1Comparison of the toxicity of silver nanoparticles and silver ions on the growth of terrestrial plant model Arabidopsis thaliana显示文摘Silver nanoparticles (AgNPs) are one of the most widely used nanomaterials, but the mechanism of AgNP toxicity in terrestrial plants is still unclear. We compared the toxic effects of AgNPs and Ag+ on Arabidopsis thaliana at the physiological, ultrastructural and molecular levels. AgNPs did not affect seed germination; however, they showed stronger inhibitory effect on root elongation than Ag+ . The results of transmission electron microscopy and metal content analysis showed that AgNPs could be accumulated in leaves. These absorbed AgNPs disrupted the thylakoid membrane structure and decreased chlorophyll content, which can inhibit plant growth. By comparison, a small amount of Ag+ was absorbed by seedlings, and it did not pronouncedly affect chloroplast structure and other metal ion absorption as AgNPs did. Compared with Ag+ , AgNPs could alter the transcription of antioxidant and aquaporin genes, indicating that AgNPs changed the balance between the oxidant and antioxidant systems, and also affected the homeostasis of water and other small molecules within the plant body. 2Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 3The High Energy X-ray telescope (HE) onboard the Insight-HXMT astronomy satellite显示文摘The Insight-Hard X-ray Modulation Telescope(Insight-HXMT) is a broadband X-ray and γ-ray(1-3000 ke V) astronomy satellite. One of its three main telescopes is the High Energy X-ray telescope(HE). The main detector plane of HE comprises 18 Na I(Tl)/Cs I(Na) phoswich detectors, where Na I(Tl) is used as the primary detector to measure ~ 20-250 ke V photons incident from the field of view(FOV) defined by collimators, and Cs I(Na) is used as the active shielding detector to Na I(Tl) by pulse shape discrimination. Additionally, Cs I(Na) is used as an omnidirectional γ-ray monitor. The HE collimators have a diverse FOV,i.e. 1.1°×5.7°(15 units), 5.7°×5.7°(2 units), and blocked(1 unit). Therefore, the combined FOV of HE is approximately5.7°×5.7°. Each HE detector has a diameter of 190 mm resulting in a total geometrical area of approximately 5100 cm2, and the energy resolution is ~15% at 60 ke V. For each recorded X-ray event by HE, the timing accuracy is less than 10 μs and the deadtime is less than 10 μs. HE is used for observing spectra and temporal variability of X-ray sources in the 20-250 ke V band either by pointing observations for known sources or scanning observations to unveil new sources. Additionally, HE is used for monitoring the γ-ray burst in 0.2-3 Me V band. 4Diclofop-methyl affects microbial rhizosphere community and induces systemic acquired resistance in rice显示文摘Diclofop-methyl(DM),a widely used herbicide in food crops,may partly contaminate the soil surface of natural ecosystems in agricultural area and exert toxic effects at low dose to nontarget plants.Even though rhizosphere microorganisms strongly interact with root cells,little is known regarding their potential modulating effect on herbicide toxicity in plants.Here we exposed rice seedlings(Xiushui 63) to 100 μg/L DM for 2 to 8 days and studied the effects of DM on rice rhizosphere microorganisms,rice systemic acquired resistance(SAR) and rice-microorganisms interactions.The results of metagenomic 16 S rDNA Illumina tags show that DM increases bacterial biomass and affects their community structure in the rice rhizosphere.After DM treatment,the relative abundance of the bacterium genera Massilia and Anderseniella increased the most relative to the control.In parallel,malate and oxalate exudation by rice roots increased,potentially acting as a carbon source for several rhizosphere bacteria.Transcriptomic analyses suggest that DM induced SAR in rice seedlings through the salicylic acid(but not the jasmonic acid) signal pathway.This response to DM stress conferred resistance to infection by a pathogenic bacterium,but was not influenced by the presence of bacteria in the rhizosphere since SAR transcripts did not change significantly in xenic and axenic plant roots exposed to DM.The present study provides new insights on the response of rice and its associated microorganisms to DM stress.Si Chen Xingxing Li Michel Lavoie Yujian Jin Jiahui Xu Zhengwei Fu Haifeng Qian 2017Journal of Environmental Sciences2017,29,1:8
5Short-term propamocarb exposure induces hepatic metabolism disorder associated with gut microbiota dysbiosis in adult male zebrafish显示文摘Propamocarb (PM) is a pesticide that is widely used to protect cucumbers and other plants from downy mildew.Recently,some studies indicated that PM exposure had potential toxic effects in animals.In this study,adult male zebrafish were exposed to 100 and 1000 μg/l PM for 7 days to assess its effects on metabolism and the gut microbiota.We observed a significant decrease in triglyceride (TG) in the livers of zebrafish that were exposed to 1000 μg/l PM for 7 days.At the same time,some genes related to glycolysis and lipid metabolism in the livers of zebrafish,including hexokinase-1 (HK1),pyruvate kinase (PK),acyi-CoA oxidase (Aco),peroxisome proliferator activated receptor alpha (Ppar-α),apolipoprotein A-Ⅳ-like (Apo),Acetyl CoA carboxylase-1 (Acc1),diacylglycerol acyltransferase (Dgat),and fatty acid synthase (Fas),were also decreased significantly after PM exposure.Based on GC-MS metabolomics analysis,a total of 48 metabolites changed significantly in the 1000 μg/l PM treatment group in comparison with the control group.These altered metabolites were mainly associated with the glycolysis,amino acid metabolism,and lipid metabolism pathways.Interestingly,we further found that the 1000 μg/l PM treatment group also showed significant elevations in Proteobacteria,Bacteroidetes,and Firmicutes at the phylum level.Sequencing of the 16S rRNA gene in the V3-V4 region also showed a significant change in the abundance and diversity of the gut microbiota in the 1000 μg/l PM treatment group.Our results indicated that exposure to PM for a short time could induce hepatic metabolic disorders and gut microbiota dysbiosis in adult male zebrafish.Rui Zhang Zihong Pan Xiaoyu Wang Manlu Shen Jiajie Zhou Zhengwei Fu Yuanxiang Jin 2019Acta Biochimica et Biophysica Sinica2019,51,1:6
6β-Cypermethrin promotes the adipogenesis of 3T3-L1 cells via inducing autophagy and shaping an adipogenesis-friendly microenvironment显示文摘The toxicity of synthetic pyrethroids has garnered attention, and studies have revealed that pyrethroids promote fat accumulation and lead to obesity in mice. Nevertheless, the effect of β-cypermethrin (β-CYP) on adipogenesis and its underlying mechanism remains largely unknown. In this study, mouse embryo fibroblasts 3T3-L1 cells were exposed to β-CYP, and the cell viability, intracellular reactive oxygen species (ROS) level, autophagy, and adipogenesis were assessed to investigate the roles of oxidative stress and autophagy in the toxic effects of β-CYP on adipogenesis. The results demonstrated that treatment with 100 μΜ β-CYP elevated the ROS level, decreased mitochondrion membrane potential, stimulated autophagy, and enhanced the adipogenesis induced by the mixture of insulin, dexamethasone, and 3-isobutyl-1-methylxanthine. However, co-treatment with N-acetyl-L-cysteine partially blocked the abovementioned effects of β-CYP in 3T3-L1 cells. In addition, co-treatment with rapamycin, an autophagy agonist, enhanced the inductive effect of β-CYP on adipogenesis, whereas co-treatment with 3-methyladenine blocked the enhancement of adipogenesis caused by β-CYP. Moreover, β-CYP also altered the microenvironment of 3T3-L1 cells to an adipogenesis-friendly one by reducing the extracellular expression of miR-34a, suggesting that the culture media of β-CYP-treated 3T3-L1 cells could shift macrophages to M2 type. Taken together, the data obtained in the present study demonstrated that β-CYP promoted adipogenesis via oxidative stress-mediated autophagy disturbance, and it caused macrophage M2 polarization via the alteration of miR-34a level in the microenvironment. 8Effect of chronic corticosterone-induced depression on circadian rhythms and age-related phenotypes in mice显示文摘Disrupted circadian rhythms are a recognized effect of depression,and our previous article demonstrated an association between depression and premature aging,but the underlying mechanisms are not well understood.In the present study,we used a mouse model of chronic corticosterone (CORT)-treated depression to elucidate a mechanism by which depression may be associated with the circadian clock and mediate age-related phenotypes.Mice received a daily injection of 20 mg/kg CORT for 21 consecutive days,and the depression-like behaviors of mice were identified by the sucrose intake test,tail suspension test and open field test.Our findings indicated that CORT injection may be correlatedwith the circadian clock by impairing circadian rhythms or shifting the phase values of clock genes.We also showed that CORT-treated mice exhibited a significant gradual reduction in body weight gain with increased oxidative stress, including reduced activity of antioxidant-related enzymes,reduced glutathione:glutathione disulfide ratio and cytochrome (Cyt)-C level,and elevated reactive oxygen species content.Moreover, chronic CORT injection affected inflammatory responses,the production of mitochondrial ATP and telomere shortening,which may be associated with the Sirtuin 3 (SIRT3)signaling pathway. 12Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 14Evidence for a Dirac nodal-line semimetal in SrAs_3显示文摘Dirac nodal-line semimetals with the linear bands crossing along a line or loop, represent a new topological state of matter. Here, by carrying out magnetotransport measurements and performing first-principle calculations, we demonstrate that such a state has been realized in high-quality single crystals of SrAs_3.We obtain the nontrivial ∏ Berry phase by analysing the Shubnikov-de Haas quantum oscillations. 15Exposure to dibutyl phthalate impairs lipid metabolism and causes inflammation via disturbing microbiota-related gut-liver axis显示文摘Dibutyl phthalate(DBP),a kind of typical environm ental pollutant,is widely used as plasticizers,and its neurotoxicity and developm ental toxicity have been found in recent years.However,whether oral DBP exposure w ill affect the homeostasis of gut m icrobiota and its adverse response in liver of m am m alians remain unclear.In the present study,10-week experimental cycles of vehicle or DBP(0.1 and 1mg/kg)were given to 6-week-old C57BL/6J mice by oral gavage.Our results revealed that the body weight of mice was increased after exposure to both low and high doses of DBP.The serum levels of hepatic triglyceride and total cholesterol were significantly increased in response to both doses of DBP.In addition,some pivotal genes related to lipogenesis were also increased in liver at the mRNA level.Evaluation of gut m icrobiota by 16S rRNA sequencing technology showed that 0.1 mg/kg DBP exposure significantly affected gut microbiota at the phylum and genus levels.Moreover,DBP exposure decreased mucus secretion and caused inflam m ation in the gut,leading to the im pairm ent of intestinal barrier function.Exposure to OBP inhibited the expression of peroxisome proliferator-activated receptor-y and activated the expression of nuclear factor kappa B.In addition,DBP exposure increased the level of lipopolysaccharide in serum,and increased the expression of toll-like receptor 4 and the levels of inflam m atory cytokines,such as interleukin(IL)-1β,IL-6,and tumor necrosis factor alpha,in the liver.These results indicated that exposure to DBP disturbed the homeostasis of gut m icrobiota,induced hepatic lipid m etabolism disorder,and caused liver inflam mation in mice via the related gut-liver axis signaling pathways.Ze Xiong Yuyao Zeng Jiafeng Zhou Ruonan Shu Xiaoxian Xie Zhengwei Fu 2020Acta Biochimica et Biophysica Sinica2020,52,12:2
16Enterorenal crosstalks in diabetic nephropathy and novel therapeutics targeting the gut microbiota显示文摘The role of gut-kidney crosstalk in the progression of diabetic nephropathy(DN)is receiving increasing concern.On one hand,the decline in renal function increases circulating uremic toxins and affects the composition and function of gut microbiota.On the other hand,intestinal dysbiosis destroys the epithelial barrier,leading to increased exposure to endotoxins,thereby exacerbating kidney damage by inducing systemic inflammation.Dietary inventions,such as higher fiber intake,prebiotics,probiotics,postbiotics,fecal microbial transplantation(FMT),and engineering bacteria and phages,are potential microbiota-based therapies for DN.Furthermore,novel diabetic agents,such as glucagon-like peptide-1(GLP-1)receptor agonists,dipeptidyl peptidase-4(DPP-4)inhibitors,and sodium-dependent glucose transporter-2(SGLT-2)inhibitors,may affect the progression of DN partly through gut microbiota.In the current review,we mainly summarize the evidence concerning the gut-kidney axis in the advancement of DN and discuss therapies targeting the gut microbiota,expecting to provide new insight into the clinical treatment of DN.Yinhua Ni Liujie Zheng Sujie Nan Lehui Ke Zhengwei Fu Juan Jin 2022Acta Biochimica et Biophysica Sinica2022,54,10:2

18The metallogenic environment of the Dounan manganese deposit,Southeast Yunnan, China: evidence from geochemistry and Mössbauer spectroscopic显示文摘The Dounan manganese deposit is a typical large-scale marine sedimentary manganese deposit of the Middle Triassic in China. The metallogenic environment and change process directly dictate the migration, enrichment, and precipitation of Mn. To better understand its metallogenetic environment, a detailed study was undertaken involving field observation, mineralogical and geochemical and M?ssbauer spectroscopic analyses. The major findings are as follows:(1) Lithofacies paleogeography, sedimentary structural characteristics, and geochemical indexes indicate that the deposits were formed in an epicontinental marine sedimentary basin environment of normal salinity;(2) there were three ore phases including Mn oxides, Mn carbonates, and mixed Mn ores. The ore minerals found were braunite, manganite, Ca-rhodochrosite, manganocalcite, and kutnahorite. Petrographic and mineralogical information indicates that the metallogenic environment was a weakly alkaline and weakly oxidized to weakly reduced environment, and the mineralization occurred near the redox interface;(3) the V/(V + Ni)ratios, δCe and Fe^(2+)/Fe^(3+) found in profiles of Baigu and Gake ore sections show that the redox conditions of the ore-forming environment were continuously changing; and(4) three Fe species, α-Fe_2O_3, para-Fe^(3+), and para-Fe^(2+),were found in hematite and clay mineral samples using M?ssbauer spectrum analysis. The presence and distribution of these Fe species indicate that the deposit was formed in a typical sedimentary environment during the mineralization process. In summary, our study showed that redox was a key factor controlling the mineralization of the Dounan manganese deposit. Our results have led us to the conclusion that transgression and regression caused fluctuations in sea level, which in turn caused the change of the redox environment. 19Investigation of High Catalyteal Durability for Porous Pt Films Deposited via Magnetron Sputtering显示文摘Porous Pt thin films were prepared on carbon papers by a single-step ultra-high dc magnetron sputtering method to obtain ideal electrodes for proton exchange membrane fuel cells.The platinum loading of the electrocatalyst layer is controlled at about 0.1 mg·cm^(-2).Structural characteristics and catalytic activities of the films were analyzed by scanning electron microscopy,atomic force microscopy,X-ray diffraction,cyclic voltammetry,and stress durability testing methods.The effect of treatment conditions of a substrate on the structural and performance characteristics of the catalytic films was shown as well.Films produced on acid-treated carbon papers at the argon pressure of 0.01 mbar possessed a homogeneous,highly developed surface along with a porous structure.Compared to Pt/TCPW(Toray carbon papers soaked in ultrapure water)electrodes,the film obtained on the acid-treated substrate had a larger electrochemical surface area(163.33 m^(2)·g^(-1))and exhibited better catalytic stability and durability due to a porous structure as a result of Pt particle accumulation.刘广权 PENG Liping FAN Long WANG Jin FU Yajun XIONG Zhengwei REN Xuetan 曹林洪 吴卫东 2022Journal of Wuhan University of Technology(Materials Science)2022,37,2:1
