Advances on strategies for searching for next generation thermal barrier coating materials显示文摘Thermal barrier coating(TBC) materials play important roles in gas turbine engines to protect the Nibased super-alloys from the high temperature airflow damage. High melting point, ultra-low thermal conductivity, large thermal expansion coefficient, excellent damage tolerance and moderate mechanical properties are the main requirements of promising TBC materials. In order to improve the efficiency of jet and/or gas turbine engines, which is the key of improved thrust-to-weight ratios and the energysaving, significant efforts have been made on searching for enhanced TBC materials. Theoretically, density functional theory has been successfully used in scanning the structure and properties of materials, and at the same time predicting the mechanical and thermal properties of promising TBC materials for high and ultrahigh temperature applications, which are validated by subsequent experiments. Experimentally, doping and/or alloying are also widely applied to further decrease their thermal conductivities. Now, the strategy through combining theoretical calculations and experiments on searching for next generation thermal insulator materials is widely adopted. In this review, the common used techniques and the recent advantages on searching for promising TBC materials in both theory and experiments are summarized. | Bin Liu Yuchen Liu Changhua Zhu Huimin Xiang Hongfei Chen Luchao Sun Yanfeng Gao Yanchun Zhou | 2019 | Journal of Materials Science & Technology2019,35,5: | 24
Role of pyroptosis in inflammation and cancer显示文摘Pyroptosis is a form of programmed cell death mediated by gasdermin and is a product of continuous cell expansion until the cytomembrane ruptures, resulting in the release of cellular contents that can activate strong inflammatory and immune responses.Pyroptosis,an innate immune response, can be triggered by the activation of inflammasomes by various influencing factors.Activation of these inflammasomes can induce the maturation of caspase-1 or caspase-4/5/11,both of which cleave gasdermin D to release its N-terminal domain, which can bind membrane lipids and perforate the cell membrane. Here,we review the latest advancements in research on the mechanisms of pyroptosis, newly discovered influencing factors, antitumoral properties, and applications in various diseases. Moreover, this review also provides updates on potential targeted therapies for inflammation and cancers, methods for clinical prevention, and finally challenges and future directions in the field. | Xiang Wei Feng Xie Xiaoxue Zhou Yuchen Wu Haiyan Yan Ting Liu Jun Huang Fangwei Wang Fangfang Zhou Long Zhang | 2022 | Cellular & Molecular Immunology2022,19,9: | 11
Global Dynamic Molecular Profiling of Stomatal Lineage Cell Development by Single-Cell RNA Sequencing显示文摘The regulation of stomatal lineage cell development has been extensively investigated. However,a comprehensive characterization of this biological process based on single-cell transcriptome analysis has not yet been reported. In this study,we performed RNA sequencing on 12844 individual cells from the cotyledons of 5-day-old Arabidopsis seedlings. We identified 11 cell clusters corresponding mostly to cells at specific stomatal developmental stages using a series of marker genes.Comparative analysis of genes with the highest variable expression among these cell clusters revealed transcriptional networks that regulate development from meristemoid mother cells to guard mother cells.Examination of the developmental dynamics of marker genes via pseudo-time analysis revealed potential interactions between these genes.Collectively, our study opens the door for understanding how the identified novel marker genes participate in the regulation of stomatal lineage cell development. | Zhixin Liu Yaping Zhou Jinggong Guo Jiaoai Li Zixia Tian Zhinan Zhu Jiajing Wang Rui Wu Bo Zhang Yongjian Hu Yijing Sun Yan Shangguan Weiqiang Li Tao Li Yunhe Hu Chenxi Guo Jean-David Rochaix Yuchen Miao Xuwu Sun | 2020 | Molecular Plant2020,13,8: | 11
Combination of anti-PD-1 antibody with P-GEMOX as a potentially effective immunochemotherapy for advanced natural killer/T cell lymphoma显示文摘Advanced natural killer/T cell lymphoma(NKTL)has demonstrated poor prognosis with currently available therapies. Here,we report the efficacy of anti-programmed death 1(PD-1)antibody with the P-GEMOX(pegaspargase,gemcitabine,and oxaliplatin)regimen in advanced NKTL.Nine patients underwent six 21-day cycles of anti-PD-1 antibody(day 1),pegaspargase 2000 U/m^(2)(day 1),gemcitabine 1 g/m^(2)(days 1 and 8)and oxaliplatin 130 mg/m^(2)(day 1),followed by anti-PD-1 antibody maintenance every 3 weeks.Programmed death-ligand 1(PD-L1)expression and genetic alterations were determined in paraffin-embedded pretreatment tissue samples using immunohistochemistry and next-generation sequencing(NGS)analysis.Responses were assessed using 18F-fluorodeoxyglucose positron emission tomography(18FDG-PET)and computed tomography or magnetic resonance imaging.Eight patients exhibited significant responses, comprising of seven complete remissions and one partial remission(overall response rate:88.9%).After a median follow-up of 10.6 months,6/9 patients(66.7%)remained in complete remission. The most common grade 3/4 adverse events were anemia(33.3%),neutropenia(33.3%),and thrombocytopenia(33.3%);all of which were manageable and resolved.Immunochemotherapy produced a high response rate in patients with positive PD-L1 expression(5/6,83.3%).NGS analysis suggested that STAT3/JAK3/PD-L1 alterations and ARID1A mutation were associated with immunochemotherapy efficacy.Mutation in DDX3X and alteration in epigenetic modifiers of KMT2D,TET2,and BCORL1 might indicate a poor response to immunochemotherapy. In conclusion, the anti-PD-1 antibody plus P-GEMOX regimen demonstrated promising efficacy in advanced NKTL.PD-L1 expression combined with specific genetic alterations could be used as potential biomarkers to predict therapeutic responses to immunochemotherapy. | Jun Cai Panpan Liu Huiqiang Huang Yajun Li Shuyun Ma Hui Zhou Xiaopeng Tian Yuchen Zhang Yan Gao Yi Xia Xuanye Zhang Hang Yang Lirong Li Qingqing Cai | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 6
PD-L1 expression and its effect on clinical outcomes of EGFRmutant NSCLC patients treated with EGFR-TKIs显示文摘Objective:Epidermal growth factor receptor(EGFR)activation was reported to upregulate programmed death-ligand 1(PD-L1)expression in lung cancer cells and subsequently contribute to immune escape, indicating its critical role in EGFR-driven lung tumors. This study characterized PD-L1 expression in patients with surgically resected EGFR-mutant non-small cell lung cancer(NSCLC).The effect of PD-L1 expression on clinical outcomes was also investigated in advanced EGFR-mutant NSCLC treated with EGFR-tyrosine kinase inhibitors(TKIs).Methods:In total,73 patients with surgically resected NSCLC and EGFR mutations were identified.PD-L1 expression and CD8+tumor-infiltrating lymphocyte(TIL)density were assessed by immunohistochemistry.A literature review of publications that assessed the predictive and prognostic value of PD-L1 expression in advanced EGFR-mutant NSCLC patients treated with EGFR-TKIs was performed.Results:Nineteen(26.0%)patients were positive for PD-L1 expression, which was significantly associated with concomitant KRAS mutation(P=0.020)and marginally associated with higher CD8+TILs density(P=0.056).Positive PD-L1 expression was associated with markedly inferior overall survival(OS)in multivariate analysis(P=0.032).The combination of PD-L1 and CD8+TILs expression could be used to stratify the population into three groups with distinct prognoses.A meta-analysis of six publications showed that positive PD-L1 expression was not associated with OS[hazard ratio(HR)=0.90;95%confidence interval(CI),0.42–1.38]or progression-free survival(HR=1.03;95 CI,0.73–1.33)in advanced EGFR-mutant NSCLC patients receiving EGFR-TKIs.Conclusions:PD-L1 expression tended to correlate with CD8+TIL expression, concomitant KRAS mutation, and poor survival in surgically resected EGFR-mutant NSCLC.PD-L1 expression was neither the predictive nor the prognostic factor in advanced EGFR-mutant NSCLC patients treated with EGFR-TKIs. | Yuchen Bai Xiaoxia Chen Likun Hou Jun Qian Tao Jiang Caicun Zhou Maciej Ciebiada | 2018 | Cancer Biology & Medicine2018,15,4: | 5
Spatiotemporal variations of nitrate sources and dynamics in a typical agricultural riverine system under monsoon climate显示文摘Nitrogen pollution is a serious environmental issue in the Danjiangkou Reservoir region(DRR),the water source of the South-to-North Water Diversion Project of China. In this research, seasonal surveys and a bi-weekly time series survey were conducted in the Qihe River Basin, one of the most densely populated agricultural basins in the DRR.Hydrochemical compositions(NO3^-Cl^-),dual isotopes(δD-H2O,δ^18O-H2O,δ^15N-NO3^-,andδ^18O-NO3^-),and a Markov Chain Monte Carlo isotope mixing model were jointly applied to unravel the sources, migrations, and transformations of the nitrate(NO3^-)in the basin. It was revealed that the mixing between different sources was the main process controlling the isotopic compositions of the riverine NO3the upper-middle reaches. In contrast, denitrification occurred in the lower reaches. For the first time, the sources of NO3^-quantified at a basin scale in the DRR.Overall, the river transported 484.2 tons/year of NO3^-N to the reservoir,of which 32.6%,36.4%,28.0%,and 3.0%was from soil organic nitrogen, chemical fertilizer, residential sewage and atmospheric precipitation, respectively. The NO3^-N fluxes of the different sources were regulated by the monsoon climate and anthropogenic activities. For example, high precipitation and intense fertilization resulted in severe nonpoint source pollution.Denitrification thrived in soils and reservoirs in wet seasons.Temperature could regulate the migration, nitrification and denitrification processes.Based on the results,we suggest that the management strategies dealing with nitrogen pollution issue in the DRR should follow the specific spatiotemporal characteristics of NO3migration and transformation mechanisms. | Hao Jiang Wenjing Liu Jiangyi Zhang Li Zhou Xiaode Zhou Ke Pan Tong Zhao Yuchen Wang Zhifang Xu | 2020 | Journal of Environmental Sciences2020,32,7: | 5
Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers. However, its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity. It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment. Here, through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically, metformin promotes DOCK1 phosphorylation, which activates RAC1 to facilitate cell survival, leading to metformin resistance. The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids, and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably, metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression. This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients. | Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang | 2022 | Protein & Cell2022,13,11: | 5
Direct observation of epitaxial alignment of Au on M0S2 at atomic resolution显示文摘The morphology and structural stability of metal/2D semic on ductor interfaces strongly affect the performa nee of 2D electronic devices and synergistic catalysis. However, the structural evolution at the interfaces has not been well explored particularly at atomic resolution. In this work, we study the structural evoluti on of Au nan oparticles (NPs) on few-layer MoS2 by high resol utio n transmissi on electro n microscopy (HRTEM) an d quan titative high-angle annular dark field seanning TEM. It is found that in the transition of Au from nan oparticles to den drites, a dynamically epitaxial align ment betwee n Au and MoS2 lattices is formed, and Moirc patter ns can be directly observed in HRTEM images due to the mismatch between Au and M0S2 lattices. This epitaxial alignment can occur in ambient conditions, and can also be accelerated by the irradiation of high-energy electron beam. In situ observation clearly reveals the rotation of Au NPs, the atom migration inside Au NPs, and the transfer of Au atoms between neighboring Au NPs, finally leading to the formation of epitaxially aligned Au dendrites on M0S2. The structural evoluti on of metal/2D semico nductor in terfaces at atomic scale can provide valuable information for the design and fabricatio n of the metal/2D semicon ductorn ano-devices with desired physical and chemical performa nces. | Yinghui Sun Haofei Zhao Dan Zhou Yuchen Zhu Huanyu Ye Yan Aung Moe | 2019 | Nano Research2019,12,4: | 4
Application of high-throughput first-principles calculations in ceramic innovation显示文摘Recent technical progress in the industry has led to an urgent requirement on new materials with enhanced multi-properties.To meet this multi-property requirement, the materials consisting of three and more elements have attracted increasing attention. However, facing to the nearly unknown huge multi-component materials system, the traditional trial and error method cannot provide sufficient data efficiently. Therefore,an efficient material innovation strategy is significant. The first-principles calculation based on the density functional theory is a powerful tool for both the accurate prediction of material properties and the identification of its underlying thermodynamics and dynamics. At the same time, the advances of computational methods and computer calculation abilities that are orders of magnitude faster than before make the high throughput first-principles calculations popular. At present, the simulation-assisted material design has become a main branch in the material research field and a great many successes have been made.In this article, the advances of the high throughput first-principles calculations are reviewed to show the achievements of the first-principles calculations and guide the future directions of its applications in ceramics. | Bin Liu Juanli Zhao Yuchen Liu Jianqi Xi Qian Li Huimin Xiang Yanchun Zhou | 2021 | Journal of Materials Science & Technology2021,,29: | 4
A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults. Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors, built and examined the operating characteristics of a point-based algorithm for HCC risk prediction. Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells, concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. At 4.5 points, only 20.86% of participants in training cohort and 23.73% in validation cohort were recognized at high risk, with positive prediction value of 22.85% and 12.35%, respectively. Conclusions: Male-ABCD algorithm identified individual's risk for HCC occurrence within short term for their HCC precision surveillance. | Yuting Wang Minjie Wang He Li Kun Chen Hongmei Zeng Xinyu Bi Zheng Zhu Yuchen Jiao Yong Wang Jian Zhu Hui Zhao Xiang Liu Chunyun Dai Chunsun Fan Can Zhao Deyin Guo Hong Zhao Jianguo Zhou Dongmei Wang Zhiyuan Wu Xinming Zhao Wei Cui Xuehong Zhang Jianqiang Cai Wanqing Chen Chunfeng Qu | 2021 | Chinese Journal of Cancer Research2021,33,3: | 3
An adaptive sequential experiment design method for model validation显示文摘Efficient experiment design is of great significance for the validation of simulation model with high nonlinearity and large input space.Excessive validation experiment raises the cost while insufficient test increases the risks of accepting an invalid model. In this paper,an adaptive sequential experiment design method combining global exploration criterion and local exploitation criterion is proposed. The exploration criterion utilizes discrepancy metric to improve the space-filling property of the design points while the exploitation criterion employs the leave one out error to discover informative points. To avoid the clustering of samples in the local region,an adaptive weight updating approach is provided to maintain the balance between exploration and exploitation.Besides, the credibility distribution function characterizing the relationship between the input and result credibility is introduced to support the model validation experiment design. Finally, six benchmark problems and an engineering case are applied to examine the performance of the proposed method. The experiments indicate that the proposed method achieves satisfactory performance for function approximation in accuracy and convergence. | Ke FANG Yuchen ZHOU Ping MA | 2020 | Chinese Journal of Aeronautics2020,33,6: | 2
On the Negative Effects of Trend Noise and Its Applications in Side-Channel Cryptanalysis显示文摘Most existing noise reduction methods in side-channel cryptanalysis treat all noises as a whole, instead of identifying and dealing with each of them individually. Motivated by this, this paper investigates the feasibility and implications of identifying trend noise from any other noises in side-channel acquisitions and then dealing with it accordingly. We discuss the effectiveness of applying least square method to remove inherent trend noise in side-channel leakages, and also clarify the limited capability of existing noise reduction methods in dealing with trend noise. | 14 | Ultrasmall Ga-ICG nanoparticles based gallium ion/photodynamic synergistic therapy to eradicate biofilms and against drug-resistant bacterial liver abscess显示文摘Pyogenic liver abscess and keratitis are aggressive bacterial infections and the treatment has failed to eradicate bacteria in infectious sites completely owing to the currently severe drug resistance to existing antibiotics. Here,we report a simple and efficient one-step development of ultrasmall non-antibiotic nanoparticles(ICG-Ga NPs)containing clinically approved gallium(Ⅲ)(Ga^(3+))and liver targeting indocyanine green(ICG)molecules to eradicate multi-drug resistant(MDR)bacteria thought the synergetic effect of photodynamic therapy and iron metabolism blocking. The ICG-Ga NPs induced photodynamic effect could destroy the bacterial membrane, further boost the endocytosis of Ga^(3+),then replace iron in bacteria cells to disrupt bacterial iron metabolism, and demonstrate the synergetic bacterial killing and biofilm disrupting effects. The ICG-Ga NPs show an excellent therapeutic effect against extended spectrumβ-lactamases Escherichia coli(ESBL E.coli)and significantly improve treatment outcomes in infected liver abscess and keratitis.Meanwhile, the ultrasmall size of ICG-Ga NPs could be cleared rapid via renal clearance route, guaranteeing the biocompatibility. The protective effect and good biocompatibility of ICG-Ga NPs will facilitate clinical treatment of bacteria infected diseases and enable the development of next-generation non-antibiotic antibacterial agents. | Tingting Xie Yuchen Qi Yangyang Li Feilu Zhang Wanlin Li Danni Zhong Zhe Tang Min Zhou | 2021 | Bioactive Materials2021,6,11: | 2
Discovery of ABO_(4)scheelites with the extra low thermal conductivity through high-throughput calculations显示文摘Searching for new materials with extra low thermal conductivities is significant in numerous fields like thermal barrier coatings and thermoelectric devices.Traditional multiple-component design has successfully reduced the thermal conductivity, but it also dramatically increases the complexity of manufactural technologies and the risk of material failures. In this work,a specific category known as ABO_(4) scheelites that with both simple crystal structure and the structural signature of the low lattice thermal conductivity is explored.High-throughput calculations are employed to screen for the materials with the targeted performance by multi-dimensional mechanical/thermal property criteria and a database of 46 stable scheelites is constructed.Seven scheelites with both ultra-low thermal conductivities(<1.2 W/(m∙K))and quasi-ductility are predicted to be novel thermal insulation materials.Low thermal conductivities prefer the scheelites with large valence disparity between“A”and“B”cations and/or small ionic radius ratio. The adopted strategy starting from the structural fingerprint and the data-driven material selection is expected to be a reference of future structural and functional ceramics design. | Yuchen Liu Dechang Jia Yu Zhou Yanchun Zhou Juanli Zhao Qian Li Bin Liu | 2020 | Journal of Materiomics2020,6,4: | 2
Review of flash sintering with strong electric field显示文摘Flash sintering is a novel field‐assisted sintering technology for ceramics that allows a dramatic reduction in processing time and temperature.Since 2010,when flash sintering was first reported,it has been a focus of research interest in the field of materials science.Recent study results have confirmed that the sintering temperature decreases with the strength of the applied electric field;for some ceramics, the sintering temperature can be lowered even to room temperature. This represents an innovative breakthrough for ceramic sintering at ultra‐low temperatures. However, once the electric field strength in flash sintering is increased, new questions and challenges arise, such as whether or not partial discharge occurs in the green body if under electro‐thermal coupling stress, how to quantitatively analyse the impact of discharge on sintering, and, moreover, whether the sintering mechanism under a strong electric field is consistent with that under a weak electric field. These research questions require knowledge of partial discharge detection, dielectric theory, and other topics that are beyond the scope of materials science. To address this need, this review summarises the work carried out in flash sintering with a strong electric field from the perspective of high voltage and insulation technology.First, the flash sintering process and mechanism are briefly introduced. Then, the published literature on flash sintering with a strong electric field for various ceramic materials is summarised in depth. In addition, experimental phenomena that are observed in flash sintering with a strong electric field, such as flash and blackening, are discussed. Finally, some suggestions for future work are presented. It is anticipated that the knowledge gap between different areas of study can be filled by this review. | Hongyang Zhou Xiang Li Yuchen Zhu Jieming Liu Angxuan Wu Guoming Ma Xilin Wang Zhidong Jia Liming Wang | 2022 | High Voltage2022,7,1: | 2
