1Multigene editing via CRISPR/Cas9 guided by a single-sgRNA seed in Arabidopsis显示文摘We report that a solo single-guide RNA(sg RNA) seed is capable of guiding Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPRàassociated 9(CRISRP/Cas_9) to simultaneously edit multiple genes At RPL_(10)A, At RPL_(10)B and At RPL_(10)C in Arabidopsis. Our results also demonstrate that it is possible to use CRISPR/Cas_9 technology to create At RPL_(10) triple mutants which otherwise cannot be generated by conventional genetic crossing. Compared to other conventional multiplex CRISPR/Cas systems, a single sg RNA seed has the advantage of reducing off-target gene-editing. 2Review on flexible photonics/electronics integrated devices and fabrication strategy显示文摘In recent years, to meet the greater demand for next generation electronic devices that are transplantable, lightweight and portable, flexible and large-scale integrated electronics attract much more attention have been of interest in both industry and academia. Organic electronics and stretchable inorganic electronics are the two major branches of flexible electronics. With the semiconductive and flexible properties of the organic semiconductor materials, flexible organic electronics have become a mainstay of our technology.Compared to organic electronics, stretchable and flexible inorganic electronics are fabricated via mechanical design with inorganic electronic components on flexible substrates, which have stretchability and flexibility to enable very large deformations without degradation of performance. This review summarizes the recent progress on fabrication strategies, such as hydrodynamic organic nanowire printing and inkjet-assisted nanotransfer printing of flexible organic electronics, and screen printing, soft lithography and transfer printing of flexible inorganic electronics. 3Serum ficolin-2 concentrations are significantly changed in patients with hepatitis B virus infection and liver diseases显示文摘Human ficolin-2 is an important lectin complement pathway activator that is secreted from liver cells and has been implicated as an anti-infection innate immune molecule. However, the role of ficolin-2 protein and its dynamic changes over the course of and in the prognosis of chronic hepatitis B(CHB) and hepatocellular carcinoma(HCC) remain unclear. In this study, we analyzed ficolin-2 protein expression in a cohort of individuals with CHB infection, HCC and cirrhosis. A sandwich enzyme-linked immunosorbent assay(ELISA) method was used to measure serum ficolin-2 concentrations. Ficolin-2 expression in liver tissues was detected by immunohistochemical staining. Serum ficolin-2 concentrations in CHB patients were significantly higher than in healthy controls and HBV carriers. After 48 weeks of routine amelioration liver function treatment, serum ficolin-2 concentrations decreased and were positively correlated with favorable alanine aminotransferase(ALT), HBV DNA and HBe Ag-seroconversion outcomes. Interestingly, we observed much lower expression of serum and intrahepatic ficolin-2 in HCC and cirrhosis compared with healthy controls. 4Ectopic expression of antifreeze protein gene from Ammopiptanthus nanus confers chilling tolerance in maize显示文摘Improved chilling tolerance is important for maize production. Previous efforts in transgenics and marker-assisted breeding have not achieved practical results. In this study, the antifreeze protein(AnAFP) from the super-xerophyte Ammopiptanthus nanus was aligned to KnS-type dehydrins.Phosphorylation in vitro and subcellular localization showed that AnAFP was phosphorylated by maize casein kinase II and translocated from nucleus to cytoplasm under chilling stress. AnAFP also increased lactate dehydrogenase activity. A parent line of commercial maize hybrids was transformed with the AnAFP gene. Based on thermal asymmetric interlaced PCR, one hemizygous and two homozygous integration sites were identified in one T_(1) line. Ectopic expression of AnAFP in transgenic lines was confirmed by quantitative real-time PCR, RNA-seq, and Western blotting. After chilling treatment, the transgenic lines showed significantly improved phenotype, higher kernel production, survival rate and biomass, and lower relative electrolyte leakage and superoxide dismutation than the untransformed line. 18Assessing the U^(K′)_(37)-sea surface temperature relationship in shallow marine waters显示文摘To investigate the relationship between the alkenone unsaturation index(U^(K′)_(37)) and sea surface temperature(SST) in coastal and continental shelf waters, 58 surface sediment samples were collected from the South China Sea(SCS), Taiwan Strait,and East China Sea(ECS). We combined the new results with the previously published 71 data points from the SCS, the shallow water areas of the Yellow Sea(YS) and northern ECS, to form a dataset with sample sites spanning across 6°N and 37°N(including annual SST calibration between 14.3℃ and 28.6℃). With this dataset, we examined the U^(K′)_(37)-SST relationship based on 129 samples from the Western North Pacific(WNP) margin as well as using 85 samples from specific WNP shallow water.The U^(K′)_(37)index from the low-mid latitudinal WNP margin demonstrated a good correlation with the surface annual mean SST(0–50 m water depth;R^(2)=0.89). The slope of linear regression(U^(K′)_(37)-SST) based on the coastal-continental shelf samples with water depth less than 200 m is similar to that of the published global open ocean regression equation. These results confirm that U^(K′)_(37)can be used as a shallow sea water SST proxy in mid-low latitudes of the WNP marginal seas. In addition, our reintegrated U^(K′)_(37)-SST results based on 172 global shallow water samples are similar(similar slopes and intercepts) to the shallow ocean results in the WNP marginal seas. However, the similarity of the regression formula to the open ocean does not imply that the formula is applicable. For example, of the 85 data in the shallow waters from the marginal sea in this study, the majority of data points lie above the regression line, showing positive residuals for U^(K′)_(37). This regression bias appears to be caused by specific marine environments, such as warm ocean currents and/or high nutrient conditions that result in positive U^(K′)_(37)residuals. Taken together,considering the specific temperature and environmental factors in the shallow waters of the low-mid-latitude WNP margin, we propose a nonlinear U^(K′)_(37)-SST regression formula: U^(K′)_(37)=-1.2488+0.1740×SST-0.0035×(SST)2, R2=0.93, N=85, specifically for the environments with SST below 24℃.Mengyuan WANG Haixian XIONG Shouye YANG Fengling YU Deming KONG Yongqiang ZONG Zhonghui LIU 2023Science China Earth Sciences2023,66,4:0
