| 1 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 2 | Variations in carbon isotopic composition in the subcontinental lithospheric mantle beneath the Yangtze and North China Cratons:Evidence from in-situ analysis of diamonds using SIMS显示文摘The components and evolution of subcontinental lithospheric mantle beneath the North China Craton and the Yangtze Craton is a current topic in the geological study of China and the carbon isotopic composition of diamond is one of the most direct probes into cratonic lithospheric mantle processes.In this paper,in-situ SIMS(Secondary Ion Mass Spectrometry) techniques were used to analyze the carbon isotope compositions at different internal growth zones of diamonds from Shandong and Liaoning in the North China Craton and Hunan in the Yangtze Craton.It was found that the carbon isotopic range of diamonds from the North China Craton are rather distinct from those of the Yangtze Craton;the former has a range of 6.0‰ to 2.0‰(relative to VPDB) with an average value of 3.0‰ in their core areas,which is consistent with global peridotitic diamonds;the diamonds from the Yangtze Craton,however,have a carbon isotopic range from 8.6‰ to 3.0‰ with an average value of 7.4‰ in their core areas,being more consistent with global eclogitic diamonds.The variations of carbon isotope ratios between different internal growth zones in individual diamonds were different in the three diamond localities studied.There was a clear correlation between changes in carbon isotopic composition and phases of diamond dissolution and new growth,while no correlation was observed between δ13C and internal inclusions.The variations suggest that the carbon isotopic compositions of mantle fluids were changing during the process of diamond crystallization,and that the heterogeneity of the carbon isotopic composition in mantle carbon reservoirs was a more important factor than carbon isotope fractionation in controlling the carbon isotopic compositions and their variation in diamonds.In addition,the preliminary results of in-situ nitrogen analyses demonstrated that the variation of carbon isotopic compositions between the core and outer growth zones does not correlate with nitrogen abundances,implying either that diamonds crystallized in an open environment or that the carbon isotopic composition and nitrogen contents in mantle fluids were controlled by other,not yet understood factors.The experimental results provide hints that the isotopic composition of carbon and its original sources were different in metasomatic fluids controlling diamond formation in the mantle beneath the North China Craton and the Yangtze Craton. | CHEN Hua QIU ZhiLi LU TaiJin STERN Richard STACHEL Thomas SUN Yuan ZHANG Jian KE Jie PENG ShuYi QIN SheCai | 2013 | Chinese Science Bulletin2013,58,1: | 7 |

Several molecular genetic tools have been developed in the past few decades to study this microorganism. These tools have been instrumental in understanding how M. tuberculosis became a successful pathogen. Advanced molecular genetic tools have played a significant role in exploring the complex pathways involved in M. tuberculosis pathogenesis. Here, we review various molecular genetic tools used in the study of M. tuberculosis. Further, we discuss the applications of clustered regularly interspaced short palindromic repeat interference(CRISPRi), a novel technology recently applied in M. tuberculosis research to study target gene functions. | 14 | Improved method for synthesis of low molecular weight protamine–siRNA conjugate显示文摘RNAi technology has aroused wide public interest due to its high efficiency and specificity to treat multiple types of diseases. However, the effective delivery of siRNA remains a challenge due to its large molecular weight and strong anionic charge. Considering their remarkable functions in vivo and features that are often desired in drug delivery carriers, biomimetic systems for siRNA delivery become an effective and promising strategy. Based on this, covalent attachment of synthetic cell penetrating peptides(CPP) to siRNA has become of great interest. We developed a monomeric covalent conjugate of low molecular weight protamine(LMWP, a well-established CPP) and siRNA via a cytosol-cleavable disulfide linkage using PEG as a crosslinker. Results showed that the conjugates didn't generate coagulation, and exhibited much better RNAi potency and intracellular delivery compared with the conventional charge-complexed CPP/siRNA aggregates. Three different synthetic and purification methods were compared in order to optimize synthesis efficiency and product yield. The methodology using hetero-bifunctional NHS–PEG–OPSS as a crosslinker to synthesize LMWP–siRNA simplified the synthesis and purification process and produced the highest yield. | 20 | The morphodynamic responses to deposition-promoting projects in island and reef coasts of the Zhoushan Archipelago, China显示文摘China's island and reef coasts are mainly distributed along the coasts of Zhejiang and Fujian provinces.These areas have favorable deep-water conditions and great development potential. The Zhoushan Archipelago is a typical island and reef coast. In the midst of the islands, there are many tidal channels with high water depth and high current velocity. Daishan Island is located on the northern part of the Zhoushan Archipelago. The sea north of Daishan Island can be characterized as having great water depth(generally between 10 and 20 m), high current velocity(the maximum measured value is about 2 m/s),and hyperconcentrated suspended sediment(annual average concentration under normal weather conditions is about 0.6 kg/m^3). Sediment transport is influenced by the co-action of waves and currents.Land reclamation can ease the contradiction between the desire for development and the land resource shortage on island and reef coasts. Land reclamation generally starts from deposition-promoting projects on island and reef coasts with hyperconcentration of suspended sediment with the aim of reducing the reclamation cost. Based on the morphodynamic characteristics of the sea area near north of Daishan Island, a two-dimensional mathematical model was developed to analyze the co-action of current and waves and the model was verified for spring as well as neap tides in 2007 and 2012. The simulated values of tidal stages, tidal current velocities, tidal current directions, and suspended sediment concentration at15 stations in 2007 and 9 stations in 2012 were in good agreement with the in-situ measured values.Furthermore, the simulated magnitude and pattern of erosion and sedimentation agreed well with the observations. This model was used to evaluate the effects of a proposed siltation scheme on the hydrodynamic environment and morphological change of the study area.
