1Effects of Grazing Exclusion on Plant Productivity and Soil Carbon, Nitrogen Storage in Alpine Meadows in Northern Tibet, China显示文摘Grazing exclusion is widely adopted in restoring degraded alpine grasslands on the Qinghai-Tibetan Plateau. However, its effectiveness remains poorly understood. In this study, we investigated the effects of grazing exclusion on plant productivity, species diversity and soil organic carbon(SOC) and soil total nitrogen(STN) storage along a transect spanning from east to west of alpine meadows in northern Tibet, China. After six years of grazing exclusion, plant cover, aboveground biomass(AGB), belowground biomass(BGB), SOC and STN were increased, but species diversity indices declined. The enhancement of AGB and SOC caused by grazing exclusion was correlated positively with mean annual precipitation(MAP). Grazing exclusion led to remarkable biomass increase of sedge species, especially Kobresia pygmaea, whereas decrease of biomass in forbs and no obvious change in grass, leguminous and noxious species. Root biomass was concentrated in the near surface layer(10 cm) after grazing exclusion. The effects of grazing exclusion on SOC storage were confined to shallow soil layer in sites with lower MAP. It is indicated that grazing exclusion is an effective measure to increase forage production and enhance soil carbon sequestration in the studied region. The effect is more efficient in sites with higher precipitation. However, the results revealed a tradeoff between vegetation restoration and ecological biodiversity. Therefore, carbon pools recover more quickly than plant biodiversity in the alpine meadows. 2Effects of livestock exclusion and climate change on aboveground biomass accumulation in alpine pastures across the Northern Tibetan Plateau显示文摘To better understand the ecological and economic benefits of short-term grazing exclusion on the aboveground net primary productivity(ANPP)of alpine pastures,we conducted annual multi-site transect surveys in the summers from 2009 to 2011 and calculated the aboveground biomass discrepancy(ABD)between grazed and ungrazed pastures at plant community and economic group levels for three zonal alpine grassland types—meadow,steppe,and desert-steppe—across the northern Tibetan Plateau.Our results indicated that aboveground biomass(AGB)significantly differed among grassland types and declined northwesterly from 64.07 to11.44 g m-2with decreasing precipitation and increasing temperature.The mean ABD exhibited considerable community dependency,with meadow(12.47 g m-2)[steppe(6.91 g m-2)[desert steppe(2.54 g m-2),and it declined from 25.42 to 1.29 g m-2with decreasing precipitation and increasing temperature.‘Good forage’,i.e.grasses and sedges,benefited most from grazing exclusion,followed by edible forbs.With longer grazing exclusion durations(GEDs),the aboveground biomass of poisonous locoweeds initially decreased and then increased compared with the adjacent grazed sites.In the nested analysis of co-variances with a general linear model,growing season precipitation(GSP;from May to September)accounted for 52.67%ofthe observed variation in AGB,followed by AGT(9.77%)and pasture management systems(PMSs;grazing or grazing-excluded,5.31%).The variation in ABD was explained primarily by AGT(16.52%),GED(20.25%),and the interaction of AGT 9 GED(19.58%).Our results confirm that precipitation is the primary factor controlling the ANPP of alpine grasslands on the Northern Tibetan Plateau and that the ecological benefits arising from grazing exclusion are also partly dependent on grassland type and exclusion duration.Therefore,spatial and temporal variations in growing season precipitation and plant functional traits or economic group composition should be jointly considered when developing policies concerning the management and spatial layouts of grazing exclosures in this region.Jianshuang Wu Xianzhou Zhang Zhenxi Shen Peili Shi Chengqun Yu Baoxiong Chen 2014Chinese Science Bulletin2014,59,32:11
3Nine compounds from the root bark of Lycium chinense and their anti-inflammatory activities显示文摘Two new compounds, named lyciumlignan D(1) and lyciumphenyl propanoid A(2), along with seven known compounds, were isolated from the root bark of Lycium chinense. Their structures were elucidated using spectroscopic data(UV, IR, HR-ESI-MS, 1D and 2D NMR, CD), as well as by comparison with those of the literature. Compounds 3–9 were isolated from this genus for the first time. 4Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8
5Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery.Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang 2020Light(Science & Applications)2020,9,1:6

7Bioactive thionic compounds and aromatic glycosides from Ligusticum chuanxiong显示文摘Three new thionic compounds,(S)-2-(2-carboxyl-2-hydroxyethylthio)-ferulic acid(1),(E)-2-methoxy-4-(3-(methylsulfonyl)prop-1-en-1-yl)phenol(2), and thiosenkyunolide C(3), together with two new aromatic glycosides(4 and 5) were isolated from the rhizome of Ligusticum chuanxiong Hort. Two known compounds(6 and 7) were also obtained. Their structures were elucidated based on extensive spectroscopic data(UV, IR, 1D and 2D NMR, and HR-ESI-MS). Furthermore the absolute configurations were established by comparison of their calculated and experimental circular dichroism spectra and by a dimolybdenum tetraacetate [Mo_2(AcO)_4]-induced circular dichroism procedure. All compounds were evaluated against lipopolysaccharide(LPS)-induced NO production in BV2 cells, and compounds 4 and 5 showed strong inhibitory activities with IC_(50) values of 2.03 and 3.09 μmol/L, respectively(positive control curcumin, IC_(50)=6.17 μmol/L). 8Establishment and Verification of An Efficient Virus-induced Gene Silencing System in Forsythia显示文摘To understand the functional identification of large-scale genomic sequences in Forsythia,tobacco rattle virus(TRV)-mediated virus-induced gene silencing(VIGS),suitable for the plant,was explored in this study.The results showed that the TRV-mediated VIGS system could be successfully used in Forsythia for silencing the reporter gene FsPDS(Forsythia phytoene desaturase)using stem infiltration and leaf infiltrationmethods.All the treated plants were pruned below the injection site after 7–15 d infection;the FsPDS was silenced and typical photobleaching symptoms were observed in newly sprouted leaves at the whole-plant level.Meanwhile,this system has been successfully tested and verified through virus detection and qRT-PCR analysis.After the optimization,Forsythia magnesium chelatase subunit H(FsChlH)was silenced successfully in Forsythia using this system,resulting in yellow leaveswith decreased chlorophyll content.The system was stable,highly efficient and had greater rapidity and convenience,which made it suitable to study the function of genes related to physiological pathways such as growth and development,and metabolic regulation in Forsythia.Jianshuang Shen Weijia Si Yutong Wu Yang Xu Jia Wang Tangren Cheng Qixiang Zhang Huitang Pan 2021Horticultural Plant Journal2021,7,1:3
9Application of five-element music therapy in pain coping skills training in patients with knee osteoarthritis显示文摘Objective:The objective of this study is to assess the application effect of five elements music therapy introduced in the pain coping skills training of knee osteoarthritis(KOA).Materials and Methods:Totally,80 patients with KOA were selected and randomly divided into the experimental group(39 cases)and the control group(41 cases).The control group was only given routine nursing measures,and the experimental group was additionally treated with five-element music therapy on the basis of the control group,twice a day,28 days in total.The Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC)was used to evaluate the functional status of the knee joint of the two groups.The clinical efficacy of the two groups was evaluated by Guiding Principles for Clinical Research of New Chinese Medicine in the Treatment of Osteoarthritis.Results:WOMAC score statistically significantly decreased in the experimental group(35.92±9.48 vs.16.17±5.43,P<0.01)and the control group(36.73±6.42 vs.22.53±7.51,P<0.01)after 28 days of intervention when compared with that before intervention;WOMAC score in the experimental group was lower than that of the control group after 28 days of intervention(16.17±5.43 vs.22.53±7.51,P<0.01).The total effective rate of the experimental group was statistically higher than that of the control group(82.0%vs.51.2%,χ2=11.97,P=0.003).Conclusion:The combination of five-element music therapy and routine nursing measures has better effect in relieving pain and bad emotions of patients with KOA when compared with routine nursing measures alone.Suqian LI Jingjin XU Ling TANG Ye LI Huaxin WANG Lixue ZHAO Jianshuang YAO Shuying WANG Nan LI 2021Journal of Integrative Nursing2021,3,4:2

11Improved and sustainable agroecosystem, food security andenvironmental resilience through zero tillage with emphasis on soilsof temperate and subtropical climate regions: A review显示文摘Tillage is one of the agricultural management practices that significantly impacts agroecosystems,crop production,and the environment.Conventional tillage(CT)practices alter the soil environment and induce organic constituents’decomposition and the emission of greenhouse gases(GHGs),which contribute to the greenhouse effect and global warming.Low organic matter,biological diversity,aggregate stability,high erosion and degradation of the soil environment,and sequestration strength are additional factors associated with CT,which negatively affect food security and environmental sustainability.As a result,CT is no more beneficial in the long run;consequently,zero tillage(ZT)could be a viable candidate for sustainable agriculture.The review to establish this systematically compared and summarized the effect of tillage systems(i.e.CT and ZT)by synthesizing and interpreting published data(>150 peer-reviewed articles)with>200 observations on soil ecosystem services and properties/agroecosystem,crop yield/food security,GHG emission,and carbon sequestration/environmental resilience.The review established that ZT improves soil structure,aggregate stability,biological diversity,organic matter and nutrients,water and water use efficiency,and reduces soil degradation,erosion,tillage machinery impacts,and GHG emissions.It allows timely seeding and better crop growth,increases yield and food security,improves carbon sequestration,strengthens soil storage potential,and helps to mitigate the adverse effects of climate change on environmental resilience.Based on various latent direct and indirect benefits,resource-saving ability,and broad adoption scope of ZT,it is corroborated that ZT is a practical and potential approach for improved and sustainable agroecosystem,food security,and environmental resilience.Waseem Hassan Yu'e Li Tahseen Saba Fanta Jabbi Bin Wang Andong Cai Jianshuang Wu 2022International Soil and Water Conservation Research2022,10,3:1

14Human Disturbance Reduces Plant Species Diversity and Stability of Phyllostachys pubescens Forests显示文摘Plant species diversity is an important index reflecting the functional complexity and stability of ecosystems.Human activity can completely alter plant species diversity and cause serious degradation of ecosystems but its impact on bamboo forest still lacks of systematic evaluation.In this study,we performed a field investigation to reveal the influences of human disturbances on the plant diversity and stability of Moso bamboo forests at Southern China.The selected bamboo fields contained different intensities of human activities that could be classified as slight,moderate and severe disturbance level.Species richness index S,Shannon-Wienner index H,Simpson index D,Pielou index Jsw,community similarity index IS and community stability index were employed to quantitatively evaluate the plant species diversity and stability.The survey revealed that there were 203 species belonging to 83 families and 108 genera in Moso bamboo forests.The number of plant species in the Moso bamboo forests decreased with the increasing of disturbance intensity.The species diversity indexes generally followed the order of slight>moderate>severe disturbance,as well as the richness index S,Shannon-Wienner index H and Pielou index Jsw.The similarity and species stability of the bamboo forest communities also decreased with the increase of the disturbance intensity.Under the severe disturbance,plant species replacement occurred strongly.The obtained results provide some a guideline for the sustainable management of bamboo forest.Ji Lei Rong Chen Renyi Gui Jianshuang Gao 2020Research in Ecology2020,2,1:1

18Alopecuroidines A-C, three matrine-derived alkaloids from the seeds of Sophora alopecuroides显示文摘Three matrine-derived alkaloids, alopecuroidine A(1), alopecuroidine B(2 a) and alopecuroidine C(2 b)were isolated from the seeds of Sophora alopecuroides. Their structures were elucidated by extensive spectroscopic analyses and X-ray diffraction. Three compounds possess an unprecedented rearranged fused7/6/5/6 tetracyclic skeleton with a diazacycloheptane structure. Their plausible biosynthetic pathway was also proposed. The anti-proliferative activities of compounds 1 and 2 a were examined by the MTT assay.Compound 1 inhibited the viability of human lung cancer A549 cells, having a half maximal inhibitory concentration(IC50) of 7.58 ± 2.47 μmol/L at 72 h. The flow cytometric analysis suggested that 1 inhibited A549 cell growth by inducing apoptosis and cell-cycle arrest. Additionally, 1 induced the loss of mitochondrial membrane potential, elevated intracellular reactive oxygen species, increased the Bax/Bcl-2 ratio, stimulated cleaved-caspase-3 and P53 protein levels, and suppressed the pro-caspase-3 level. 19Mating behaviors in ovoviviparous black rockfish(Sebastes schlegelii):molecular function of prostaglandin E2 as both a hormone and pheromone显示文摘Prostaglandins(PGs)are profound hormones in teleost sexual behavior,especially in mating.PGs act as pheromones that affect the olfactory sensory neurons of males,inducing the initiation of a series of mating behaviors.However,the molecular mechanism by which PGs trigger mating behavior in ovoviviparous teleosts is still unclear.In the present study,we employed the ovoviviparous black rockfish(Sebastes schlegelii),an economically important marine species whose reproductive production is limited by incomplete fertilization,as a model species.The results showed that when the dose of PGE2 was higher than 10 nmol/L,a significant(P<0.05)increase in mating behaviors was observed.Dual-fluorescence in situ hybridization indicated that PGE2 could fire specific neurons in different brain regions and receptor cells in the olfactory sac.After combining with specific neurons in the central nervous system(CNS),a series of genes related to reproduction are activated.The intracerebroventricular administration of PGE_(2) significantly increased lhb levels(P<0.05)in both sexes.Moreover,steroidogenesis in gonads was also affected,inducing an increase(P<0.05)in E_(2) levels in males and T levels in females.PGE_(2) levels were also increased significantly(P<0.05)in both sexes.The present study revealed that PGE2 can activate mating behavior in black rockfish in both hormone and pheromone pathways,leading to variations in sex steroid levels and activation of reproductive behaviors.Our results provide not only novel insight into the onset of mating behaviors in ovoviviparous teleosts but also solutions for the incomplete fertilization caused by natural mating in cage aquaculture.Likang Lyu Yijia Yao Songyang Xie Xiaojie Wang Haishen Wen Yun Li Jianshuang Li Chenpeng Zuo Shaojing Yan Jingyi Dong Xin Qi 2024Marine Life Science & Technology2024,6,1:0
20A NIR ratiometric fluorescent biosensor for sensitive detection and imaging of α-L-fucosidase in living cells and HCC tumor-bearing mice显示文摘Detection and imaging of α-L-fucosidase(AFU)is of great value to understand its roles in hepatocellular carcinoma(HCC)and tumor early diagnosis,but ideal assays are still lacking.Herein,a near-infrared(NIR)fluorescent biosensor(α-Fuc-DCM)was elaborately designed and synthesized for rapid and ratiometric detection of AFU activity in cells and HCC tumor mouse models.In the presence of AFU,this biosensor shows an enhancement in NIR emission in a ratiometric manner,which significantly improves the detection accuracy with the limit of detection as low as 4.8 mU/mL.Taking advantage of these merits,the activity of AFU in lysosomes could be visualized using ratiometric and NIR dual modality in living cells.Furthermore,its remarkable application for monitoring of endogenous AFU activity in HCC tumor-bearing mouse model is also demonstrated with bright fluorescence signal,which indicated that the biosensor could clearly monitor the liver tumor in the early stage.Importantly,the α-Fuc-DCM probe can be utilized to detect the AFU in serum from HCC patients.This strategy offers a promising biosensor system for early diagnosis of HCC and studying the roles of AFU in cancers.Guo-Qiang Zhang Weiwei Feng Zhiyuan Gao Gao-Lan Zhang Xinying Wu Yating Xiao Xun Li Lei Zheng Dan Ding Jianshuang Guo Bo Situ 2023Aggregate2023,4,2:0
