specificity and sensitivity of these markers.Mohammad Reza Abbaszadegan Alireza Tavasoli Arash Velayati Hamid Reza Sima Hassan Vosooghinia Mehdi Farzadnia Hamid Asadzedeh Mehran Gholamin Ezzat Dadkhah Azadeh Aarabi 2007World Journal of Gastroenterology2007,13,10:14
4Risk assessment for the daily intake of polycyclic aromatic hydrocarbons from the ingestion of cockle (Anadara granosa) and exposure to contaminated water and sediments along the west coast of Peninsular Malaysia显示文摘The concentration of carcinogenic polycyclic aromatic hydrocarbons (c-PAHs) present in the sediment and water of Peninsular Malaysia as well as in the cockle Anadara granosa was investigated.Samples were extracted and analysed with gas chromatographymass spectrometry.The concentrations of total carcinogenic polycyclic aromatic hydrocarbons (t-PAHs) were measured between 0.80 ± 0.04 to 162.96 ± 14.74 ng/g wet weight (ww) in sediment,between 21.85 ± 2.18 to 76.2 ± 10.82 ng/L in water samples and between 3.34 ± 0.77 to 46.85 ± 5.50 ng/g ww in the cockle tissue.The risk assessment of probable human carcinogens in the Group B2 PAHs was calculated and assessed in accordance with the standards of the United States Environmental Protection Agency (US EPA).Case I in the toxicity assessment analysed the cancer risk to consumers of Malaysian blood cockle.Case II assessed the risk of cancer from exposure to PAHs from multiple pathways.The average cancer risk of case I and case II were found to be classifiable as unsafe according to the US EPA standard.The cancer risk due to c-PAHs acquired by the ingestion of blood cockle was (8.82 ± 0.54) × 10-6 to (2.67 ± 0.06) × 10-2,higher than the US EPA risk management criterion.The non-cancer risks associated with multiple pathways in Kuala Gula,Kuala Juru and Kuala Perlis were higher than the US EPA safe level,but the non-cancer risk for eating blood cockle was below the level of US EPA concern.Seiedeh Aghileh Mirsadeghi Mohamad Pauzi Zakaria Chee Kong Yap Azadeh Shahbazi 2011Journal of Environmental Sciences2011,23,2:11
5Therapeutic potential of flavonoids in inflammatory bowel disease: A comprehensive review显示文摘The inflammatory process plays a central role in the development and progression of numerous pathological situations,such as inflammatory bowel disease(IBD),autoimmune and neurodegenerative diseases,metabolic syndrome,and cardiovascular disorders. IBDs involve inflammation of the gastrointestinal area and mainly comprise Crohn's disease(CD) and ulcerative colitis(UC). Both pathological situations usually involve recurring or bloody diarrhea,pain,fatigue and weight loss. There is at present no pharmacological cure for CD or UC. However,surgery may be curative for UC patients. The prescribed treatment aims to ameliorate the symptoms and prevent and/or delay new painful episodes. Flavonoid compounds are a large family of hydroxylated polyphenolic molecules abundant in plants,including vegetables and fruits which are the major dietary sources of these compounds for humans,together with wine and tea. Flavonoids are becoming very popular because they have many health-promoting and disease-preventive effects. Most interest has been directed towards the antioxidant activity of flavonoids,evidencing a remarkable free-radical scavenging capacity. However,accumulating evidence suggests that flavonoids have many other biological properties,including anti-inflammatory,antiviral,anticancer,and neuroprotective activities through different mechanisms of action. 6Using Kalman filter algorithm for short-term traffic flow prediction in a connected vehicle environment显示文摘We develop a Kalman filter for predicting traffic flow at urban arterials based on data obtained from con-nected vehicles. The proposed algorithm is computationally efficient and offers a real-time prediction since it invokes the connected vehicle data just before the prediction period. Moreover, it can predict the traffic flow for various pene-tration rates of connected vehicles (the ratio of the number of connected vehicles to the total number of vehicles). At first, the Kalman filter equations are calibrated using data derived from Vissim traffic simulator for different penetra-tion rates, different fluctuating arrival rates of vehicles and various signal settings. Then the filter is evaluated for a variety of traffic scenarios generated in Vissim simulator. We evaluate the performance of the algorithm for different penetration rates under several traffic situations using some statistical measures. 7轴向和循环荷载组合下夹角对纤维复合材料包裹钢筋混凝土梁柱节点性能的影响显示文摘通过数字分析,研究轴向和循环荷载作用下夹角对采用碳纤维复合包裹的钢筋混凝土梁柱节点的剪力和延性的影响。对于梁柱节点,建立了三维非线性有限元模型并采用Marc.MentatTM2001有限元分析软件进行模拟。采用已有文献中纤维增强复合材料包裹梁柱节点试验研究所提供的资料,对有限元模型的精度进行评估。用有限元方法分析不同夹角配置的碳纤维增强复合聚合物包裹加固的3个梁-柱节点的性能。分析结果显示:与水平轴成±45°,连续4层包裹纤维增强复合聚合物,对于提高轴向和循环荷载组合下梁-柱节点的剪力和延性,是最好的设计方案。Azadeh Parvin 2008钢结构2008,23,7:9
8Long-term impact of municipal sewage irrigation on treated soil and black locust trees in a semi-arid suburban area of Iran显示文摘The effects of municipal sewage irrigation on the soil and black locust (Robinia pseudoacacia L.) tree were studied. For this purpose, two artificial black locust stands under irrigation of municipal sewage and well water were selected in south of Tehran, Iran. Data were collected using technique of systematic random sampling with 4 replicates in each stand. It was found that the growth of black locust tree, as indicated by diameter at breast height, total height, crown length, average crown diameter, basal area and volume, in sewage irrigation stand was much higher than that of well water irrigation stand (P < 0.01). Plant analysis indicated that concentrations of leaf nutrients (N, P, K, Ca, Mg, Na, Fe, Mn, Cu and Zn) were greater in sewage-irrigated trees, without toxicity to the minerals of tree leaf, than those of well waterirrigated trees, and positively correlated with their respective value in soil. Ni, Cr and Pb were not detected in leaf samples. Application of sewage resulted in a 1.5-fold increase in the concentrations of soil nutrients, Ni, Cr and Pb. Among these minerals only Pb and Ni in some soil samples exceeded the toxicity limit. The increase in pH, electrical conductivity (EC) and organic carbon of soil was also observed in sewage irrigation. Results confirm that besides the use as irrigation water, municipal sewages are also a potential source of plant nutrients. 9Cattle toxoplasmosis in Iran:a systematic review and meta-analysis显示文摘Objective:To analyze and review the overall seroprevalence rate of Toxoplasma gondii(T.gondii) infection in cattle from Iran.Methods:In the current study,data collection(published and unpublished papers,abstracts of national scientific congresses and dissertations) using particular terms was carried out systematically on the following electronic databases like PubMed,Google Scholar,Ebsco,Science Direct,Scopus,Magiran,Irandoc,IranMedex and SID(Scientific Information Database).Results:A total of 22 studies since 1983 to 2012 reporting the seroprevalence of toxoplasmosis in cattle from different regions of Iran met our eligibility criteria.The pooled proportion of toxoplasmosis,using random effect model,among cattle in Iran from over the 30-year period was estimated 18.1%(95%CI:9.9%to 28.2%).Conclusions:This study firstly establishes a crude seroprevalence rate of Toxoplasma infection in cattle which can lead us to understand the condition of eattle toxoplasmosis,which have to take into accounted fur an appropriate and effective prevention and controls.Secondly,it compares and discusses elaborately the role of risk factors including sex,age and breed in the epidemiology of the disease.Thus,it determines gaps and drawbacks in the prior studies which are greatly useful to design more accurate investigations in the future.Shahabeddin Sarvi Ahmad Daryani Mohammad Taghi Rahimi Mohsen Aarabi Azar Shokri Ehsan Ahmadpour Azadeh Mizani Mahdi Sharif 2015Asian Pacific Journal of Tropical Medicine2015,8,2:4
10Effect of Stimulating the BL67 Point on Fetal Correction from Breech to Cephalic Presentation and Natural Delivery after the 36 Weeks of Pregnancy: A Randomized Clinical Trial显示文摘Objective:Various techniques are proposed for changing fetal presentation.We aimed to assess the effect of BL67 point stimulation on correcting breech presentation and natural delivery in women at 36-38 gestational weeks.Methods:A parallel single blinded randomized clinical trial was conducted on 72 eligible pregnant women with breech presentation at the 36 weeks of pregnancy.The subjects were divided into two groups-intervention(n=36)and control groups(n=36)by block randomization method.The intervention group stimulated the BL67 point by self-administration for 20 min once a day for 2 weeks.Finally,the appearance of cephalic presentation and rate of vaginal delivery was compared between the groups(n1=n2=32)using the Chi-square test and multivariate logistic regression.Results:The correction of breech to cephalic presentation occurred in 53.1% of patients in the intervention group.The adjusted relative risk(RR)for fetal correction from breech to cephalic was 1.80(RR=1.80,95%confidence interval[CI],1.13-5.17).It was shown that the stimulation of the BL67 point increased the correction of breech to cephalic presentation.In addition,the rate of vaginal delivery increased by>4-fold(RR=4.16,95%CI,2.54-6.82)by correction to cephalic presentation.Moreover,65.6%of mothers in intervention group and 90.6%in the control group underwent cesarean section.Conclusions:The stimulation of the BL67 point is a safe,inexpensive,and effective method that can be self-administered at home for fetal correction from breech to cephalic presentation in women with breech presentations during 36-38 gestational weeks.This promotes uncomplicated natural childbirth.Khatereh Sourani Abolfazl Mohammadbeigi Nasim Khademi Azadeh Asgarian Imaneh Khaki Zohreh Ahmadi 2020Reproductive and Developmental Medicine2020,4,2:3
11The Electrochemical Behavior of Au/Au NPs/PNA/ZnS e-QD/ACA Electrode Towards Cy SH Oxidation显示文摘This work describes the electrochemical behavior of azodicarboxamide(ACA) film immobilized on the surface of penicillamine(PNA)/Zn Se-quantum dot(Zn Se-QD) gold nanoparticle(Au NPs) Au electrode. Electrocatalytic activity of modified electrode toward the oxidation of cysteine(Cy SH) was investigated. The surface structure and composition of the sensor were characterized by scanning electron microscopy(SEM). Oxidation of Cy SH on the surface of modified electrode was investigated with cyclic voltammetry, electrochemical impedance spectroscopy(EIS),hydrodynamic voltammetry and chronoamperometry methods. The results show that the PNA/Zn Se-QD/ACA film displays excellent electrochemical catalytic activities towards Cy SH oxidation. The modified electrode shows reproducible behavior and high level of stability during the electrochemical experiments. Also it has short response time, low detection limit, high sensitivity and low operation potential, which can be used as an amperometric sensor for monitoring of Cy SH. 12Fabrication of a Highly Sensitive Hydrazine Electrochemical Sensor Based on Bimetallic Au-Pt Hybrid Nanocomposite onto Modified Electrode显示文摘In this research a novel nickel complex was used as electrocatalyst for electrooxidation of hydrazine.A nano-structured nickel-complex was electrodeposited on a bimetallic Au-Pt inorganic-organic hybrid nanocomposite modified electrode.The electrode possesses a three-dimensional(3D) porous network nanoarchitecture,in which the bimetallic Au-Pt NPs serving as metal nanoparticle based microelectrode ensembles are distributed in the matrix of interlaced 3,3′,5,5′-Tetramethylbenzidine(TMB) organic nanofibers(NFs).Surface structure and composition of the sensor was characterized by scanning electron microscopy.Electrocatalytic oxidation of hydrazine on the surface of modified electrode was investigated with cyclic voltammetry method.The results showed that the nickelcomplex films displayed excellent electrochemical catalytic activities towards hydrazine oxidation.The hydrodynamic amperometry at rotating modified electrode at constant potential versus reference electrode was used for detection of hydrazine.Under optimized conditions the calibration plots were linear in the concentration range of 0.2-85 μM and detection limit was found to be 0.1 μM.The modified electrode exhibited reproducible behavior and a high level stability during the electrochemical experiments,making it particularly suitable for the analytical purposes.Azadeh Azadbakht Amir Reza Abbasi 2010Nano-Micro Letters2010,2,4:3
13MicroRNA in rectal cancer显示文摘In rectal cancer,one of the most common cancers worldwide,the proper staging of the disease determines the subsequent therapy.For those with locally advancedrectal cancer,a neoadjuvant chemoradiotherapy(CRT) is recommended before any surgery.However,response to CRT ranges from complete response(responders) to complete resistance(non-responders).To date we are not able to separate in advance the first group from the second,due to the absence of a valid biomarker.Therefore all patients receive the same therapy regardless of whether they reap benefits.On the other hand almost all patients receive a surgical resection after the CRT,although a watch-and-wait procedure or an endoscopic resection might be sufficient for those who responded well to the CRT.Being highly conserved regulators of gene expression,micro RNAs(mi RNAs) seem to be promising candidates for biomarkers.Many studies have been analyzing the mi RNAs expressed in rectal cancer tissue to determine a specific mi RNA profile for the ailment.Unfortunately,there is only a small overlap of identified mi RNAs between different studies,posing the question as to whether different methods or differences in tissue storage may contribute to that fact or if the results simply are not reproducible,due to unknown factors with undetected influences on mi RNA expression.Other studies sought to find mi RNAs which correlate to clinical parameters(tumor grade,nodal stage,metastasis,survival) and therapy response.Although several mi RNAs seem to have an impact on the response to CRT or might predict nodal stage,there is still only little overlap between different studies.We here aimed to summarize the current literature on rectal cancer and mi RNA expression with respect to the different relevant clinical parameters.azadeh azizian jens gruber b michael ghadimi jochen gaedcke 2016World Journal of Gastrointestinal Oncology2016,8,5:3
14Hydrothermal Alteration Mapping in Northern Khur, Iran, Using ASTER Image Processing: a New Insight to the Type of Copper Mineralization显示文摘Eastern Iran has great potential for the discovery of different types of mineralization. The study area encompasses Tertiary magmatism in the northern Lut block located in northern Khur, South Khorasan, eastern Iran and is mostly covered by volcanic rocks, which are intruded by porphyritic subvolcanic intrusions in some places. Application of the spectral angle mapper (SAM) technique to Advanced Spaceborne Thermal Emission and Reflection Radiometer (ASTER) images detected sericitic, argillic, and propylitic alterations, silicification, and secondary iron oxides. The alteration is linear and associated within vein-type mineralization. Twelve prospective areas are selected for detailed exploration and based on our processing results, in addition to NW-SE faults, which are associated with Cu mineralization indications, NE-SW faults are also shown to be important. Based on the presence of subvolcanic rocks and numerous Cu ± Pb-Zn vein-type mineralizations, extensive alteration, high anomaly of Cu and Zn (up to 100 ppm), the age (43.6 to 31.4 Ma) and the initial 87Sr/86Sr ratio (0.7047 to 0.7065) of the igneous rocks, and the metallogenic epoch of the Lut block (middle Eocene-lower Oligocene) for the formation of porphyry Cu and epithermal deposits, the studied area shows great potential for porphyry copper deposits.Azadeh Malekzadeh SHAFAROUDI Mohammad Hassan KARIMPOUR 2013Acta Geologica Sinica(English Edition)2013,87,3:3

16玻璃体腔组织纤溶酶原激活剂治疗玻璃体黄斑粘连(英文)显示文摘目的:评估仅玻璃体腔注射组织纤溶酶原激活剂(TPA)对玻璃体黄斑牵引以及玻璃体腔注射TPA和贝伐单抗对视网膜血管疾病的改善作用。方法:前瞻性研究。对24例24眼患者进行干预性系统研究。其中5眼玻璃体黄斑牵引综合征(VMT),19眼视网膜血管疾病包括:糖尿病黄斑水肿(DME)眼,糖尿病性玻璃体出血(VH)眼,视网膜中央静脉阻塞(CRVO)和新生血管年龄相关性黄斑变性眼(AMD)。在注射前及注射1mo后分别进行视力,B超和OCT检查。3眼VMT接受玻璃体腔单次注射TPA50μg,2眼接受100μg注射。19例视网膜血管疾病患者接受玻璃体腔组织纤溶酶原激活剂(50μg)和贝伐单抗(1.25 mg)联合治疗。结果:纳入病例中男性10眼视网膜血管疾病和VMT患者平均年龄分别为56.8y和60.4y。纳入病例中男性10眼(41.7%),女性14眼(58.3%)。22眼(91.7%)晶状体眼,2眼(8.3%)人工晶状体眼。VMT和视网膜血管疾病的玻璃体后部脱离(PVD)分别为0(0/5)和57.8%(11/19)(P=0.04)。在改善最佳矫正视力(BCVA)和降低黄斑中心凹厚度(CMT)方面,与无PVD眼相比,有PVD眼改善更多。结论:在VMT患者中,单独玻璃体腔内注射TPA不能成功诱导玻璃体后部完全脱离。玻璃体腔联合注射TPA和贝伐单抗可引起视网膜血管疾病患者玻璃体后部脱离,提高最佳矫正视力以及降低黄斑中心凹厚度。Saeed Karimi Masoud Soheilian Homayoun Nikkhah Azadeh Haseli Mofrad 2018国际眼科杂志2018,18,2:3

18A simple method for inducing nonalcoholic steatohepatitis with fibrosis显示文摘Background: Nonalcoholic fatty liver disease(NAFLD) is increasingly occurring in sedentary people, and may progress to NASH and hepatocellular carcinoma. It is essential to design affordable animal models for the study of various diseases, including fatty liver, which was the aim of the study. In this study, a high-fat diet was devised that triggers NASH’s animal model quickly and easily. High-fat diet(HFD) was used both with intra-mouth oral gavage and in combination with animal pellets.Methods: Twenty-four male C57 BL/6 J mice were divided into HFD and ND groups, which received a high-fat diet and a normal diet, respectively. At the end of the experiment(fourth week of treatment), body and liver weights, biochemical parameters, PPAR-α gene expression and histopathologic characteristics of the liver were evaluated.Results: During 4 weeks, body weight of mice did not show a significant increase in the HFD group compared to the ND group, while weight gain of the liver was significant. Histological assessment of the HFD group’s liver confirmed NASH symptoms.
